Background Unravelling the relationship between trichome density and resistance to jassids in upland cotton,nine parental lines,viz.MCU 5,CO 14,CO 17,TCH 1828,KC 2,KC 3,GISV 323,GTHV 15–34,and RHC 1409 were obtained from the Tamilnadu Agricultural University.These genotypes were subjected to molecular analysis using 27 primers,merely the JESPR 154 primer amplifying a 150-bp fragment in genotypes exhibiting the pubescence.Result This finding validated the association between pubescence and jassid resistance.Further analysis revealed that resistant genotypes(KC 3,GTHV 15–34,GISV 323,and RHC 1409)exhibited significantly higher trichome densities and length compared with susceptible genotypes.These results stalwartly support the hypothesis that trichomes play a pivotal role in conferring resistance to jassids in upland cotton.Conclusion By breeding cotton varieties with increased trichome density and length,it is possible to reduce jassid infestations,thereby decreasing the reliance on chemical pesticides and promoting a more sustainable agricultural environment. 展开更多

施氮量对不同开花期棉(Gossypium hirsutum L.)铃纤维细度和成熟度形成的影响 被引量:2

为兼顾试验的重复性和生态区域性,选用高品质棉(科棉1号)和常规棉(美棉33B)品种为材料,于2005年分别在江苏南京(118°50'E,32°02'N,长江流域下游棉区)和江苏徐州(117°11'E,34°15'N,黄河流域黄淮棉区... 为兼顾试验的重复性和生态区域性,选用高品质棉(科棉1号)和常规棉(美棉33B)品种为材料,于2005年分别在江苏南京(118°50'E,32°02'N,长江流域下游棉区)和江苏徐州(117°11'E,34°15'N,黄河流域黄淮棉区)设置施氮量(低氮N 0 kg/hm2;适氮N 240 kg/hm2;高氮N 480 kg/hm2)试验,研究施氮量对不同开花期棉铃纤维细度、成熟度和马克隆值形成的影响。结果表明,(1)施氮量显著影响棉纤维细度、成熟度和马克隆值的形成过程,但三者在不同开花期对氮素水平的响应不同,施氮量与开花期对棉纤维细度、成熟度和马克隆值的形成存在互作效应。8月10日前开花的棉铃,铃期[花后0~50 d(DPA)]日均温在23.3℃以上,纤维细度、马克隆值以N 0 kg/hm2施氮量下最大,棉纤维马克隆值与纤维细度的相关性较大;8月25日开花的棉铃(铃期日均温在20.8~23.3℃之间),纤维成熟度、马克隆值以N 240 kg/hm2施氮量下最大;9月10日开花棉铃(铃期日均温低于20.8℃),纤维细度、成熟度和马克隆值均以N 480 kg/hm2最大,棉纤维马克隆值与纤维成熟度的相关性增强。(2)影响不同开花期间纤维细度、成熟度和马克隆值的主要因素是铃期日均温;最终纤维细度、成熟度和马克隆值在不同施氮量之间的变异与不同开花期(铃期日均温不同)间的变异比较,前者显著小于后者。综上,因开花期不同而形成的铃期日均温是决定纤维细度、成熟度和马克隆值的最重要因素,施氮量可通过对位叶叶氮浓度NA影响棉纤维细度、成熟度和马克隆值的形成过程,增加施氮量可减小上述指标在不同开花期间的差异。 展开更多

Background As the most widely cultivated fiber crop,cotton production depends on hybridization to unlock the yield potential of current varieties.A deep understanding of genetic dissection is crucial for the cultivation of enhanced hybrid plants with desired traits,such as high yield and fine fiber quality.In this study,the general combining ability(GCA)and specific combining ability(SCA)of yield and fiber quality of nine cotton parents(six lines and three testers)and eighteen F1 crosses produced using a line×tester mating design were analyzed.Results The results revealed significant effects of genotypes,parents,crosses,and interactions between parents and crosses for most of the studied traits.Moreover,the effects of both additive and non-additive gene actions played a notably significant role in the inheritance of most of the yield and fiber quality attributes.The F1 hybrids of(Giza 90×Aust)×Giza 86,Uzbekistan 1×Giza 97,and Giza 96×Giza 97 demonstrated superior performance due to their favorable integration of high yield attributes and premium fiber quality characteristics.Path analysis revealed that lint yield has the highest positive direct effect on seed cotton yield,while lint percentage showed the highest negative direct effect on seed cotton yield.Principal component analysis identified specific parents and hybrids associated with higher cotton yield,fiber quality,and other agronomic traits.Conclusion This study provides insights into identifying potential single-and three-way cross hybrids with superior cotton yield and fiber quality characteristics,laying a foundation for future research on improving fiber quality in cotton. 展开更多

