This type of callus induction occurred at a lower fre-quency with either a-naphthaleneacetic acid (NAA) or IBA (both 8 mg/L). White, translucent, glossy mucilaginous callus was embryogenic and mainly developed from the cotyledons of the mature zygotic embryo. Somatic embryos were formed on dif-ferentiation medium. Desiccation tolerance can be induced by culturing somatic embryos of loblolly pine (Pinus taeda L.) on medium supplemented with 50 mm abscisic acid (ABA) and/or 8.5% polyethylene glycol (PEG6000). Scanning electron micros-copy of desiccated somatic embryos showed that the size and external morphology of the desiccation tolerant somatic embryos recovered to the pre-desiccation state within 24-36 h, whereas the sensitive somatic embryos did not recover and remained shriveled, after the desiccated somatic embryos had been rehydrated. Peroxidase activity of desiccated somatic embryos in-creased sharply after 3 days of desiccation treatment, and desiccation tolerant somatic embryos had higher peroxidase activity compared to sensitive somatic embryos. Higher peroxidase activity of desiccation tolerant somatic embryos was possibly ad-vantage of catalyzing the reduction of H2O2 which was produced by drought stress, and protecting somatic embryos from oxida-tive damage.展开更多

Cronobacter spp.has strong resistance to desiccation and high permeability in Enterobacteriaceae,and powdered infant formula(PIF)is one of the main contamination routes.In recent years,the contamination of Cronobacter...Cronobacter spp.has strong resistance to desiccation and high permeability in Enterobacteriaceae,and powdered infant formula(PIF)is one of the main contamination routes.In recent years,the contamination of Cronobacter spp.in PIF incidents occurs from time to time,causing infant serious diseases or death.In this investigation,matrix-assisted laser desorption/ionization time of flight mass spectrometry was used to identify the phenotypes of 35 Cronobacter strains isolated from PIF and its processing environment.Subsequently,the isolates were evaluated for drying and osmotic pressure tolerance.The results showed that the deactivation rate of the strains ranged from 9.01%to 77.57%,and the highest osmotic pressure condition the strains could tolerate was 6 g/100 mL Na Cl.In addition,there was a positive correlation between biofilm formation ability and desiccation resistance.Combined with transcriptomics,Cronobacter spp.could activate biofilm synthesis,produce more trehalose,accumulate betaine and electrolytes to stabilize intracellular structure under the two treatment conditions.A total of 31 and 43 genes were found related to desiccation and permeability resistance,respectively.And some genes(cysM,thuF,ycjO,etc.)were found to be associated with two tolerances for the first time.展开更多

Seeds with(intact fruit nut)and without the pericarp were subjected to an imbibition test, and water uptake was measured. After nuts had been stored for 5 months at 3–5℃, seeds and desiccated seeds were cold-stratified at 3–5 ℃(for 0, 1, 2 or 3 months),then tested for germination. Nuts were warm-stratified at 20–25℃(for 0, 1 or 2 months), then cold-stratified(for 0,1, 2 or 3 months) and tested for seed germination. The nuts were sown outdoors on three dates in the same year of their collection: 15 September(immediately after collection), 2 November, and 17 December, and emerged seedlings were counted the following spring. The pericarp was not a barrier to water uptake by the seed;however, the seeds imbibed water faster and had more mass when the pericarp was removed. Seeds tolerated desiccation;germinationpercentage was not reduced when seed moisture content was reduced to 8.71%. Germination of seeds without the pericarp(regardless of the moisture content) was the highest when the seeds were cold-stratified for 2 months.When seeds with the pericarp were tested for germination,2 months of warm stratification before a 2-month cold stratification maximized seed germination. For outdoor sowing of nuts, sowing date affected percentage of seedling emergence the following spring;early autumn sowing(immediately after collection) resulted in the best emergence.展开更多
Water relation parameters of bare-root seedlings of Chinese fir (Cunninghamia lanceolata Hook.) and Masson pine (Pinus massoniana Lamb.) were measured and changes of root growth potential as well as field survival rat...Water relation parameters of bare-root seedlings of Chinese fir (Cunninghamia lanceolata Hook.) and Masson pine (Pinus massoniana Lamb.) were measured and changes of root growth potential as well as field survival rate of both species were studied after the bare-root seedlings were exposed in a sunny field condition. The results showed that Masson pine had a lower osmotic potential (-2.07Mpa) at turgor loss point and at full turgor (-1.29Mpa), compared with Chinese fir (-1.80Mpa and -1.08Mpa respectively). The parameter Vp/V0 (63.27%) of Masson pine was higher than that of Chinese fir (58.03%). This means that Masson pine has a stronger ability to tolerate desiccation, compared to Chinese fir according to analysis of above water relation parameters. Root growth potential and field survival rate decreased with prolonging duration of exposure. The field survival rate of both species was reduced to less than 40% after the seedling being exposed only two hours. Water poten-tials of 1.60 Mpa and -1.70 Mpa were suggested to be critical values for Chinese fir and Masson pine respectively in successful reforestation.展开更多

