Lead(Pb)and zinc(Zn)are widely recognized as common environmental contaminants,contributing to soil degradation and posing risks to environmental health.Combining functional carbon-based materials with microorganisms ...Lead(Pb)and zinc(Zn)are widely recognized as common environmental contaminants,contributing to soil degradation and posing risks to environmental health.Combining functional carbon-based materials with microorganisms has been considered as an effective and environmentally friendly strategy for remediating Pb/Zn-contaminated soil.However,there is still a lack of understanding the connection between heavy metal immobilization and plant responses,which hampers practical applications.Here,a 90-day pot experiment was conducted to investigate the integrated effects of biochar(WS700)and microorganisms including inorganic phosphate-solubilizing bacteria(IPSB)and sulfate reducing bacteria(SRB)on Pb and Zn synchronous immobilization and the physiological responses of Brassica rapa var.chinensis(Brassica).Compared with CK,bacteria-loaded biochar treatment declined the exchangeable Pb and Zn fraction by 94.69%−98.37%and 94.55%−99.52%,while increasing the residual state Pb and Zn by 75.50%−208.58%and 96.71%−110.85%,respectively.Three amendments enhanced Brassica growth by improving total chlorophyll content and superoxide dismutase(SOD)and peroxidase(POD)activities.The bacteria-loaded biochar treatment effectively regulated stomatal conductance and reduced intercellular CO_(2) concentration.Moreover,compared with CK,three amendments reduced MDA content by 28.84%,28.30%and 41.60%,respectively,under the high concentration of Pb and Zn.The findings demonstrated the significant role of bacterial-biochar consortia in immobilizing Pb and Zn and mitigating Pb and Zn-induced stress in plants by regulating photosynthetic characteristics and antioxidant enzyme activities.展开更多
Acetobacteraceae has garnered significant attention because of its unique properties and the broad applications of the bacterial cellulose it produces.However,unlike model strains,Acetobacteraceae have few synthetic b...Acetobacteraceae has garnered significant attention because of its unique properties and the broad applications of the bacterial cellulose it produces.However,unlike model strains,Acetobacteraceae have few synthetic biology applications because they are difficult to manipulate genetically and have insufficient genetic regulatory elements,among other factors.To address this limitation,this study characterized the fundamental properties and synthetic biology elements of three commonly used bacterial cellulose-producing strains.First,the basic characteristics of the three strains,including their cellulose film production ability,division time,antibiotic susceptibility,and plasmid features,were analyzed.Two inducible promoters(pTrc and pLux101)were subsequently characterized within the three strains.The inducibility of the pTrc promoter was relatively low across the three strains(induction ratio:1.98–6.39),whereas the pLux101 promoter demonstrated a significantly greater level of inducibility within the three strains(induction ratio:87.28–216.71).Finally,through gene knockout experiments,this study identified four genes essential for bacterial cellulose film production in the genome of the Gluconacetobacter hansenii ATCC 5358 strain.This study not only enriches the library of synthetic biology elements in nonmodel strains,but also lays the foundation for the synthetic biology applications of Acetobacteraceae.展开更多
Copper-nickel alloys can suffer severe localized corrosion in marine environments containing sulfate-reducing bacteria(SRB),but the effect of SRB on the under-deposit corrosion of copper-nickel alloys is unknown.In th...Copper-nickel alloys can suffer severe localized corrosion in marine environments containing sulfate-reducing bacteria(SRB),but the effect of SRB on the under-deposit corrosion of copper-nickel alloys is unknown.In this work,the corrosion behavior of B10 copper-nickel alloy beneath a deposit caused by SRB with carbon source starvation in artificial seawater was studied based on electrochemical measurements and surface analysis.Results demonstrate that SRB with an organic carbon starvation can survive in artificial water but most SRB cells have died.The survived SRB cells can attach to the bare and deposit-covered B10 copper-nickel alloy,leading to the corrosion acceleration.Due to the limitation of organic carbon source,the pitting corrosion of B10 copper-nickel alloy caused by SRB is not serious.However,serious pitting corrosion of the deposit-covered B10 copper-nickel alloy can be found both in abiotic and biotic conditions,and the pitting corrosion and uniform corrosion are further accelerated by SRB.There is a galvanic effect between the bare and deposit-covered specimens in the presence of SRB in the early stage but the galvanic effect after 5 d of testing can be neglected due to the low OCP difference values.展开更多
Profiling the protein composition of bacteria is essential for understanding their biology,physiology and interaction with environment.Mass spectrometry has become a pivotal tool for protein analysis,facilitating the ...Profiling the protein composition of bacteria is essential for understanding their biology,physiology and interaction with environment.Mass spectrometry has become a pivotal tool for protein analysis,facilitating the examination of expression levels,molecular masses and structural modifications.In this study,we compared the performance of three widely-used mass spectrometry methods,i.e.,matrix-assisted laser desorption/ionization(MALDI)protein fingerprinting,top-down proteomics and bottom-up proteomics,in the profiling of bacterial protein composition.It was revealed that bottom-up proteomics provided the highest protein coverage and exhibited the greatest protein profile overlap between bacterial species.In contrast,MALDI protein fingerprinting demonstrated superior detection reproducibility and effectiveness in distinguishing between bacterial species.Although top-down proteomics identified fewer proteins than bottom-up approach,it complemented MALDI fingerprinting in the discovery of bacterial protein markers,both favoring abundant,stable,and hydrophilic bacterial ribosomal proteins.This study represents the most systematic and comprehensive comparison of mass spectrometry-based protein profiling methodologies to date.It provides valuable guidelines for the selection of appropriate profiling strategies for specific analytical purposes.This will facilitate studies across various fields,including infection diagnosis,antimicrobial resistance detection and pharmaceutical target discovery.展开更多

