In recent years,the prevalence of rice sheath blight caused by Rhizoctonia solani has significantly increased in Heilongjiang Province.Chemical control has become the primary control method.To cope with this,a novel m...In recent years,the prevalence of rice sheath blight caused by Rhizoctonia solani has significantly increased in Heilongjiang Province.Chemical control has become the primary control method.To cope with this,a novel mycelium growth rate method was employed to assess the toxicity of 13 fungicides,including a combination of 45%prochloraz and 125 g·mL^(-1)epoxiconazole,against R.solani.Additionally,the resistance of 99 R.solani strains to thifluzamide across various regions was also evaluated.The findings indicated that 75%trifloxystrobin-tebuconazole exhibited the most effective inhibitory effect,with an effective inhibitory medium concentration(EC50)value of 0.0101μg·mL^(-1).The EC50 values for 20%prothioconazole,125 g·mL^(-1)epoxiconazole,24%thifluzamide,and 50%hexaconazole were all less than 10μg·mL^(-1),indicating a better inhibitory effect on R.solani.The strongest synergistic effect was noted in the mixture of prochloraz and epoxiconazole at a 1:2 ratio,resulting in an EC50 value of 2.9917μg·mL^(-1),and a co-toxicity coefficient of 213.38.Among the 34 strains from Harbin City,the average EC50 value was 196.9341μg·mL^(-1)indicating the highest susceptiblility to thifluzamide.Conversely,15 strains from Shuangyashan City exhibited an average EC50 value of 364.7323μg·mL^(-1),reflecting the lowest sensitivity to thifluzamide.The sensitivity baseline EC50 value for R.solani was 253.8854μg·mL^(-1),with an overall resistance level between 0.1567 and 3.3292,indicating that the resistance level of R.solani in Heilongjiang Province remained low.Therefore,R.solani was still sensitive to thifluzamide in most areas of Heilongjiang Province,but there was a certain risk of resistance in Qitaihe City,which needed to be continuously monitored.At the same time,this study might provide a theoretical foundation for enhancing the prevention and management of the rice sheath blight.展开更多
Background Cotton(Gossypium hirsutum L.)is one of the most significant fibre and cash crops and plays an important role in Indian industrial and agricultural economies.However,over the years quantity and quality have ...Background Cotton(Gossypium hirsutum L.)is one of the most significant fibre and cash crops and plays an important role in Indian industrial and agricultural economies.However,over the years quantity and quality have been hampered by the pest leafhopper.Leafhopper alone has been shown to cause yield losses of up to 40%.In this study,screening and evaluation were performed to identify and categorize 100 cotton genotypes along with 5 checks as resistant,moderately resistant,sensitive and highly sensitive to leafhoppers.Results A total of hundred genotypes were evaluated along with five checks for leafhopper resistance.Based on the screening results,a total of 19 genotypes were resistant to leafhoppers,which was on par with the findings of the check KC 3.The contents of total soluble sugar,total soluble protein,and total free amino acids were significantly positively correlated with the mean grade,whereas total phenols content and trichome density were significantly negatively correlated with the susceptibility grade.However,based on screening and biochemical analysis,the genotypes KC 2,JR-23,Samaru-26-T,D 4,TCH 1728,RS 253,and B-61-1862 exhibited high resistance to leafhopper.Conclusion According to the findings of this study,choosing genotypes with high total phenolics content together with high trichome density and low contents of total soluble sugar,total soluble protein,and free amino acids may aid in the development of resistant genotypes.展开更多

Heavy metals in domestic rubbish were controlled by source screening before composting. The study consisted of a control with garden soil. The contents of main mineral elements and heavy metals in rubbish compost and control were determined by the method of ICP-AES. The results showed that heavy metal concentrations in rubbish were lower than those in garden soil, and little difference occurred between rubbish and garden soil in main mineral element concentrations. Based on this, it was concluded that rubbish compost was favorable for using as turfgrass medium and heavy metal control in rubbish by source screening was effective.展开更多

