【Objective】This study aimed to clarify the key pathways and related genes of taro leaves in response to drought stress,analyze the gene expression patterns under drought conditions,and explore the molecular response...【Objective】This study aimed to clarify the key pathways and related genes of taro leaves in response to drought stress,analyze the gene expression patterns under drought conditions,and explore the molecular response mecha‐nisms.The findings would provide theoretical references for understanding the molecular mechanisms of taro’s drought regulation and for breeding different drought tolerant taro varieties in the future.【Method】Using Lipu taro as the experi‐mental material,leaf samples were collected after consecutive 7 d of drought treatment as the treatment group,while leaf samples from plants watered daily served as the control group.Transcriptome sequencing was performed to identify dif‐ferentially expressed genes,which were then subjected to GO functional annotation and KEGG pathway enrichment analysis.【Result】Under drought stress,there were 1613 differentially expressed genes(DEGs),including 1043 upregulated and 570 down-regulated genes.GO functional annotation analysis revealed that the DEGs were categorized into three major functional groups:molecular function,cellular component,and biological process.In the molecular function category,DEGs were annotated to binding and catalytic activity.In the cellular component category,DEGs were anno‐tated to cellular anatomical entities and protein-containing complexes.In the biological process category,DEGs were an‐notated to cellular processes and metabolic processes.KEGG signaling pathway enrichment analysis showed that 85.00%of the DEGs were enriched in metabolic pathway.Among these,the DEGs were primarily enriched in the glutathione me‐tabolism pathway and the starch and sucrose metabolism pathway,with 11 and 19 DEGs identified in each pathway re‐spectively.Under drought stress,a total of 112 differentially expressed transcription factors(TFs)were identified,mainly including members of the bHLH,ERF,WRKY and NAC families.Among all differentially expressed TFs,82.14%showed up-regulated transcription levels under drought conditions.Plant hormone signal transduction,carotenoid biosynthesis,and the MAPK signaling pathway-plant were identified as key abscisic acid-responsive pathways involved in drought response,influencing stomatal closure in taro leaves and seed dormancy to cope with drought stress.The reliability of the transcriptome data was confirmed by quantitative real-time PCR analysis.【Conclusion】Under drought stress,the gene expression in the glutathione metabolism pathway,the starch and sucrose metabolism pathway,and transcription factors in taro leaves is affected.Most TFs are positively involved in regulating taro plant’s drought response.展开更多

