The aim of this work was to develop an automated on-line solid phase extraction(SPE)with liquid chromatography-tandem mass spectrometry method for the detection of fifteen sulfonamides in pork and fish samples.Samples were extracted with 0.2%formic acid acetonitrile solution,purified by on-line SPE device with HLB column,then separated by XBridge C18 column,using 0.1%formic acid solution and acetonitrile as the mobile phase.Mass spectrometric data was acquired under multiple reaction monitoring(MRM)mode using positive ionization electrospray.Internal standard method was used in the quantification,good linear relationship was got in range of 0.1–100 ng/mL and correlation coefficient was higher than 0.9990.The limits of detection were in the range of 0.125–2.00g/kg and the limits of quantitation were in the range of 0.250–5.00g/kg.Recoveries of the method were in range of 78.3%–99.3%,relative standard deviation were lower than 10%.The method was simple,sensitivity,and could be used for routine supervision and analysis of fifteen sulfonamides in pork and fish. 展开更多

In this experiment,a liquid chromatography tandem mass spectrometry method was built to determine 15 pesticide residues in Chinese cabbage and cucumber samples based on online turbulent flow chromatography purification.After modified quick,easy,cheap,effective,rugged,and safe(QuEChERS)extraction,extracts were directly injected to the TLX(TurboFlow Liquid Xcalibur)system and brought to TurboFlow™columns for on-line purification and then transferred to analytical column for further separation and analysis.TurboFlow™columns types,transfer flow rate,and transfer time were optimized.Limits of detection and limits of quantification of the method obtained for 15 pesticide residues were ranged between 0.2–1.0μg/kg and 0.5–2.0μg/kg in Chinese cabbage and cucumber samples.Recoveries of pesticide residues were in range of 75.3%–103.7%.Matrix effects for 15 pesticides were in range of 5.6%–106.6%.The developed method has been successfully used for the determination of 15 pesticide residues in real samples. 展开更多

The measurement of urine catecholamine and metanephrine concentrations is important for biochemical screening and diagnosis of pheochromocytoma.The goal of this work was to develop a simple liquid chromatography-tandem mass spectrometry(LC-MS/MS)method for determining catecholamines and metanephrines in urine to replace an existing liquid chromatographic method using electrochemical detection.Urine samples were prepared using Oasis weak-cation-exchange cartridges.The eluate was analyzed on an Agilent ZORBAX Eclipse Plus Phenyl-Hexyl column in 3 min.Adrenaline,noradrenaline,dopamine,metanephrine,normetanephrine,and their deuterated internal standards were monitored in positive electrospray ionization mode by multiple reaction monitoring(MRM).No evidence of ion suppression was observed.The assay was linear up to 5μmol/L for adrenaline,5μmol/L for noradrenaline,6.1μmol/L for dopamine,5.6μmol/L for metanephrine,and 34.6μmol/L for normetanephrine,with lower limits of quantification of 5,5,12,6 and 7nmol/L,respectively.The intra-day and inter-day precisions for all analytes ranged from 0.59%to 4.64%and1.98%to 4.80%,respectively.External quality assurance samples were assayed and showed excellent agreement with the target values.This simple method provides an improved assay for determining urine catecholamines and metanephrines. 展开更多

Taraxacum kok-saghyz(TKS)is rich in natural rubber(NR),a natural organic macromolecular compound composed of cis-1,4-polyisoprene,and may become the second NR-bearing plant for biochemical engineering development.In this paper,a rapid and quantitative ultra-high performance liquid chromatography tandem mass spectrometry(UHPLCMS/MS)method was established for determination of macromolecular biosynthesis substrate(dimethylallyl pyrophosphate,DMAPP)and initiator(farnesyl pyrophosphate,FPP)contained in TKS.A Kromasil C18 chromatographic column was used for separation,and the multi-reaction monitoring mode(MRM)of triple quadrupole mass spectrometry was used for detection.Quantification was performed by external calibration method.The results showed that the limit of detection(LOD)and the limit of quantitation(LOQ)of DMAPP were 2.42μg/L and 7.26μg/L,respectively,and the LOQ and the LOD of FPP were 1.02μg/L and 3.05μg/L,respectively.At a concentration of 1—1000μg/L,both analytes had good determination coefficients(>0.999)of calibration curve.The recoveries of DMAPP and FPP were between 99.0%and 117.1%.In real samples detection,the contents of DMAPP and FPP in TKS samples were between 23.32—82.77μg/L and 12.03—85.67μg/L,respectively.Thus,this approach is a reliable method to quantify DMAPP and FPP in TKS. 展开更多

