Marginal Selenium Deficiency Alternates Inflammatory Response through NF-κB Pathway in LPS-induced Mouse Mastitis Model

The trace element selenium(Se)occurs naturally throughout the earth.Se deficiency has been linked to impaired breast health and other diseases in human and animals.Compared to severe Se deficiency,marginal dietary Se deficiency accusers more frequently in low-Se regions.Therefore,to investigate the Se status and inflammatory response of the mammary gland under marginal dietary Se levels,an lipopolysaccharide(LPS)induced mouse mastitis model was established.Mice were fed with moderate Se diet(0.087 mg•kg^(-1) Se),adequate Se diet(0.15 mg•kg^(-1) Se)or excessive Se diet(1.5 mg•kg^(-1) Se)for 60 days.Se status and inflammatory factors were investigated.Results showed that the Se status of mammary gland correlated with dietary Se levels.Marginal Se deficiency exacerbated mammary tissue histopathology;increased the mRNA level of inflammatory genes tumor necrosis factor alpha(TNF-α),interleukin-1β(IL-1β)and cyclooxygenase-2(COX-2);and enhanced the phosphorylation of NF-κB p65 in mammary gland tissues.Supplementation of Se in diet higher than recommended levels reduced the inflammatory reaction of mammary glands in LPS-induced mastitis model and provided a protective effect. 展开更多

In recent years,growth hormone and insulin-like growth factors have become key regulators of bone metabolism and remodeling,crucial for maintaining healthy bone mass throughout life.Studies have shown that adult growth hormone deficiency leads to alterations in bone remodeling,significantly affecting bone microarchitecture and increasing fracture risk.Although recombinant human growth hormone replacement therapy can mitigate these adverse effects,improving bone density,and reduce fracture risk,its effectiveness in treating osteoporosis,especially in adults with established growth hormone deficiency,seems limited.Bisphosphonates inhibit bone resorption by targeting farnesyl pyrophosphate synthase in osteoclasts,and clinical trials have confirmed their efficacy in improving osteoporosis.Therefore,for adult growth hormone deficiency patients with osteoporosis,the use of bisphosphonates alongside growth hormone replacement therapy is recommended. 展开更多

Severe hyperhomocysteinemia due to MTHFR deficiency caused by a new mutation:A case report and literature review

Methylenetetrahydrofolate reductase(MTHFR)deficiency is a rare autosomal recessive genetic disorder caused by mutations in the MTHFR gene,leading to a variety of clinical manifestations.In October 2022,the Second Xiangya Hospital of Central South University admitted a 21-year-old male patient with neuropsychiatric disorders,presenting primarily with cognitive decline,limb tremors,abnormal mental and behavioral symptoms,seizures,and gait disturbances.These symptoms had gradually developed over 5 years,worsening significantly in the past year.The patient’s plasma homocysteine levels were 10 times higher than normal,and brain MRI revealed brain atrophy and significant abnormal signals in the bilateral paraventricular nuclei and heads of the bilateral caudate nuclei.Further genetic testing identified a paternal mutation c.1604G>A(p.R535Q)and a maternal mutation c.227T>G(p.L76R)of the MTHFR gene.After betaine supplementation,the plasma homocysteine levels decreased within a week,and the symptoms improved.The patient was ultimately diagnosed with severe hyperhomocysteinemia due to MTHFR deficiency.The c.227T>G(p.L76R)mutation represents a novel missense mutation in the MTHFR gene associated with MTHFR deficiency,but further research is needed to confirm its potential pathogenicity.Early treatment with betaine can fully reverse the symptoms. 展开更多

This objective of the study was to develop a model for the determination of N deficiency in cherry trees using a combination of visible near infrared methods and spectro-radiometric measurement.In our experimental design,cherry seedlings were grown under various N deficiency conditions in nutrient-controlled containers.The reflectance values of plant leaves were measured using a spectro-radiometer.Plant leaves samples were simultaneously collected.Their nutrient contents were determined in the laboratory.Afterwards,we performed a statistical comparison of the reflectance values.Sample analysis results established the significant wavelengths.Moreover,we received accurate regression models for predicting N deficiency in cherry leaves that were grown in nutrient solutions.Next,we verified the model validity by measuring the reflectance of the leaves collected from cherry orchards at various locations using a spectroradiometer.Nutrient deficiencies were calculated using the developed model,and then,the predicted and measured data were compared to evaluate model validity.From these results,we determined the wavelengths that yielded the most accurate results for N prediction,selected from the blue and green regions of the spectrum.We established that for N prediction in cherry trees,the simplest model can be created using 560and 570nm wavelengths.However,the evaluated model can be applicable only under certain conditions.We concluded that in order to develop aprediction method with sufficient application capacity,as well as the ability to assess nutritional and physiological characteristics,the ecology condition of the plant should be properly considered based on the model. 展开更多