Background Cotton is extremely affected by severe natural stresses.Drought is one of the most serious abiotic stress that adversely influences cotton growth,productivity,and fiber quality.Previous studies indicate tha...Background Cotton is extremely affected by severe natural stresses.Drought is one of the most serious abiotic stress that adversely influences cotton growth,productivity,and fiber quality.Previous studies indicate that basic leucinezipper(bZIP)transcription factors are involved in the response of plants to various stresses.However,the molecular function and regulatory mechanism of GhVIP1 in response to drought stress are still unknown.Results In this research,GhVIP1 was cloned from a drought-tolerant variety.Expression of GhVIP1 was up-regulated in response to multiple abiotic stresses,especially under drought stress.And GhVIP1 was highly expressed in the root,stem,and 10 days post-anthesis ovule.Inhibiting the expression of GhVIP1 in cotton using the virus-induced gene silencing method resulted in higher electrical conductivity in leaves,but lower water content under drought stress compared with the WT plant.Overexpression of GhVIP1 in Arabidopsis enhanced plant drought tolerance through increasing the seed germination rate and improving the development of root.The exogenous expression of GhVIP1 up-regulated the transcription of genes associated with drought response and proline biosynthesis during drought stress in Arabidopsis.Conclusion In summary,these results indicated that GhVIP1 played a positive role in plants’response to drought stress.The use of GhVIP1 via modern biotechnology might facilitate the improvement of drought tolerance in cotton cultivars.展开更多
In order to reveal the photosynthetic characteristics of C. trichotomum responses to drought, salt and water-logging stresses, one-year-old potted seedlings were taken as materials, and the several stresses including ...In order to reveal the photosynthetic characteristics of C. trichotomum responses to drought, salt and water-logging stresses, one-year-old potted seedlings were taken as materials, and the several stresses including natural drought, submergence stress, water-logging and different salt treatments (0.2%, 0.4%, 0.6% and 0.8% NaCl) were carried out on August 15, 2012. The morphological and photosynthetic characteristics were observed and determined. The results showed that adverse enviromental stress had a significant effect on the morphological changes and photosynthetic characteristics of C. trichotomum. On the 14th day after natural drought, the leaves wilted and could not recovery at night, and 60% of the seedlings could recover after re-watering. From the 7th day to the 10th day after submergence stress treatment, the 2nd and the 3rd leaves at the base of 60% seedling turned yellow and the lenticels were observed. At the early stage of water-logging stress, white lenticels appeared at the base of seedlings, and the leaves wilted, chlorina and fallen off on the 8th day. A large number of leaves fallen off under 0.6% NaCl or more salt stress, and even the whole plant died. The chlorophyll content, net photosynthetic rate (Pn) and transpiration rate (Tr) decreased gradually with the stress process, such as 8 days after natural drought, less than 0.4% salt stress and water-logging stress, but the changes were not significant compared with those of the control. With the increase of the stress intensity and the prolonged time, the changes of photosynthetic index were significant. All the results indicated that C. trichotomum had a certain degree of tolerance to drought, water and salt, but it was not suitable for living, in water-logging condition for a long time.展开更多
Background:Cotton is mainly grown for its natural fiber and edible oil.The fiber obtained from cotton is the indispensable raw material for the textile industries.The ever changing climatic condition,threatens cotton ...Background:Cotton is mainly grown for its natural fiber and edible oil.The fiber obtained from cotton is the indispensable raw material for the textile industries.The ever changing climatic condition,threatens cotton production due to a lack of sufficient water for its cultivation.Effects of drought stress are estimated to affect more than 50%of the cotton growing regions.To elucidate the drought tolerance phenomenon in cotton,a backcross population was developed from G.tomentosum,a drought tolerant donor parent and G.hirsutum which is highly susceptible to drought stress.Results:A genetic map of 10888 SNP markers was developed from 200 BC_2F_2 populations.The map spanned 4191.3 centi-Morgan(c M),with an average distance of 0.1047 c M,covering 51%and 49%of At and Dt sub genomes,respectively.Thirty stable Quantitative trait loci(QTLs)were detected,in which more than a half were detected in the At subgenome.Eighty-nine candidate genes were mined within the QTL regions for three traits:cell membrane stability(CMS),saturated leaf weight(SLW)and chlorophyll content.The genes had varied physiochemical properties.A majority of the genes were interrupted by introns,and only 15 genes were intronless,accounting for 17%of the mined genes.The genes were found to be involved molecular function(MF),cellular component(CC)and biological process(BP),which are the main gene ontological(GO)functions.A number of mi RNAs were detected,such as mi R164,which is associated with NAC and MYB genes,with a profound role in enhancing drought tolerance in plants.Through RT-q PCR analysis,5 genes were found to be the key genes involved in enhancing drought tolerance in cotton.Wild cotton harbors a number of favorable alleles,which can be exploited to aid in improving the narrow genetic base of the elite cotton cultivars.The detection of 30 stable QTLs and 89 candidate genes found to be contributed by the donor parent,G.tomentosum,showed the significant genes harbored by the wild progenitors which can be exploited in developing more robust cotton genotypes with diverse tolerance levels to various environmental stresses.Conclusion:This was the first study involving genome wide association mapping for drought tolerance traits in semi wild cotton genotypes.It offers an opportunity for future exploration of these genes in developing highly tolerant cotton cultivars to boost cotton production.展开更多
In this experiment,the drought-tolerant soybean variety Heinong 44 and the sensitive soybean variety Heinong 65 were used as experimental materials to study the effects of drought stress and re-watering on the osmotic...In this experiment,the drought-tolerant soybean variety Heinong 44 and the sensitive soybean variety Heinong 65 were used as experimental materials to study the effects of drought stress and re-watering on the osmotic adjustment ability and yield of soybean.The results showed that given a normal water supply,the proline and soluble protein contents of Heinong 44 and Heinong65 showed an increasing trend during the growth process,while the soluble sugar content increased and then decreased.The contents of proline,soluble protein and soluble sugar increased significantly with increasing drought stress,and the magnitude of increase of Heinong 44 was higher than that of Heinong 65.The osmotic adjustment ability of the drought-tolerant variety was significantly higher than that of the sensitive variety and showed consistent performance at different stages.After re-watering,the proline and soluble sugar contents of the flowering and pod-setting stages and the soluble protein content of the seedling stage were restored to the control levels.Drought stress resulted in a decrease in the effective pod number,pod number per pod,100-grain weight and yield.Severe drought had the greatest impact by increasing stress and decreasing soybean yields,which were followed by the flowering,bulging and seedling stages.展开更多
Background:The cotton crop is universally considered as protein and edible oil source besides the major contributor of natural fiber and is grown in tropical and subtropical regions around the world Unpredicted enviro...Background:The cotton crop is universally considered as protein and edible oil source besides the major contributor of natural fiber and is grown in tropical and subtropical regions around the world Unpredicted environmental stresses are becoming significant threats to sustainable cotton production,ultimately leading to a substantial irreversible economic loss.Mitogen-activated protein kinase(MAPK)is generally considered essential for recognizing environmental stresses through phosphorylating downstream signal pathways and plays a vital role in numerous biological processes.Results:We have identified 74 MAPK genes across cotton,41 from G.hirsutum,19 from G.raimondii,whereas 14 have been identified from G.arboreum.The MAPK gene-proteins have been further studied to determine their physicochemical characteristics and other essential features.In this perspective,characterization,phylogenetic relationship,chromosomal mapping,gene motif,cis-regulatory element,and subcellular localization were carried out.Based on phylogenetic analysis,the MAPK family in cotton is usually categorized as A,B,C,D,and E clade.According to the results of the phylogenic relationship,cotton has more MAPKS genes in Clade A than Clade B.The cis-elements identified were classified into five groups(hormone responsiveness,light responsiveness,stress responsiveness,cellular development,and binding site).The prevalence of such elements across the promoter region of these genes signifies their role in the growth and development of plants.Seven GHMAPK genes(GH_A07G1527,GH_D02G1138,GH_D03G0121,GH_D03G1517,GH_D05G1003,GH_D11G0040,and GH_D12G2528)were selected,and specific tissue expression and profiling were performed across drought and salt stresses.Results expressed that six genes were upregulated under drought treatment except for GH_D11G0040 which is downregulated.Whereas all the seven genes have been upregulated at various hours of salt stress treatment.Conclusions:RNA sequence and qPCR results showed that genes as differentially expressed across both vegetative and reproductive plant parts.Similarly,the qPCR analysis showed that six genes had been upregulated substantially through drought treatment while all the seven genes were upregulated across salt treatments.The results of this study showed that cotton GHMPK3 genes play an important role in improving cotton resistance to drought and salt stresses.MAPKs are thought to play a significant regulatory function in plants’responses to abiotic stresses according to various studies.MAPKs’involvement in abiotic stress signaling and innovation is a key goal for crop species research,especially in crop breeding.展开更多

