Lycii Radicis Cortex(LRC)is a medicinal and food homologous plant with various pharmacological activities,including anti-tumor effects.This study explores the anti-tumor effect of LRC on non-small cell lung cancer(NSCLC)and its molecular mechanism using mice bearing Lewis lung carcinoma cells.LRC significantly suppressed the growth of NSCLC.Besides,RNA sequencing of mice tumors and hematoxylin&eosin and immunofluorescence staining revealed that LRC promoted the infiltration of T lymphocytes,specifically GZMB~+CD8~+T lymphocytes,in tumor tissues.The Gene Set Enrichment Analysis of spleen RNA indicated that LRC up-regulated PD-1-downstream pathways,suggesting that LRC exerted its effects through the PDL1/PD-1 pathway.Further experiments revealed that LRC interacted with PD-L1,blocking PD-L1/PD-1 binding and thus restoring the T cell killing activity on tumor cells.Together,these results support using LRC as healthy food to improve anti-tumor immunity in patients with NSCLC. 展开更多

Obesity-induced insulin resistance is the hallmark of metabolic syndrome,and chronic,low-grade tissue inflammation links obesity to insulin resistance through the activation of tissue-infiltrating immune cells.Current therapeutic approaches lack efficacy and immunomodulatory capacity.Thus,a new therapeutic approach is needed to prevent chronic inflammation and alleviate insulin resistance.Here,we synthesized a tetrahedral framework nucleic acid(tFNA)nanoparticle that carried resveratrol(RSV)to inhibit tissue inflammation and improve insulin sensitivity in obese mice.The prepared nanoparticles,namely tFNAs-RSV,possessed the characteristics of simple synthesis,stable properties,good water solubility,and superior biocompatibility.The tFNA-based delivery ameliorated the lability of RSV and enhanced its therapeutic efficacy.In high-fat diet(HFD)-fed mice,the administration of tFNAs-RSV ameliorated insulin resistance by alleviating inflammation status.tFNAs-RSV could reverse M1 phenotype macrophages in tissues to M2 phenotype macrophages.As for adaptive immunity,the prepared nanoparticles could repress the activation of Th1 and Th17 and promote Th2 and Treg,leading to the alleviation of insulin resistance.Furthermore,this study is the first to demonstrate that tFNAs,a nucleic acid material,possess immunomodulatory capacity.Collectively,our findings demonstrate that tFNAs-RSV alleviate insulin resistance and ameliorate inflammation in HFD mice,suggesting that nucleic acid materials or nucleic acid-based delivery systems may be a potential agent for the treatment of insulin resistance and obesity-related metabolic diseases. 展开更多

In order to eliminate false alarms,issued by gas sensors in coal mining,caused by Electromagnetic Interference(EMI),both computer simulation and field measurements were introduced to analyze the underground EMI distribution.A simplified model of a sensor with metal enclosure was established and the effects of shielding properties about the enclosure aperture were studied.Because the haulage motor is the moving EMI source,varying with time,the onsite flameproof measuring instruments cannot accomplish synchronous measurements of electromagnetic field vectors.To simplify the field measurements,two sensors,one with a lead and the other without a lead,were chosen to conduct the contrasting measurements.The EMI current caused by the perforation lead was comparatively strong and therefore nickel zinc ferrite beads were used to cut off the EMI propagation paths.The peak value of the interference current was reduced by 20%-70% with the beads.After switching on the sensor power,the sen-sors still occasionally gave false alarms when the switch of nearby large-scale electric equipment was operated.A complex EMI filter was used and the EMI attenuated markedly.The running results demonstrated that false alarms had been eliminated.We con-clude that the improved shielding and filtering are highly significant in enhancing the immunity of the gas sensor. 展开更多

The effects of dietary zinc on the immune organs and cellular immunity were investigated in the rats of zinc-deficient(ZD), zinc-excessive(ZE), paired feeding(PF) and ad libitus(AL) groups. The findings were as follows: (1) The weight of the thymus and spleen and the ratio with the body weight, the size of the cells,the production of thymic peptides and thymin activity were significantly lower in ZD and ZE groups than in PF and AL groups. (2) The total number of the cells in G_0/G_1 phase of T-and B-cells and bone marrow cells were significantly higher in ZD and ZF groups than in PF and AL groups while that of the cells in S and G_2/M phases were significantly lower in ZD and ZE groups than in PF and AL groups. (3) The cellular DNA, RNA and protein content in the thymus, spleen and bone marrow were significantly lower in ZD and ZE groups than in PF and AL groups.(4) The  ̄3H-TdR incorporation rate of splenic T-and B-cells , IL-2 activity, plasma level of TNF and nitrogen oxide(NO) were significantly lower in ZD group than in PF group. (5) The leucocyte count of the perpheral blood,the percentage of lymphocytes, the bacteriocidal activity of phagocytes, and the phagocytic rate and index of abdominal phagocytes were significantly lower in ZD and ZE groups than in PF and AL groups. These findings indicate that an adequate amount of dietary zinc is essential for the development of immune organs and the promotion of cellular immunity while deficiency or excess of zinc is able to damage the immune function. 展开更多

