It is shown that the migration that the (tr) is a function of electric (Q), relative molecular mass (Mr), conformation parameter (Rc) of peptides and electrophoretic condition parameter(A). The conformation parameter is introduced to characterize multifarious shapes owing to the complex conlormation and the various kinds of macromolecules, where Rc≥1/3. The parameters A and Rc can be obtained from experimental data. The times of migration of the nine standard peptides were measured in pH 2.5buffer on different electrophoretic conditions in CZE. Methods Mixture of β cyclodextrin and glutamate β cyclodextrin was used as a chiral selector to the separation of chlorpheniramine. The effects of the concentrations of β cyclodextrin and glutamate β cyclodextrin, the voltage and the pH of buffers on the separation of chlorpheniramine were studied. Results The chiral recognization of the mixed chiral selector was better than that of a single selector. At pH below 4, the pH of buffers did not affect the separation and the separation of chlorpheniramine had good reproducibility. Conclusion Using a mixture of β CD and Glu β CD as chiral selector is useful for the enantioseparation of Chp in CE.展开更多

Another species, A. cathayana Fu et al., one of six ...The genus Armeniaca Scop. is well known for its popular cultivated edible fruit trees such as Armeniaca vul- garis Lam. and ornamental flowers such as A. mume Sieb. Another species, A. cathayana Fu et al., one of six important dry fruits (kernel-using apricot), is cultivated for its edible seeds in North China. In the present study, DNA from 70 individuals of A rmeniaca, including 38 of A. cathayana, 18 ofA. vulgaris, 12 ofA. sibirica, 1 ofA. dasycarpa and 1 of A. mume, was extracted and analyzed using microsatellites and capillary electrophoresis. For 20 polymorphic loci selected, 339 alleles and 140.7 effective alleles were detected. The number of alleles per locus ranged from 8 to 28, with an average of 16.95 alleles per locus. The observed heterozygosity (Ho) and the expected heterozygosity (He) ranged from 0.427 to 0.971 and from 0.737 to 0.912, respectively. The polymorphism information content varied from 0.708 to0.905, with an average of 0.827. Based on the genetic similarity among 70 individuals, a UPGMA was used to establish the phylogenetic relationships. The taxonomic positions among five species were clearly revealed, and A. cathayana was more closely related to A. vulgaris than to A. sibirica. The results will provide a scientific basis for research on the taxonomy, germplasm resources and breeding ofArmeniaca, especially for A. cathayana.展开更多

Methods The extracts of Dermatophagoides pteronyssinus were prepared with Coca's solution, lysis buffer of 2-DE, and Trizol reagent, respectively. Bicinchoninic acid (BCA) assay was used to determine the total protein concentration of the samples. The efficiency of different protein extraction methods were evaluated with 2-DE analysis. Results The concentrations of extracted protein by methods of Coca's solution, lysis buffer, and Trizol reagent were 0.63 g/L, 0.90 g/L, and 0.80 g/L, respectively. The 2-DE analysis results showed that some protein spots in low molecular weight (LMW) range could be detected with the Coca's solution method. With the lysis buffer of 2-DE method, more protein spots in LMW range could be detected, while the medium molecular weight (MMW) protein spots were absent. Several MMW protein spots (174-178 kD and 133 kD) and more LMW protein spots were detected withTrizol reagent method. Conclusions Among Coca's solution, lysis buffer of 2-DE, and Trizol reagent, the concentration of extracted protein of Dermatophagoides pteronyssinus by lysis buffer of 2-DE is the highest. However, most protein components of Dermatophagoides pteronyssinus purified mite bodies can be extracted by Trizol reagent, which may generally reflect the whole profile of Dermatophagoides pteronyssinus allergens.展开更多

To optimize the conditions for the establishment of two-dimensional gel electrophoresis(2-DE)of soybean protein isolate(SPI),we investigated Ampholine mixture,anodic and cathodic electrolytes,loading amount of sample,...To optimize the conditions for the establishment of two-dimensional gel electrophoresis(2-DE)of soybean protein isolate(SPI),we investigated Ampholine mixture,anodic and cathodic electrolytes,loading amount of sample,acrylamide concentration,p H gradient and gel staining method in twodimensional gel electrophoresis to optimize the protein imaging conditions in two-dimensional gel.The results of mixed-level design experiments showed that Ampholine,loading amount and gel staining method had significant effect(P<0.05)on 2-DE of SPI.The optimal conditions were Ampholine mixture(pH 3–10+pH 5–7 or pH 4–6+pH 5–7),loading amount of 2 mg sample and silver staining.Although the acrylamide concentration of the gel,the p H gradient,the anodic and cathodic electrolyte solutions had significant statistical effects on the protein separation degree,the complexity of the protein composition and the visibility of the gel images were more inclined to the 12%gel,the 3–10 pH gradient and the H3PO4/NaOH electrolyte.According to the established conditions,the hydrolyzed products of SPI emulsion were determined by 2-DE,and the dynamic changes of protein in the process of enzymatic hydrolysis were described.展开更多