They concluded that to maintain improved growth under phosphorus defioiency oonditions plants develop two major mechanisms: (i) Phosphorus acquisition (root morphology, root exudation and phosphorus uptake mechanisms), (ii) Phosphorus utilization (internal mechanisms associated with better use of absorbed phosphorus at cellular level). OBJECTIVE To explore the curative effect and mechanism of Yiqi Huoxue decoction in the treatment of coronary heart disease with Qi deficiency and blood stasis syndrome.METHODS The patients with coronary heart disease of Qi deficiency and blood stasis syndrome were treated with Yiqi Huoxue decoction for 3 months,and the changes of cardiac function were observed.61 serum samples(including 29 cases of disease group and 32 cases of Yiqi Huoxue expression group)were analyzed by non labeled proteomics.The disease group was used as the control group,and the protein with expression level difference of more than 1.2 folds(P<0.05)was screened.The molecular function,biological pathway and protein interaction of the different proteins were analyzed by bioinformatics,so as to identify the molecular and biological pathway of Yiqi Huoxue decoction in the treatment of coronary heart disease with Qi deficiency and blood stasis syndrome.RESULTS Clinical treatment found that Yiqi Huoxue decoction can improve TCM syndrome score and left ventricular ejection fraction,regulate blood glucose and blood lipid levels,prolong thrombin time,and improve heart function.The results of proteomic quantitative analysis showed that there were 69 proteins with different expression levels in the disease group.Bioinformatics analysis results showed that Yiqi Huoxue decoction may regulate ApoA1,alpha-2 and other proteins to act on HDL assembly,platelet degradation,PI3K Akt signaling pathway,and then play a therapeutic role in coronary heart disease with Qi deficiency and blood stasis syndrome.CONCLUSION Yiqi Huoxue decoction can effectively improved the heart function decline caused by Qi deficiency and blood stasis syndrome of coronary heart disease.It mainly act on energy metabolism and platelet activation pathway by activating HDL assembly and platelet degradation signal pathway proteins.This study can provide reference for the follow-up treatment mechanism of Qi deficiency and blood stasis syndrome of coronary heart disease. 展开更多

OBJECTIVE To observe the regulating effect of Weiliuan on digestive enzyme and relevant gastrointestinal hormone in spleen-deficiency rats.METHODS The rats were given the decoction of Radix et Rhizoma combine with dietary disorder to induce spleen-deficiency syndrome.Spleen-deficiency rats were divided into 8groups:normal control group,spleen deficiency model group,Mengtuoshi San group(0.15g·kg-1),Weiliuan 31.5and 15.75g·kg-1 groups,Weiliuan splited prescriptionⅠ 31.5 and 15.75g·kg-1groups,Weiliuan splited prescriptionⅡ 31.5and 15.75g·kg-1 groups.At the end of experiment,the level of amylase(AMS),lactic dehydrogenase(LDH),succinate dehydrogenase(SDH),motilin(MTL),somatostatin(SS),gastrin(GAS)and organ coefficient were detected.RESULTS Weiliuan group,Weiliuan splited prescriptionⅠ group and Weiliuan splited prescriptionⅡ group can significantly reduce the spleen index,the level of SDH and LDH,it can also improve the level of the AMS(P<0.05,P<0.01)In the regulation of hormone levels,Weiliuan group,Weiliuan splited prescriptionⅠ group and Weiliuan splited prescriptionⅡ group can significantly improve the levels MTL and GAS(P<0.05),but had no obvious effect on SS.CONCLUSION Weiliuan group,Weiliuan splited prescriptionⅠ group and Weiliuan splited prescriptionⅡ group can improve the symptoms of spleen deficiency syndrome significantly.Its mechanism may be to regulate the levels of AMS,SDH,LDH,MTL and GAS. 展开更多