Previous studies concluded that s a polymorphic SNP associated with NAT12 was significant di erent between salt-tolerant and salt-sensitive materials of upland cotton. In current study, a comprehensive analysis of NAT family genes was conducted for the first time in cotton.Results: In this study, we discovered 32, 32, 18, and 16 NAT genes in Gossypium hirsutum, G. barbadense, G. raimondii and G. arboreum, respectively, which were classified into four groups(groups I–IV) based on the multiple sequence analysis. These GhNAT genes were unevenly distributed on At and Dt sub-genome in G. hirsutum. Most GhNAT members in the same group had similar gene structure characteristics and motif composition. The collinearity analysis revealed segmental duplication as well as tandem duplication contributing to the expansion of the GhNATs. The analysis of cis-acting regulatory elements of GhNATs showed that the function of GhNAT genes in cotton might be related to plant hormone and stress response. Under di erent conditions, the expression levels further suggested the GhNAT family genes were associated with plant response to various abiotic stresses. GhNAT12 was detected in the plasma membrane. And it was validated that the GhNAT12 gene played an important role in regulating cotton resistance to salt and drought stress through the virus-induced gene silencing(VIGS) analysis.Conclusions: A comprehensive analysis of NAT gene family was performed in cotton, including phylogenetic analysis, chromosomal location, collinearity analysis, motifs, gene structure and so on. Our results will further broaden the insight into the evolution and potential functions of NAT genes in cotton. Current findings could make significant contribution towards screening more candidate genes related to biotic and abiotic resistance for the improvement in cotton.展开更多
Background: Cotton is an important commercial crop for being a valuable source of natural fiber.Its production has undergone a sharp decline because of abiotic stresses,etc.Drought is one of the major abiotic stress c...Background: Cotton is an important commercial crop for being a valuable source of natural fiber.Its production has undergone a sharp decline because of abiotic stresses,etc.Drought is one of the major abiotic stress causing significant yield losses in cotton.However,plants have evolved self-defense mechanisms to cope abiotic factors like drought,salt,cold,etc.The evolution of stress responsive transcription factors such as the trihelix,a nodule-inception-like protein(NLP),and the late embryogenesis abundant proteins have shown positive response in the resistance improvement to several abiotic stresses.Results: Genome wide identification and characterization of the effects of Light-Harvesting Chloro a/b binding(LHC)genes were carried out in cotton under drought stress conditions.A hundred and nine proteins encoded by the LHC genes were found in the cotton genome,with 55,27,and 27 genes found to be distributed in Gossypium hirsutum,G.arboreum,and G.raimondii,respectively.The proteins encoded by the genes were unevenly distributed on various chromosomes.The Ka/Ks(Non-synonymous substitution rate/Synonymous substitution rate)values were less than one,an indication of negative selection of the gene family.Differential expressions of genes showed that majority of the genes are being highly upregulated in the roots as compared with leaves and stem tissues.Most genes were found to be highly expressed in MR-85,a relative drought tolerant germplasm.Conclusion: The results provide proofs of the possible role of the LHC genes in improving drought stress tolerance,and can be explored by cotton breeders in releasing a more drought tolerant cotton varieties.展开更多

