The genus Armeniaca Scop. is well known for its popular cultivated edible fruit trees such as Armeniaca vul- garis Lam. and ornamental flowers such as A. mume Sieb. Another species, A. cathayana Fu et al., one of six important dry fruits (kernel-using apricot), is cultivated for its edible seeds in North China. In the present study, DNA from 70 individuals of A rmeniaca, including 38 of A. cathayana, 18 ofA. vulgaris, 12 ofA. sibirica, 1 ofA. dasycarpa and 1 of A. mume, was extracted and analyzed using microsatellites and capillary electrophoresis. For 20 polymorphic loci selected, 339 alleles and 140.7 effective alleles were detected. The number of alleles per locus ranged from 8 to 28, with an average of 16.95 alleles per locus. The observed heterozygosity (Ho) and the expected heterozygosity (He) ranged from 0.427 to 0.971 and from 0.737 to 0.912, respectively. The polymorphism information content varied from 0.708 to0.905, with an average of 0.827. Based on the genetic similarity among 70 individuals, a UPGMA was used to establish the phylogenetic relationships. The taxonomic positions among five species were clearly revealed, and A. cathayana was more closely related to A. vulgaris than to A. sibirica. The results will provide a scientific basis for research on the taxonomy, germplasm resources and breeding ofArmeniaca, especially for A. cathayana. 展开更多

It is shown that the migration that the (tr) is a function of electric (Q), relative molecular mass (Mr), conformation parameter (Rc) of peptides and electrophoretic condition parameter(A). The conformation parameter is introduced to characterize multifarious shapes owing to the complex conlormation and the various kinds of macromolecules, where Rc≥1/3. The parameters A and Rc can be obtained from experimental data. The times of migration of the nine standard peptides were measured in pH 2.5buffer on different electrophoretic conditions in CZE. Methods Mixture of β cyclodextrin and glutamate β cyclodextrin was used as a chiral selector to the separation of chlorpheniramine. The effects of the concentrations of β cyclodextrin and glutamate β cyclodextrin, the voltage and the pH of buffers on the separation of chlorpheniramine were studied. Results The chiral recognization of the mixed chiral selector was better than that of a single selector. At pH below 4, the pH of buffers did not affect the separation and the separation of chlorpheniramine had good reproducibility. Conclusion Using a mixture of β CD and Glu β CD as chiral selector is useful for the enantioseparation of Chp in CE. 展开更多

Capillary electrophoresis (CE) has become a powerful tool for enantiomer separations during the last decade. Since 1993, the author has investigated enantiomer separations by affinity capillary electrophoresis (affinity CE) with some proteins and by cyclodextrin electrokinetic chromatography (CDEKC) with some charged cyclodextrins (CDs). Many successful enantiomer separations are demonstrated from our study in this review article. In the enantiomer separations by affinity CE, the deterioration of detection sensitivity was observed under high concentration of the protein in running solutions. The partial filling technique was practically useful to solve the serious problem. It allowed operation at high protein concentrations, such as 500 μmol/L, without the detection problem. Charged CDs had several advantages for the enantiomer separations over neutral ones. Strong electrostatic interactions between a charged CD and oppositely charged analytes should be effective for the formation of the complex. A large difference in electrophoretic mobility between the free analyte and the inclusion complex should also enhance the enantiomeric resolution. In CE mass spectrometry (CE MS), the partial filling technique was applied to avoid the introduction of nonvolatile chiral selectors into the CE MS interface. By replacing the nonvolatile electrolytes in the running buffer by volatile ones, the separation conditions employed in CE with the UV detection method could be transferred to CE MS. 展开更多

This work presents an approach to build a high-performance, low-viscous and replaceable separation matrix, semi-crosslinked polyacrylamide (semi-CPA) capillary gel electrophoresis. Non- denatured basic proteins, such as lysozyme, cytochrome C, ribonuclease A and trypsin were separa- ted. The impacts of monomer and cross-linker concentrations on protein separation were studied, and the ability of dynamic capillary inner wall coating was demonstrated. The UV absorption interfer- ence by semi-CPA gel matrix was successfully overcome by a partial filling technique, which results in sensitivity 20 times higher than other protein separation method. The excellent separation ability, reproducibility and dynamic coating ability made semi-CPA an ideal separation media in both capillar- y electrophoresis and microfluidic chip separation scheme. 展开更多

Microchip capillary electrophoresis (MCE) has significant impact on diagnostic testing. One of the most importance in clinical analysis test is the determination of cholesterol level in blood, because its increase is associated with coronary heart disease which is a major cause of death world over. Polydimethylsiloxane (PDMS) was material used to make a MCE for determination of cholesterol. Hydrogen peroxide was generated from the oxidation of cholesterol with cholesterol oxidase (ChOx) and was detected electrochemically at a downstream gold (Au) wire electrode by amperometry. Various parameters, such as the detection potential, concentration of running buffer, pH of running buffer, separation voltage, injection time, and concentration of ChOx, were studied. The migration time of cholesterol is less than 100s and the calibration curve is linear from 50 mg.dL-1 to 250 mg-dL-1 with the coefficient of determination (R2) of 0.9955. Therefore, this proposed assay is very rapid and sensitive for the detection of cholesterol. 展开更多

It has been shown that a significant decrease in adsorption was obtained and eletroosmotic flow was the diminished in the pH range 3-5. However with higher pH values, appreciable peak deformation and decreases in the resolving power were observed. Under pH 5, the epoxy polymer coating was shown to be quite stable and exhibited reproducible separations from run-to-run and day-to-day over a period of time. 展开更多

