Cardiac arrest(CA)is a critical condition in the field of cardiovascular medicine.Despite successful resuscitation,patients continue to have a high mortality rate,largely due to post CA syndrome(PCAS).However,the inju...Cardiac arrest(CA)is a critical condition in the field of cardiovascular medicine.Despite successful resuscitation,patients continue to have a high mortality rate,largely due to post CA syndrome(PCAS).However,the injury and pathophysiological mechanisms underlying PCAS remain unclear.Experimental animal models are valuable tools for exploring the etiology,pathogenesis,and potential interventions for CA and PCAS.Current CA animal models include electrical induction of ventricular fibrillation(VF),myocardial infarction,high potassium,asphyxia,and hemorrhagic shock.Although these models do not fully replicate the complexity of clinical CA,the mechanistic insights they provide remain highly relevant,including post-CA brain injury(PCABI),post-CA myocardial dysfunction(PAMD),systemic ischaemia/reperfusion injury(IRI),and the persistent precipitating pathology.Summarizing the methods of establishing CA models,the challenges encountered in the modeling process,and the mechanisms of PCAS can provide a foundation for developing standardized CA modeling protocols.展开更多

OBJECTIVE Nowadays,with the living habits and diet structure changes,the morbidity and mortality of hypertension has been rising yearly,which induced by adverse lifestyles,excessive alcohol and poor diet.However,the a...OBJECTIVE Nowadays,with the living habits and diet structure changes,the morbidity and mortality of hypertension has been rising yearly,which induced by adverse lifestyles,excessive alcohol and poor diet.However,the animal models of hypertension resulted from adverse lifestyles have rarely been reported.It is essential to set up some more systematic models to study the characters of hypertension and evaluate the efficacy of traditional Chinese medicine(TCM).METHODS To build the rat model of human lifehabitual hypertension,SD rats had free access to alcohol with high-sucrose-fat diet(AHSFD),alcohol with high-fat diet(AHFD),high-sucrose-fat diet(HSFD),high-fat diet(HFD)and alcohol etc.Various of indicators were detected systematically to explore the features of hypertension models which included blood pressure(BP),liver and kidney functions,the lipid profiles and indexes quantifying TCM symptoms.Then we choose one of the stable and sustained animal models,studying the effects of RPA.E(which extracted from a TCM)on modulating the level of hypertension and the preliminary mechanism in this abstract.RESULTS ①The BP level,serum TC,TG,LDL-C of AHSFD-induced and AHFD-induced rats increased significantly,while the HDL-C reduced in the 4th week.② The BP level HSFD-induced rats increased significantly in the 6th week,serum TC,TG increased in 10 th week.③ The BP level of alcohol-induced rats increased significantly in the 9th week,serum TC,TG increased in 24 th week.④ The level of serum ALT,AST,UA and Cr of all model rats increased significantly after 12 weeks.Meanwhile the microcirculation increased significantly after8 weeks.⑤ After 4-week administration,RPA.E could significantly reduce BP of AHFD-induced hypertensive rats,and could reduce the serum TC,LDL-C,HDL-C,ALT,AST level after 8-week.CONCLUSION The rats induced by AHSFD,AHFD,HSFD and alcohol can raise BP obviously with underlying hepatic and kidney damage,the lipid profiles disorder and TCM symptoms change,while the AH(S)FD-induced hypertension models are earlier and more stable.RPA.E could mildly reduce BP level,and improve the lipid profiles disorders,hepatic damage,which reflects the characteristics of TCM on antihypertensive effects.The above models have their own characteristics,can be used to study the causes and pathogenesis of hypertension complicating metabolic disorder and the related treatment drug screening.展开更多
Well known for its abundant and extraordinary soft-tissue fossils,the Chengjiang fauna has witnessed the main phase of the Cambrian explosion and the first great congress of the ancestors of nearly all major phyla of ...Well known for its abundant and extraordinary soft-tissue fossils,the Chengjiang fauna has witnessed the main phase of the Cambrian explosion and the first great congress of the ancestors of nearly all major phyla of animals on Earth.The large-scale survey and exploration of the fauna by Chinese paleontologists,partly in collaboration with international scientists,over the past 30 years can be broadly divided into three stages.In the first decade since 1984(19841994),a large number of invertebrates,including basal animals and the protostomes of the early animal tree were discovered,but the subkingdom Deuterostomia was completely unknown.Then in the second decade(19952005),the great discoveries of various deuterostomes led to the construction of the basic framework of the Deuterostomia and then to the formation of the tripartite phylogenetic trees of early animals(TPTEA,including basal animals,protostomes and deuterostomes),for the first time.In the third decade(after 2005),the academic community began