Silicon is considered to be a necessary element for plant growth, and soluble silicon can improve plant resilience. To improve the drought resilience of Petunia hybrida, the silicon transporter protein OsLsi1 and OsLsi2 genes cloned from rice(Oryza sative) were transferred into Petunia hybrida by Agrobacterium-mediated method, and finally got 26 and 32 positive plants, respectively by PCR and RT-PCR detections. With a control of non-transgenic plants, the obtained transgenic plants were taken by drought treatment stress for 0, 4, 7, 10 and 14 days, then re-watered and measured physiological indexes as malondialdehyde(MDA) content, free proline(Pro) content, superoxide dismutase(SOD) activity and peroxidase(POD) activity to study the effect of Petunia's drought resistance. All the results proved that the silicon transporter protein OsLsi1 and OsLsi2 genes were normally transcripted and expressed in transgenic Petunia hybrida; OsLsi1 gene could improve the abilities of plants' drought resistance and recover after drought stress, while OsLsi2 gene could reduce the above abilities. The order of the drought resistance ability of the three strains from strong to weak was OsLsi1〉CK〉OsLsi2; and silicon indeed improved the ability of drought resistance as well. All these results provided a new way to improve the drought resistance of Petunia, and laid a foundation to improve the ability of garden plants' drought resistance and water saving.展开更多

In order to determine the physiological mechanism of drought resistance of northern wheat in China,six drought resistant wheat and one sensitivity to drought wheat were planted in pots.They were subjected to drought t...In order to determine the physiological mechanism of drought resistance of northern wheat in China,six drought resistant wheat and one sensitivity to drought wheat were planted in pots.They were subjected to drought treatment and normal water when the plants grew to the 3-leaf stage.Samples were collected at 10,20,30,and 40 days after the drought treatment,respectively.The electrical conductivity,photosynthetic parameters,chlorophyll fluorescence parameters,sugar content,proline content,protein content,and active oxygen scavenging enzyme activity of the plants were detected,and the agronomic traits of the wheat varieties were investigated at maturity.The results indicated that the phenotype and yield-related factors of Darkhan 144 changed little under the drought stress.The relative electrical conductivity of Kefeng 6 and Darkhan 166 was lower under the drought stress,and their cell membrane was less damaged.The Darkhan 144 and Darkhan 166 had higher drought resistance coefficients,and were the wheat varieties with stronger drought resistance.However,the physiological mechanisms of drought resistance of these three wheat were different:Darkhan 144 maintained a higher photosynthetic activity under the drought stress;Darkhan 166 maintained a higher protein content,photosynthetic activity and active oxygen scavenging enzyme activity.In addition,other drought-resistant varieties Kefeng 6,Kefeng 10 and Longmai 26 had a higher content of osmoregulatory substances under the drought stress.展开更多

文摘【Objective】This study aimed to clarify the key pathways and related genes of taro leaves in response to drought stress,analyze the gene expression patterns under drought conditions,and explore the molecular response mecha‐nisms.The findings would provide theoretical references for understanding the molecular mechanisms of taro’s drought regulation and for breeding different drought tolerant taro varieties in the future.【Method】Using Lipu taro as the experi‐mental material,leaf samples were collected after consecutive 7 d of drought treatment as the treatment group,while leaf samples from plants watered daily served as the control group.Transcriptome sequencing was performed to identify dif‐ferentially expressed genes,which were then subjected to GO functional annotation and KEGG pathway enrichment analysis.【Result】Under drought stress,there were 1613 differentially expressed genes(DEGs),including 1043 upregulated and 570 down-regulated genes.GO functional annotation analysis revealed that the DEGs were categorized into three major functional groups:molecular function,cellular component,and biological process.In the molecular function category,DEGs were annotated to binding and catalytic activity.In the cellular component category,DEGs were anno‐tated to cellular anatomical entities and protein-containing complexes.In the biological process category,DEGs were an‐notated to cellular processes and metabolic processes.KEGG signaling pathway enrichment analysis showed that 85.00%of the DEGs were enriched in metabolic pathway.Among these,the DEGs were primarily enriched in the glutathione me‐tabolism pathway and the starch and sucrose metabolism pathway,with 11 and 19 DEGs identified in each pathway re‐spectively.Under drought stress,a total of 112 differentially expressed transcription factors(TFs)were identified,mainly including members of the bHLH,ERF,WRKY and NAC families.Among all differentially expressed TFs,82.14%showed up-regulated transcription levels under drought conditions.Plant hormone signal transduction,carotenoid biosynthesis,and the MAPK signaling pathway-plant were identified as key abscisic acid-responsive pathways involved in drought response,influencing stomatal closure in taro leaves and seed dormancy to cope with drought stress.The reliability of the transcriptome data was confirmed by quantitative real-time PCR analysis.【Conclusion】Under drought stress,the gene expression in the glutathione metabolism pathway,the starch and sucrose metabolism pathway,and transcription factors in taro leaves is affected.Most TFs are positively involved in regulating taro plant’s drought response.