The influences of temperature, initial pH value, the volume of medium, shaking speed and illite concentration on the desilicating ability of the strain Lv(1- z) were investigated. The results show that the bacterium is a Gram-negative rod-shaped bacterium with oval endspores and thick capsule, but without flagellum. The biochemical and physiological tests indicate that the strain Lv(1- z) is similar to Bacillus rnucilaginosus. In GenBank the 16S rDNA sequence similarity of the strain Lv(1- z) and the B. rnucilaginosus YNUCC0001(AY571332) is more than 99%. Based on the above results, the strain Lv(1- z) is identified as B. rnucilaginosus. The optimum conditions for the strain L(1- z) to remove silicon from illite are as follows., temperature is 30℃ ;initial pH value is 7.5; medium volume in 200 mL bottle is 60 mL; shaking speed of rotary shaker is 220 r/m ; illite concentration is 1%.展开更多

The development ...Background: Large quantities of nitrogen (N) fertilizer applied to cotton cropping systems support high yields but cause adverse environmental impacts such as N20 emission and water eutrophication. The development of cotton cultivars with higher N use efficiencies suitable for low-N conditions is therefore important for sustainable production. In this study, we evaluated 100 cotton genotypes in 2016 for N use efficiency and related traits at the seedling stage. Methods: Sand culture experiment was conducted with low N levels (0.01 g.kg i) or normal N levels (0.1 g.kg i). We investigated plant height, SPAD value (soil plant analysis development chlorophyll meter), dry weight, N accumulation, N utilization efficiency, and N uptake efficiency. Through descriptive statistics, principal component analysis and heatmap clustering analysis, we confirmed the evaluation index system of N-efficient genotypes and the classification of N-efficient genotypes. Results: Significant differences were observed among N levels and genotypes for all agronomic traits and N levels. Coefficients of variation varied greatly and ranged from 6.7N28.8 and 7.4N20.8 under low-N and normal-N treatment, respectively. All traits showed highly significant positive correlations with each other, except SPAD value. The principal components under both N levels were similar, showing that total dry weight, aboveground dry weight, total N accumulation, and N uptake efficiency were important components. We confirmed these four traits as suitable screening indexes for low N tolerance. Based on the results of heatmap clustering and scatter diagram analysis of N efficiency value, 10 genotypes were found low-N tolerant, in which five varieties were inefficient under both low and normal N conditions, while four varieties were found efficient under low-N conditions but inefficient under normal-N conditions. Only one variety was efficient under both low and normal-N conditions. Meanwhile, 20 genotypes were identified as low-N sensitive ones, in which 19 genotypes were inefficient under low-N conditions but efficient under normal-N conditions, one variety was inefficient under both low and normal-N conditions. Conclusion: We preliminarily identified Kashi as a low-N tolerant and N-efficient cotton genotype, and CCRI 64 as a low- N sensitive and N-inefficient cotton genotype. Further studies should be carried out to verify the yield and heritability effect of specific genotypes in the field.展开更多
Background:Salt stress significantly inhibits the growth,development,and productivity of cotton because of osmotic,ionic,and oxidative stresses.Therefore,the screening and development of salt tolerant cotton cultivars...Background:Salt stress significantly inhibits the growth,development,and productivity of cotton because of osmotic,ionic,and oxidative stresses.Therefore,the screening and development of salt tolerant cotton cultivars is a key issue towards sustainable agriculture.This study subjected 11 upland cotton genotypes at the seedling growth stage to five different salt concentrations and evaluated their salt tolerance and reliable traits.Results:Several morpho-physiological traits were measured after 10 days of salinity treatment and the salt tolerance performance varied significantly among the tested cotton genotypes.The optimal Na Cl concentration for the evaluation of salt tolerance was 200 mmol·L-1.Membership function value and salt tolerance index were used to identify the most consistent salt tolerance traits.Leaf relative water content and photosynthesis were identified as reliable indicators for salt tolerance at the seedling stage.All considered traits related to salt tolerance indices were significantly and positively correlated with each other except for malondialdehyde.Cluster heat map analysis based on the morpho-physiological salt tolerance-indices clearly discriminated the 11 cotton genotypes into three different salt tolerance clusters.Cluster I represented the salt-tolerant genotypes(Z9807,Z0228,and Z7526)whereas clusters II(Z0710,Z7514,Z1910,and Z7516)and III(Z0102,Z7780,Z9648,and Z9612)represented moderately salttolerant and salt-sensitive genotypes,respectively.Conclusions:A hydroponic screening system was established.Leaf relative water content and photosynthesis were identified as two reliable traits that adequately represented the salt tolerance of cotton genotypes at the seedling growth stage.Furthermore,three salt-tolerant genotypes were identified,which might be used as genetic resources for the salt-tolerance breeding of cotton.展开更多