Determination of 19 polyphenolic compounds in tea by ultra-high performance liquid chromatography combined with quadrupole-time of flight mass spectrometry

A rapid method was presented for the determination of 19 polyphenols in tea by ultra-high performance liquid chromatography coupled with quadrupole-time of flight mass spectrometry(UPLC-Q-TOF MS).Tea samples were extracted by 50%(V/V)ethanol,then separated by Waters Acquity BEH C18 column using a binary solvent system composed of acetonitrile and water(0.1%formic acid)by gradient elution.The analytes were determined by Q-TOF MS in TOF MS and information dependent acquisition(IDA)-MS/MS mode.The results showed that mass accuracy error of the 19 polyphenols were lower than 5.0×10^(-6),good linear relationship was got in range of 0.2–500μg/L and correlation coefficient was higher than 0.9990.The LOD was in the range of 0.002–0.100 mg/kg and the LOQ was in the range of 0.004–0.200 mg/kg.Recovery of the method was in range of 78.4%–109.2%with spike levels of 0.004–2.000 mg/kg,relative standard deviations were lower than 10%.The method was simple,rapid and accurate.It could be used for the rapid screening and quantitative analysis of 19 polyphenols in tea. 展开更多

Five thyreostats(TSs),namely tapazole,thiouracil,methylthiouracil,propylthiouracil,and phenylthiouracil,were determined in bovine urine using ultra-high performance liquid chromatography-tandem mass spectrometry(UHPLC-MS/MS)in positive electrospray ionization mode.Extraction and clean-up were achieved using a ChemElut cartridge with tert-butyl methyl ether,without a derivatization step.Separation was achieved on an Acquity UPLC SS T3 column.The mobile phase was acetonitrile and water containing 0.2%(v/v)formic acid.The mass spectrometer was operated in multiple reaction monitoring mode.Urine samples were spiked with TS solution at levels corresponding to 5,10,15,and 20μg/L.The accuracy(internal standard corrected)ranged from 92%to 107%,with a repeatability precision(relative standard deviation,RSD)less than 15%for all five analytes.The RSDs within-laboratory reproducibility was less than 26%.The decision limits(CCα)and detection capabilities(CCβ)were obtained from a calibration curve and were in the ranges of 3.1-6.1μg/L and 4.0-7.4μg/L,respectively.The CCαand CCβvalues were below the recommended concentration,which was set at 10μg/L.The results show that the described method is suitable for the direct detection of TSs in bovine urine.This method can also be used to determine TSs in porcine urine. 展开更多

麦角生物碱(EAs)是由麦角菌属(Claviceps)产生的一类真菌毒素,当人类摄入被EAs污染的谷物及其制品时,可能会遭受慢性或急性中毒,从而威胁身体健康。鉴于此,本文基于超高效液相色谱-串联质谱(UPLC-MS/MS)建立了谷物及其制品中11种EAs的... 麦角生物碱(EAs)是由麦角菌属(Claviceps)产生的一类真菌毒素,当人类摄入被EAs污染的谷物及其制品时,可能会遭受慢性或急性中毒,从而威胁身体健康。鉴于此,本文基于超高效液相色谱-串联质谱(UPLC-MS/MS)建立了谷物及其制品中11种EAs的灵敏、快速测定方法。采用20 mL乙腈-200 mg/L碳酸铵溶液(80∶20,v/v)对谷物及其制品中的11种EAs进行提取,在10000 r/min下离心10 min,用通过式Captiva EMR-Lipid净化柱对上清液进行净化。采用ACQUITY UPLC HSS T3色谱柱(100 mm×3 mm,1.8μm)分离,以1 mmol/L乙酸铵溶液和乙腈为流动相进行梯度洗脱。在电喷雾电离(ESI)正离子模式下,采用多反应监测(MRM)模式采集数据,用基质匹配标准曲线进行外标法定量。方法学验证表明,11种EAs在各自的线性范围内具有良好的线性关系,相关系数(r^(2))为0.9933~0.9999,检出限为0.006~0.2μg/kg,定量限为0.02~0.6μg/kg。以小麦粉、薏苡仁、小麦粉制品和玉米粉为基质样品,在低、中、高3个加标水平下,11种EAs的回收率为80.1%~118%,相对标准偏差(RSD)为0.2%~13.3%。将所建方法用于市售谷物及其制品(240份小麦粉、80份玉米粉、30份大米、30份薏苡仁及146份小麦粉制品)中EAs的含量测定。结果表明,每种样品均有EAs检出,11种EAs的检出率为0.57%~20.3%。该方法具有简单、快速、准确等优势,适用于谷物及其制品中多种EAs的快速同时测定。 展开更多