The pre-see...Y-type zeolite membranes were synthesized by a two-step approach in which a particle seed layer was prepared by electrophoresis deposition(EPD) at first, followed by densification through secondary growth. The pre-seeding adopted the directing agent for Y-type zeolite synthesis serving as seeds. The effects of aging time of the directing agent, electrophoresis voltage and electrophoresis deposition time on seed layers quality as well as the quality of zeolite membranes were investigated. The results indicated that the zeolite seeds derived from the directing agent could be evenly deposited on substrate under certain EPD conditions. The XRD patterns of the seeded substrates after the secondary growth showed that the pure as-synthesized Y-type zeolite membranes had successfully grown on the substrates. The SEM images indicated that the substrate was covered by the highly intergrown zeolite crystals when the seeding solution employed the directing agent with an aging time of 2 days. The separation performance of zeolite membrane was evaluated using a CO_2/N2 mixture(with a mole ratio of 1:1) at different temperatures. Furthermore, the pervaporation measurements were carried out for the dehydration of isopropanol aqueous solutions with different mass fractions. The as-synthesized Y-type zeolite membranes exhibited a relatively high selectivity of water from isopropanol and sustainable permeation flux.展开更多

In this study, BDD films are taken as a two-dimensional (2D...Chemically robust conductive p-type boron-doped diamond (BDD) films are an important electrode material and have been widely applied in electrochemistry. In this study, BDD films are taken as a two-dimensional (2D) electrode in a eleetrophoresis tank system instead of the conventional one-dimensional platinum wire electrode. The theoretical simulations by finite element numerical analysis reveal that the 2D BDD electrodes have relatively high intensity and uniformity of electric field in the tank. Experimentally, the 2D BDD electrodes with smaller size show excellent properties for the separation of DNA fragments. The advantages of the 2D BDD electrodes with chemical inertness, sustainability, high intensity and uniformity electronic field, as well as reduced small size of electrophoresis tank would open a possibility for realizing new generation, high-performance biological devices.展开更多
We measured the intrinsic electrophoretic drag coefficient of a single charged particle by optically trapping the particle and applying an AC electric field,and found it to be markedly different from that of the Stoke...We measured the intrinsic electrophoretic drag coefficient of a single charged particle by optically trapping the particle and applying an AC electric field,and found it to be markedly different from that of the Stokes drag.The drag coefficient,along with the measured electrical force,yield a mobility-zeta potential relation that agrees with the literature.By using the measured mobility as input,numerical calculations based on the Poisson-Nernst-Planck equations,coupled to the Navier-Stokes equation,reveal an intriguing microscopic electroosmotic flow near the particle surface,with a well-defined transition between an inner flow field and an outer flow field in the vicinity of electric double layer’s outer boundary.This distinctive interface delineates the surface that gives the correct drag coefficient and the effective electric charge.The consistency between experiments and theoretical predictions provides new insights into the classic electrophoresis problem,and can shed light on new applications of electrophoresis to investigate biological nanoparticles.展开更多

Methods: Two-dimensional gel electrophoresis was performed on 4 normal sperm samples from healthy men and 4 sperm samples from 4 asthenospermia patients. After silver staining, the differential expression proteins were analyzed by PDQuest 2D analysis software. Results: Six differential protein spots were identified. Four spots showed increased expression in the control gels compared with the patient gels. Conclusion: The protein profiles of differential expression between the normal spermatozoa and idiopathic asthenospermia were established and some differential proteins were found. The data of this study would establish the better fundament for further isolation and identification of differentially expressed proteins in human asthenospermia sperm.展开更多