Background:Potassium(K)deficiency has become a common field production problem following the widespread adoption of Bacillus thuringiensis(Bt)transgenic cotton(Gossypium hirsutum L.)worldwide.The purpose of this study was to clarify whether the introduction of Bt gene directly reduces the K-use efficiency of cotton to induce K deficiency.Results:The cotton variety,Jihe 321(wild type,WT)and its two Bt(Cry1Ac)-transgenic overexpression lines(OE-29317,OE-29312)were studied in field with low soil-test K+(47.8 mg·kg^(−1)).In the field with low soil-test K+,only OE-29317 had less biomass and K+accumulation than the WT at some growth stages.Both Bt lines produced similar or even greater seed cotton yield than WT in the field.When the Bt gene(~70%)in OE-29317 and OE-29312 plants was silenced by virus-induced gene silencing(VIGS),the VIGS-Bt plants did not produce more biomass than VIGSgreen fluorescent protein(control)plants.Conclusions:The introduction of Bt gene did not necessarily hinder the K use efficiency of the cotton lines under this study. 展开更多

DnaJ蛋白不仅在生物体应对热激胁迫方面起作用,而且能响应盐、重金属和氧化等多种胁迫,因此,DnaJ蛋白的相关研究对植物抗逆研究具有重要意义。在前期的研究中,笔者所在课题组已经利用电子克隆及逆转录-聚合酶链式反应(RT-PCR)的方法从番茄中克隆了全长为465 bp的热激蛋白基因LeDnaJ。为了进一步验证其功能,针对LeDnaJ基因构建了含有35S启动子及NOS终止子的植物表达载体,通过农杆菌介导法转化到陆地棉(Gossypium hirsutum Linn.)品系R15中。目的基因的PCR检测结果表明,LeDnaJ基因已经整合到陆地棉R15基因组中;耐盐性鉴定结果表明,LeDnaJ基因的表达,提高了陆地棉R15萌发期、苗期的耐盐性。该研究结果为棉花耐盐性的改良提供了新的基因资源。 展开更多

Background:Calmodulin(CaM)is one of the most important Ca^(2+)signaling receptors because it regulates diverse physiological and biochemical reactions in plants.CaM functions by interacting with CaM-binding proteins(CaMBPs)to modulate Ca^(2+)signaling.IQ domain(IQD)proteins are plant-specific CaMBPs that bind to CaM by their specific CaM binding sites.Results:In this study,we identified 102 GhIQD genes in the Gossypium hirsutum L.genome.The GhIQD gene family was classified into four clusters(Ⅰ,Ⅱ,Ⅲ,andⅣ),and we then mapped the GhIQD genes to the G.hirsutum L.chromosomes.Moreover,we found that 100 of the 102 GhIQD genes resulted from segmental duplication events,indicating that segmental duplication is the main force driving GhIQD gene expansion.Gene expression pattern analysis showed that a total of 89 GhIQD genes expressed in the elongation stage and second cell wall biosynthesis stage of the fiber cells,suggesting that GhIQD genes may contribute to fiber cell development in cotton.In addition,we found that 20 selected GhIQD genes were highly expressed in various tissues.Exogenous application of MeJA significantly enhanced the expression levels of GhIQD genes.Conclusions:Our study shows that GhIQD genes are involved in fiber cell development in cotton and are also widely induced by MeJA.Thw results provide bases to systematically characterize the evolution and biological functions of GhIQD genes,as well as clues to breed better cotton varieties in the future. 展开更多

