Cardiac arrest(CA)is a critical condition in the field of cardiovascular medicine.Despite successful resuscitation,patients continue to have a high mortality rate,largely due to post CA syndrome(PCAS).However,the inju...Cardiac arrest(CA)is a critical condition in the field of cardiovascular medicine.Despite successful resuscitation,patients continue to have a high mortality rate,largely due to post CA syndrome(PCAS).However,the injury and pathophysiological mechanisms underlying PCAS remain unclear.Experimental animal models are valuable tools for exploring the etiology,pathogenesis,and potential interventions for CA and PCAS.Current CA animal models include electrical induction of ventricular fibrillation(VF),myocardial infarction,high potassium,asphyxia,and hemorrhagic shock.Although these models do not fully replicate the complexity of clinical CA,the mechanistic insights they provide remain highly relevant,including post-CA brain injury(PCABI),post-CA myocardial dysfunction(PAMD),systemic ischaemia/reperfusion injury(IRI),and the persistent precipitating pathology.Summarizing the methods of establishing CA models,the challenges encountered in the modeling process,and the mechanisms of PCAS can provide a foundation for developing standardized CA modeling protocols.展开更多
OBJECTIVE CYP2 family including CYP2C and CYP2J is the predominant arachidonic acid(AA)epoxygenase,and the epoxidation of AA produces four regioisomeric cis-epoxyeicosatrienoic acids(5,6-,8,9-,11,12-,and 14,15-EET).Hu...OBJECTIVE CYP2 family including CYP2C and CYP2J is the predominant arachidonic acid(AA)epoxygenase,and the epoxidation of AA produces four regioisomeric cis-epoxyeicosatrienoic acids(5,6-,8,9-,11,12-,and 14,15-EET).Human CYP2J2 is one of the main CYP isoforms expressed in brain,but CYP2C8 was present at a low level.The aim of this study is to investigate the roles of brain CYP2J in Parkinson disease.METHODS Rats received the right-unilateral y injection with concentrated LV-CYP2J3 or LV-EGFP in the substantia nigra(SN)at 3 d before LPS or 6-OHDA treatment.The animals were tested for rotational behavior with the dopaminergic agonist apomorphine dissolved in sterile saline at 14 and 21 d after LPS injection.The influence of CYP2J-dependent derivative,14,15-EET,on the genes related with oxidative stress was assayed in SH-SY5Y cells.RESULTS CYP2J overexpression or 14,15-EET treatment significantly increased the levels of SOD1,CAT,GPX1,NRF2 and KEAP1 in neurons.TLR4-My D88 signaling pathway was involved the down-regulation of CYP2J by LPS.The binding of p-CREB with the promoter of CYP2J was inhibited by the LPS treatment.The loss of dopaminergic neurons in the right SN induced by LPS or 6-OHDA was significantly decreased by CYP2J3 transfection at 21 d after LPS injection.Compared with LPS or 6-OHDA group,the number of the rotation of rats was decreased by 42.6% and 60.7%by CYP2J3 transfection at 14 d after LPS or 6-OHDA injection;meanwhile,the rotation number was decreased by 12.7%and 21.3%at 21 d.The accumulation of alpha synuclein induced by LPS was significantly decreased by CYP2J3 transfection.The mR NA levels of SOD1,CAT,GPX1,NRF2 and KEAP1 in SN were decreased by LPS,which was attenuated by the injection of LV-CYP2J3.CONCLUSION Brain CYP2J can play a protective role in the damage of the inflammation and oxidative stress to the dopaminergic neurons.Brain CYP2J-dependent derivatives from AA may have therapeutic effects in Parkinson disease via the up-regulation of the antioxidant system in neurons.展开更多
OBJECTIVE Nowadays,with the living habits and diet structure changes,the morbidity and mortality of hypertension has been rising yearly,which induced by adverse lifestyles,excessive alcohol and poor diet.However,the a...OBJECTIVE Nowadays,with the living habits and diet structure changes,the morbidity and mortality of hypertension has been rising yearly,which induced by adverse lifestyles,excessive alcohol and poor diet.However,the animal models of hypertension resulted from adverse lifestyles have rarely been reported.It is essential to set up some more systematic models to study the characters of hypertension and evaluate the efficacy of traditional Chinese medicine(TCM).METHODS To build the rat model of human lifehabitual hypertension,SD rats had free access to alcohol with high-sucrose-fat diet(AHSFD),alcohol with high-fat diet(AHFD),high-sucrose-fat diet(HSFD),high-fat diet(HFD)and alcohol etc.Various of indicators were detected systematically to explore the features of hypertension models which included blood pressure(BP),liver and kidney functions,the lipid profiles and indexes quantifying TCM symptoms.Then we choose one of the stable and sustained animal models,studying the effects of RPA.E(which extracted from a TCM)on modulating the level of hypertension and the preliminary mechanism in this abstract.RESULTS ①The BP level,serum TC,TG,LDL-C of AHSFD-induced and AHFD-induced rats increased significantly,while the HDL-C reduced in the 4th week.② The BP level HSFD-induced rats increased significantly in the 6th week,serum TC,TG increased in 10 th week.