利用毛细管电泳(capillary electrophoresis,CE)仪结合紫外-可见光二极管阵列检测器检测技术建立水体中痕量微囊藻毒素的检测新方法。通过与GB/T 20466—2006《水中微囊藻毒素的测定》高效液相色谱(high performance liquid chromato... 利用毛细管电泳(capillary electrophoresis,CE)仪结合紫外-可见光二极管阵列检测器检测技术建立水体中痕量微囊藻毒素的检测新方法。通过与GB/T 20466—2006《水中微囊藻毒素的测定》高效液相色谱(high performance liquid chromatography,HPLC)方法对比分析,进行2种检测技术的评价。CE检测条件为:毛细管柱(60 cm×75μm i.d.),有效长度为44 cm,分离缓冲溶液为12 mmol/L硼酸盐(p H 9.0),分离电压25 kV,检测波长238 nm,压力6 895 Pa流体动力学进样;优化后的HPLC检测条件为:C18色谱柱(250 mm×4.6 mm i.d.,5μm),甲醇-磷酸盐缓冲溶液(60∶40,V/V)为流动相,检测波长238 nm,柱温度35℃,流速1 m L/min。结果表明:CE对3种微囊藻毒素MC-RR、MC-LR和MC-YR的检出限分别是0.16、0.20μg/m L和0.24μg/m L,HPLC对3种微囊藻毒素的检出限分别为0.020、0.079μg/m L和0.052μg/m L,这2个方法的灵敏度相差1个数量级;加标回收率分别92.5%-106.0%和99.6%-102.5%,CE对应的保留时间和峰面积的精密度相对标准偏差为0.53%-0.64%和2.67%-3.29%;HPLC法的保留时间和峰面积精密度相对标准偏差为0.16%-0.53%和0.80%-1.53%。检测同一水样中微囊藻毒素含量,CE检测结果和HPLC结果之间差异不显著(P〉0.05)。 展开更多

[目的]为加强孔雀石绿的管理和监控提供理论依据。[方法]采用高效毛细管电泳法(检测波长为265 nm),研究孔雀石绿在鲫鱼体内的主要代谢物隐性孔雀石绿的残留情况。[结果]不同浓度的隐性孔雀石绿标准品在0.062 5-1.000 0μg/ml范围内与相应的峰面积呈良好的线性关系,回归直线方程为:y=610.230x-23.345(R=0.9994)。加样回收率试验表明,隐性孔雀石绿的回收率为95%-102%。隐性孔雀石绿的保留时间为6.732 min,其在鱼体内的残留时间较长,呈非常平缓的下降趋势,不易消除,药浴30 d后隐性孔雀石绿的残留量为0.028μg/ml。[结论]高效毛细管电泳法检测鱼体内隐性孔雀石绿的残留具有消耗少、分离速度快、灵敏度高、重复性好等优点,且可以直接检测出隐性孔雀石绿。 展开更多

目的建立毛细管电泳-电化学发光法分离并检测复方磷酸可待因中三种成分,磷酸可待因、马来酸溴苯那敏和盐酸麻黄碱含量的新方法。方法基于碱性介质中,复方磷酸可待因溶液中三种主要成分对联吡啶钌在铂电极上的电化学发光信号的增敏作用,... 目的建立毛细管电泳-电化学发光法分离并检测复方磷酸可待因中三种成分,磷酸可待因、马来酸溴苯那敏和盐酸麻黄碱含量的新方法。方法基于碱性介质中,复方磷酸可待因溶液中三种主要成分对联吡啶钌在铂电极上的电化学发光信号的增敏作用,且增敏强度和药物浓度成线性关系,与毛细管电泳联用,建立了毛细管电泳-电化学发光法分离并检测其含量的新方法。结果在优化的实验条件下,确定三种药物对照品的线性范围、线性方程和检出限。通过对浓度为1.0×10-5kg/L的磷酸可待因、马来酸溴苯那敏和盐酸麻黄碱对照品进行12次平行测定,其ECL发光强度的RSD值分别为2.89%,3.76%,3.32%。结论该方法可用于复方磷酸可待因口服溶液中三种主要成分的含量测定,待测样品中的基质不干扰测定,方法的回收率分别为100.4%,101.9%,99.8%(n=5)。 展开更多

在区段-区段动力学毛细管电泳(ppKCE)的理论基础上,以兔红细胞膜和人红细胞膜为受体,研究了药物跨膜转运的动力学过程,同时测得了表征药物与细胞膜间结合快慢的正向结合速率常数kon、反向解离速率常数koff及配体与受体间相互作用的结合... 在区段-区段动力学毛细管电泳(ppKCE)的理论基础上,以兔红细胞膜和人红细胞膜为受体,研究了药物跨膜转运的动力学过程,同时测得了表征药物与细胞膜间结合快慢的正向结合速率常数kon、反向解离速率常数koff及配体与受体间相互作用的结合常数Kb.考察了迁移时间及峰高的稳定性、pH值和温度对药物与细胞膜相互作用动力学参数的影响.结果表明,与人体生理pH值一致时,细胞膜具有最大的生物活性,细胞膜与药物的Kb及kon均达到最大值.测定了细胞膜与西酞普兰、依匹斯汀、加替沙星及罗沙替丁相互作用的kon,koff和Kb.采用"时间比法"和紫外-可见吸收光谱法对该方法的准确性进行了验证.本研究为评价药物疗效、研究毒性及药物动力学提供了一种新方法. 展开更多

The results showed that the licorice plants come from various cultivated areas of China has different contents of GA. The GA content of licorice from Zhaodong in Heilongjiang Province is the highest, followed by those from E抰uoke, Chifeng, and Hangjin Banner in Inner Mongolia. Some suggestions for establishing the production base of licorice were put forward based on the study. 展开更多