to think about the internal relationships between the formation of TPTEA and the multiepisode Cambrian explosion,leading to the new hypothesis of the three-episode Cambrian explosion.The Chengjiang fauna is important for deciphering the fauna structure,paleoecological environment,and so on.However,its core academic values mainly rest on two aspects.Firstly,The Chengjiang fauna,as the main phase witness of the Cambrian explosion,has created a nearly complete phylogenetic framework of the TPTEA on Earth for the first time.The three-phase radiation hypothesis reveals the essential connotation of the Cambrian explosion:a step-wise divergent evolution of animals,from basal to highly advanced groups,lasting about 40 million years.In the first phase,it gave birth to a bulk of basal animals(including some now extinct“animal”groups)in the latest Ediacaran,probably including some pioneer protostomes.The second phase took place in the first epoch of the Cambrian period(Terreneuvian),giving rise to the main invertebrate protostomes with a persistent prosperity of basal animals.The third phase proceeded in Cambrian Epoch 2(represented by the Chengjiang fauna),which not only maintained the prosperity of basal animals and protostomes,but also,more importantly,gave birth to all the main phyla of the subkingdom Deuterostomia.Thus,the rudimental framework of the whole TPTEA has been shaped,with the termination of the major innovation events of the Cambrian explosion.Here,we discussed the evolutionary properties of Ediacaran biota,small shelly fossils and the Chengjiang fauna in the Cambrian explosion with emphasis on the biological properties of several important animal groups.The order Myllokunmingiida is the only known oldest vertebrate,while Yunnanozoon and Haikouella are neither vertebrates nor stem-group chordates but a special group of basal deuterostomes;Cheungkongella is a credible ancestor of the urochordate and it supports the classical hypothesis on the origin of the urochordates;and the gill slits were first invented in the members of the phylum Vetulicolia to provide key information on the origin of the deuterostomes.The second core value of the Chengjiang fauna is of profound humanistic and philosophical significance:the discoveries of the‘first gill openings’,‘first brain’,‘first vertebrae’and‘first heart’provide the pivotal evidence for solving the unsolved mystery of the origin of the main basic human organs as described in Darwin’s“The Descent of Man”.In addition,the morphological and anatomical information of the Chengjiang fauna can provide important clues for a better understanding of most components of Ediacaran and Cambrian metazoans.展开更多

We found that considering the influence of the resistance to ciprofloxacin hydrochloride, enrofloxacin, ofloxacin, pefloacin mesylate and norfloxacin nicotinate of the five types of fluoroquinolones to GyrA mutation of animal Salmonella, the resistance of pefloacin mesylate had the most significant effect, while the resistance of ciprofloxacin hydrochloride and enrofloxacin were the least significant factors. Nearly half of the Salmonella supports such a rule that the MIC of norfloxacin nicotinate reaching 64 or 128 might lead to the mutation Ser83→Phe, MIC exceeding 512 might lead to the mutation Ser83→Gly; 60% of the sample supported that the MIC of enrofloxacin reaching 32 or 64 might lead to the mutation Asp87 →Asn. 80% of them agreed to the fact that the MIC of neither ciprofloxacin hydrochloride under 64 nor pefloacin mesylate below 512 might result in the gene mutation in 119 site. All Salmonella isolates supported the conclusion that the mutation Alal19→Val took place, if and only if the MIC of norfloxacin nicotinate exceeded 512.展开更多
OBJECTIVE In recent years,the frequency of hyperuricemia has gradually risen along with the improvement of living standards,the irregular of diet and overfeeding greasy and surfeit flavor.However,hyperuricemia is a di...OBJECTIVE In recent years,the frequency of hyperuricemia has gradually risen along with the improvement of living standards,the irregular of diet and overfeeding greasy and surfeit flavor.However,hyperuricemia is a disorder of purine metabolism,and is strongly associated with insulin resistance and abnormal glucose metabolism.It is important to obtain a more stable and sustained animal model for the efficacy evaluation of traditional Chinese medicine(TCM).METHODS To manufacture the rodent model of hyperuricemia,the theory of increasing the source of the uric acid,reducing uric acid excretion and inhibiting uricase were used.We observed the influence on the serum uric acid and other indicators of rats induced by some factors:the lipid emulsion,high purine diet,beer with sugar,beer with sugar and high purine,and so on.Then we choose one of the stable and sustained animal models,studying the effects of Plo.E(which extracted from a TCM)on modulating the level of serum uric acid and the preliminary mechanism in this abstract.RESULTS ① At the 2nd week,the level of serum UA,BUN,Cr,TC,LDL-c of rats in the lipid emulsion group raised significantly.