A series of experimental conditions were optimized. It is revealed that the optimum measurement procedure is as follows: adding 50μL of diluted enzyme sample and 50 μL substrate, incubating at 45 ℃ for exactly 5 min in micro-plate, mixing with 100 μL 3,5-dinitrosalicylic acid (DNS) reagent, maintaining at boiling point for 15 rain, cooling down to room temperature before determining the ABS value at 540 nm using an ELISA micro-plate reader. The reaction volume of the optimized microplate-assay is reduced to 200μL from 2 500 μL used in the standard β-mannanase macro-assay. The optimized micro-assay is significantly more sensitive in all of the 643 candidates during endo-1,4-β-mannosidase screening. Statistical analyses show that the sensitivity of the optimized micro-method is significantly greater than that of the macro-assay. The optimized method is convenient, fast, and cheap for high throughput enzyme screening.展开更多
OBJECTIVE To utilize a structure-based lead optimization approach to generate novel natural product-like typeⅡ inhibitors of JAK2 using the DOLPHIN protocol.METHODS Initially,the DOLHPIN computational protocol was em...OBJECTIVE To utilize a structure-based lead optimization approach to generate novel natural product-like typeⅡ inhibitors of JAK2 using the DOLPHIN protocol.METHODS Initially,the DOLHPIN computational protocol was employed to convert an active(DFG-in)conformation of JAK2 into a typeⅡ-compatible conformation,which was used as a model for the structure-based virtual screening of 150 000 natural product-like compounds in silico.The novel biflavonoid analogues were designed and synthesized based on the structure of lead compound and then tested for JAK2 and STAT3 inhibitory activity,cytotoxicity and HCV antiviral activity.RESULTS The top eleven highest-scoring compounds were generated from the initial high-throughput virtual screening.Amentoflavone 1a,a biflavonoid from the Chinese plant Gingko biloba,emerged as a promising candidate for further biological evaluation.In dose-response experiments,amentoflavone 1ainhibited JAK2 activity in a concentration dependent fashion with an IC50 value of 5μmol·L-1.As a proof-of-concept,we designed nine analogues 1b-1j with the addition of one or more aliphatic side chains to the biflavonoid scaffold of 1a.The octyl(C8)analogue 1bdisplayed superior potency against JAK2 activity and HCV activity compared to the parent compound 1a,validating the structure-based lead optimization approach used in this study.Moreover,kinetic analysis indicated that analogue 1bexhibited a non-competitive mode of inhibition,suggesting that this compound may be a putative typeⅡ inhibitor of JAK2.CONCLUSION Amentoflavone 1ahas been identified as a JAK2 inhibitor by structure-based virtual screening of a natural product library.In silico optimization using the DOLPHIN model yielded analogues with enhanced potency against JAK2 activity and HCV activity in cellulo.Molecular modeling and kinetic experiments suggested that the analogues may function as typeⅡ inhibitors of JAK2.展开更多
OBJECTIVE To apply molecular docking techniques to identify STAT3 inhibitors from a database of over 90 000 natural product and natural product-like compounds.METHODS Molecular docking was used for the virtual screeni...OBJECTIVE To apply molecular docking techniques to identify STAT3 inhibitors from a database of over 90 000 natural product and natural product-like compounds.METHODS Molecular docking was used for the virtual screening campaign and hit validation of STAT3 inhibitor.To further evaluate the potency of candidates at inhibiting STAT3-DNA binding activity,a STAT3 and STAT1transcription factor ELISA was performed.A dual-luciferase reporter assay,co-immunoprecipitation assay and Western blotting were carried out for the investigation of effect of compound 1 on STAT3-driven transcription,STAT3 dimerization and STAT3 phosphorylation.Finally,the cell toxicity of compound 1 was assessed by using MTT assay on different cell lines.RESULTS The virtual screening campaign furnished fourteen hit compounds,from which compound 1 emerged as a top candidate.Compound 1inhibited STAT3DNA-binding activity in vitro and attenuated STAT3-directed transcription in cellulo with selectivity over STAT1 and comparable potency to the wellknown STAT3 inhibitor S3I-201.Furthermore,compound 1 inhibited STAT3 dimerization and decreased STAT3 phosphorylation in cells without affecting STAT1 dimerization and phosphorylation.Compound 1 also exhibited selective anti-proliferative activity against cancer cells over normal cells in vitro.CONCLUSION The benzofuran derivative 1 was identified as a potential inhibitor of STAT3 dimerization using in silico screening.Molecular docking analysis suggested that compound 1 might putatively function as an inhibitor of STAT3 dimerization by binding to the SH2 domain.To the best of our knowledge,compound 1 has not been reported as a STAT3 inhibitor and no biological activity of compound 1 has been presented in the literature.展开更多