Mature seeds with moisture content (MC) of 31% exhibited 73% germination. During the period of desiccation (open lab condition) seeds with MC 23% showed 40% germination. After further drying to MC 21% germination was reduced to 16%. Complete loss in viability resulted when seed moisture was reduced to 18%. The seeds stored at -10℃, 0℃, 10℃ and 28±2℃ (open lab condition) lost their viability within 10 days. Seeds stored in sealed polythene hags and moist sand retained viability for more days than did seeds stored under all other storage conditions. Sensitivity of seeds to lower temperature and desiccation suggest that the storage behavior of K. attenuata seeds is recalcitrant. Seeds stored in moist conditions can, at best, be stored for a period of two months.展开更多
Araucaria angustifolia(Bertol.)Kuntze is a representative species of the Mixed Ombrophilous Forest in the Atlantic Forest Biome of Brazil.The development of a germplasm conservation protocol for long-term seed bank st...Araucaria angustifolia(Bertol.)Kuntze is a representative species of the Mixed Ombrophilous Forest in the Atlantic Forest Biome of Brazil.The development of a germplasm conservation protocol for long-term seed bank storage is compromised for this species,as it is sensitive to desiccation.Furthermore,in situ establishment of a soil seed bank in its natural habitat may be limited.This study evaluates the storability of two provenances of A.angustifolia seeds and their behavior in an artificial soil seed bank in two forest environments(understory and edge).Results show that both seed provenances may be stored at 5C for approximately 12 months,retaining high viability.The subsequent decrease in germination was associated with a reduction and an increase in seed water content,as well as with increased electrical conductivity.In the understory environment,seed viability was above 85%for the first 60 days,and at the end of the experiment(270 days),seedlings emerged.However,at the forest edge,there was a total loss of seed viability after 120 days associated with a reduction in water content and high predation.It is concluded,therefore,that short-term storage of A.angustifolia seeds is possible in a cold room,which is fundamental to supply seed demand outside the production period.Forest cover conservation is important for regeneration and conservation of the species.展开更多

However, the present dehumidification system has many weak points. The liquid desiccant air-conditioning system has recently gained growing interest from the stand point of reducing energy consumption during dehumidification. In order to find the appropriate ionic liquids(ILs) as a desiccant for the liquid desiccant air-conditioner system, we conducted a systematic evaluation of the humidification capability of 16 types of ILs. Among the tested ILs, tributyl(methyl)phosphonium dimethyl phosphate([P4441][DMPO_4]) exhibited the best dehumidification capacity and had a less corrosive effect on four types of metals as possible piping materials. It should be noted that this [P_(4441)][DMPO_4] has a very stable nature and produced no odor while conducting the experiment and storing for over 1 year at room temperature under ambient conditions. Furthermore, it was revealed that a 77%(w/w) aqueous solution of [P4441][DMPO_4] worked as an efficient desiccant liquid for the liquid desiccant air-conditioner system.展开更多

A chamber of 28.4 m3 was installed to simulate a mobile home. The formaldehyde off-gassing properties of particleboard were evaluated using the chamber. The relationship between the concentration obtained by the chamber and the values by the desiccator test was discussed in this study under different conditions of conditioning day, air exchange, and loading ratio. These two methods were compared and discussed regarding the formaldehyde emission level. Three loading ratios, 0.429 m2/m3, 0.264 m2/m3, and 0.132 m2/m3, were chosen to represent different applications of particleboard products. There was strong correlation between emissions and air exchange rates at equal product-loading ratios in the large chamber, the related coefficient R 2 exceeded 0.90. There was also an indication of a generic correlation between the large chamber and the two-hour desiccator test with a single product designated loading ratio, air exchange rate, and climatic conditions.展开更多
