Cooperative activation of sodium channels for downgrading the energy efficiency in neuronal information processing

The Hodgkin–Huxley model assumes independent ion channel activation,although mutual interactions are common in biological systems.This raises the problem why neurons would favor independent over cooperative channel activation.In this study,we evaluate how cooperative activation of sodium channels affects the neuron’s information processing and energy consumption.Simulations of the stochastic Hodgkin–Huxley model with cooperative activation of sodium channels show that,while cooperative activation enhances neuronal information processing capacity,it greatly increases the neuron’s energy consumption.As a result,cooperative activation of sodium channel degrades the energy efficiency for neuronal information processing.This discovery improves our understanding of the design principles for neural systems,and may provide insights into future designs of the neuromorphic computing devices as well as systematic understanding of pathological mechanisms for neural diseases. 展开更多

The development of the solid-state polymer electrolytes (SPEs) for Li-ion batteries (LIBs) can effectively address the hidden safety issues of commercially used liquid electrolytes.Nevertheless,the unsatisfactory room temperature ion conductivity and inferior mechanical strength for linear PEO-based SPEs are still the immense obstacles impeding the further applications of SPEs for large-scale commercialization.Herein,we fabricate a series of semi-interpenetrating-network (semi-IPN) polymer electrolytes based on a novel liquid crystal (C6M LC) and poly(ethylene glycol) diglycidyl ether (PEGDE) via UV-irradiation at the first time.The LCs not only highly improve the mechanical properties of electrolyte membranes via the construction of network structure with PEGDE,but also create stable ion transport channels for ion conduction.As a result,a free-standing flexible SPE shows outstanding ionic conductivity(5.93×10^(-5) S cm^(-1) at 30℃),a very wide electrochemical stability window of 5.5 V,and excellent thermal stability at thermal decomposition temperatures above 360℃ as well as the capacity of suppressing lithium dendrite growth.Moreover,the LiFePO_(4)/Li battery assembled with the semi-IPN electrolyte membranes exhibits good cycle performance and admirable reversible specific capacity.This work highlights the obvious advantages of LCs applied to the electrolyte for the advanced solid lithium battery. 展开更多

Outward and inward single channels in plasma membrane were observed using cell\|attached recording of patch\|clamp technique. In this study, single channel records showed that more than one species of channel were obtained. Despite the enormous interest in inorganic/polymer composite solid-state electrolytes(CSEs)for solid-state batteries(SSBs),the underlying ion transport phenomena in CSEs have not yet been elucidated.Here,we address this issue by formulating a mechanistic understanding of bi-percolating ion channels formation and ion conduction across inorganic-polymer electrolyte interfaces in CSEs.A model CSE is composed of argyrodite-type Li_6PS_5Cl(LPSCl)and gel polymer electrolyte(GPE,including Li~+-glyme complex as an ion-conducting medium).The percolation threshold of the LPSCl phase in the CSE strongly depends on the elasticity of the GPE phase.Additionally,manipulating the solvation/desolvation behavior of the Li~+-glyme complex in the GPE facilitates ion conduction across the LPSCl-GPE interface.The resulting scalable CSE(area=8×6(cm×cm),thickness~40μm)can be assembled with a high-mass-loading LiNi_(0.7)Co_(0.15)Mn_(0.15)O_(2)cathode(areal-mass-loading=39 mg cm~(-2))and a graphite anode(negative(N)/positive(P)capacity ratio=1.1)in order to fabricate an SSB full cell with bi-cell configuration.Under this constrained cell condition,the SSB full cell exhibits high volumetric energy density(480 Wh L_(cell)~(-1))and stable cyclability at 25℃,far exceeding the values reported by previous CSE-based SSBs. 展开更多