Parts of the international research on the safety of GMO focus on its effect on soil ecosystem, especially on microbial communities changing and process in soil that are essential to key terrestrial ecosystem functions. This paper studied the dynamic change of soil microbe after cultivating Roundup Ready soybean (RRS) and the effect on biochemical processing of nitrogen cycle. According to the variance analysis, the ammonifying bacteria and nitrifying bacteria quantities of RRS in the rhizosphere soil were much lower than that of other genotype soybeans. The effects of different genotype soybeans on ammoniation intensity and nitrification intensity were remarkable. The nitrification intensity and the nitrifying bacteria had the great positive correlation and sustainable development.展开更多
Bioleaching experiments combined with X-ray photoelectron spectroscopy(XPS),X-ray diffraction(XRD)and scanning electron microscopy(SEM)were conducted to investigate three kinds of bornites from different regions leach...Bioleaching experiments combined with X-ray photoelectron spectroscopy(XPS),X-ray diffraction(XRD)and scanning electron microscopy(SEM)were conducted to investigate three kinds of bornites from different regions leached by moderately thermophilic mixed bacteria of Leptospirillum ferriphilum YSK,Acidithiobacillus caldus D1 and Sulfobacillus thermosulfidooxidans ST.The results of bioleaching experiments showed that the leaching efficiency and the redox potential were significantly increased.The copper extraction efficiencies of three kinds of bornite maintained rapid growth until around the 12th day and no longer increased after the 18th,reaching 83.7%,96.5%and 86.6%,respectively.The XRD results of the leaching residue indicated that three kinds of bornites all produced jarosite in the late stage of leaching,and the leaching residues from of Daye Museum and Yunnan Geological Museum contained a mass of elemental sulfur.XPS analysis and scanning electron microscopy experiments showed that the surface of mineral particles was jarosite and the copper in the leaching residue was almost dissolved.展开更多

The recombinant pNZ8148-BSH was transferred into lactic acid bacteria NZ9000 with electrotransformation method. And the recombinant which could express BSH protein was obtained. The potential biodegradation of ETC was evaluated b...Ethylthionocarbamates (ETC), which is the most widely used as collectors in the flotation of sulfide, is known to cause serious pollution to soil and groundwater. The potential biodegradation of ETC was evaluated by applying a mixed culture of iron-reducing bacteria (IRB) enriched from tailings dam sediments. The results showed that ETC can be degraded by IRB coupled to Fe(III) reduction, both of which can be increased in the presence of anthraquinone-2,6-disulfonate (AQDS). Moreover, Fe(III)-EDTA was found to be a more favorable terminal electron acceptor compared to α-Fe2O3, e.g., within 30 d, 72% of ETC was degraded when α-Fe2O3+AQDS was applied, while it is 82.67% when Fe(III)-EDTA+AQDS is added. The dynamic models indicated that the kETC degradation was decreased in the order of Fe(III)-EDTA+AQDS〉α-Fe2O3+AQDS〉Fe(III)-EDTA〉α-Fe2O3, with the corresponding maximum biodegradation rates being 2.6, 2.45, 2.4 and 2.0 mg/(L·d), respectively, and positive parallel correlations could be observed between kFe(III) and kETC. These findings demonstrate that IRB has a good application prospect in flotation wastewater.展开更多