A monitoring system is an important guarantee of safety in a production mine. However, not all monitoring substations pass the electrical fast transient/burst (EFT/B) immunity test and the explosion-proof test simultaneously. To enhance the immunity, the values of capacitance and inductance should be increased, which are actually limited by coal mine explosion-proof standards. Hence, for the first time, an active filter was applied in an electromagnetic interference (EMI) output filter. As a result, the interference within 30 MHz clearly weakened, but the frequency spectrum had a wide range. An EMI input filter and ferrite beads were adopted to restrain higher frequency interference. An output interference spectrogram of the substation was obtained with an analyzer. The results indicate that the improved complex filtering markedly help to control interference. With the support of improved complex filtering and other enhancing immunity means about I/O ports, the substation managed to pass both the EFT/B immunity test and the explosion-proof test synchronously. We conclude that improved complex filtering is of vital importance in enhancing the electromagnetic compatibilitv (EMC) of the coal mine monitoring system. 展开更多

Mushrooms are experiencing a kind of renaissance as a part of the contemporary human diet.These valuable organisms are more than food,they fit in perfectly as a novel market group known as nutra-mycoceuticals.Immune-balancing mushroom dietary fibers and secondary metabolites such as polyphenols are the main focus of the healthcare industry.Wellness and cosmetic companies are increasingly using mushroom extracts rich in these ingredients.This review considers the basic molecular immunomodulatory mechanisms of action of the most commonly used mushroom dietary fibers,β-glucans.The literature data on their bioavailability,metabolic transformations,preclinical and human clinical research,and safety are discussed.Immunomodulatory mechanisms of polyphenol ingredients are also considered.These molecules present great potential in the design of the new immunity balancer formulations according to their widespread structural diversity.Finally,we draw attention to the perspectives of modern trends in mushroom nutraceutical and cosmeceutical formulations to strengthen and balance immunity. 展开更多

The index of thymus and spleen, total supperoxide dismutase activity (T-SOD), serum malondialdehyde (MDA), content of nitrogen oxide (NO), the anti-superoxide anion ability, the suppressing hydroxy radical ability, contents of reduced glutathione (GSH) in thymus, total antioxidationabiUty (T-AOC) and the level of mouse ly- sozyme (LZM) in plasma were measured. LPS-induced B thymocytes proliferation was measured by MTT assay. In the low-immunity model group, MDA and NO levels were decreased (p 〈0. 001). Ju- glone improved lysozyme (LZM), GSH contents in thymus, T-AOC, T-SOD activity, the anti-super- oxide anion ability (anti- O2-) (p 〈0. 001). In the stimulation immunity model group, MDA and NO levels (p 〈0. 05), anti- 02- and T-SOD activity (p 〈0. 05) were up-regulated, whereas LZM, T-AOC contents were down-regulated, juglone has a significant effect, which promotes cell regeneration and function recovery, also may alleviate oxidative damage; juglone possesses a dual regulating effect on humoral immunity in mice. 展开更多

Based on negative selection mechanism of the immune system, a local network worm detection system that detects worms was proposed. Normal network service requests were represented by self-strings, and the detection system used self-strings to monitor the network for anomaly. According to the properties of worm propagation, a control center correlated the anomalies detected in the form of binary trees to ensure the accuracy of worm detection. Experiments show the system to be effective in detecting the traditional as well as multi-vector polymorphic worms. 展开更多