② At the 6th weeks,the serum UA in both the high purine diet group and lipid emulsion group raised obviously.③ The effects of Plo.E on hyperuricemia rats induced by high purine diet:After 8d administration,the Plo.E(three dosages)can reduce the UA level,and the middle and low dosages can reduce the TG level.After 25 dadministration,Plo.E can reduce the plasma viscosity,UA,TG,the whole blood viscosity level,the high dosage also can reduce the TC level.CONCLUSION The rats induced by the high purine diet and lipid emulsion can raised the serum UA obviously,while the way of lipid emulsion is earlier and more stable.Plo.E has a therapeutic effect on hyperuricemia,with an activity of reducing plasma viscosity and blood lipid.The above models conform to the pathogenesis of humans,can be used to study the causes and pathogenesis of hyperuricemia complicating metabolic disorder and the related treatment drug screening.展开更多

文摘OBJECTIVE Nowadays,with the living habits and diet structure changes,the morbidity and mortality of hypertension has been rising yearly,which induced by adverse lifestyles,excessive alcohol and poor diet.However,the animal models of hypertension resulted from adverse lifestyles have rarely been reported.It is essential to set up some more systematic models to study the characters of hypertension and evaluate the efficacy of traditional Chinese medicine(TCM).METHODS To build the rat model of human lifehabitual hypertension,SD rats had free access to alcohol with high-sucrose-fat diet(AHSFD),alcohol with high-fat diet(AHFD),high-sucrose-fat diet(HSFD),high-fat diet(HFD)and alcohol etc.Various of indicators were detected systematically to explore the features of hypertension models which included blood pressure(BP),liver and kidney functions,the lipid profiles and indexes quantifying TCM symptoms.Then we choose one of the stable and sustained animal models,studying the effects of RPA.E(which extracted from a TCM)on modulating the level of hypertension and the preliminary mechanism in this abstract.RESULTS ①The BP level,serum TC,TG,LDL-C of AHSFD-induced and AHFD-induced rats increased significantly,while the HDL-C reduced in the 4th week.② The BP level HSFD-induced rats increased significantly in the 6th week,serum TC,TG increased in 10 th week.③ The BP level of alcohol-induced rats increased significantly in the 9th week,serum TC,TG increased in 24 th week.④ The level of serum ALT,AST,UA and Cr of all model rats increased significantly after 12 weeks.Meanwhile the microcirculation increased significantly after8 weeks.⑤ After 4-week administration,RPA.E could significantly reduce BP of AHFD-induced hypertensive rats,and could reduce the serum TC,LDL-C,HDL-C,ALT,AST level after 8-week.CONCLUSION The rats induced by AHSFD,AHFD,HSFD and alcohol can raise BP obviously with underlying hepatic and kidney damage,the lipid profiles disorder and TCM symptoms change,while the AH(S)FD-induced hypertension models are earlier and more stable.RPA.E could mildly reduce BP level,and improve the lipid profiles disorders,hepatic damage,which reflects the characteristics of TCM on antihypertensive effects.The above models have their own characteristics,can be used to study the causes and pathogenesis of hypertension complicating metabolic disorder and the related treatment drug screening.

文摘Well known for its abundant and extraordinary soft-tissue fossils,the Chengjiang fauna has witnessed the main phase of the Cambrian explosion and the first great congress of the ancestors of nearly all major phyla of animals on Earth.The large-scale survey and exploration of the fauna by Chinese paleontologists,partly in collaboration with international scientists,over the past 30 years can be broadly divided into three stages.In the first decade since 1984(19841994),a large number of invertebrates,including basal animals and the protostomes of the early animal tree were discovered,but the subkingdom Deuterostomia was completely unknown.Then in the second decade(19952005),the great discoveries of various deuterostomes led to the construction of the basic framework of the Deuterostomia and then to the formation of the tripartite phylogenetic trees of early animals(TPTEA,including basal animals,protostomes and deuterostomes),for the first time.In the third decade(after 2005),the academic community began to think about the internal relationships between the formation of TPTEA and the multiepisode Cambrian explosion,leading to the new hypothesis of the three-episode Cambrian explosion.The Chengjiang fauna is important for deciphering the fauna structure,paleoecological environment,and so on.However,its core academic values mainly rest on two aspects.Firstly,The Chengjiang fauna,as the main phase witness of the Cambrian explosion,has created a nearly complete phylogenetic framework of the TPTEA on Earth for the first time.The three-phase radiation hypothesis reveals the essential connotation of the Cambrian explosion:a step-wise divergent evolution of animals,from basal to highly advanced groups,lasting about 40 million years.In the first phase,it gave birth to a bulk of basal animals(including some now extinct“animal”groups)in