The results showed that A. chinensis seeds were recalcitrant with being highly desiccation-sensitive. The seed moisture content of fresh fruits was higher than 60%. When the seeds were naturally dried for 30 days, their moisture content declined to 30.2% and their viability was completely lost. The seed germination percentage had a small increase at the beginning of desiccation and then decreased rapidly. The relative electrical conductivity of the A. chinensis seeds increased along with a decrease in seed moisture content. However, there was an abnormal increase in relative electrical conductivity when the seed moisture content was between 53.7% and 50.9%. Superoxide dismutase (SOD) activity decreased rapidly in the period of desiccation except for an abnormality when the seed moisture content was between 53.7% and 50.9%. Malondialdehyde (MDA) content increased slowly at the early stage of desiccation and then rose rapidly after the moisture content was below 50.9%. The soluble sugar content in seeds slowly increased with the increasing period of desiccation. The seed germination percentage was at the high level when seed moisture content was in range of 47%- 60%, which suggests that this was the optimum moisture content for maintaining A. chinensis seed viability.展开更多
Objective To identify the factors associated with the development of postoperative atrial fibrillation (POAF) after coronary artery bypass graft (CABG) in elderly patients with coronary artery disease (CAD). Met...Objective To identify the factors associated with the development of postoperative atrial fibrillation (POAF) after coronary artery bypass graft (CABG) in elderly patients with coronary artery disease (CAD). Methods A total of 81 patients with CAD who underwent CABG were enrolled in the study. Patients were divided into two groups: Group 1, without postoperative atrial fibrillation (59 patients, 74.6% men, mean age 65.8 ~ 4.0 years); Group 2, with early new-onset atrial fibrillation after CABG (22 patients, 90.9% men, mean age 67.7 + 5.4 years). Interleukin (IL)-6, IL-8, IL-10, C-reactive protein (CRP), fibrinogen, superoxide dismutase (SOD), N-terminal pro-B-type natriuretic peptide (NT-proBNP), and troponin I were studied. Results During the observation period, atrial fibrillation occurred in 27.2% cases, an average of 4.9 ±3.8 days after surgery. In group 2, the left atrium (LA) dimension was larger than in group 1 (43.9 ± 3.4 mm vs. 37.6 ±3.9 rnm, P 〈 0.001). Patients with POAF had significantly higher IL-6 (72.7 ±60.8 pg/mL vs. 38.0 ± 34.6 pg/mL, P = 0.04), IL-8 (11.9 ± 6.0 pg/mL vs. 7.7± 5.4 pg/mL, P = 0.01) and SOD (2462.0 ± 2029.3 units/g vs. 1515.0 ± 1292.9 units/g, P = 0.04) compared with group without POAF. The multivariate analysis showed that the odds ratio (OR) for POAF development in patients with left atrium more than 39 mm was 2.1 [95% confidence interval (CI): 1.2-3.8, P = 0.0004], IL-6 levels more than 65.18 pg/mL-1.4 (95% Ch 1.1-2.7, P = 0.009), IL-8 levels more than 9.67 pg/mL-1.2 (95% CI: 1.1-3.7, P : 0.009), SOD more than 2948 units/g-1.1 (95% Ch 1.01-2.9, P = 0.04). Conclusions In our study, the independent predictors of postoperative atrial fibrillation after CABG in elderly patients were left atrium dimension and the increased postoperative concentration of IL-6, IL-8 and superoxide dismutase.展开更多

At different time-points, the normal and hepatic neoplasm tissues of the animal models were taken out to detect the superoxide dismutase (SOD) and malondialdehyde (MDA) respectively.The results show that the products and injurious effects of oxygen free radical (OFR) of the neoplasm tissues are more serious than those of the normal hepatic tissues.展开更多