In this study,we investigated the effects of major royal jelly proteins(MRJPs)on the estrogen,gut microbiota,and immunological responses in mice.Mice given 250 or 500 mg/kg,not 125 mg/kg of MRJPs,enhanced the proliferation of splenocytes in response to mitogens.The splenocytes and mesenteric lymphocytes activated by T-cell mitogens(Con A and anti-CD3/CD28 antibodies)released high levels of IL-2 but low levels of IFN-γand IL-17A.The release of IL-4 was unaffected by MRJPs.Additionally,splenocytes and mesenteric lymphocytes activated by LPS were prevented by MRJPs at the same dose as that required for producing IL-1βand IL-6,two pro-inflammatory cytokines.The production of IL-1β,IL-6,and IFN-γwas negatively associated with estrogen levels,which were higher in the MRJP-treated animals than in the control group.Analysis of the gut microbiota revealed that feeding mice 250 mg/kg of MRJPs maintained the stability of the natural intestinal microflora of mice.Additionally,the LEf Se analysis identified biomarkers in the MRJP-treated mice,including Prevotella,Bacillales,Enterobacteriales,Gammaproteobacteria,Candidatus_Arthromitus,and Shigella.Our results showed that MRJPs are important components of royal jelly that modulate host immunity and hormone levels and help maintain gut microbiota stability. 展开更多

The role of B cells in anti-tumour immunity is still debated and,accordingly,immunotherapies have focused on targeting T and natural killer cells to inhibit tumour growth1,2.Here,using high-throughput flow cytometry as well as bulk and single-cell RNA-sequencing and B-cell-receptor-sequencing analysis of B cells temporally during B16F10 melanoma growth,we identified a subset of B cells that expands specifically in the draining lymph node over time in tumour-bearing mice.The expanding B cell subset expresses the cell surface molecule T cell immunoglobulin and mucin domain 1(TIM-1,encoded by Havcr1)and a unique transcriptional signature,including multiple co-inhibitory molecules such as PD-1,TIM-3,TIGIT and LAG-3.Although conditional deletion of these co-inhibitory molecules on B cells had little or no effect on tumour burden,selective deletion of Havcr1 in B cells both substantially inhibited tumour growth and enhanced effector T cell responses.Loss of TIM-1 enhanced the type 1 interferon response in B cells,which augmented B cell activation and increased antigen presentation and co-stimulation,resulting in increased expansion of tumour-specific effector T cells.Our results demonstrate that manipulation of TIM-1-expressing B cells enables engagement of the second arm of adaptive immunity to promote anti-tumour immunity and inhibit tumour growth. 展开更多

Toll-like receptors(TLRs) are a central component of innate immune system and play a major role as the initiator of the innate immune responses to defend against bacteria,viruses,parasite and other pathogens.During malaria infection,TLRs signaling pathways are initialed with the recognition of Plasmodium glycosylphosphatidylinositols(GPI) and hemozoin as pathogen-associated molecular patterns(PAMPs).And then,activation of TLRs signaling induces specific biological responses against malaria parasites invasion.However,TLRs are also involved in malaria pathogenesis and enhancement of immune tolerance and evasion for malaria infection.Moreover,malaria parasites regulate selectively TLRs expression on immune cells.Thus,these evidences indicated that TLRs have contrary roles on malaria infection.Understanding the complicated roles of TLRs on malaria infection will contribute us to design more effective anti-malaria drugs or vaccines. 展开更多

In this paper, an efficient multi-conductor simplification technique is proposed to model the electromagnetic immunity on cable bundles within a braid shielding structure over a large frequency range. By grouping together the conductors based on the knowledge of Z-Smith chart, the required computation time is markedly reduced and the complexity of modeling the completely shielding cable bundles is significantly simplified with a good accuracy. After a brief description of the immunity problems in shielding structure, a six-phase procedure is detailed to generate the geometrical characteristics of the reduced cable bundles. Numerical simulation is carried out by using a commercial software CST to validate the efficiency and advantages of the proposed approach. The research addressed in this paper is considered as a simplified modeling technique for the electromagnetic immunity within a shielding structure. 展开更多

Objective: To explore the effect of cellular immunity in halothane hepatitis. Methods: Hepatotoxicity model was established by exposing male Hartley guinea pigs to 1% halothane via inspiration for 4 h each time for 1 or 3 times within a 42-day interval. Then their hepatocytes and lymphocytes were collected and divided into 2 parts for different cultures. Hepatocytes were cultivated with or without 1% halothane for 4 h and lymphocytes were cultivated with or without 12.5 μg/ml trifluoroacetylated guinea pig serum albumin (TFA-GSA). Then the 2 kinds of hepatocytes were co-cultivated with lymphocytes (1:100) with or without TFA-GSA induction respectively and the supernatant fluid was taken after 24, 48 and 72 h to determine the concentration of alanine aminotransferase (ALT). The halothane cultivated hepatocytes were co-cultivated with various proportion of TFA-GSA antigen induced lymphocytes and ALT was determined after 48 h to determine the proper proportion of hepatocytes and lymphocyte. Results: Lymphocytes of 3 times halothane induced guinea pigs caused a significant increase of ALT in hepatocytes with or without halothane induction. But the lymphocytes of 1 time halothane induced guinea pigs only caused a significant increase of ALT in hepatocytes with induction of halothane. The increase of ALT was only seen after 48- and 72-hour co-culture. The proper proportion of hepatocytes and lymphocytes was 1:100 for lymphocytes cytotoxicity. Conclusion: Lymphocytes is sensitized after inhalation of halothane and generates cytotoxicity to hepatocytes. The immune response of lymphocytes to hepatocytes will be enhanced by repeated inhalation of halothane. The cellular immunity may be one of the mechanisms of halothane induced hepatotoxicity. 展开更多