Objectives This study aimed at investigating the cellular mechanism of isoproterenol (ISO) on delayed afterdepolarizations (DADs) and triggered activity (TA) of the noninfarcted myocardium in the myocardial infarcted rabbit model.Methods Rabbits with the left anterior descending coronary artery occlusion were prepared and recovered for 8 wk (healed myocardial infarction, HMI). Myocytes were isolated from regions of the noninfarcted left ventricular free wall. ISO was added to cellular surface by perfusion way. Action potentials and ion currents were recorded with whole-cell patch clamp. Results The results showed that treatment with ISO induced more DADs and TA events in HMI myocytes. Iti and IC,_L of myocytes treated with ISO were increased significantly compared with HMI cells, which contributed to DADs-related triggered arrhythmia. Conclusions The results suggested that more arrhythmia events of DADs and TA developed in myocytes with ISO treatment. An electromagnetic view of relay time in propagation of neural signals

We review the experimental and computational data about the propagation of neural signals in myelinated axons in mice,cats,rabbits,and frogs published in the past five decades.In contrast to the natural assumption that neural signals occur one by one in time and in space,we figure out that neural signals are highly overlapped in time between neighboring nodes.This phenomenon was occasionally illustrated in some early reports,but seemed to have been overlooked for some time.The shift in time between two successive neural signals from neighboring nodes,defined as relay timeτ,was calculated to be only 16.3μs-87.0μs,i.e.,0.8%-4.4%of the average duration of an action potential peak(roughly 2 ms).We present a clearer picture of the exact physical process about how the information transmits along a myelinated axon,rather than a whole action potential peak,what is transmitted is only a rising electric field caused by transmembrane ion flows.Here in the paper,τrepresents the waiting time until the neighboring node senses an attenuated electric field reaching the threshold to trigger the open state.The mechanisms addressed in this work have the potential to be universal,and may hold clues to revealing the exact triggering processes of voltage-gated ion channels and various brain functions. 展开更多

This study aimed at investigating the effects of nervous growth factor (NGF) on delayed afterdepolarizations (DADs) and triggered activity (TA) of the noninfarcted myocardium in the myocardial infarcted rabbit model. Rabbits with the left anterior descending coronary artery occlusion were prepared and recovered for 8 weeks (HMI group, n=9). Other rabbits with myocardial infarction were infused NGF to the left stellate ganglion (HMI+NGF group, 400 U/day for 8 weeks, n=8). Myocytes were isolated from regions of the noninfarcted left ventricular free wall. Action potentials and ion currents were recorded with whole-cell patch clamp. The results showed that more DADs and TA events of HMI+NGF myocytes than that of HMI and Ctrl group. Iti and ICa-L of NGF+HMI myocytes were increased significantly compared with HMI and Ctrl cells, which contributed to DADs-related triggered arrhythmia. Comparing with HM1 and Ctrl myocytes, significant prolongations of APD50 and APD90 in HMI+NGF myocytes were found. The results indicated the electrophysiological change of HMI myocytes with NGF infusion. It suggested that more events of DADs and TA in HMI myocytes with NGF treatment. The underlying mechanism may be involved in the increase of Iti and ICa-L. 展开更多

Objectives To evaluate the association between a KCNQ 1 mutation, R259H, and short QT syndrome (SQTS) and to explore the elec- trophysiological mechanisms underlying their association. Methods We performed genetic screening of SQTS genes in 25 probands and their family members (63 patients). We used direct sequencing to screen the exons and intron-exon boundaries of candidate genes that en- code ion channels which contribute to the repolarization of the ventricular action potential, including KCNQI, KCNH2, KCNE1, KCNE2, KCNJ2, CACNAlc, CACNB2b and CACNA2D1. In one of the 25 SQTS probands screened, we discovered a KCNQ1 mutation, R259H. We cloned R259H and transiently expressed it in HEK-293 cells; then, currents were recorded using whole cell patch clamp techniques. Results R259H-KCNQ 1 showed significantly increased current density, which was approximately 3-fold larger than that of wild type (WT) after a depolarizing pulse at 1 s. The steady state voltage dependence of the activation and inactivation did not show significant differences between the WT and R259H mutation (P 〉 0.05), whereas the time constant of deactivation was markedly prolonged in the mutant compared with the WT in terms of the test potentials, which indicated that the deactivation of R259H was markedly slower than that of the WT. These results suggested that the R259H mutation can effectively increase the slowly activated delayed rectifier potassium current (Irs) in phase 3 of the cardiac action potential, which may be an infrequent cause of QT interval shortening. Conclusions R259H is a gain-of-function muta- tion of the KCNQ1 channel that is responsible for SQTS2. This is the first time that the R259H mutation was detected in Chinese people. 展开更多