合成大麻素(SCs)是数量最多的一类新精神活性物质(NPS)。我国于2021年7月起对SCs实行了整类列管。然后,经过结构修饰后产生的新型SCs仍然层出不穷,给法庭科学实验室检测分析带来巨大挑战。因此,亟需构建高效、绿色和自动化的分析检测方... 合成大麻素(SCs)是数量最多的一类新精神活性物质(NPS)。我国于2021年7月起对SCs实行了整类列管。然后,经过结构修饰后产生的新型SCs仍然层出不穷,给法庭科学实验室检测分析带来巨大挑战。因此,亟需构建高效、绿色和自动化的分析检测方法为真实案件样品的准确筛查提供技术支持。同时,吲唑类SCs因药效更强,自2013年后数量急剧增加,是SCs中需要重点关注的一个亚类,也是法庭科学实验室检测分析SCs的主要类别。本文开发了在线固相萃取前处理技术结合液相色谱-线性离子阱质谱(online SPE/LC-LIT-MS)从人体尿液和血液中检测51种吲唑类SCs的分析方法。将加入了51种吲唑类SCs混合标准溶液和内标溶液的人血液或尿液试样经乙腈沉淀蛋白后,用含0.1%甲酸的10 mmol/L醋酸铵溶液(pH=4.8)稀释,过滤后直接进样;以含0.1%甲酸(v/v)的乙腈-含0.1%甲酸的10 mmol/L醋酸铵溶液为流动相进行分析。在全扫描模式下,选择目标分析物的分子离子([M+H]^(+))和保留时间监测其二级离子,共实现了51种吲唑类SCs的定量分析;结合色谱和多级质谱数据也实现了的定性筛查(包括5组同分异构体)。各分析物检出限为0.02~1 ng/mL,定量限为0.04~4 ng/mL(尿液)和0.1~4 ng/mL(血液)。采用线性拟合时(权重因子1/x),各分析物在各自的线性范围内线性关系良好。同时在定量限、低、中、高4个水平下考察了方法的回收率和精密度,回收率为83.47%~116.95%,精密度为0.58%~13.79%。本文方法通过阀切换在动态模式下实现样品的提取、富集和分析,不仅操作简单,还实现了样品的自动化和高通量分析;同时,具有较好的灵敏度和更宽的适用范围,为实际案件中SCs的快速筛查和定量分析提供了科学依据和技术支持。 展开更多