This paper deals with the ex...Denitrification has been long thought to be a unique characteristic of prokaryotes, but in recent years, several filamentous fungi and yeasts were found to exhibit denitrifying activities. This paper deals with the examination of denitrification capabilities by mix-cultures of the fungus (Fusarium oxysporum) and the bacterium (Pseudomonas stutzeri TR2) in combination with a specific medium and using a synthetic wastewater of defined quality. The results revealed that P. stutzeri TR2 has strong and fast denitrifying capabilities under anaerobic conditions, and that co-denitrification of mix-cultures with F. oxysporum and P. stutzeri TR2 was more effective to remove nitrate under limited oxygen conditions. P. stutzeri TR2 was able to remove nitrate completely during cultivation for 12 hr in the specific medium and in mixed culture with F. oxysporum. A rapid N 2 evolution by mixed culture with F. oxysporum and P. stutzeri TR2 was observed in both mixed culture medium and synthetic wastewater. Using synthetic wastewater with a defined composition, about 87% of the nitrate was eliminated to form about 420μmol of N 2 from 1.0mmol of NO-3 by co-denitrification of F. oxysporum and P. stutzeri TR2 after incubation for 6days. In co-cultures of F. oxysporum and P. stutzeri TR2, N2O produced by F. oxysporum was rapidly consumed by P. stutzeri TR2. This indicated that mixed culture of F. oxysporum and P. stutzeri TR2 can be used to remove nitrate and nitrite from wastewater effectively.展开更多

It utilizes the micro-organisms to replace or to interact with the chemical reagents to increase the gap between surface properties of similar minerals and to enhance the separation selectivity. In this work, dolomite-phosphate separation was investigated using amphoteric collector (dodecyl-N-carboxyethyl-N-hyroxyethyl-imidazoline) in presence of bacteria. Two types of bacteria, Corynebacterium- diphtheriae-intermedius (CDI), and Pseudomonas aeruginosa (PA), were used. The collector-bacteria interaction was characterized by Fourier transform infra-red (FTIR), frothing height and Zeta potential. The results show that the collector-bacteria interaction improves the flotation selectivity. Although, the PA positively affects the separation results, the CDI cannot lower the MgO to less than 1%. A phosphate content of 0.7% MgO and 31.77% P205 with a recovery of 68% at pH 11, 3.0 kg/t amphoteric collector, 4× 10^7 cells of PA is obtained.展开更多

Approximately 136 strains of bacteria were isolated from phyllospheres of tobacco and 9 of these possessed high levels of antagonistic properties, They significantly reduced brown spot in detached tobacco leaves when artificially inoculated with Alternaria alternata. Culture filtrate of the most effective bacterial isolate which designated as Tpb88 was shown to be very efficient in inhibiting mycelial growth of A alternata in dual cultures. Culture filtrate of Tpb88 inhibited germination and germ tube elongation of A alternata. The results showed that the culture filtrate directly inhibited spore germination of A alternata, especially during the first hours of the paired cultivation. The rate of antagonistic activity of culture filtrate of Tpb88 depended on its concentration in the mixture. The greatest inhibition of spore germination was ob served at the highest concentration of filtrate (filtrate to fungal spores inocula mixed in proportion 1 : 50). These suggest that the hypothetic mechanism of Tpb88 against tobacco brown spot is to produce antagonistic substances.展开更多