文摘Total RNA from leaves of Poapratensis cultivars under drought stress was extracted for reversing transcription to cDNA and then cDNA as template for PCR reaction by designing primer of cds of Hordeum valgare HVA1 drought resistance gene from GenBank. The amplified products were positive recon identified by using procedures of recovery, connection, transformation and enzyme separation. The length of cloned gene sequence was 324 bp, identity reached 79.27% with Barley HVA1 gene that meaned the cloned gene sequence was the partial HVA1 gene of Poapratensis.
文摘Water stress treatment at different developmental stages of a winter wheat varietywas made in outdoor pot experiments from 1987 to 1988.It was found that the most sensitivestage was the booting stage and the second most sensitive stage was the flowering stage.Waterstress at most development stages affects the number of grains.Wheat water stress can be diag-nosed effectively and quickly by means of leaf temperature difference between the drought fieldand the well watered control plot measured by an infra-red thermometer.

文摘Drought is one of the most important environmental constraints limiting plant growth, development and crop yield. Many drought-inducible genes have been identified by molecular and genomic analyses in Arabidopsis, rice and other crops. To better understand reaction mechanism of plant to drought tolerance, we mainly focused on introducing the research of transcription factors (TFs) in signal transduction and regulatory network of gene expression conferring drought. A TF could bind multiple target genes to increase one or more kinds of stress tolerance. Sometimes, several TFs might act together with a target gene. So drought-tolerance genes or TFs might respond to high-salinity, cold or other stresses. The crosstalk of multiple stresses signal pathways is a crucial aspect of understanding stress signaling.

文摘The myeloblastosis oncogenes(MYB)are important transcription factors that facilitate induction of variously develop-mental and stress responsive genes.They are hence,emerging as key players in improving stress tolerance of plants in response to several abiotic stresses.It was predicted that DfMYB2 contained an open reading frame(ORF)of 1621 bp coding 377 amino acid residues with molecular weight of 41.5 ku.It had 50 potential phosphorylation sites,44 potential N-glycosylation sites and one trans-membrane domain.In neighbor-joining(NJ)phylogenetic tree,DfMYB2 was close to the branch of angiosperms.The subcellular localization of DfMYB2 was in nucleus.The expressions of DfMYB2 increased gradually in the filament,prothalli and sporophyte.The results showed that DfMYB2 played a more and more important role in its growth and development.And the expression levels in rhizomes were significantly higher than those in roots and leaves.After abscisic acid(ABA)and PEG600 treatment,DfMYB2 showed a downward trend,followed by an upward trend.The expression of DfMYB2 was inhibited in a short time under drought stress,the strong induction of DfMYB2 expression by ABA indicated that it was possibly involved in abiotic stress responses in an ABA-dependent manner.

文摘In order to develop the resources of native turfgrass,the morphological traits and drought resistance of native Siberian bluegrass(Poa sibirica,abbreviated as PS)was evaluated using the introduced Kentucky bluegrass'Midnight'(Poa pratensis,abbreviated as PP)as a control.Two water schemes were imposed to plants in this pot culture study in greenhouse.One was with drought stress persistent limiting water supply for 20 days,the other was re-hydrated until 14 days after drought.The leaf shape,turf color,water status and cell plasma membrane permeability were evaluated.Similar changing trends with these parameters were shown for both species,and there were not significant differences with most evaluations during drought and re-water periods.The values leaf width and length of PS were higher while leaf color intensity was slightly lower than that of PP,but the greenness of PS leaf was still visually acceptable.There were not significant differences with cell membrane stability between the two species.In comparison,the native wild species PS possessed the potential for to be domesticated into a new cultivar for turf industry.
文摘There is evindence showing that stress susceptibility index(SSI)(1一Yd/Yp)/(1—(?)d/(?)p)used as a measure of drought resistance of crop on the field is an altered form of droughtresistance coefficient(DRC)(Yd/Yp).The correlative coefficient SSI and DRC is r=-1.Therefore,the SSI doesn’t improve the defect of the DRC.After two years experiments per-formed by using thirty winter wheat varieties as trial materials,the concept of drought resistanceindex in crops was put forward.Its expressing equation is:the yield in drylan×drought resis-tance coefficient/average yield in dryland.It makes the drought resistance coefficient(physicalindex)correlate well with the yield in dryland(agronomy index)and is suitable for breeder.