Methods: Adult rat ventricular myocytes were isolated, cultured, and identified, and pretreated without or with 100 μmol/L diazoxide for 10 min. Phosphoproteins prepared and enriched from the control and diazoxide-pretreated cells were separated by two-dimensional gel electrophoresis (2-DE) followed by sliver staining. The obtained interesting phosphoproteins were further identified by mass spectrometry. Results. Associated with diazoxide preconditioning, the proteins of chaperonin containing TCP-1 and hypothetical protein XP-346548 were phosphorylated significantly (P〈0. 01), while the 94-kDa glucose-regulated protein, calpactin I heavy chain and ferritin were dephosphorylated markedly (P〈0. 01). Conclusion: These findings suggest that cardiomyocytes undergo significant posttranslational modification via phosphorylation in a multitude of proteins in order to respond diazoxide preconditioning, and these phosphorylated protein may mediate the downstream signaling of cardioprotection by mitochondrial KATp channel opening induced by ischemic preconditioning.展开更多

Non- denatured basic proteins, suc...This work presents an approach to build a high-performance, low-viscous and replaceable separation matrix, semi-crosslinked polyacrylamide (semi-CPA) capillary gel electrophoresis. Non- denatured basic proteins, such as lysozyme, cytochrome C, ribonuclease A and trypsin were separa- ted. The impacts of monomer and cross-linker concentrations on protein separation were studied, and the ability of dynamic capillary inner wall coating was demonstrated. The UV absorption interfer- ence by semi-CPA gel matrix was successfully overcome by a partial filling technique, which results in sensitivity 20 times higher than other protein separation method. The excellent separation ability, reproducibility and dynamic coating ability made semi-CPA an ideal separation media in both capillar- y electrophoresis and microfluidic chip separation scheme.展开更多

One of the most importance in clinical analysis test is the determination of cholesterol level in blood, because its increase is associated with coronary heart disease which is a major cause of death world over. Polydimethylsiloxane (PDMS) was material used to make a MCE for determination of cholesterol. Hydrogen peroxide was generated from the oxidation of cholesterol with cholesterol oxidase (ChOx) and was detected electrochemically at a downstream gold (Au) wire electrode by amperometry. Various parameters, such as the detection potential, concentration of running buffer, pH of running buffer, separation voltage, injection time, and concentration of ChOx, were studied. The migration time of cholesterol is less than 100s and the calibration curve is linear from 50 mg.dL-1 to 250 mg-dL-1 with the coefficient of determination (R2) of 0.9955. Therefore, this proposed assay is very rapid and sensitive for the detection of cholesterol.展开更多

They are heptakis{2,6-di-O-[3-L-(1-isopropyl carboxyl methyl amino)-2-(hydroxy) propyl]}-β-cyclodextrin (i.e. L-Val-β-CD), heptakis{2,6-di-O-[3-L-(1-benzyl carboxyl methyl amino)-2-hydroxy propyl]}-β-cyclodextrin (i.e. L-Phe-β-CD), heptakis{2,6-di-O-[3-(D, L-1-benzyl carboxyl methyl amino)-2-hydroxy propyl]}-β-cyclodextrin (i.e. D,L-Phe-β-CD), heptakis{2,6-di-O-[3-(L-1-hydroxymethyl carboxyl methyl amino)-2-hydroxy propyl]}-β-cyclodextrin (i.e. L-Ser-β-CD), heptakis{2,6-di-O-[3-(L-1-carboxylmethyl carboxyl methyl amino)- 2-hydroxy propyl]}-β-cyclodextrin (i.e. L-Asp-β-CD), heptakis{2,6-di-O-[3-(L-2-carboxyl tetramethylene amino)-2-hydroxy propyl]}-β-cyclodextrin (i.e. L-Pro-β-CD). Their chemical structures were certified using FTIR and ()~1H NMR. Except for L-Phe-β-CD and D,L-Phe-β-CD, that are in soluble in water, the other amino acid derived β-CDs all have good water solubility. D,L-tyrosine and promethazine were baselinely separated by L-Val-β-CD in capillary electrophoresis.
文摘Pulsed field gel electrophoresis(PFGE) has been firstly introduced in characterization of the pathogenic fungi Penicillium marne f fei and Exophiala dermatitidis genomes.The numbers and sizes of their chromosomes have been detected.Polymorphism was identified on the smallest chromosome of E.dermatitidis.The result shows that PFGE for characterization of large molecular DNA pathogenic fungi is very suitable,it is more simple and more efficacy.The result also shows the diversity of pathogenic fungi is relative common even in rare occurred pathogenic fungi such as E.dermatitidis.