We mainly studied the effects of culture time, inoculation amount, initial p H and different sources of carbon, nitrogen and inorganic salt on the cellulase production of strain WR-C1. The advancement of planar micro-supercapacitors(PMSCs)for micro-electromechanical systems(MEMS)has been significantly hindered by the challenge of achieving high energy and power densities.This study addresses this is...The advancement of planar micro-supercapacitors(PMSCs)for micro-electromechanical systems(MEMS)has been significantly hindered by the challenge of achieving high energy and power densities.This study addresses this issue by leveraging screen-printing technology to fabricate high-performance PMSCs using innovative composite ink.The ink,a synergistic blend of few-layer graphene(Gt),carbon black(CB),and NiCo_(2)O_(4),was meticulously mixed to form a conductive and robust coating that enhanced the capacitive performance of the PMSCs.The optimized ink formulation and printing process result in a micro-supercapacitor with an exceptional areal capacitance of 18.95 mF/cm^(2)and an areal energy density of 2.63μW·h/cm^(2)at a current density of 0.05 mA/cm^(2),along with an areal power density of 0.025 mW/cm^(2).The devices demonstrated impressive durability with a capacitance retention rate of 94.7%after a stringent 20000-cycle test,demonstrating their potential for long-term applications.Moreover,the PMSCs displayed excellent mechanical flexibility,with a capacitance decrease of only 3.43%after 5000 bending cycles,highlighting their suitability for flexible electronic devices.The ease of integrating these PMSCs into series and parallel configurations for customized power further underscores their practicality for integrated power supply solutions in various technologies.展开更多
High purity and ultrafine DAAF(u-DAAF)is an emerging insensitive charge in initiators.Although there are many ways to obtain u-DAAF,developing a preparation method with stable operation,accurate control,good quality c...High purity and ultrafine DAAF(u-DAAF)is an emerging insensitive charge in initiators.Although there are many ways to obtain u-DAAF,developing a preparation method with stable operation,accurate control,good quality consistency,equipment miniaturization,and minimum manpower is an inevitable requirement to adapt to the current social technology development trend.Here reported is the microfluidic preparation of u-DAAF with tunable particle size by a passive swirling microreactor.Under the guidance of recrystallization growth kinetics and mixing behavior of fluids in the swirling microreactor,the key parameters(liquid flow rate,explosive concentration and crystallization temperature)were screened and optimized through screening experiments.Under the condition that no surfactant is added and only experimental parameters are controlled,the particle size of recrystallized DAAF can be adjusted from 98 nm to 785 nm,and the corresponding specific surface area is 8.45 m^(2)·g^(-1)to 1.33 m^(2)·g^(-1).In addition,the preparation method has good batch stability,high yield(90.8%-92.6%)and high purity(99.0%-99.4%),indicating a high practical application potential.Electric explosion derived flyer initiation tests demonstrate that the u-DAAF shows an initiation sensitivity much lower than that of the raw DAAF,and comparable to that of the refined DAAF by conventional spraying crystallization method.This study provides an efficient method to fabricate u-DAAF with narrow particle size distribution and high reproducibility as well as a theoretical reference for fabrication of other ultrafine explosives.展开更多