建立了通过式固相萃取-超高效液相色谱-串联质谱法测定沉积物中草铵膦、草甘膦及其6种代谢物(3-甲基磷酸亚基丙酸、N-乙酰草铵膦、氨甲基膦酸、N-乙酰氨甲基膦酸、N-乙酰草甘膦、N-甲基草甘膦)的方法。样品采用4%(体积分数)氨水溶液提取... 建立了通过式固相萃取-超高效液相色谱-串联质谱法测定沉积物中草铵膦、草甘膦及其6种代谢物(3-甲基磷酸亚基丙酸、N-乙酰草铵膦、氨甲基膦酸、N-乙酰氨甲基膦酸、N-乙酰草甘膦、N-甲基草甘膦)的方法。样品采用4%(体积分数)氨水溶液提取,提取液经PRiME HLB固相萃取柱净化,过0.22μm聚醚砜滤膜后供超高效液相色谱-串联质谱测定。目标化合物使用Metrosep A Supp 5阴离子色谱柱(150 mm×4.0 mm,5μm)分离,以水和200 mmol/L碳酸氢铵溶液(含0.05%(v/v)氨水)作为流动相进行梯度洗脱,在电喷雾离子源(ESI)、负离子扫描和多反应监测(MRM)模式下进行测定,基质匹配外标法定量。结果表明,草铵膦、草甘膦及其代谢物在15 min内即可完成色谱分离,色谱峰形良好,响应值高,目标化合物在2.0~200.0μg/L范围内线性关系良好,相关系数均大于0.995。草铵膦、3-甲基磷酸亚基丙酸、N-乙酰草铵膦、N-乙酰氨甲基膦酸、N-乙酰草甘膦、N-甲基草甘膦的检出限为5μg/kg,定量限为20μg/kg;草甘膦、氨甲基膦酸的检出限为10μg/kg,定量限为30μg/kg。以空白沉积物为基质样品,在3个加标水平(定量限、5倍定量限和10倍定量限)下,低有机质含量的沉积物中目标化合物的平均回收率为78.5%~107%,相对标准偏差为1.32%~14.7%(n=6);高有机质含量的沉积物中目标化合物的平均回收率为76.4%~113%,相对标准偏差为2.60%~11.2%(n=6)。采用本方法对池塘、湖泊、水库、河流等不同类型的沉积物样品进行测定,结果显示,湖泊、水库、河流沉积物样品中未检出草铵膦、草甘膦及其代谢物,1个池塘沉积物样品中检出草甘膦和氨甲基膦酸,检出含量分别为31.7μg/kg和52.3μg/kg。本研究建立的方法具有简单、快速、绿色环保、准确度和灵敏度高、重复性好等优势,适用于沉积物中草铵膦、草甘膦及其代谢物的快速检测,为研究其在沉积物中的残留特征和环境行为提供了技术支持。 展开更多

目的建立一种有机溶剂提取、固相萃取柱净化、超高效液相色谱⁃串联质谱法(ultra performance liquid chromatography-tandem mass spectrometry,UPLC-MS/MS)同时测定淡水鱼中全氟烷基羧酸、全氟烷基磺酸及多氟调聚磺酸3类23种全氟及多... 目的建立一种有机溶剂提取、固相萃取柱净化、超高效液相色谱⁃串联质谱法(ultra performance liquid chromatography-tandem mass spectrometry,UPLC-MS/MS)同时测定淡水鱼中全氟烷基羧酸、全氟烷基磺酸及多氟调聚磺酸3类23种全氟及多氟烷基化合物(perfluorinated and polyfluoroalkyl substances,PFASs)的分析方法。方法优化了色谱分离条件、质谱检测参数,并考察了提取溶剂及固相萃取柱填料对样品加标回收率和净化效果的影响,确定了最佳样品前处理及仪器条件。淡水鱼样品经2%甲酸/乙腈提取后,采用去磷脂PPR Pro小柱净化,以5 mmol/L甲酸铵水溶液和甲醇为流动相,通过C_(18)反相色谱柱分离目标物;质谱采用电喷雾离子源负离子模式(electrospray ionization,ESI-),以多反应监测模式(multiple reaction monitoring,MRM)检测,稳定同位素内标法定量。结果23种PFASs目标物在0.5∼100.0 ng/mL质量浓度范围内线性关系良好(r^(2)>0.980),检出限为0.030~0.170μg/kg,定量限为0.100~0.567μg/kg,方法回收率为72.9%~111.0%,相对标准偏差为3.0%~17.5%(n=9)。结论该方法操作简便,灵敏度高,抗干扰性强,精密度好,适用于淡水鱼中23种全氟烷基羧酸、全氟烷基磺酸及多氟调聚磺酸的快速检测分析。 展开更多