Signs began to appear at the age of one week, and became seri... 20 one-day-old Tianfu ducklings were fed on a natural diet deficient in phosphorus (Ca 0.80%, P 0.366%) for three weeks and examined for signs and lesions. Signs began to appear at the age of one week, and became serious at two weeks. 13 ducklings died during the experiment. Morbidity was 100% and mortality was 65%. The affected ducklings mainly showed leg weakness, severe lameness, depression, lack of appetite and stunted growth. The serum alkaline phosphatasc activities increased markedly. The scrum phosphorus concentration, tibial ash, ash calcium and phosphorus content decreased obviously. At necropsy, maxillae and ribs were soft, and the latter was crooked. Long bones were soft and broke easily. The hypcrtrophic zone of the growth- plate in the epiphysis of long bones was lengthened and ostcoid tissue increased in the mctaphyseal spongiosa histopathologically. The above mentioned symptoms and lesions could be prevented by adding phosphorus to the natural deficient diet (up to 0.65%). The relationship between lesions and signs, pathomorphological characterisation and pathogcnsis were also discussed in this paper. 展开更多

The results showed that the prevalence of left ventricular hypertrophy in cardiac deficiency of the Qi (CDQ) was obviously higher than that of cardiac deficiency of the Yin (CDY). The higher prevalence of tachycardia occurred in the patients with CDY.The prevalence of the Q - T interval prolongation and abnormalities of ST - T segment in the patients with CDY was also higher than that of CDQ. The difference between two groups suggests that the diagnosis of two syndromes by ECG may be of important value. 展开更多

目的探讨乌灵胶囊对心血亏虚、肾精不足型失眠症血清SP、NPY水平及睡眠质量的影响。方法选取医院收治的心血亏虚、肾精不足型失眠症患者80例,将纳入患者随机分为两组,每组各40例。对照组使用常规西药治疗,观察组在对照组基础上联合乌灵... 目的探讨乌灵胶囊对心血亏虚、肾精不足型失眠症血清SP、NPY水平及睡眠质量的影响。方法选取医院收治的心血亏虚、肾精不足型失眠症患者80例,将纳入患者随机分为两组,每组各40例。对照组使用常规西药治疗,观察组在对照组基础上联合乌灵胶囊治疗,疗程为4周,对比两组患者临床治疗效果、血清P物质(substance P,SP)、神经肽类物质神经肽Y(neuropeptide Y,NPY)水平及睡眠质量情况。结果在治疗过程中,对照组表现出了明显低于观察组的临床治疗效果,对照组的有效率为70.00%,观察组总有效率92.50%,两组比较差异有统计学意义(P<0.05);治疗后,两组的SP水平显著降低(P<0.05),而观察组的SP水平则显著低于对照组(P<0.05)。与此同时,两组的NPY水平显著增加(P<0.05),观察组的NPY水平也显著高于对照组(P<0.05);此外,治疗前后,两组的阿森斯失眠量表(Athens insomnia scale,AIS)、匹兹堡睡眠质量指数量表(Pittsburgh sleep quality index,PSQI)以及汉密尔顿焦虑量表(Hamilton anxiety rating scale,HAMA)评分都呈现出显著降低的趋势(P<0.05),而观察组在这些评分指标上的表现也明显低于对照组(P<0.05);两组患者入睡艰难、易醒、多梦、疲乏、腰酸症状明显改善(P<0.05),观察组显著低于对照组(P<0.05)。结论乌灵胶囊对心血亏虚、肾精不足型失眠症患者具有良好的治疗效果,可提高患者睡眠质量,改善患者临床症状,安全性高,值得临床应用。 展开更多