the latest Ediacaran,probably including some pioneer protostomes.The second phase took place in the first epoch of the Cambrian period(Terreneuvian),giving rise to the main invertebrate protostomes with a persistent prosperity of basal animals.The third phase proceeded in Cambrian Epoch 2(represented by the Chengjiang fauna),which not only maintained the prosperity of basal animals and protostomes,but also,more importantly,gave birth to all the main phyla of the subkingdom Deuterostomia.Thus,the rudimental framework of the whole TPTEA has been shaped,with the termination of the major innovation events of the Cambrian explosion.Here,we discussed the evolutionary properties of Ediacaran biota,small shelly fossils and the Chengjiang fauna in the Cambrian explosion with emphasis on the biological properties of several important animal groups.The order Myllokunmingiida is the only known oldest vertebrate,while Yunnanozoon and Haikouella are neither vertebrates nor stem-group chordates but a special group of basal deuterostomes;Cheungkongella is a credible ancestor of the urochordate and it supports the classical hypothesis on the origin of the urochordates;and the gill slits were first invented in the members of the phylum Vetulicolia to provide key information on the origin of the deuterostomes.The second core value of the Chengjiang fauna is of profound humanistic and philosophical significance:the discoveries of the‘first gill openings’,‘first brain’,‘first vertebrae’and‘first heart’provide the pivotal evidence for solving the unsolved mystery of the origin of the main basic human organs as described in Darwin’s“The Descent of Man”.In addition,the morphological and anatomical information of the Chengjiang fauna can provide important clues for a better understanding of most components of Ediacaran and Cambrian metazoans.

文摘By means of mathematical modeling methods, we analyzed the relationship between the resistance to fluoroquinolones and GyrA mutation of Salmonella from animal isolates. We found that considering the influence of the resistance to ciprofloxacin hydrochloride, enrofloxacin, ofloxacin, pefloacin mesylate and norfloxacin nicotinate of the five types of fluoroquinolones to GyrA mutation of animal Salmonella, the resistance of pefloacin mesylate had the most significant effect, while the resistance of ciprofloxacin hydrochloride and enrofloxacin were the least significant factors. Nearly half of the Salmonella supports such a rule that the MIC of norfloxacin nicotinate reaching 64 or 128 might lead to the mutation Ser83→Phe, MIC exceeding 512 might lead to the mutation Ser83→Gly; 60% of the sample supported that the MIC of enrofloxacin reaching 32 or 64 might lead to the mutation Asp87 →Asn. 80% of them agreed to the fact that the MIC of neither ciprofloxacin hydrochloride under 64 nor pefloacin mesylate below 512 might result in the gene mutation in 119 site. All Salmonella isolates supported the conclusion that the mutation Alal19→Val took place, if and only if the MIC of norfloxacin nicotinate exceeded 512.

文摘OBJECTIVE In recent years,the frequency of hyperuricemia has gradually risen along with the improvement of living standards,the irregular of diet and overfeeding greasy and surfeit flavor.However,hyperuricemia is a disorder of purine metabolism,and is strongly associated with insulin resistance and abnormal glucose metabolism.It is important to obtain a more stable and sustained animal model for the efficacy evaluation of traditional Chinese medicine(TCM).METHODS To manufacture the rodent model of hyperuricemia,the theory of increasing the source of the uric acid,reducing uric acid excretion and inhibiting uricase were used.We observed the influence on the serum uric acid and other indicators of rats induced by some factors:the lipid emulsion,high purine diet,beer with sugar,beer with sugar and high purine,and so on.Then we choose one of the stable and sustained animal models,studying the effects of Plo.E(which extracted from a TCM)on modulating the level of serum uric acid and the preliminary mechanism in this abstract.RESULTS ① At the 2nd week,the level of serum UA,BUN,Cr,TC,LDL-c of rats in the lipid emulsion group raised significantly.② At the 6th weeks,the serum UA in both the high purine diet group and lipid emulsion group raised obviously.③ The effects of Plo.E on hyperuricemia rats induced by high purine diet:After 8d administration,the Plo.E(three dosages)can reduce the UA level,and the middle and low dosages can reduce the TG level.After 25 dadministration,Plo.E can reduce the plasma viscosity,UA,TG,the whole blood viscosity level,the high dosage also can reduce the TC level.CONCLUSION The rats induced by the high purine diet and lipid emulsion can raised the serum UA obviously,while the way of lipid emulsion is earlier and more stable.Plo.E has a therapeutic effect on hyperuricemia,with an activity of reducing plasma viscosity and blood lipid.The above models conform to the pathogenesis of humans,can be used to study the causes and pathogenesis of hyperuricemia complicating metabolic disorder and the related treatment drug screening.