Methods: The hepatocarc...Objective:To study the effect of reactive oxygen species of inducing apoptosis on the heptocacinoma tissues following ischemia and reperfusion and perfusion hyperoxia liquid of hepatocarcinoma. Methods: The hepatocarcinoma animal models ware established by implantation of VX2 tumor constitution mass into the left middle lobe of liver of rabbits. The animals were subjected to 60 min clamp-induced ischemia of hepatic artery distributing in the left middle lobe followed by reperfusion at 1 h, 1 d, 3 d and 7 d, respectively, and perfusion hyperoxia liquid (partial pressure of oxygen, PO2>80 kPa) at the same time with reperfusion beginning. The concentration of MDA and NO ware tested. Apoptotic changes in the hepatocarcinoma and normal hepatic tissues were observed by means of HE staining and terminal deoxynucleotidyl transferase-mediated dUTP nick end-labeling (TUNEL). Results:The concentration of MDA in normal hepatic tissues and hepatocarcinoma tissue increased followed ischemia and reperfusion especially for reperfusion 1 h (4.61±0.40, 3.10±0.23) and restored to normal level at reperfusion 7 d in normal hepatic tissues but still kept high concentration in the hepatocarcinoma tissue. Even though concentration of MDA in normal hepatic tissues is higher than that of before ischemia and reperfusion, no difference have been found after perfusion of hyperoxia liquid, and in the hepatocarcinoma tissue, the increasing of concentration of MDA was obvious after simply ischemia and reperfusion at reperfusion 1 d (4.25±0.45). The concentration of NO in normal hepatic tissues increased for reperfusion 3 d and 7 d(18.17±0.13, 17.45±0.23),while that of hepatocarcinoma tissue decreased at reperfusion 3 d(15.95±043). After perfusion of hyperoxia liquid, the concentration of NO in normal hepatic tissues kept increasing and that decreased in the hepatocarcinoma tissues in all time point and reached the lowest level at reperfusion 1 d(14.62±0.45).The result demonstrated the changes of concentration of NO and MDA in the hepatocarcinoma tissues ware more obvious than that of normal hepatic tissues(P<0.01). Conclusion:Perfusion of hyperoxia liquid from hepatic portal vein can intensify ischemia and reperfusion injury but less so for normal hepatic tissues.展开更多
Objective To investigate the radiosensitizing effect of nitric oxide(NO) combined with radiation on esophageal cancer cell line TE-1.Methods Methyl thiazolyl tetrazolium(MTT) assay was used to assess the effects of NO...Objective To investigate the radiosensitizing effect of nitric oxide(NO) combined with radiation on esophageal cancer cell line TE-1.Methods Methyl thiazolyl tetrazolium(MTT) assay was used to assess the effects of NO and radiation on TE-1 cells regarding inhibition of cell proliferation.Flow cytometry was used to examine the effect of NO and radiation on cell apoptosis and cycle.Reverse transcription polymerase chine reaction and Western blot were used to evaluete the effect of NO on mRNA and protein expression of manganese superoxide dismutase(MnSOD).Results NO inhibited the proliferation of TE-1 cells while significantly enhancing their radiosensitivity.The application of NO combined with radiation significantly increased the apoptosis rate and G2/M phase proportion of TE-1 cells,with substantial decreases in the MnSOD mRNA and protein expression levels.Conclusions NO reduces the MnSOD mRNA and protein expression levels by affecting TE-1 cell cycle,further inhibiting the apoptosis of esophageal cancer cells and enhancing the killing effect of radiation on esophageal cancer cells.展开更多

Meth...Objective To investigate the correlation between drinking behavior combined with polymorphisms of extracellular superoxide dismutase (EC-SOD) and aldehyde dehydrogenase-2 (ALDH2) genes and pancreatic cancer. Methods The genetic polymorphisms of EC-SOD and ALDH2 were analyzed by polymerase chain reaction restriction fragment length polymorphism in the peripheral blood leukocytes obtained from 680 pancreatic cancer cases and 680 non-cancer controls. Subsequently the frequency of genotype was compared between the pancreatic cancer patients and the healthy controls.The relationship of drinking with pancreatic cancer was analyzed. Results The frequencies of EC-SOD (C/G) and ALDH2 variant genotypes were 37.35% and 68.82% respectively in the pancreatic cancer cases, and were significantly higher than those in the healthy controls (21.03% and 44.56%, all P〈0.01). People who carried EC-SOD (C/G) (0R=2.24, 95% C1= 1.81-4.03, P〈0.01) or ALDH2 variant genotypes (OR=2.75, 95% CI=1.92-4.47, P〈0.01) had a high risk to develop pancreatic cancer. Those who carried EC-SOD (C/G) genotype combined with ALDH2 variant genotype had a high risk for pancreatic cancer (29.56% vs. 6.76%, 0R=7.69, 95% CI=3.58-10.51, P〈0.01). The drinking rate of the pancreatic cancer group (64.12%) was significantly higher than that of the control group (40.15%; OR=2.66, 95% CI=1.30-4.42, P〈0.01). An interaction between drinking and EC-SOD (C/G)/ALDH2 variant genotypes increased the risk of occurrence of pancreatic cancer (OR=25.00, 95% CI= 11.87-35.64, P〈0.01). Conclusion EC-SOD (C/G), ALDH2 variant genotypes and drinking might be the risk factors of pancreatic cancer.展开更多

The goal of this study was to prepare anti-CD31 monoclonal antibody conjugated PEG-modified liposomes containing the AU-rich region connecting factor 1 (AUF1) gene (CD31-PILs-AUF1) and to explore the effects of targeting CD31-PILs-AUF1 to aging VECs. Methods The mean particle sizes of various PEGylated immunoliposomes (PILs) were measured using a Zetasizer Nano ZS. Gel retardation assay was used to confirm whether PILs had encapsulated the AUF1 plasmid successfully. Fluorescence microscopy and flow cytometry were used to quantify binding of CD31-PILs-AUF1 to target cells. Flow cytometry was also used to analyze the cell cycles of aging bEnd3 cells treated with CD31-PILs-AUF1. We also developed an aging mouse model by treating mice with D-galactose. Enzyme-linked immunosorbent assay (ELISA) was used to evaluate the levels of interleuldn-6 (IL-6) and tumor necrosis factor-or (TNF-ct). The malondialdehyde (MDA) and the superoxide dismutase (SOD) levels were detected by commercial kits. Hematoxylin-eosin (HE) staining was used to determine whether treatment with CD31-PILs-AUF 1 was toxic to the mice. Results CD31-PILs-AUF 1 specifically could targeted bEnd3 VECs and increased the percentage of cells in the S and G2/M phases of aging bEnd3 cells. ELISA showed that content of the IL-6 and TNF-ct decreased in CD31-PILs-AUF1 group. The level of SOD increased, whereas MDA decreased in the CD31-PILs-AUF1 group. Additionally, CD31-PILs-AUF 1 was not toxic to the mice. Conclusion CD31-PILs-AUF 1 targets VECs and may delay their senescence.展开更多