③ The BP level of alcohol-induced rats increased significantly in the 9th week,serum TC,TG increased in 24 th week.④ The level of serum ALT,AST,UA and Cr of all model rats increased significantly after 12 weeks.Meanwhile the microcirculation increased significantly after8 weeks.⑤ After 4-week administration,RPA.E could significantly reduce BP of AHFD-induced hypertensive rats,and could reduce the serum TC,LDL-C,HDL-C,ALT,AST level after 8-week.CONCLUSION The rats induced by AHSFD,AHFD,HSFD and alcohol can raise BP obviously with underlying hepatic and kidney damage,the lipid profiles disorder and TCM symptoms change,while the AH(S)FD-induced hypertension models are earlier and more stable.RPA.E could mildly reduce BP level,and improve the lipid profiles disorders,hepatic damage,which reflects the characteristics of TCM on antihypertensive effects.The above models have their own characteristics,can be used to study the causes and pathogenesis of hypertension complicating metabolic disorder and the related treatment drug screening.展开更多
OBJECTIVE In recent years,the frequency of hyperuricemia has gradually risen along with the improvement of living standards,the irregular of diet and overfeeding greasy and surfeit flavor.However,hyperuricemia is a di...OBJECTIVE In recent years,the frequency of hyperuricemia has gradually risen along with the improvement of living standards,the irregular of diet and overfeeding greasy and surfeit flavor.However,hyperuricemia is a disorder of purine metabolism,and is strongly associated with insulin resistance and abnormal glucose metabolism.It is important to obtain a more stable and sustained animal model for the efficacy evaluation of traditional Chinese medicine(TCM).METHODS To manufacture the rodent model of hyperuricemia,the theory of increasing the source of the uric acid,reducing uric acid excretion and inhibiting uricase were used.We observed the influence on the serum uric acid and other indicators of rats induced by some factors:the lipid emulsion,high purine diet,beer with sugar,beer with sugar and high purine,and so on.Then we choose one of the stable and sustained animal models,studying the effects of Plo.E(which extracted from a TCM)on modulating the level of serum uric acid and the preliminary mechanism in this abstract.RESULTS ① At the 2nd week,the level of serum UA,BUN,Cr,TC,LDL-c of rats in the lipid emulsion group raised significantly.② At the 6th weeks,the serum UA in both the high purine diet group and lipid emulsion group raised obviously.③ The effects of Plo.E on hyperuricemia rats induced by high purine diet:After 8d administration,the Plo.E(three dosages)can reduce the UA level,and the middle and low dosages can reduce the TG level.After 25 dadministration,Plo.E can reduce the plasma viscosity,UA,TG,the whole blood viscosity level,the high dosage also can reduce the TC level.CONCLUSION The rats induced by the high purine diet and lipid emulsion can raised the serum UA obviously,while the way of lipid emulsion is earlier and more stable.Plo.E has a therapeutic effect on hyperuricemia,with an activity of reducing plasma viscosity and blood lipid.The above models conform to the pathogenesis of humans,can be used to study the causes and pathogenesis of hyperuricemia complicating metabolic disorder and the related treatment drug screening.展开更多

Background Cotton is one of the most important commercial crops after food crops,especially in countries like India,where it’s grown extensively under rainfed conditions.Because of its usage in multiple industries,su...Background Cotton is one of the most important commercial crops after food crops,especially in countries like India,where it’s grown extensively under rainfed conditions.Because of its usage in multiple industries,such as textile,medicine,and automobile industries,it has greater commercial importance.The crop’s performance is greatly influenced by prevailing weather dynamics.As climate changes,assessing how weather changes affect crop performance is essential.Among various techniques that are available,crop models are the most effective and widely used tools for predicting yields.Results This study compares statistical and machine learning models to assess their ability to predict cotton yield across major producing districts of Karnataka,India,utilizing a long-term dataset spanning from 1990 to 2023 that includes yield and weather factors.The artificial neural networks(ANNs)performed superiorly with acceptable yield deviations ranging within±10%during both vegetative stage(F1)and mid stage(F2)for cotton.The model evaluation metrics such as root mean square error(RMSE),normalized root mean square error(nRMSE),and modelling efficiency(EF)were also within the acceptance limits in most districts.Furthermore,the tested ANN model was used to assess the importance of the dominant weather factors influencing crop yield in each district.Specifically,the use of morning relative humidity as an individual parameter and its interaction with maximum and minimum tempera-ture had a major influence on cotton yield in most of the yield