妊娠晚期缺铁性贫血的危险因素和列线图预测模型 被引量:2

【目的】对孕妇妊娠期缺铁性贫血(IDA)的相关因素进行分析,根据其独立的危险因素,构建预测妊娠晚期缺铁性贫血的列线图预测模型,以期获得IDA的有效干预方案。【方法】将2022年7月—2023年12月在扬州市妇幼保健院定期产检的孕妇为调查对... 【目的】对孕妇妊娠期缺铁性贫血(IDA)的相关因素进行分析,根据其独立的危险因素,构建预测妊娠晚期缺铁性贫血的列线图预测模型,以期获得IDA的有效干预方案。【方法】将2022年7月—2023年12月在扬州市妇幼保健院定期产检的孕妇为调查对象,采取统一调查问卷结合监测血常规的方法跟踪孕妇直至分娩,入组500例,最终完成482例。根据孕晚期有无IDA分为IDA组与非IDA组。对可能影响缺铁性贫血的危险因素进行单因素分析及多因素logistic回归分析,并采用R软件建立预测发生的列线图模型。【结果】482例妊娠晚期女性发生IDA有96例,发生率为19.92%;单因素分析显示,经济独立情况、孕前体质指数、孕次及孕期有无正规补铁与妊娠晚期IDA的发生有关,差异均有统计学意义(P<0.05)。多因素Logistic回归分析显示,经济独立(P=0.031,OR=0.583)、孕期正规补铁(P<0.001,OR=5.337)是IDA的保护因素;孕前低体质指数(P=0.021,OR=2.375),孕次≥3次(P=0.015,OR=2.253)是IDA的危险因素。ROC曲线显示,列线图模型预测IDA发生的曲线下面积为0.84,最佳截断值为-1.481,预测灵敏度为81.2%,特异度分别为75.1%。【结论】妊娠晚期IDA发病率较高;孕前低体质指数、孕次大于3次是妊娠晚期IDA的危险因素;经济独立、正规补铁是妊娠晚期IDA的保护因素。可利用上述因素构建预测模型、采取预防性措施研究降低妊娠晚期IDA发病率的可行性方案。 展开更多

目的:分析SMARCA4缺失型非小细胞肺癌中分泌性磷蛋白1(SPP1)及细胞程序性死亡-配体1(PD-L1)的表达,为罕见病理类型肺癌后续治疗研究提供科学依据。方法:回顾性分析12例SMARCA4缺失型非小细胞肺癌患者的临床与病理特征,根据其形态学分为... 目的:分析SMARCA4缺失型非小细胞肺癌中分泌性磷蛋白1(SPP1)及细胞程序性死亡-配体1(PD-L1)的表达,为罕见病理类型肺癌后续治疗研究提供科学依据。方法:回顾性分析12例SMARCA4缺失型非小细胞肺癌患者的临床与病理特征,根据其形态学分为腺癌与低分化癌两组,分析SPP1与PD-L1在两组的表达与关系。结果:所有患者均检出SPP1表达且其在低分化癌组表达水平较腺癌组显著升高(P=0.015);PD-L1表达者占6/7(5例未测),较之腺癌组,PD-L1亦在低分化癌组高表达(P=0.048),两组的PD-L1表达差异与SPP1表达差异一致。结论:SMARCA4缺失型非小细胞肺癌具有较高的SPP1及PD-L1阳性表达,病理形态为低分化癌者更甚,SPP1与PD-L1在SMARCA4缺失型非小细胞肺癌中的表达情况可能具有正相关性,其关联机制及免疫治疗中作用有待后续研究进一步探讨。 展开更多

目的观察益气聪明汤联合针刺治疗气血两虚型眩晕的效果。方法选择医院2020年1月—2023年1月收治气血两虚型眩晕患者100例作为研究对象,使用随机数字表通过简单随机的方法将其分为观察组与对照组,每组50例。两组均接受针刺治疗4周,观察... 目的观察益气聪明汤联合针刺治疗气血两虚型眩晕的效果。方法选择医院2020年1月—2023年1月收治气血两虚型眩晕患者100例作为研究对象,使用随机数字表通过简单随机的方法将其分为观察组与对照组,每组50例。两组均接受针刺治疗4周,观察组在此基础上给予益气聪明汤治疗4周,比较两组治疗前后椎-基底动脉血流收缩期最大流速、血清总胆固醇(total cholesterol,TC)、血清纤维蛋白原(fibrinogen,Fib)水平,并比较两组治疗前后眩晕评分,在疗程结束时,按照《中医新药临床研究指导原则》相关标准比较两组治疗疗效,比较两组治疗期间不良反应发生情况。结果观察组与对照组临床痊愈率分别为38.00%(19/50)和18.00%(9/50),差异具有统计学意义(P<0.05);治疗后两组血清TC、Fib水平均较本组治疗前降低,其中观察组低于对照组,差异具有统计学意义(P<0.05);治疗后两组左椎动脉、右椎动脉以及基底动脉血流收缩期最大流速均较本组治疗前明显升高,其中观察组明显高于对照组,差异具有统计学意义(P<0.05);治疗后两组眩晕评分均高于本组治疗前,且观察组明显高于对照组,差异具有统计学意义(P<0.05)。两组治疗期间均无不良反应发生。结论益气聪明汤联合针刺治疗气血两虚型眩晕较单纯应用针刺治疗效果更佳,能显著提升患者椎-基底动脉血流收缩期最大流速,且具有较高安全性,适合推广。 展开更多