文摘Background Cotton is extremely affected by severe natural stresses.Drought is one of the most serious abiotic stress that adversely influences cotton growth,productivity,and fiber quality.Previous studies indicate that basic leucinezipper(bZIP)transcription factors are involved in the response of plants to various stresses.However,the molecular function and regulatory mechanism of GhVIP1 in response to drought stress are still unknown.Results In this research,GhVIP1 was cloned from a drought-tolerant variety.Expression of GhVIP1 was up-regulated in response to multiple abiotic stresses,especially under drought stress.And GhVIP1 was highly expressed in the root,stem,and 10 days post-anthesis ovule.Inhibiting the expression of GhVIP1 in cotton using the virus-induced gene silencing method resulted in higher electrical conductivity in leaves,but lower water content under drought stress compared with the WT plant.Overexpression of GhVIP1 in Arabidopsis enhanced plant drought tolerance through increasing the seed germination rate and improving the development of root.The exogenous expression of GhVIP1 up-regulated the transcription of genes associated with drought response and proline biosynthesis during drought stress in Arabidopsis.Conclusion In summary,these results indicated that GhVIP1 played a positive role in plants’response to drought stress.The use of GhVIP1 via modern biotechnology might facilitate the improvement of drought tolerance in cotton cultivars.

文摘In order to reveal the photosynthetic characteristics of C. trichotomum responses to drought, salt and water-logging stresses, one-year-old potted seedlings were taken as materials, and the several stresses including natural drought, submergence stress, water-logging and different salt treatments (0.2%, 0.4%, 0.6% and 0.8% NaCl) were carried out on August 15, 2012. The morphological and photosynthetic characteristics were observed and determined. The results showed that adverse enviromental stress had a significant effect on the morphological changes and photosynthetic characteristics of C. trichotomum. On the 14th day after natural drought, the leaves wilted and could not recovery at night, and 60% of the seedlings could recover after re-watering. From the 7th day to the 10th day after submergence stress treatment, the 2nd and the 3rd leaves at the base of 60% seedling turned yellow and the lenticels were observed. At the early stage of water-logging stress, white lenticels appeared at the base of seedlings, and the leaves wilted, chlorina and fallen off on the 8th day. A large number of leaves fallen off under 0.6% NaCl or more salt stress, and even the whole plant died. The chlorophyll content, net photosynthetic rate (Pn) and transpiration rate (Tr) decreased gradually with the stress process, such as 8 days after natural drought, less than 0.4% salt stress and water-logging stress, but the changes were not significant compared with those of the control. With the increase of the stress intensity and the prolonged time, the changes of photosynthetic index were significant. All the results indicated that C. trichotomum had a certain degree of tolerance to drought, water and salt, but it was not suitable for living, in water-logging condition for a long time.

文摘Background:Cotton is mainly grown for its natural fiber and edible oil.The fiber obtained from cotton is the indispensable raw material for the textile industries.The ever changing climatic condition,threatens cotton production due to a lack of sufficient water for its cultivation.Effects of drought stress are estimated to affect more than 50%of the cotton growing regions.To elucidate the drought tolerance phenomenon in cotton,a backcross population was developed from G.tomentosum,a drought tolerant donor parent and G.hirsutum which is highly susceptible to drought stress.Results:A genetic map of 10888 SNP markers was developed from 200 BC_2F_2 populations.The map spanned 4191.3 centi-Morgan(c M),with an average distance of 0.1047 c M,covering 51%and 49%of At and Dt sub genomes,respectively.Thirty stable Quantitative trait loci(QTLs)were detected,in which more than a half were detected in the At subgenome.Eighty-nine candidate genes were mined within the QTL regions for three traits:cell membrane stability(CMS),saturated leaf weight(SLW)and chlorophyll content.The genes had varied physiochemical properties.A majority of the genes were interrupted by introns,and only 15 genes were intronless,accounting for 17%of the mined genes.The genes were found to be involved molecular function(MF),cellular component(CC)and biological process(BP),which are the main gene ontological(GO)functions.A number of mi RNAs were detected,such as mi R164,which is associated with NAC and MYB genes,with a profound role in enhancing drought tolerance in plants.Through RT-q PCR analysis,5 genes were found to be the key genes involved in enhancing drought tolerance in cotton.Wild cotton harbors a number of favorable alleles,which can be exploited to aid in improving the narrow genetic base of the elite cotton cultivars.The detection of 30 stable QTLs and 89 candidate genes found to be contributed by the donor parent,G.tomentosum,showed the significant genes harbored by the wild progenitors which can be exploited in developing more robust cotton genotypes with diverse tolerance levels to various environmental stresses.Conclusion:This was the first study involving genome wide association mapping for drought tolerance traits in semi wild cotton genotypes.It offers an opportunity for future exploration of these genes in developing highly tolerant cotton cultivars to boost cotton production.