This paper addresses the formulae and numerical issues related to the possibility that fast wave may be grown when a relativistic electron beam through an ion channel in a cylindrical metal waveguide. To derive the dispersion equations of the beam-wave interaction, it solves relativistic Lorentz equation and Maxwell's equations for appropriate boundary conditions. It has been found in this waveguide structure that the TM0m modes are the rational operating modes of coupling between the electromagnetic modes and the betatron modes. The interaction of the dispersion curves of the electromagnetic TM0m modes and the upper betatron modes is studied. The growth rates of the wave are obtained, and the effects of the beam radius, the beam energy, the plasma frequency, and the beam plasma frequency on the wave growth rate are numerically calculated and discussed. 展开更多

In this paper, by discussing a simple channel model, we show that there are two different schemes to understand the biexponential distribution of open times. One scheme is mathematically strict based on generator matrix theory, while the other one has a clear physical explanation according to an approximation process with numerical simulation of Markovian channel dynamics. Our comparison results suggest that even for biologically complex channels, in addition to carrying out a stochastic simulation, the strict theoretical analysis should be considered to understand the multiple exponential distributions of open times. 展开更多

It is known that ion channel can effectively limit the radial expansion of an artificial electron beam during its longrange propagation in the space plasma environment.Most prior studies discussed the focusing characteristics of the beam in the ion channel,but the establishment process and transient properties of the ion channel itself,which also plays a crucial role during the propagation of the relativistic electron beam in the plasma environment,were commonly neglected.In this study,a series of two-dimensional(2D)particle-in-cell simulations is performed and an analytical model of ion channel oscillation is constructed according to the single-particle motion.The results showed that when the beam density is higher than the density of plasma environment,ion channel can be established and always continues to oscillate periodically over the entire propagation.Multiple factors,including the beam electron density,initial beam radius,and the plasma density can affect the oscillation properties of ion channel.Axial velocity of the beam oscillates synchronously with the ion channel and this phenomenon will finally develop into a two-stream instability which can seriously affect the effective transport for relativistic electron beam.Choosing appropriate beam parameters based on various plasma environments may contribute to the improvement of the stability of ion channel.Additionally,radial expansion of the beam can be limited by ion channel and a stable long-range propagation in terrestrial atmosphere may be achieved. 展开更多

Considering the self-fields of relativistic electron beam (REB), the electron beam injected in plasma will form an ion-channel, which can stably guide REB transmitted in plasma. Based on the effect of ion-channel, the EM dispersion characteristics in a dielectric waveguide filled with plasmas is studied by using linear electromagnetic (EM) hydrodynamics perturbation theory. Both the TM mode dispersion equation and the dispersion relation are derived. After that, a discussion on the conditions of Cherenkov radiation in this system is given. It was found that the betatron oscillation of the REB is one of the important factors for the slow-wave EM instability in this system. The dispersion relation is of the form of electromagnetic-electrostatic (EM-ES) hybrid mode, which is different from the case where the effect of the ion channel is neglected. 展开更多

Taking self-fields into consideration, dispersion properties of two types of electromagnetic modes for a high energy electron beam guided with an ion channel are investigated by using the linear perturbation theory. The dependences of the dispersion frequencies of electromagnetic waves on the electron beam radius, betatron frequency and boundary current are revealed. It is found that the electron beam radius and betatron frequency have different influences on the electromagnetic waves dispersion behavior by compared with the previous works. As the boundary current is taken into account, the TM modes will have two branches and a lowfrequency branch emerged as the new branch in strong ion channel case. This new branch has similar dispersion behavior to the betatron modes. For TE modes, there are two branches and they have different dispersion behaviors in strong ion channel case. However, in weak ion channel case, the dispersion behaviors for both of the low frequency and high frequency branches are similar. 展开更多

ResultsCVB 3 infection increased I Ca and I out , while decreased I KI ; but it had no obvious effect on I Na . Conclusion.The effects of CVB 3 on I Ca 、 I out 、 I KI may be one of the mechanisms of myocytes damage and the occurrence of abnormal electroactivities induced by CVB 3 infection. 展开更多