Effects of part...Bench-scale soil column experiments were carried out to evaluate the effectiveness of Cr(VI) bioremediation process in soils by using indigenous bacteria with the addition of bacteria nutrient media. Effects of particle size, spray intensity, initial Cr(VI) concentration, circulation mode and soil depth on Cr(VI) remediation were studied. Results show that soils after 6 d remediation with spray intensity controlled in the range of 29.6-59.2 mL/min could well fulfill the requirement of concrete aggregate and roadbed material usage, for the leaching toxicity concentration of the Cr(VI) in treated soils under the chosen condition is far less than 5 mg/L The leaching toxicity and fractions of both hexavalent chromium and trivalent chromium from remediated soils were determined and compared with that of untreated soil. The results show that water soluble Cr(VI) declines from 1520.54 mg/kg to 0.68 mg/kg, exchangeable Cr(VI) decreases from 34.83 mg/kg to 0.01 mg/kg and carbonates-bonded Cr(V1) falls from 13.55 mg/kg to 0.68 mg/kg. Meanwhile, a corresponding increase in carbonate-bonded Cr(III), Fe and Mn oxides-bonded Cr(III) and organic matter-bonded Cr(III) are found. It reveals that indigenous bacteria can leach out water soluble Cr(VI), exchangeable Cr(VI) and carbonates-bonded Cr(VI) from contaminated soil followed by converting into carbonate-bonded Cr(III), Fe and Mn oxides-bonded Cr(IlI), organic matter-bonded Cr(III) and residual Cr(III).展开更多

The s...Bioleaching of sulfide minerals by bacteria, mainly Thiobacillus ferrooxidans(T.f.) and Thiobacillus thiooxidans, plays an important role in hydrometallurgy because of its economic and environmental attractions. The surveys of production process and the bacterial oxidation activity in the heap bioleaching were investigated. The results show that pH value is high, bacteria biomass and ferric concentration are low, generation time (above 7.13 h) is long in leachate, and less bacteria are adsorbed on the ores. The bacteria in the leachate exposing on the surface and connecting with mineral, have much faster oxidation rate of Fe(Ⅱ) and shorter generation time, compared with those which are in the reservoir for a long time. There is diversity for oxidation activity of Fe(Ⅱ), while there is no diversity for oxidation of sulfur. So it is advisable to add sulfuric acid to degrade pH value to 2.0, add nutrients and shorten recycling time of leachate, so as to enhance bacteria concentration of leachate and the leaching efficiency.展开更多
Background Apolygus lucorum is a worldwide omnivorous pest damaging a range of crops and causing great economic losses.Symbiotic bacteria living in insects play a key role in the nutrition,physiology,and behavior of h...Background Apolygus lucorum is a worldwide omnivorous pest damaging a range of crops and causing great economic losses.Symbiotic bacteria living in insects play a key role in the nutrition,physiology,and behavior of hosts.Here,we present an experiment using Illumina HiSeq sequencing targeting the V3–V4 regions of bacteria’s 16S rRNA throughout the entire life cycle of A.lucorum.Results The first and second instar nymphs have the largest alpha diversity compared with other life stages of the insect.Bacterial phyla Proteobacteria(72.29%),Firmicutes(15.24%),Actinobacteria(7.76%)exhibit the largest relative abundance in all developmental stages.Erwinia(23.97%)and Lactococcus(10.62%)are the two genera with the high-est relative abundance.The relative abundance of Erwinia in the nymph stage is significantly greater than the adult stage,and the relative abundance of Lactococcus in 6-day-old and 9-day-old adult females is higher compared with adult males.Conclusions These results reveal that microbial community composition and relative abundance shift dynamically at different life stages,implying that different bacterial phyla and genera may have specific roles in specific life stages such as metabolism,nutrition absorption,detoxification,and reproduction.This study reveals for the first time the community composition and ecological dynamics of symbiotic bacteria throughout the life stages of A.lucorum,and thus may provide insight to new strategies for pest control.展开更多