文摘In this paper,we introduce the censored composite conditional quantile coefficient(cC-CQC)to rank the relative importance of each predictor in high-dimensional censored regression.The cCCQC takes advantage of all useful information across quantiles and can detect nonlinear effects including interactions and heterogeneity,effectively.Furthermore,the proposed screening method based on cCCQC is robust to the existence of outliers and enjoys the sure screening property.Simulation results demonstrate that the proposed method performs competitively on survival datasets of high-dimensional predictors,particularly when the variables are highly correlated.

文摘This paper, probing into heavy metal control in domestic rubbish by source screening and nutrient element analysis, revealed the feasibility of source control of heavy metals and the suitability of rubbish as turfgrass medium. Heavy metals in domestic rubbish were controlled by source screening before composting. The study consisted of a control with garden soil. The contents of main mineral elements and heavy metals in rubbish compost and control were determined by the method of ICP-AES. The results showed that heavy metal concentrations in rubbish were lower than those in garden soil, and little difference occurred between rubbish and garden soil in main mineral element concentrations. Based on this, it was concluded that rubbish compost was favorable for using as turfgrass medium and heavy metal control in rubbish by source screening was effective.

文摘The strain Lv(1- z) isolated from the Henan bauxite was characterized by morphological observation, biochemical and physiological identification, and 16S rDNA sequence analysis. The influences of temperature, initial pH value, the volume of medium, shaking speed and illite concentration on the desilicating ability of the strain Lv(1- z) were investigated. The results show that the bacterium is a Gram-negative rod-shaped bacterium with oval endspores and thick capsule, but without flagellum. The biochemical and physiological tests indicate that the strain Lv(1- z) is similar to Bacillus rnucilaginosus. In GenBank the 16S rDNA sequence similarity of the strain Lv(1- z) and the B. rnucilaginosus YNUCC0001(AY571332) is more than 99%. Based on the above results, the strain Lv(1- z) is identified as B. rnucilaginosus. The optimum conditions for the strain L(1- z) to remove silicon from illite are as follows., temperature is 30℃ ;initial pH value is 7.5; medium volume in 200 mL bottle is 60 mL; shaking speed of rotary shaker is 220 r/m ; illite concentration is 1%.