predicted districts.These differences highlighted the differential interactions of weather factors in each district for cotton yield formation,highlighting individual response of each weather factor under different soils and management conditions over the major cotton growing districts of Karnataka.Conclusions Compared with statistical models,machine learning models such as ANNs proved higher efficiency in forecasting the cotton yield due to their ability to consider the interactive effects of weather factors on yield forma-tion at different growth stages.This highlights the best suitability of ANNs for yield forecasting in rainfed conditions and for the study on relative impacts of weather factors on yield.Thus,the study aims to provide valuable insights to support stakeholders in planning effective crop management strategies and formulating relevant policies.展开更多

Since the beginning of the 21st century,advances in big data and artificial intelligence have driven a paradigm shift in the geosciences,moving the field from qualitative descriptions toward quantitative analysis,from...Since the beginning of the 21st century,advances in big data and artificial intelligence have driven a paradigm shift in the geosciences,moving the field from qualitative descriptions toward quantitative analysis,from observing phenomena to uncovering underlying mechanisms,from regional-scale investigations to global perspectives,and from experience-based inference toward data-and model-enabled intelligent prediction.AlphaEarth Foundations(AEF)is a next-generation geospatial intelligence platform that addresses these changes by introducing a unified 64-dimensional shared embedding space,enabling-for the first time-standardized representation and seamless integration of 12 distinct types of Earth observation data,including optical,radar,and lidar.This framework significantly improves data assimilation efficiency and resolves the persistent problem of“data silos”in geoscience research.AEF is helping redefine research methodologies and fostering breakthroughs,particularly in quantitative Earth system science.This paper systematically examines how AEF’s innovative architecture-featuring multi-source data fusion,high-dimensional feature representation learning,and a scalable computational framework-facilitates intelligent,precise,and realtime data-driven geoscientific research.Using case studies from resource and environmental applications,we demonstrate AEF’s broad potential and identify emerging innovation needs.Our findings show that AEF not only enhances the efficiency of solving traditional geoscientific problems but also stimulates novel research directions and methodological approaches.展开更多

文摘OBJECTIVE Nowadays,with the living habits and diet structure changes,the morbidity and mortality of hypertension has been rising yearly,which induced by adverse lifestyles,excessive alcohol and poor diet.However,the animal models of hypertension resulted from adverse lifestyles have rarely been reported.It is essential to set up some more systematic models to study the characters of hypertension and evaluate the efficacy of traditional Chinese medicine(TCM).METHODS To build the rat model of human lifehabitual hypertension,SD rats had free access to alcohol with high-sucrose-fat diet(AHSFD),alcohol with high-fat diet(AHFD),high-sucrose-fat diet(HSFD),high-fat diet(HFD)and alcohol etc.Various of indicators were detected systematically to explore the features of hypertension models which included blood pressure(BP),liver and kidney functions,the lipid profiles and indexes quantifying TCM symptoms.Then we choose one of the stable and sustained animal models,studying the effects of RPA.E(which extracted from a TCM)on modulating the level of hypertension and the preliminary mechanism in this abstract.RESULTS ①The BP level,serum TC,TG,LDL-C of AHSFD-induced and AHFD-induced rats increased significantly,while the HDL-C reduced in the 4th week.② The BP level HSFD-induced rats increased significantly in the 6th week,serum TC,TG increased in 10 th week.③ The BP level of alcohol-induced rats increased significantly in the 9th week,serum TC,TG increased in 24 th week.④ The level of serum ALT,AST,UA and Cr of all model rats increased significantly after 12 weeks.Meanwhile the microcirculation increased significantly after8 weeks.⑤ After 4-week administration,RPA.E could significantly reduce BP of AHFD-induced hypertensive rats,and could reduce the serum TC,LDL-C,HDL-C,ALT,AST level after 8-week.CONCLUSION The rats induced by AHSFD,AHFD,HSFD and alcohol can raise BP obviously with underlying hepatic and kidney damage,the lipid profiles disorder and TCM symptoms change,while the AH(S)FD-induced hypertension models are earlier and more stable.RPA.E could mildly reduce BP level,and improve the lipid profiles disorders,hepatic damage,which reflects the characteristics of TCM on antihypertensive effects.The above models have their own characteristics,can be used to study the causes and pathogenesis of hypertension complicating metabolic disorder and the related treatment drug screening.