文摘Profiling the protein composition of bacteria is essential for understanding their biology,physiology and interaction with environment.Mass spectrometry has become a pivotal tool for protein analysis,facilitating the examination of expression levels,molecular masses and structural modifications.In this study,we compared the performance of three widely-used mass spectrometry methods,i.e.,matrix-assisted laser desorption/ionization(MALDI)protein fingerprinting,top-down proteomics and bottom-up proteomics,in the profiling of bacterial protein composition.It was revealed that bottom-up proteomics provided the highest protein coverage and exhibited the greatest protein profile overlap between bacterial species.In contrast,MALDI protein fingerprinting demonstrated superior detection reproducibility and effectiveness in distinguishing between bacterial species.Although top-down proteomics identified fewer proteins than bottom-up approach,it complemented MALDI fingerprinting in the discovery of bacterial protein markers,both favoring abundant,stable,and hydrophilic bacterial ribosomal proteins.This study represents the most systematic and comprehensive comparison of mass spectrometry-based protein profiling methodologies to date.It provides valuable guidelines for the selection of appropriate profiling strategies for specific analytical purposes.This will facilitate studies across various fields,including infection diagnosis,antimicrobial resistance detection and pharmaceutical target discovery.
文摘A novel compound(H_(2)L)SCN(5⁃methyl⁃3⁃phenyl⁃2H⁃pyrazol⁃1⁃ium thiocyanate)has been obtained by the reaction of thiosemicarbazide with benzoylacetone in ethanol.Two zinccomplexes[Zn(HL)_(2)(NCS)(CH_(3)COO)](1)and[Zn_(2)(L)_(2)(HL)_(2)(NCS)_(2)]_(2)·2CH_(3)OH(2)have been synthesized by the coordination reactions of Zn(OAc)_(2)·2H_(2)O or ZnCl_(2)with(H_(2)L)SCN under reflux conditions.Elemental analyses and single⁃crystal X⁃ray diffraction have con⁃firmed the structures of the synthesized compounds.The(H_(2)L)SCN ligand and complex 1 pertain to the triclinic sys⁃tem with space group P1,while complex 2 belongs to the monoclinic system with space group P2_(1)/n.Additionally,the antibacterial activities of the compounds were evaluated in vitro using the agar diffusion method against the bac⁃terial strains(Candida albicans,Staphylococcus aureus,and Escherichia coli).The results showed that the ligand exhibited relatively good antibacterial activities against the bacteria,and the complexes possessed stronger antibac⁃terial activities against the same bacteria than the free ligand.CCDC:2190252,(H2L)SCN;2190253,1;2190256,2.

文摘The biology safety of genetically modified organisms (GMO) has been a topic of considerable public debate in recent years. Parts of the international research on the safety of GMO focus on its effect on soil ecosystem, especially on microbial communities changing and process in soil that are essential to key terrestrial ecosystem functions. This paper studied the dynamic change of soil microbe after cultivating Roundup Ready soybean (RRS) and the effect on biochemical processing of nitrogen cycle. According to the variance analysis, the ammonifying bacteria and nitrifying bacteria quantities of RRS in the rhizosphere soil were much lower than that of other genotype soybeans. The effects of different genotype soybeans on ammoniation intensity and nitrification intensity were remarkable. The nitrification intensity and the nitrifying bacteria had the great positive correlation and sustainable development.

文摘Bioleaching experiments combined with X-ray photoelectron spectroscopy(XPS),X-ray diffraction(XRD)and scanning electron microscopy(SEM)were conducted to investigate three kinds of bornites from different regions leached by moderately thermophilic mixed bacteria of Leptospirillum ferriphilum YSK,Acidithiobacillus caldus D1 and Sulfobacillus thermosulfidooxidans ST.The results of bioleaching experiments showed that the leaching efficiency and the redox potential were significantly increased.The copper extraction efficiencies of three kinds of bornite maintained rapid growth until around the 12th day and no longer increased after the 18th,reaching 83.7%,96.5%and 86.6%,respectively.The XRD results of the leaching residue indicated that three kinds of bornites all produced jarosite in the late stage of leaching,and the leaching residues from of Daye Museum and Yunnan Geological Museum contained a mass of elemental sulfur.XPS analysis and scanning electron microscopy experiments showed that the surface of mineral particles was jarosite and the copper in the leaching residue was almost dissolved.

文摘According to the sequence of the bile salt hydrolase (BSH) gene of Bifidobacterium and the restriction enzyme cutting sites of expression vector pNZ8148, primers were designed and the bile salt hydrolase (BSH) gene was gotten from Bacillus bifidus ATCC 29521 by PCR. BSH gene was inserted into lactic acid bacteria expression vector pNZ8148 to construct the recombinant pNZ8148-BSH. The recombinant pNZ8148-BSH was transferred into lactic acid bacteria NZ9000 with electrotransformation method. And the recombinant which could express BSH protein was obtained. It was identified by SDS-PAGE electrophoresis and activity verification. The result could provide a rationale reference for expressing BSH in lactic acid bacteria.