Background:Fiber,as the main product of cotton,provides main raw material for the textile industry.Many key factors have been revealed a significant role in fiber cell development including Myb proteins,phytohormones,fatty acid metabolites,and epigenetic modifications.DNA methylation is one of the important epigenetic modifications to regulate plant development and responses to abiotic or biotic stimuli.In general,DNA methylation consisting of 5mC and 6mA regulates the chromatin structure and gene transcription to affect plant development,however,the detailed role and underlying mechanism of DNA methylation in the fiber development of cotton are yet vague.Results:Here,systematical study of the 5mC and 6mA DNA methylation profiles during the fiber initiation period of Xu142 and its glabrous mutant Xu142fl represented a clear alteration of global DNA methylation associated with fiber cell initiation.Then,the genome-wide identification of genes responsible for methylation regulation at the fifth carbon of cytosine and the sixth carbon of adenine of DNA was operated in Gossypium hirsutum.As a result,13,10,6,and 17 genes were identified for 5mC methylation,5mC demethylation,6mA methylation,and 6mA demethylation,respectively.We then investigated the tissue expression pattern of all these genes,and some genes showed higher expression levels in fiber initiation,among which some displayed a significant change in transcription between Xu142 and Xu142fl.The possible interaction between histone acetylation and DNA methylation in fiber initiation through in vitro culture was studied by dot blot,and the results showed that repressed histone deacetylation by Trichostatin A(TSA)inhibited the global DNA methylation,and some causal genes(e.g.,GhDMT13,GhDAMT2,GhALKBH12,GhDM7)were also identified.Conclusions:In this study,all the findings indicated the interplay between histone acetylation and DNA methylation,supporting their important roles and providing precious clues for the epigenetic modifications associated with DNA methylation in the fiber development of cotton. 展开更多

Background:Cotton(Gossypium hirsutum),the major textile fiber crop ofthe world,is negatively affected by salinity.It leads to the induction of adverse effects on growth and development of cott on.The overall yield of cotton faces major drawback once they are grown in saline soil.To improve cotton salt tolerance,tunsgenic approach offers a fast and effective way but it relies on the availability of salt tolerance genes.Results:In this study,we have reported the evaluation of ThST103,a homologue of Arabidopsis ozone-induced protein(AtOZI1)in Thellungiellahalophila,in enhancing salt tolerance in cotton.Overexpression ofThST103 enabled cotton plants to germinate and grow better than the wild types under salt stress.The transgenic lines showed enhanced survival rate in the saline environment and experienced less oxidative damage compared with the wild types.In the field,the transgenic cotton lines produced higher yield than the wild type in saline soil.Transcriptomic comparison analyses of ThST103 overexpression lines versus the wild type revealed upregulated genes enriched in salt stress tolerance and ion homeostasis.Conclusions:Our results dem on strate that ThST103 has the capability to improve salt tolerance in cotton.It can be used in cotton breeding for salt tolerance cultivars. 展开更多

Background: Nucleobase-ascorbate transporters(NAT), synonymously called nucleobase-cation symporter 2(NCS2) proteins, were earlier reported to be involved in plant growth, development and resistance to stress. Previous studies concluded that s a polymorphic SNP associated with NAT12 was significant di erent between salt-tolerant and salt-sensitive materials of upland cotton. In current study, a comprehensive analysis of NAT family genes was conducted for the first time in cotton.Results: In this study, we discovered 32, 32, 18, and 16 NAT genes in Gossypium hirsutum, G. barbadense, G. raimondii and G. arboreum, respectively, which were classified into four groups(groups I–IV) based on the multiple sequence analysis. These GhNAT genes were unevenly distributed on At and Dt sub-genome in G. hirsutum. Most GhNAT members in the same group had similar gene structure characteristics and motif composition. The collinearity analysis revealed segmental duplication as well as tandem duplication contributing to the expansion of the GhNATs. The analysis of cis-acting regulatory elements of GhNATs showed that the function of GhNAT genes in cotton might be related to plant hormone and stress response. Under di erent conditions, the expression levels further suggested the GhNAT family genes were associated with plant response to various abiotic stresses. GhNAT12 was detected in the plasma membrane. And it was validated that the GhNAT12 gene played an important role in regulating cotton resistance to salt and drought stress through the virus-induced gene silencing(VIGS) analysis.Conclusions: A comprehensive analysis of NAT gene family was performed in cotton, including phylogenetic analysis, chromosomal location, collinearity analysis, motifs, gene structure and so on. Our results will further broaden the insight into the evolution and potential functions of NAT genes in cotton.