文摘Background: Large quantities of nitrogen (N) fertilizer applied to cotton cropping systems support high yields but cause adverse environmental impacts such as N20 emission and water eutrophication. The development of cotton cultivars with higher N use efficiencies suitable for low-N conditions is therefore important for sustainable production. In this study, we evaluated 100 cotton genotypes in 2016 for N use efficiency and related traits at the seedling stage. Methods: Sand culture experiment was conducted with low N levels (0.01 g.kg i) or normal N levels (0.1 g.kg i). We investigated plant height, SPAD value (soil plant analysis development chlorophyll meter), dry weight, N accumulation, N utilization efficiency, and N uptake efficiency. Through descriptive statistics, principal component analysis and heatmap clustering analysis, we confirmed the evaluation index system of N-efficient genotypes and the classification of N-efficient genotypes. Results: Significant differences were observed among N levels and genotypes for all agronomic traits and N levels. Coefficients of variation varied greatly and ranged from 6.7N28.8 and 7.4N20.8 under low-N and normal-N treatment, respectively. All traits showed highly significant positive correlations with each other, except SPAD value. The principal components under both N levels were similar, showing that total dry weight, aboveground dry weight, total N accumulation, and N uptake efficiency were important components. We confirmed these four traits as suitable screening indexes for low N tolerance. Based on the results of heatmap clustering and scatter diagram analysis of N efficiency value, 10 genotypes were found low-N tolerant, in which five varieties were inefficient under both low and normal N conditions, while four varieties were found efficient under low-N conditions but inefficient under normal-N conditions. Only one variety was efficient under both low and normal-N conditions. Meanwhile, 20 genotypes were identified as low-N sensitive ones, in which 19 genotypes were inefficient under low-N conditions but efficient under normal-N conditions, one variety was inefficient under both low and normal-N conditions. Conclusion: We preliminarily identified Kashi as a low-N tolerant and N-efficient cotton genotype, and CCRI 64 as a low- N sensitive and N-inefficient cotton genotype. Further studies should be carried out to verify the yield and heritability effect of specific genotypes in the field.

文摘Background:Salt stress significantly inhibits the growth,development,and productivity of cotton because of osmotic,ionic,and oxidative stresses.Therefore,the screening and development of salt tolerant cotton cultivars is a key issue towards sustainable agriculture.This study subjected 11 upland cotton genotypes at the seedling growth stage to five different salt concentrations and evaluated their salt tolerance and reliable traits.Results:Several morpho-physiological traits were measured after 10 days of salinity treatment and the salt tolerance performance varied significantly among the tested cotton genotypes.The optimal Na Cl concentration for the evaluation of salt tolerance was 200 mmol·L-1.Membership function value and salt tolerance index were used to identify the most consistent salt tolerance traits.Leaf relative water content and photosynthesis were identified as reliable indicators for salt tolerance at the seedling stage.All considered traits related to salt tolerance indices were significantly and positively correlated with each other except for malondialdehyde.Cluster heat map analysis based on the morpho-physiological salt tolerance-indices clearly discriminated the 11 cotton genotypes into three different salt tolerance clusters.Cluster I represented the salt-tolerant genotypes(Z9807,Z0228,and Z7526)whereas clusters II(Z0710,Z7514,Z1910,and Z7516)and III(Z0102,Z7780,Z9648,and Z9612)represented moderately salttolerant and salt-sensitive genotypes,respectively.Conclusions:A hydroponic screening system was established.Leaf relative water content and photosynthesis were identified as two reliable traits that adequately represented the salt tolerance of cotton genotypes at the seedling growth stage.Furthermore,three salt-tolerant genotypes were identified,which might be used as genetic resources for the salt-tolerance breeding of cotton.

文摘Abstract: A simple optimized microplate-based method to assay endo-1,4-β-mannosidase activity was described as an improved high-throughput screening method. A series of experimental conditions were optimized. It is revealed that the optimum measurement procedure is as follows: adding 50μL of diluted enzyme sample and 50 μL substrate, incubating at 45 ℃ for exactly 5 min in micro-plate, mixing with 100 μL 3,5-dinitrosalicylic acid (DNS) reagent, maintaining at boiling point for 15 rain, cooling down to room temperature before determining the ABS value at 540 nm using an ELISA micro-plate reader. The reaction volume of the optimized microplate-assay is reduced to 200μL from 2 500 μL used in the standard β-mannanase macro-assay. The optimized micro-assay is significantly more sensitive in all of the 643 candidates during endo-1,4-β-mannosidase screening. Statistical analyses show that the sensitivity of the optimized micro-method is significantly greater than that of the macro-assay. The optimized method is convenient, fast, and cheap for high throughput enzyme screening.

