The response of glutathione(GSH) content and glutathione S-transferease(GST) activity to the acetochlor in roots and shoots of the maize 'Dongnong248',the sorghum 'Aoza No.2' and millet'Yugu' was evaluated.The concentrations of pre-emergence acetochlor causing a 50% inhibition of plant shoot height were 25 μmol·L^(-1) for the tolerant 'Dongnong248' maize,5 μmol·L^(-1) for the sensitive 'Aoza No.2' sorghum and 0.5 μmol·L^(-1) for the very sensitive 'Yugu'millet.Pre-treatment with 10 μmol·L^(-1) of acetochlor induced the root GST activities and nonprotein thiol content of all three cultivars.The induction of root GST activities and nonprotein thiol content compared to controls are observed on the fourth day after acetochlor treatment,The extents of activity and content increase from the higher to the lower were:tolerant maize cultivar 'Dongnong248'>sorghum cultivar'Aoza No.2'>millet cultivar 'Yugu'.The activities and contents induced in shoots were similar to that in roots,but the degrees of increase were less.Under different concentration treatment,the thiol content and GST activities increased with the herbicide concentration rising,then reached their peaks and began to decrease in all tested crop seedlings.The extent of induced GST activities and thiol content correlated well with differential cultivar resistance to acetochlor,so their protective mechanism appears to be strongly dependent on the endogenous levels of GSH and activities of GST. 展开更多

The effects of various toxic carbonyls such as malondialdehyde(MDA),a secondary product of lipid peroxidation,and other aldehydes on rheological parameters and their relationship with aging-associated alterations were studied.Both MDA and glutaraldehyde(Glu) in different concentrations significantly increase viscosity,plastic viscosity and yield stress of human plasma and erythrocyte suspensions.MDA(20 mmol/L) reduces sharply the typical fluorescence of proteins(excitation 280 nm/emission 350 nm),and produces age pigment-like fluorescence with a strong emission peak at 460 nm when excites at 395 nm by only being incubated for some hours.In contrast,Glu decreases merely the fluorescence of proteins without producing age pigment-like fluorescence.These data suggest interestingly that the MDA-induced gradual protein cross linking seems to form from different mechanisms compared to the fast rheological changes of blood materials which may take place either in acute and chronic diseases or during aging.On the other hand,MDA induces various deleterious alterations of erythrocytes whereas glutathione(GSH) inhibits the MDA-related carbonyl stress in a concentration-dependent manner.The results indicate that carbonyl-amino reaction exists in the blood widely and GSH has the ability to interrupt or reverse this reaction in a certain way.It implies that carbonyl stress may be one of the important factors in blood stasis and suggests a theoretical and practical approach in anti-stresses and anti-aging. 展开更多