文摘In this experiment,the drought-tolerant soybean variety Heinong 44 and the sensitive soybean variety Heinong 65 were used as experimental materials to study the effects of drought stress and re-watering on the osmotic adjustment ability and yield of soybean.The results showed that given a normal water supply,the proline and soluble protein contents of Heinong 44 and Heinong65 showed an increasing trend during the growth process,while the soluble sugar content increased and then decreased.The contents of proline,soluble protein and soluble sugar increased significantly with increasing drought stress,and the magnitude of increase of Heinong 44 was higher than that of Heinong 65.The osmotic adjustment ability of the drought-tolerant variety was significantly higher than that of the sensitive variety and showed consistent performance at different stages.After re-watering,the proline and soluble sugar contents of the flowering and pod-setting stages and the soluble protein content of the seedling stage were restored to the control levels.Drought stress resulted in a decrease in the effective pod number,pod number per pod,100-grain weight and yield.Severe drought had the greatest impact by increasing stress and decreasing soybean yields,which were followed by the flowering,bulging and seedling stages.

文摘Background:The cotton crop is universally considered as protein and edible oil source besides the major contributor of natural fiber and is grown in tropical and subtropical regions around the world Unpredicted environmental stresses are becoming significant threats to sustainable cotton production,ultimately leading to a substantial irreversible economic loss.Mitogen-activated protein kinase(MAPK)is generally considered essential for recognizing environmental stresses through phosphorylating downstream signal pathways and plays a vital role in numerous biological processes.Results:We have identified 74 MAPK genes across cotton,41 from G.hirsutum,19 from G.raimondii,whereas 14 have been identified from G.arboreum.The MAPK gene-proteins have been further studied to determine their physicochemical characteristics and other essential features.In this perspective,characterization,phylogenetic relationship,chromosomal mapping,gene motif,cis-regulatory element,and subcellular localization were carried out.Based on phylogenetic analysis,the MAPK family in cotton is usually categorized as A,B,C,D,and E clade.According to the results of the phylogenic relationship,cotton has more MAPKS genes in Clade A than Clade B.The cis-elements identified were classified into five groups(hormone responsiveness,light responsiveness,stress responsiveness,cellular development,and binding site).The prevalence of such elements across the promoter region of these genes signifies their role in the growth and development of plants.Seven GHMAPK genes(GH_A07G1527,GH_D02G1138,GH_D03G0121,GH_D03G1517,GH_D05G1003,GH_D11G0040,and GH_D12G2528)were selected,and specific tissue expression and profiling were performed across drought and salt stresses.Results expressed that six genes were upregulated under drought treatment except for GH_D11G0040 which is downregulated.Whereas all the seven genes have been upregulated at various hours of salt stress treatment.Conclusions:RNA sequence and qPCR results showed that genes as differentially expressed across both vegetative and reproductive plant parts.Similarly,the qPCR analysis showed that six genes had been upregulated substantially through drought treatment while all the seven genes were upregulated across salt treatments.The results of this study showed that cotton GHMPK3 genes play an important role in improving cotton resistance to drought and salt stresses.MAPKs are thought to play a significant regulatory function in plants’responses to abiotic stresses according to various studies.MAPKs’involvement in abiotic stress signaling and innovation is a key goal for crop species research,especially in crop breeding.

文摘Background: Nucleobase-ascorbate transporters(NAT), synonymously called nucleobase-cation symporter 2(NCS2) proteins, were earlier reported to be involved in plant growth, development and resistance to stress. Previous studies concluded that s a polymorphic SNP associated with NAT12 was significant di erent between salt-tolerant and salt-sensitive materials of upland cotton. In current study, a comprehensive analysis of NAT family genes was conducted for the first time in cotton.Results: In this study, we discovered 32, 32, 18, and 16 NAT genes in Gossypium hirsutum, G. barbadense, G. raimondii and G. arboreum, respectively, which were classified into four groups(groups I–IV) based on the multiple sequence analysis. These GhNAT genes were unevenly distributed on At and Dt sub-genome in G. hirsutum. Most GhNAT members in the same group had similar gene structure characteristics and motif composition. The collinearity analysis revealed segmental duplication as well as tandem duplication contributing to the expansion of the GhNATs. The analysis of cis-acting regulatory elements of GhNATs showed that the function of GhNAT genes in cotton might be related to plant hormone and stress response. Under di erent conditions, the expression levels further suggested the GhNAT family genes were associated with plant response to various abiotic stresses. GhNAT12 was detected in the plasma membrane. And it was validated that the GhNAT12 gene played an important role in regulating cotton resistance to salt and drought stress through the virus-induced gene silencing(VIGS) analysis.Conclusions: A comprehensive analysis of NAT gene family was performed in cotton, including phylogenetic analysis, chromosomal location, collinearity analysis, motifs, gene structure and so on. Our results will further broaden the insight into the evolution and potential functions of NAT genes in cotton. Current findings could make significant contribution towards screening more candidate genes related to biotic and abiotic resistance for the improvement in cotton.