文摘Ethylthionocarbamates (ETC), which is the most widely used as collectors in the flotation of sulfide, is known to cause serious pollution to soil and groundwater. The potential biodegradation of ETC was evaluated by applying a mixed culture of iron-reducing bacteria (IRB) enriched from tailings dam sediments. The results showed that ETC can be degraded by IRB coupled to Fe(III) reduction, both of which can be increased in the presence of anthraquinone-2,6-disulfonate (AQDS). Moreover, Fe(III)-EDTA was found to be a more favorable terminal electron acceptor compared to α-Fe2O3, e.g., within 30 d, 72% of ETC was degraded when α-Fe2O3+AQDS was applied, while it is 82.67% when Fe(III)-EDTA+AQDS is added. The dynamic models indicated that the kETC degradation was decreased in the order of Fe(III)-EDTA+AQDS〉α-Fe2O3+AQDS〉Fe(III)-EDTA〉α-Fe2O3, with the corresponding maximum biodegradation rates being 2.6, 2.45, 2.4 and 2.0 mg/(L·d), respectively, and positive parallel correlations could be observed between kFe(III) and kETC. These findings demonstrate that IRB has a good application prospect in flotation wastewater.

文摘Denitrification has been long thought to be a unique characteristic of prokaryotes, but in recent years, several filamentous fungi and yeasts were found to exhibit denitrifying activities. This paper deals with the examination of denitrification capabilities by mix-cultures of the fungus (Fusarium oxysporum) and the bacterium (Pseudomonas stutzeri TR2) in combination with a specific medium and using a synthetic wastewater of defined quality. The results revealed that P. stutzeri TR2 has strong and fast denitrifying capabilities under anaerobic conditions, and that co-denitrification of mix-cultures with F. oxysporum and P. stutzeri TR2 was more effective to remove nitrate under limited oxygen conditions. P. stutzeri TR2 was able to remove nitrate completely during cultivation for 12 hr in the specific medium and in mixed culture with F. oxysporum. A rapid N 2 evolution by mixed culture with F. oxysporum and P. stutzeri TR2 was observed in both mixed culture medium and synthetic wastewater. Using synthetic wastewater with a defined composition, about 87% of the nitrate was eliminated to form about 420μmol of N 2 from 1.0mmol of NO-3 by co-denitrification of F. oxysporum and P. stutzeri TR2 after incubation for 6days. In co-cultures of F. oxysporum and P. stutzeri TR2, N2O produced by F. oxysporum was rapidly consumed by P. stutzeri TR2. This indicated that mixed culture of F. oxysporum and P. stutzeri TR2 can be used to remove nitrate and nitrite from wastewater effectively.
文摘Eight cholesterol-lowering lactic acid bacteria(LAB 1 through 8)isolated from pickles and yogurt were evaluated for probiotic potential based on their tolerance to low pH and bile salt and antagonistic activity against Staphylococcus aureusand Escherichia coli.Strain LAB 4 had excellent tolerance to low pH and bile salt and all 8 LAB strains had antagonisticactivity against Staphylococcus aureus and Escherichia coli.Moreover,strain LAB4 had synergic cholesterol-lowering effect with 0.0125%licorice root extract,0.025%Chinese wolfberry extract or 0.025%milkvetch root extract.
文摘Bioflotation represents one of the growing trends to enhance the selectivity of conventional flotation processes. It utilizes the micro-organisms to replace or to interact with the chemical reagents to increase the gap between surface properties of similar minerals and to enhance the separation selectivity. In this work, dolomite-phosphate separation was investigated using amphoteric collector (dodecyl-N-carboxyethyl-N-hyroxyethyl-imidazoline) in presence of bacteria. Two types of bacteria, Corynebacterium- diphtheriae-intermedius (CDI), and Pseudomonas aeruginosa (PA), were used. The collector-bacteria interaction was characterized by Fourier transform infra-red (FTIR), frothing height and Zeta potential. The results show that the collector-bacteria interaction improves the flotation selectivity. Although, the PA positively affects the separation results, the CDI cannot lower the MgO to less than 1%. A phosphate content of 0.7% MgO and 31.77% P205 with a recovery of 68% at pH 11, 3.0 kg/t amphoteric collector, 4× 10^7 cells of PA is obtained.