OBJECTIVE Intracellular aggre⁃gation ofα-synuclein(SNCA)is one of the core pathological features of neurodegenerative disor⁃ders including Parkinson disease,whilst the detailed mechanism for consequently neuron loss has not been fully illustrated.Since the altered phospholipid homeostasis has been suggested to play a role in synucleinopathy,this study aims to depict the fully-featured status of phospholip⁃ids and the targets reposingα-synuclein-related neurotoxicity.METHODS SNCAA53T transgenic mice were utilized as the model of Parkinson disease.Behavioral tests including pole test,rotarod test and gait analysis were conducted to assess the motor features of Parkinsonism.Tyro⁃sine hydroxylase were determined by immunohis⁃tochemistry.Glutathione,dopamine,3,4-dihy⁃droxyphenylacetic acid and homovanillic acid were determined by HPLC-ECD analysis.Assess⁃ment of lipid peroxidation included MDA assay and Liperfluo staining.Phospholipid-omics was analyzed based on LC-MS/MS.Investigation on mechanism was relied on Western blotting and qPCR assay.The injection of AAV into midbrain was achieved by ultra-micro injection pump to obtain the target genotype.RESULTS SNCAA53T transgenic mice displayed progres⁃sively deteriorated motor coordination functions and the mechanisms were related with lipid per⁃oxidation and ferroptosis,which might help to explain the parkinsonism.These hydroperoxides were observed on plasm membrane and were further characterized by LC-MS/MS-based phos⁃pholipid-omics analysis.α-synucleinA53T trans⁃genic mice displayed distinct patterns of phos⁃pholipid peroxidation in midbrain regions com⁃pared to wild type littermates.Among different subtypes of oxidized phospholipids,oxidative phosphatidylcholine(PC-ox)was more promi⁃nently elevated.Phospholipid peroxidation is believed as a biomarker of ferroptosis,which is largely a specialized death program caused by insufficiency of glutathione peroxidase-4(GPX4),the only known enzyme that can reduce lipid hydroperoxides within biological membranes.The deficiency of Gpx4 was demonstrated to be responsible forα-synuclein-induced lipid peroxi⁃dation,and the cell lines and mouse models underwent genetic Gpx4 downregulation showed exacerbated dopaminergic neuron loss and par⁃kinsonism.On the other hand,the potentiation of Gpx4 expression remarkably inhibited dopami⁃nergic ferroptotic death and behavioral deficits in a mouse model with synucleinopathy.CONCLU⁃SION A cellular pathway that Gpx4 deficit-medi⁃ated phospholipid peroxidation and behavioral consequence participated in synucleinopathy,suggesting a potential strategy targeting Gpx4 to alleviate PD symptoms. 展开更多

OBJECTIVE To assess whether N-acetylcysteine(NAC)and reduced glutathione(GSH)are effective in reversing flupirtine-induced hepatotoxicity and whether they have other beneficial effects when combined with flupirtine.METHODS The analgesic effects of NAC and flupirtine were first evaluated in carrageenaninduced inflammatory pain and paclitaxel-induced neuropathic pain.The combination subthreshold⁃ing approach was then used to determine whether the combination of NAC and flupirtine produced synergistic analgesic effects.Hepatotoxicity markers and histopathological examination of the liver were used to assess the efficacy of NAC and GSH in reversing flupirtine-induced hepato⁃toxicity.Finally,the effect of GSH on the safe range of flupirtine was assessed in an acute tox⁃icity assay.RESULTS Flupirtine and NAC pro⁃duced dose-dependent antiallodynic effects evoked by carrageenan and paclitaxel in mice.In the above model,the combination of NAC and flupirtine produced an unexpected synergistic analgesic effect.There were no significant differ⁃ences observed in the hepatotoxicity markers and liver histopathology between the experimen⁃tal group and the control group under NAC and GSH treatment.Finally,GSH(200 mg·kg^(-1))expanded the therapeutic index of flupirtine by 1.77 times.CONCLUSION NAC and GSH are effective in preventing liver damage caused by long-term flupirtine use,which provides a solu⁃tion for the safe and effective treatment of chronic pain with flupirtine.In addition,the other benefi⁃cial effects of NAC and GSH when combined with flupirtine may provide the basis for the devel⁃opment of a new therapy with minimal sideeffects and good efficacy. 展开更多

Coacervation of oxidized glutathione(GSSG)and a cationic surfactant,didodecyldimethylammonium bromide(DDAB),was constructed mainly driven by the electrostatic and hydrophobic interactions.The pH-dependent coacervate of GSSG-DDAB(1∶4,mol/mol)was analyzed.Under acidic and neutral conditions,a turbid suspension of droplets is observed,and alkaline pH results in the phase separation of coacervates as the top phase.The coacervate phase exhibits good performance(extraction efficiency>85%)in extracting several dyes from water,including brilliant yellow,acid red 13,cresyl violet acetate,eriochrom blue SE,and 4-hydroxyazobenzene.The dyes are added into the suspension in acidic conditions.Then,the dyes are enriched and extracted along with the coacervates as the top phase when pH is adjusted to~10.Coacervation of GSSG with DDAB provides a simple approach to extract organic pollutants in wastewater treatment. 展开更多

Effect of Glutathione and Storage Time on Rheological Properties of Per-proofed Frozen Dough