文摘To optimize the conditions for the establishment of two-dimensional gel electrophoresis(2-DE)of soybean protein isolate(SPI),we investigated Ampholine mixture,anodic and cathodic electrolytes,loading amount of sample,acrylamide concentration,p H gradient and gel staining method in twodimensional gel electrophoresis to optimize the protein imaging conditions in two-dimensional gel.The results of mixed-level design experiments showed that Ampholine,loading amount and gel staining method had significant effect(P<0.05)on 2-DE of SPI.The optimal conditions were Ampholine mixture(pH 3–10+pH 5–7 or pH 4–6+pH 5–7),loading amount of 2 mg sample and silver staining.Although the acrylamide concentration of the gel,the p H gradient,the anodic and cathodic electrolyte solutions had significant statistical effects on the protein separation degree,the complexity of the protein composition and the visibility of the gel images were more inclined to the 12%gel,the 3–10 pH gradient and the H3PO4/NaOH electrolyte.According to the established conditions,the hydrolyzed products of SPI emulsion were determined by 2-DE,and the dynamic changes of protein in the process of enzymatic hydrolysis were described.

文摘Y-type zeolite membranes were synthesized by a two-step approach in which a particle seed layer was prepared by electrophoresis deposition(EPD) at first, followed by densification through secondary growth. The pre-seeding adopted the directing agent for Y-type zeolite synthesis serving as seeds. The effects of aging time of the directing agent, electrophoresis voltage and electrophoresis deposition time on seed layers quality as well as the quality of zeolite membranes were investigated. The results indicated that the zeolite seeds derived from the directing agent could be evenly deposited on substrate under certain EPD conditions. The XRD patterns of the seeded substrates after the secondary growth showed that the pure as-synthesized Y-type zeolite membranes had successfully grown on the substrates. The SEM images indicated that the substrate was covered by the highly intergrown zeolite crystals when the seeding solution employed the directing agent with an aging time of 2 days. The separation performance of zeolite membrane was evaluated using a CO_2/N2 mixture(with a mole ratio of 1:1) at different temperatures. Furthermore, the pervaporation measurements were carried out for the dehydration of isopropanol aqueous solutions with different mass fractions. The as-synthesized Y-type zeolite membranes exhibited a relatively high selectivity of water from isopropanol and sustainable permeation flux.

文摘Chemically robust conductive p-type boron-doped diamond (BDD) films are an important electrode material and have been widely applied in electrochemistry. In this study, BDD films are taken as a two-dimensional (2D) electrode in a eleetrophoresis tank system instead of the conventional one-dimensional platinum wire electrode. The theoretical simulations by finite element numerical analysis reveal that the 2D BDD electrodes have relatively high intensity and uniformity of electric field in the tank. Experimentally, the 2D BDD electrodes with smaller size show excellent properties for the separation of DNA fragments. The advantages of the 2D BDD electrodes with chemical inertness, sustainability, high intensity and uniformity electronic field, as well as reduced small size of electrophoresis tank would open a possibility for realizing new generation, high-performance biological devices.
文摘We measured the intrinsic electrophoretic drag coefficient of a single charged particle by optically trapping the particle and applying an AC electric field,and found it to be markedly different from that of the Stokes drag.The drag coefficient,along with the measured electrical force,yield a mobility-zeta potential relation that agrees with the literature.By using the measured mobility as input,numerical calculations based on the Poisson-Nernst-Planck equations,coupled to the Navier-Stokes equation,reveal an intriguing microscopic electroosmotic flow near the particle surface,with a well-defined transition between an inner flow field and an outer flow field in the vicinity of electric double layer’s outer boundary.This distinctive interface delineates the surface that gives the correct drag coefficient and the effective electric charge.The consistency between experiments and theoretical predictions provides new insights into the classic electrophoresis problem,and can shed light on new applications of electrophoresis to investigate biological nanoparticles.
文摘Objective: To evaluate the application of two-dimensional electrophoresis in the research of differentially expressed proteins in the human asthenospermia. Methods: Two-dimensional gel electrophoresis was performed on 4 normal sperm samples from healthy men and 4 sperm samples from 4 asthenospermia patients. After silver staining, the differential expression proteins were analyzed by PDQuest 2D analysis software. Results: Six differential protein spots were identified. Four spots showed increased expression in the control gels compared with the patient gels. Conclusion: The protein profiles of differential expression between the normal spermatozoa and idiopathic asthenospermia were established and some differential proteins were found. The data of this study would establish the better fundament for further isolation and identification of differentially expressed proteins in human asthenospermia sperm.
文摘Four isozymes, such as Malate dehydrogenase (MDH), Alchol dehydrogenase (ADH), Peroxidae (POD) and Esterase (EST) in six tissues (heart, liver, kidney, muscle, eye, gonad) ofMartes zibellina L., were analyzed by means of vertical polyacrylamide gel electrophoresis (PAGE). The results indicated that the zymograms of these four isozymes in different tissues were different from each other, i.e. there existed apparent tissue-specificitity in these isozymes inMartes zibellina L.. Characteristic enzyme band was found both in POD zymogram and in EST zymogram. Moreover, the characteristic enzyme band in POD isozyme would be of some value to sexual identification ofMartes zibellina L.