文摘The possibility of employing antagonistic bacteria for the control of tobacco brown spot was studied. Approximately 136 strains of bacteria were isolated from phyllospheres of tobacco and 9 of these possessed high levels of antagonistic properties, They significantly reduced brown spot in detached tobacco leaves when artificially inoculated with Alternaria alternata. Culture filtrate of the most effective bacterial isolate which designated as Tpb88 was shown to be very efficient in inhibiting mycelial growth of A alternata in dual cultures. Culture filtrate of Tpb88 inhibited germination and germ tube elongation of A alternata. The results showed that the culture filtrate directly inhibited spore germination of A alternata, especially during the first hours of the paired cultivation. The rate of antagonistic activity of culture filtrate of Tpb88 depended on its concentration in the mixture. The greatest inhibition of spore germination was ob served at the highest concentration of filtrate (filtrate to fungal spores inocula mixed in proportion 1 : 50). These suggest that the hypothetic mechanism of Tpb88 against tobacco brown spot is to produce antagonistic substances.

文摘Bench-scale soil column experiments were carried out to evaluate the effectiveness of Cr(VI) bioremediation process in soils by using indigenous bacteria with the addition of bacteria nutrient media. Effects of particle size, spray intensity, initial Cr(VI) concentration, circulation mode and soil depth on Cr(VI) remediation were studied. Results show that soils after 6 d remediation with spray intensity controlled in the range of 29.6-59.2 mL/min could well fulfill the requirement of concrete aggregate and roadbed material usage, for the leaching toxicity concentration of the Cr(VI) in treated soils under the chosen condition is far less than 5 mg/L The leaching toxicity and fractions of both hexavalent chromium and trivalent chromium from remediated soils were determined and compared with that of untreated soil. The results show that water soluble Cr(VI) declines from 1520.54 mg/kg to 0.68 mg/kg, exchangeable Cr(VI) decreases from 34.83 mg/kg to 0.01 mg/kg and carbonates-bonded Cr(V1) falls from 13.55 mg/kg to 0.68 mg/kg. Meanwhile, a corresponding increase in carbonate-bonded Cr(III), Fe and Mn oxides-bonded Cr(III) and organic matter-bonded Cr(III) are found. It reveals that indigenous bacteria can leach out water soluble Cr(VI), exchangeable Cr(VI) and carbonates-bonded Cr(VI) from contaminated soil followed by converting into carbonate-bonded Cr(III), Fe and Mn oxides-bonded Cr(IlI), organic matter-bonded Cr(III) and residual Cr(III).
文摘Bioleaching of sulfide minerals by bacteria, mainly Thiobacillus ferrooxidans(T.f.) and Thiobacillus thiooxidans, plays an important role in hydrometallurgy because of its economic and environmental attractions. The surveys of production process and the bacterial oxidation activity in the heap bioleaching were investigated. The results show that pH value is high, bacteria biomass and ferric concentration are low, generation time (above 7.13 h) is long in leachate, and less bacteria are adsorbed on the ores. The bacteria in the leachate exposing on the surface and connecting with mineral, have much faster oxidation rate of Fe(Ⅱ) and shorter generation time, compared with those which are in the reservoir for a long time. There is diversity for oxidation activity of Fe(Ⅱ), while there is no diversity for oxidation of sulfur. So it is advisable to add sulfuric acid to degrade pH value to 2.0, add nutrients and shorten recycling time of leachate, so as to enhance bacteria concentration of leachate and the leaching efficiency.

文摘Background Apolygus lucorum is a worldwide omnivorous pest damaging a range of crops and causing great economic losses.Symbiotic bacteria living in insects play a key role in the nutrition,physiology,and behavior of hosts.Here,we present an experiment using Illumina HiSeq sequencing targeting the V3–V4 regions of bacteria’s 16S rRNA throughout the entire life cycle of A.lucorum.Results The first and second instar nymphs have the largest alpha diversity compared with other life stages of the insect.Bacterial phyla Proteobacteria(72.29%),Firmicutes(15.24%),Actinobacteria(7.76%)exhibit the largest relative abundance in all developmental stages.Erwinia(23.97%)and Lactococcus(10.62%)are the two genera with the high-est relative abundance.The relative abundance of Erwinia in the nymph stage is significantly greater than the adult stage,and the relative abundance of Lactococcus in 6-day-old and 9-day-old adult females is higher compared with adult males.Conclusions These results reveal that microbial community composition and relative abundance shift dynamically at different life stages,implying that different bacterial phyla and genera may have specific roles in specific life stages such as metabolism,nutrition absorption,detoxification,and reproduction.This study reveals for the first time the community composition and ecological dynamics of symbiotic bacteria throughout the life stages of A.lucorum,and thus may provide insight to new strategies for pest control.