The effect of reduced glutathione (GSH) on fresh and pre-proofed frozen dough rheological properties were investigated using dynamic stress rheometry and small scale extensibility with the addition of three levels (80×10-6, 160× 10-6 and 240×10-6 GSH) and six storage times (0 and 1 day, 2, 4, 6 and 8 weeks). Three relaxation times (1, 13 and 26min) after loading the dough in the rheometer were used to determine storage (G’) and loss (G”) moduli. Correlations for G’ (r=0.678 and 0.622 at 0.05, and 10Hz, respectively) and G” (r=0.699, and 0.690 at 0.05, and 10Hz, respectively) were observed with the area under the extension curve at 26 min relaxation time. The addition of GSH to fresh dough reduced G’ (16.4% to 55.9%) and G” (13.7% to 52.2%). Freezing and frozen storage caused increase in G’ and G”. The addition of GSH reduced dough strength indicated by the reduction in maximum resistance to extension (Rmax) and the ratio of maximum resistance to extensibility (Rmax/E). The reduction in Rmax across all relaxation times ranged from 16.2% to 59.4%. An increase in dough extension (E) was observed with 240×10-6 GSH at all frozen storage and rest period times. Addition of GSH caused an increase of liquid phase (30.6% to 35.3%) in fresh dough and frozen dough (10.3% to 20.7%) after one day frozen storage. Negative correlations of water content in the solid phase with dough extensibility and area under the extensibility curve were found (r=-0.594 and-0.563, respectively, p<0.001). This suggests a loss of dough extensibility and strength as the water holding capacity of the dough components changes during frozen storage. 展开更多

Transcriptome analysis of taro(Colocasia esculenta)leaves under drought stress

【Objective】This study aimed to clarify the key pathways and related genes of taro leaves in response to drought stress,analyze the gene expression patterns under drought conditions,and explore the molecular response... 【Objective】This study aimed to clarify the key pathways and related genes of taro leaves in response to drought stress,analyze the gene expression patterns under drought conditions,and explore the molecular response mecha‐nisms.The findings would provide theoretical references for understanding the molecular mechanisms of taro’s drought regulation and for breeding different drought tolerant taro varieties in the future.【Method】Using Lipu taro as the experi‐mental material,leaf samples were collected after consecutive 7 d of drought treatment as the treatment group,while leaf samples from plants watered daily served as the control group.Transcriptome sequencing was performed to identify dif‐ferentially expressed genes,which were then subjected to GO functional annotation and KEGG pathway enrichment analysis.【Result】Under drought stress,there were 1613 differentially expressed genes(DEGs),including 1043 upregulated and 570 down-regulated genes.GO functional annotation analysis revealed that the DEGs were categorized into three major functional groups:molecular function,cellular component,and biological process.In the molecular function category,DEGs were annotated to binding and catalytic activity.In the cellular component category,DEGs were anno‐tated to cellular anatomical entities and protein-containing complexes.In the biological process category,DEGs were an‐notated to cellular processes and metabolic processes.KEGG signaling pathway enrichment analysis showed that 85.00%of the DEGs were enriched in metabolic pathway.Among these,the DEGs were primarily enriched in the glutathione me‐tabolism pathway and the starch and sucrose metabolism pathway,with 11 and 19 DEGs identified in each pathway re‐spectively.Under drought stress,a total of 112 differentially expressed transcription factors(TFs)were identified,mainly including members of the bHLH,ERF,WRKY and NAC families.Among all differentially expressed TFs,82.14%showed up-regulated transcription levels under drought conditions.Plant hormone signal transduction,carotenoid biosynthesis,and the MAPK signaling pathway-plant were identified as key abscisic acid-responsive pathways involved in drought response,influencing stomatal closure in taro leaves and seed dormancy to cope with drought stress.The reliability of the transcriptome data was confirmed by quantitative real-time PCR analysis.【Conclusion】Under drought stress,the gene expression in the glutathione metabolism pathway,the starch and sucrose metabolism pathway,and transcription factors in taro leaves is affected.Most TFs are positively involved in regulating taro plant’s drought response. 展开更多