助阳通便汤治疗功能性便秘临床观察及对便秘模型小鼠结肠组织VIP的影响 被引量:1

目的观察助阳通便汤对功能性便秘(脾肾阳虚证)患者的临床疗效和患者血清P物质(substance P,SP)、血管活性肠肽(vasoactive intestinal peptide,VIP)含量的影响,以及其对便秘模型小鼠结肠组织病理及VIP表达的影响。方法(1)将66例符合功... 目的观察助阳通便汤对功能性便秘(脾肾阳虚证)患者的临床疗效和患者血清P物质(substance P,SP)、血管活性肠肽(vasoactive intestinal peptide,VIP)含量的影响,以及其对便秘模型小鼠结肠组织病理及VIP表达的影响。方法(1)将66例符合功能性便秘诊断,且中医证型为脾肾阳虚证的患者,用随机数字表法进行分组,其中治疗组(33例)、对照组(33例),治疗组助阳通便汤130 mL日2次口服,对照组乳果糖15 mL日2次口服,均治疗2周。比较两组患者用药前后便秘症状评分、粪便性状评分、中医证候评分及血清SP、VIP含量。(2)将18只雄性BALB/c小鼠依据随机数字表法分为KB组(空白对照组)、MX组(模型组)、ZY组(助阳通便汤组)3组,每组6只,以复方地芬诺酯片混悬液80 mg/kg灌胃的方法进行造模。ZY组给予助阳通便汤0.2 mL/10 g连续2周灌胃,结束后取材,观察各组小鼠结肠组织病理及VIP免疫组化表达。结果(1)治疗后两组患者便秘症状及粪便性状的评分均比治疗前显著提高(P<0.05);治疗组中医证候评分差异有统计学意义(P<0.05);治疗组患者血清SP含量高于对照组(P<0.05),血清VIP含量低于对照组(P<0.05)。治疗组有效率90.0%,对照组有效率73.3%(P<0.05)。(2)与模型组(MX组)小鼠比较,助阳通便汤组(ZY组)小鼠结肠组织中VIP阳性表达细胞分布相对较密集,阳性细胞数量较多(P<0.05)。结论助阳通便汤对功能性便秘(脾肾阳虚证)患者的临床症状及中医证候有较好地改善,并且对患者血清SP、VIP含量有干预作用;助阳通便汤调控便秘模型小鼠结肠组织VIP含量是其治疗功能性便秘的作用机制之一。 展开更多

目的探讨开窍醒神针法联合补阳还五汤在脑出血恢复期患者(气虚血瘀证)治疗中的应用价值,观察其对炎性指标、神经因子的影响。方法166例脑出血恢复期患者(气虚血瘀证)患者均于2021年9月至2023年10月在南昌市洪都中医院就诊,纳入研究前均... 目的探讨开窍醒神针法联合补阳还五汤在脑出血恢复期患者(气虚血瘀证)治疗中的应用价值,观察其对炎性指标、神经因子的影响。方法166例脑出血恢复期患者(气虚血瘀证)患者均于2021年9月至2023年10月在南昌市洪都中医院就诊,纳入研究前均随机分组。对照组83例采用常规西医治疗,观察组83例联合开窍醒神针法及补阳还五汤治疗,治疗8周后比较疗效。结果治疗后两组血清基质金属蛋白酶-9(MMP-9)、内皮素-1(endothelin,ET-1)、白细胞介素-2(interleukin-2,IL-2)、同型半胱氨酸(homocysteine,Hcy)、Bcl-2相关X蛋白(the Bcl-2-associated X protein,bax)降低,脑源性神经营养因子(brain-derived neurotrophic factor,BDNF)升高,观察组改善更显著,差异有统计学意义(P<0.05);治疗后观察组脑血管平均流量、脑血管平均流速升高,血管外周阻力降低,观察组改善更显著(P<0.05);治疗后观察组中医总症状积分降低,Fuel-Meyer功能评分(FMA评分)升高,观察组改善更显著(P<0.05);观察组总有效率93.98%(78/83),高于对照组的80.72%(67/83),差异有统计学意义(P<0.05)。结论对于脑出血恢复期患者(气虚血瘀证)而言应用开窍醒神针法及加味补阳还五汤有助于减轻炎症反应,促进血脑屏障恢复,改善脑血流量,促进神经功能恢复,提高临床疗效。 展开更多