文摘OBJECTIVE To utilize a structure-based lead optimization approach to generate novel natural product-like typeⅡ inhibitors of JAK2 using the DOLPHIN protocol.METHODS Initially,the DOLHPIN computational protocol was employed to convert an active(DFG-in)conformation of JAK2 into a typeⅡ-compatible conformation,which was used as a model for the structure-based virtual screening of 150 000 natural product-like compounds in silico.The novel biflavonoid analogues were designed and synthesized based on the structure of lead compound and then tested for JAK2 and STAT3 inhibitory activity,cytotoxicity and HCV antiviral activity.RESULTS The top eleven highest-scoring compounds were generated from the initial high-throughput virtual screening.Amentoflavone 1a,a biflavonoid from the Chinese plant Gingko biloba,emerged as a promising candidate for further biological evaluation.In dose-response experiments,amentoflavone 1ainhibited JAK2 activity in a concentration dependent fashion with an IC50 value of 5μmol·L-1.As a proof-of-concept,we designed nine analogues 1b-1j with the addition of one or more aliphatic side chains to the biflavonoid scaffold of 1a.The octyl(C8)analogue 1bdisplayed superior potency against JAK2 activity and HCV activity compared to the parent compound 1a,validating the structure-based lead optimization approach used in this study.Moreover,kinetic analysis indicated that analogue 1bexhibited a non-competitive mode of inhibition,suggesting that this compound may be a putative typeⅡ inhibitor of JAK2.CONCLUSION Amentoflavone 1ahas been identified as a JAK2 inhibitor by structure-based virtual screening of a natural product library.In silico optimization using the DOLPHIN model yielded analogues with enhanced potency against JAK2 activity and HCV activity in cellulo.Molecular modeling and kinetic experiments suggested that the analogues may function as typeⅡ inhibitors of JAK2.

文摘OBJECTIVE To apply molecular docking techniques to identify STAT3 inhibitors from a database of over 90 000 natural product and natural product-like compounds.METHODS Molecular docking was used for the virtual screening campaign and hit validation of STAT3 inhibitor.To further evaluate the potency of candidates at inhibiting STAT3-DNA binding activity,a STAT3 and STAT1transcription factor ELISA was performed.A dual-luciferase reporter assay,co-immunoprecipitation assay and Western blotting were carried out for the investigation of effect of compound 1 on STAT3-driven transcription,STAT3 dimerization and STAT3 phosphorylation.Finally,the cell toxicity of compound 1 was assessed by using MTT assay on different cell lines.RESULTS The virtual screening campaign furnished fourteen hit compounds,from which compound 1 emerged as a top candidate.Compound 1inhibited STAT3DNA-binding activity in vitro and attenuated STAT3-directed transcription in cellulo with selectivity over STAT1 and comparable potency to the wellknown STAT3 inhibitor S3I-201.Furthermore,compound 1 inhibited STAT3 dimerization and decreased STAT3 phosphorylation in cells without affecting STAT1 dimerization and phosphorylation.Compound 1 also exhibited selective anti-proliferative activity against cancer cells over normal cells in vitro.CONCLUSION The benzofuran derivative 1 was identified as a potential inhibitor of STAT3 dimerization using in silico screening.Molecular docking analysis suggested that compound 1 might putatively function as an inhibitor of STAT3 dimerization by binding to the SH2 domain.To the best of our knowledge,compound 1 has not been reported as a STAT3 inhibitor and no biological activity of compound 1 has been presented in the literature.