Understanding of the mechanisms of neural phase transitions is crucial for clarifying cognitive processes in the brain. We investigate a neural oscillator that undergoes different bifurcation transitions from the big saddle homoclinic orbit type to the saddle node on an invariant circle type, and the saddle node on an invariant circle type to the small saddle homoclinic orbit type. The bifurcation transitions are accompanied by an increase in thermodynamic temperature that affects the voltage-gated ion channel in the neural oscillator. We show that nonlinear and thermodynamical mechanisms are responsible for different switches of the frequency in the neural oscillator. We report a dynamical role of the phase response curve in switches of the frequency, in terms of slopes of frequency-temperature curve at each bifurcation transition. Adopting the transition state theory of voltagegated ion channel dynamics, we confirm that switches of the frequency occur in the first-order phase transition temperature states and exhibit different features of their potential energy derivatives in the ion channel. Each bifurcation transition also creates a discontinuity in the Arrhenius plot used to compute the time constant of the ion channel. 展开更多

As the key component of electrochemical energy storage devices, an electrode with superior ions transport pores is the important premise for high electrochemical performance. In this paper, we developed a unique solution process to prepare freestanding TiO_2/graphene hydrogel electrode with tunable density and porous structures. By incorporating room temperature ionic liquids(RTILs), even upon drying, the non-volatile RTILs that remained in the gel film would preserve the efficient ion transport channels and prevent the electrode from closely stacking, to develop dense yet porous structures. As a result, the dense TiO_2/graphene gel film as an electrode for lithium ion battery displayed a good gravimetric electrochemical performance and more importantly a high volumetric performance. 展开更多

Use of antiarrhythmic drugs in elderly patients 被引量:3

Human aging is a global issue with important implications for current and future incidence and prevalence of health conditions and disability. Cardiac arrhythmias, including atrial fibrillation, sudden cardiac death, and bradycardia requiring pacemaker placement, all increase exponentially after the age of 60. It is important to distinguish between the normal, physiological consequences of aging on cardiac electrophysiology and the abnormal, pathological alterations. The age-related cardiac changes include ventricular hypertrophy, senile amyloidosis, cardiac valvular degenerative changes and annular calcification, fibrous infiltration of the conduction system, and loss of natural pacemaker cells and these changes could have a profound effect on the development of arrhythmias. The age-related cardiac electrophysiological changes include up- and down-regulation of specific ion channel expression and intmcellular Ca2+ overload which promote the development of cardiac arrhythmias. As ion channels are the substrates of antiarrhythmic drugs, it follows that the pharmacokinetics and pharmacodynamics of these drugs will also change with age. Aging alters the absorption, distribution, metabolism, and elimination of antiarrhythmic drugs, so liver and kidney function must be monitored to avoid potential adverse drug effects, and antiarrhythmic dosing may need to be adjusted for age. Elderly patients are also more susceptible to the side effects of many antiarrhytbanics, including bradycardia, orthostatic hypotension, urinary retention, and falls. Moreover, the choice of antiarrhythmic drugs in the elderly patient is frequently complicated by the presence of co-morbid conditions and by polypharmacy, and the astute physician must pay careful attention to potential drug-drug interactions. Finally, it is important to remember that the use of antiarrhythmic drugs in elderly patients must be individualized and tailored to each patient's physiology, disease processes, and medication regimen. 展开更多

Because a brain consists of tremendous neuronal networks with different neuron numbers ranging from tens to tens of thousands, we study the coherence resonance due to ion channel noises in globally coupled neuronal networks with different neuron numbers. We confirm that for all neuronal networks with different neuron numbers there exist the array enhanced coherence resonance and the optimal synaptic conductance to cause the maximal spiking coherence. Furthermoremore, the enhancement effects of coupling on spiking coherence and on optimal synaptic conductance are almost the same, regardless of the neuron numbers in the neuronal networks. Therefore for all the neuronal networks with different neuron numbers in the brain, relative weak synaptic conductance (0.1 mS/cm2) is sufficient to induce the maximal spiking coherence and the best sub-threshold signal encoding. 展开更多