The proposed method is used to constmct Boolean functions with an odd number of variables and with maximum algebraic immunity. We also discuss the nonlinearity of the constructed functions. With the rise of probiotics fermentation in food industry,fermented foods have attracted worldwide attention.In this study,protective effects of Rosa roxburghii&edible fungus fermentation broth(REFB)on immune function and gut health in Cyclophosphamide induced immunosuppressed mice were investigated.Results showed that REFB could improve the immune organ index,and promote the proliferation and differentiation of splenic T lymphocytes.In addition,it attenuated intestinal mucosal damage and improved intestinal cellular immunity.REFB administration also up-regulated the expression of IL-4,INF-γ,TNF-α,T-bet and GATA-3 mRNA in small intestine.Furthermore,administration of REFB modulated gut microbiota composition and increased the relative abundance of beneficial genus,such as Bacteroides.It also increased the production of fecal short-chain fatty acids.These indicate that REFB has the potential to improve immunity,alleviate intestinal injury and regulate gut microbiota in immunosuppressed mice. 展开更多

This study is to explore the mechanisms underlying the protective effects of Lactobacillus casei(L.casei)on the intestinal mucosal barrier of chicks infected with Salmonella pullorum(S.pullorum)through histological,immunological,and molecular biology methods.The results showed that L.casei significantly reduced the diarrhea rate,increased the daily weight gain,and maintained normal levels of IgA,IgM,and IgG in the serum of chicks infected with S.pullorum.Furthermore,we found that L.casei markedly improved the immunity of gut mucosa by regulating cytokine and chemokine receptor balance,elevating the number of intraepithelial lymphocytes. 展开更多

The numerous health benefits of olive oil are widely known,however,it also provides anti-allergic properties that have not yet been fully defined.In this study,the anti-allergic activity of olive oil was evaluated by analyzing the clinical symptoms and immune-related factors in BALB/c mice that had ingested600 mg/(kg·day)olive oil for two weeks prior to the evaluation.An allergy model was subsequently constructed for analysis,the results of which showed that the olive oil reduced the scores of allergic symptoms in the mice,and up-regulated the hypothermia and the decline in the immune organ index.Moreover,fewer allergy-related cytokines and reduced intestinal inflammation was discovered in the olive oil-treated group.In addition,analysis of intestinal mucosal immune-related factors revealed that the olive oil promoted the expression of intestinal tight junction proteins(Claudin-1,Occludin,and ZO-1)and IL-22,and helped maintain the integrity of the intestinal epithelial physical barrier.Increased levels of mucin 2 andβ-defensin were also found in the intestinal mucus of the olive oil-treated mice.These findings suggest that the oral administration of olive oil effectively attenuated the ovalbumin-induced allergic immune response in the mice,and had a positive effect on intestinal epithelial mucosal immunity. 展开更多

This study was to explore the immunity-promoting activity of porcine placenta as a potential raw material for functional foods.Porcine placenta was subjected to the analysis for its bioactive substances,and their immunity-promoting activity was determined in mice supplemented with porcine placenta extract(PPE)and freeze-dried porcine placenta powder at high(PPH)and low(PPL)dosage.Results showed that porcine placenta contained placental peptides and 15 free amino acids,and the amounts of estrogen and progesterone in products developed from porcine placenta were within the limit of national standard.Mice model experiment revealed that compared with the control,the PPH treatment significantly improved the spleen index(P<0.05)by increasing the phagocytic rate of macrophages from 20%to 60%and the conversion rate of T lymphocytes from 8%to 60%.The q PCR analysis disclosed that the porcine placenta powder enhanced mice immunity via promoting the expression of Th1 cytokines of interleukin-2(IL-2)and IFN-γ,especially the former,by almost 8 times in the spleens of male mice,while inhibited Th2 cytokines of IL-4 and IL-10.This investigation has provided a reference for the development of porcine placenta as a raw material applied in functional foods to improve human immunity. 展开更多