文摘Background: Cotton is an important commercial crop for being a valuable source of natural fiber.Its production has undergone a sharp decline because of abiotic stresses,etc.Drought is one of the major abiotic stress causing significant yield losses in cotton.However,plants have evolved self-defense mechanisms to cope abiotic factors like drought,salt,cold,etc.The evolution of stress responsive transcription factors such as the trihelix,a nodule-inception-like protein(NLP),and the late embryogenesis abundant proteins have shown positive response in the resistance improvement to several abiotic stresses.Results: Genome wide identification and characterization of the effects of Light-Harvesting Chloro a/b binding(LHC)genes were carried out in cotton under drought stress conditions.A hundred and nine proteins encoded by the LHC genes were found in the cotton genome,with 55,27,and 27 genes found to be distributed in Gossypium hirsutum,G.arboreum,and G.raimondii,respectively.The proteins encoded by the genes were unevenly distributed on various chromosomes.The Ka/Ks(Non-synonymous substitution rate/Synonymous substitution rate)values were less than one,an indication of negative selection of the gene family.Differential expressions of genes showed that majority of the genes are being highly upregulated in the roots as compared with leaves and stem tissues.Most genes were found to be highly expressed in MR-85,a relative drought tolerant germplasm.Conclusion: The results provide proofs of the possible role of the LHC genes in improving drought stress tolerance,and can be explored by cotton breeders in releasing a more drought tolerant cotton varieties.

文摘As one of the important materials in landscaping for flower terrace and border, Petunia hybrida needs high environmental conditions and its growth is seriously influenced by the drought. Silicon is considered to be a necessary element for plant growth, and soluble silicon can improve plant resilience. To improve the drought resilience of Petunia hybrida, the silicon transporter protein OsLsi1 and OsLsi2 genes cloned from rice(Oryza sative) were transferred into Petunia hybrida by Agrobacterium-mediated method, and finally got 26 and 32 positive plants, respectively by PCR and RT-PCR detections. With a control of non-transgenic plants, the obtained transgenic plants were taken by drought treatment stress for 0, 4, 7, 10 and 14 days, then re-watered and measured physiological indexes as malondialdehyde(MDA) content, free proline(Pro) content, superoxide dismutase(SOD) activity and peroxidase(POD) activity to study the effect of Petunia's drought resistance. All the results proved that the silicon transporter protein OsLsi1 and OsLsi2 genes were normally transcripted and expressed in transgenic Petunia hybrida; OsLsi1 gene could improve the abilities of plants' drought resistance and recover after drought stress, while OsLsi2 gene could reduce the above abilities. The order of the drought resistance ability of the three strains from strong to weak was OsLsi1〉CK〉OsLsi2; and silicon indeed improved the ability of drought resistance as well. All these results provided a new way to improve the drought resistance of Petunia, and laid a foundation to improve the ability of garden plants' drought resistance and water saving.

文摘The situation of global warming imparts negative impacts on crop growth and development.Cotton is the most important fiber crop around the globe.However,frequent drought episodes pose serious threats to cotton production worldwide.Due to the complex genetic structure of drought tolerance,the development of a tolerant cultivar is cumbersome via conventional breeding.Multiple omics techniques have appeared as successful tool for cotton improvement in drought tolerance.Advanced omics-based biotechniques have paved the way for generation of omics data like transcriptomics,genomics,metabolomics and proteomics,which greatly expand the knowledge of cotton response to drought stress.Omics methodologies and have provided ways for the identification of quantitative trait loci(QTLs),gene regulatory networks,and other regulatory pathways against drought stress in cotton.These resources could speed up the discovery and incorporation of drought tolerant traits in the elite genotypes.The genome wide association study(GWAS),gene-editing system CRISPER/Cas9,gene silencing through RNAi are efficient tools to explore the molecular mechanism of drought tolerance and facilitate the identification of mechanisms and candidate genes for the improvement of drought tolerance in cotton.

文摘In order to determine the physiological mechanism of drought resistance of northern wheat in China,six drought resistant wheat and one sensitivity to drought wheat were planted in pots.They were subjected to drought treatment and normal water when the plants grew to the 3-leaf stage.Samples were collected at 10,20,30,and 40 days after the drought treatment,respectively.The electrical conductivity,photosynthetic parameters,chlorophyll fluorescence parameters,sugar content,proline content,protein content,and active oxygen scavenging enzyme activity of the plants were detected,and the agronomic traits of the wheat varieties were investigated at maturity.The results indicated that the phenotype and yield-related factors of Darkhan 144 changed little under the drought stress.The relative electrical conductivity of Kefeng 6 and Darkhan 166 was lower under the drought stress,and their cell membrane was less damaged.The Darkhan 144 and Darkhan 166 had higher drought resistance coefficients,and were the wheat varieties with stronger drought resistance.However,the physiological mechanisms of drought resistance of these three wheat were different:Darkhan 144 maintained a higher photosynthetic activity under the drought stress;Darkhan 166 maintained a higher protein content,photosynthetic activity and active oxygen scavenging enzyme activity.In addition,other drought-resistant varieties Kefeng 6,Kefeng 10 and Longmai 26 had a higher content of osmoregulatory substances under the drought stress.