文摘OBJECTIVE In recent years,the frequency of hyperuricemia has gradually risen along with the improvement of living standards,the irregular of diet and overfeeding greasy and surfeit flavor.However,hyperuricemia is a disorder of purine metabolism,and is strongly associated with insulin resistance and abnormal glucose metabolism.It is important to obtain a more stable and sustained animal model for the efficacy evaluation of traditional Chinese medicine(TCM).METHODS To manufacture the rodent model of hyperuricemia,the theory of increasing the source of the uric acid,reducing uric acid excretion and inhibiting uricase were used.We observed the influence on the serum uric acid and other indicators of rats induced by some factors:the lipid emulsion,high purine diet,beer with sugar,beer with sugar and high purine,and so on.Then we choose one of the stable and sustained animal models,studying the effects of Plo.E(which extracted from a TCM)on modulating the level of serum uric acid and the preliminary mechanism in this abstract.RESULTS ① At the 2nd week,the level of serum UA,BUN,Cr,TC,LDL-c of rats in the lipid emulsion group raised significantly.② At the 6th weeks,the serum UA in both the high purine diet group and lipid emulsion group raised obviously.③ The effects of Plo.E on hyperuricemia rats induced by high purine diet:After 8d administration,the Plo.E(three dosages)can reduce the UA level,and the middle and low dosages can reduce the TG level.After 25 dadministration,Plo.E can reduce the plasma viscosity,UA,TG,the whole blood viscosity level,the high dosage also can reduce the TC level.CONCLUSION The rats induced by the high purine diet and lipid emulsion can raised the serum UA obviously,while the way of lipid emulsion is earlier and more stable.Plo.E has a therapeutic effect on hyperuricemia,with an activity of reducing plasma viscosity and blood lipid.The above models conform to the pathogenesis of humans,can be used to study the causes and pathogenesis of hyperuricemia complicating metabolic disorder and the related treatment drug screening.

文摘Background Cotton is one of the most important commercial crops after food crops,especially in countries like India,where it’s grown extensively under rainfed conditions.Because of its usage in multiple industries,such as textile,medicine,and automobile industries,it has greater commercial importance.The crop’s performance is greatly influenced by prevailing weather dynamics.As climate changes,assessing how weather changes affect crop performance is essential.Among various techniques that are available,crop models are the most effective and widely used tools for predicting yields.Results This study compares statistical and machine learning models to assess their ability to predict cotton yield across major producing districts of Karnataka,India,utilizing a long-term dataset spanning from 1990 to 2023 that includes yield and weather factors.The artificial neural networks(ANNs)performed superiorly with acceptable yield deviations ranging within±10%during both vegetative stage(F1)and mid stage(F2)for cotton.The model evaluation metrics such as root mean square error(RMSE),normalized root mean square error(nRMSE),and modelling efficiency(EF)were also within the acceptance limits in most districts.Furthermore,the tested ANN model was used to assess the importance of the dominant weather factors influencing crop yield in each district.Specifically,the use of morning relative humidity as an individual parameter and its interaction with maximum and minimum tempera-ture had a major influence on cotton yield in most of the yield predicted districts.These differences highlighted the differential interactions of weather factors in each district for cotton yield formation,highlighting individual response of each weather factor under different soils and management conditions over the major cotton growing districts of Karnataka.Conclusions Compared with statistical models,machine learning models such as ANNs proved higher efficiency in forecasting the cotton yield due to their ability to consider the interactive effects of weather factors on yield forma-tion at different growth stages.This highlights the best suitability of ANNs for yield forecasting in rainfed conditions and for the study on relative impacts of weather factors on yield.Thus,the study aims to provide valuable insights to support stakeholders in planning effective crop management strategies and formulating relevant policies.

文摘Since the beginning of the 21st century,advances in big data and artificial intelligence have driven a paradigm shift in the geosciences,moving the field from qualitative descriptions toward quantitative analysis,from observing phenomena to uncovering underlying mechanisms,from regional-scale investigations to global perspectives,and from experience-based inference toward data-and model-enabled intelligent prediction.AlphaEarth Foundations(AEF)is a next-generation geospatial intelligence platform that addresses these changes by introducing a unified 64-dimensional shared embedding space,enabling-for the first time-standardized representation and seamless integration of 12 distinct types of Earth observation data,including optical,radar,and lidar.This framework significantly improves data assimilation efficiency and resolves the persistent problem of“data silos”in geoscience research.AEF is helping redefine research methodologies and fostering breakthroughs,particularly in quantitative Earth system science.This paper systematically examines how AEF’s innovative architecture-featuring multi-source data fusion,high-dimensional feature representation learning,and a scalable computational framework-facilitates intelligent,precise,and realtime data-driven geoscientific research.Using case studies from resource and environmental applications,we demonstrate AEF’s broad potential and identify emerging innovation needs.Our findings show that AEF not only enhances the efficiency of solving traditional geoscientific problems but also stimulates novel research directions and methodological approaches.