文摘A fungus(WR-C1) decomposed cellulose was isolated from a hypothermal litter layer using Congo red medium as the preliminary screening culture medium and then using a filter as the secondary screening medium at low temperature. The experiment showed that the weight loss rate of filter paper on the 15 th days could reach 30.69%. A morphologic and ITS gene sequence analysis suggested that CF-C1 was Cladosporium. We mainly studied the effects of culture time, inoculation amount, initial p H and different sources of carbon, nitrogen and inorganic salt on the cellulase production of strain WR-C1. Under optimum cultural condition, the highest value of WR-C1 enzyme production and filter paper enzyme were 3.27 U · m L~(-1) and 0.51 U · m L~(-1).
文摘In order to address the current inability of screen printing to monitor printing pressure online,an online printing pressure monitoring system applied to screen printing machines was designed in this study.In this study,the consistency of printed electrodes was measured by using a confocal microscope and the pressure distribution detected by online pressure monitoring system was compared to investigate the relationship.The results demonstrated the relationship between printing pressure and the consistency of printed electrodes.As printing pressure increases,the ink layer at the corresponding position becomes thicker and that higher printing pressure enhances the consistency of the printed electrodes.The experiment confirms the feasibility of the online pressure monitoring system,which aids in predicting and controlling the consistency of printed electrodes,thereby improving their performance.
文摘In the printing industry,the common method of coloring relies on inks,which contains amounts of chemical agents,causing environment pollution.However,structural color achieves coloration through the refraction and diffraction of light by periodic structure,offering eco-friendly and fade-resistant advantages,as well as colorful.In this study,screen printing was used to create patterned mask on paper substrates.Then,coated SiO_(2)microspheres on the mask to create structural color patterns with angle-dependent color characteristics.The patterns showed color changes from rose-red to orange to green by changing the viewing angle.By changing the color grayscale,the absorption of stray light by the substrate was enhanced,thereby the brightness and saturation of the structural color improved too.This method is simple,cost-effective,and environmentally friendly,and it has highly promising for the application in printing and anti-counterfeiting.

文摘The advancement of planar micro-supercapacitors(PMSCs)for micro-electromechanical systems(MEMS)has been significantly hindered by the challenge of achieving high energy and power densities.This study addresses this issue by leveraging screen-printing technology to fabricate high-performance PMSCs using innovative composite ink.The ink,a synergistic blend of few-layer graphene(Gt),carbon black(CB),and NiCo_(2)O_(4),was meticulously mixed to form a conductive and robust coating that enhanced the capacitive performance of the PMSCs.The optimized ink formulation and printing process result in a micro-supercapacitor with an exceptional areal capacitance of 18.95 mF/cm^(2)and an areal energy density of 2.63μW·h/cm^(2)at a current density of 0.05 mA/cm^(2),along with an areal power density of 0.025 mW/cm^(2).The devices demonstrated impressive durability with a capacitance retention rate of 94.7%after a stringent 20000-cycle test,demonstrating their potential for long-term applications.Moreover,the PMSCs displayed excellent mechanical flexibility,with a capacitance decrease of only 3.43%after 5000 bending cycles,highlighting their suitability for flexible electronic devices.The ease of integrating these PMSCs into series and parallel configurations for customized power further underscores their practicality for integrated power supply solutions in various technologies.

文摘High purity and ultrafine DAAF(u-DAAF)is an emerging insensitive charge in initiators.Although there are many ways to obtain u-DAAF,developing a preparation method with stable operation,accurate control,good quality consistency,equipment miniaturization,and minimum manpower is an inevitable requirement to adapt to the current social technology development trend.Here reported is the microfluidic preparation of u-DAAF with tunable particle size by a passive swirling microreactor.Under the guidance of recrystallization growth kinetics and mixing behavior of fluids in the swirling microreactor,the key parameters(liquid flow rate,explosive concentration and crystallization temperature)were screened and optimized through screening experiments.Under the condition that no surfactant is added and only experimental parameters are controlled,the particle size of recrystallized DAAF can be adjusted from 98 nm to 785 nm,and the corresponding specific surface area is 8.45 m^(2)·g^(-1)to 1.33 m^(2)·g^(-1).In addition,the preparation method has good batch stability,high yield(90.8%-92.6%)and high purity(99.0%-99.4%),indicating a high practical application potential.Electric explosion derived flyer initiation tests demonstrate that the u-DAAF shows an initiation sensitivity much lower than that of the raw DAAF,and comparable to that of the refined DAAF by conventional spraying crystallization method.This study provides an efficient method to fabricate u-DAAF with narrow particle size distribution and high reproducibility as well as a theoretical reference for fabrication of other ultrafine explosives.