文摘Objectives To analyze and identify differentially expressed phosphoproteins associated with mitochondrial KATP channel opening. Methods: Adult rat ventricular myocytes were isolated, cultured, and identified, and pretreated without or with 100 μmol/L diazoxide for 10 min. Phosphoproteins prepared and enriched from the control and diazoxide-pretreated cells were separated by two-dimensional gel electrophoresis (2-DE) followed by sliver staining. The obtained interesting phosphoproteins were further identified by mass spectrometry. Results. Associated with diazoxide preconditioning, the proteins of chaperonin containing TCP-1 and hypothetical protein XP-346548 were phosphorylated significantly (P〈0. 01), while the 94-kDa glucose-regulated protein, calpactin I heavy chain and ferritin were dephosphorylated markedly (P〈0. 01). Conclusion: These findings suggest that cardiomyocytes undergo significant posttranslational modification via phosphorylation in a multitude of proteins in order to respond diazoxide preconditioning, and these phosphorylated protein may mediate the downstream signaling of cardioprotection by mitochondrial KATp channel opening induced by ischemic preconditioning.

文摘This work presents an approach to build a high-performance, low-viscous and replaceable separation matrix, semi-crosslinked polyacrylamide (semi-CPA) capillary gel electrophoresis. Non- denatured basic proteins, such as lysozyme, cytochrome C, ribonuclease A and trypsin were separa- ted. The impacts of monomer and cross-linker concentrations on protein separation were studied, and the ability of dynamic capillary inner wall coating was demonstrated. The UV absorption interfer- ence by semi-CPA gel matrix was successfully overcome by a partial filling technique, which results in sensitivity 20 times higher than other protein separation method. The excellent separation ability, reproducibility and dynamic coating ability made semi-CPA an ideal separation media in both capillar- y electrophoresis and microfluidic chip separation scheme.

文摘Microchip capillary electrophoresis (MCE) has significant impact on diagnostic testing. One of the most importance in clinical analysis test is the determination of cholesterol level in blood, because its increase is associated with coronary heart disease which is a major cause of death world over. Polydimethylsiloxane (PDMS) was material used to make a MCE for determination of cholesterol. Hydrogen peroxide was generated from the oxidation of cholesterol with cholesterol oxidase (ChOx) and was detected electrochemically at a downstream gold (Au) wire electrode by amperometry. Various parameters, such as the detection potential, concentration of running buffer, pH of running buffer, separation voltage, injection time, and concentration of ChOx, were studied. The migration time of cholesterol is less than 100s and the calibration curve is linear from 50 mg.dL-1 to 250 mg-dL-1 with the coefficient of determination (R2) of 0.9955. Therefore, this proposed assay is very rapid and sensitive for the detection of cholesterol.
文摘为了解不同性状窖泥细菌群落结构及酸酯代谢的差异,分别选取新窖泥、趋老熟窖泥和老熟窖泥,对其细菌16S r DNA的V3区进行变性梯度凝胶电泳分析和同源性比较,并结合窖泥主要有机酸及有机酸酯含量进行了典型相关分析。结果表明,老熟窖泥的细菌多样性指数及均匀度指数高于新窖泥和趋老熟窖泥,得到的39个优势条带,进行细菌DNA测序可分为14类;Clostridium XIVa、Aminobacterium均只在老熟窖泥中检测到;新窖泥和趋老熟窖泥与Lactococcus、Lactobacillus、乳酸、乳酸乙酯含量正相关,老熟窖泥与Clostridiales、己酸、己酸乙酯和丁酸含量正相关。

文摘Paulownia tomentosa, P. fargesii, P. lamprorhylla, P. albiphloea, P. australis, P. fortunei, P. elongata, P. elongata f.alba and P. albiphloea var c henggtuensis were classified into three groups: P. fortunei group (P. fortunei a nd P. elongata f. alba); P. australis group (P. australis and P. albiphloea var chenggtuensis) and P. tomentasa group (P. tomentasa, P. fargesii, P. albiprhlaca , P. lamproprhylia and P. elongata ) accordance to the results of the single and two-dimensional SDS-PAGE of protein in the Paulownia tree leaves. The result co uld lay a foundation for classifying the Genus Paulownia plants.
