目的比较几种不同因素组合对实验性糖尿病肾病大鼠模型建立中大鼠体质量、血糖及尿微量白蛋白的影响,确定建立糖尿病肾病大鼠模型的最佳方案。方法雄性SD大鼠30只,随机分为5组,每组6只。A组(正常对照组),B组(高糖高脂饲料),C组高糖高脂... 目的比较几种不同因素组合对实验性糖尿病肾病大鼠模型建立中大鼠体质量、血糖及尿微量白蛋白的影响,确定建立糖尿病肾病大鼠模型的最佳方案。方法雄性SD大鼠30只,随机分为5组,每组6只。A组(正常对照组),B组(高糖高脂饲料),C组高糖高脂饲料+链脲佐菌素(Streptozotocin,STZ)组,D组(高糖高脂饲料+阿霉素+STZ组),E组(高糖高脂饲料+单肾切除+STZ组)。大鼠造模后每2周测体质量、进食量、饮水量、血糖及尿微量白蛋白。造模12周后,处死大鼠。结果E组大鼠明显多食、多饮、消瘦、血糖升高,尿微量白蛋白不仅与正常组比较有显著差异,并且与前几个模型组比较也都有显著性差异。肾/体质量比显著增大。结论以大鼠高糖高脂饲料4周后单肾切除,2周后小剂量腹腔注射链脲佐菌素的模型为最佳。 展开更多

Homology modeling and docking studies of IscS from extremophile Acidithiobacillus ferrooxidans 被引量:1

The gene iscS-3 from ,4cidithiobacillus ferrooxidans may play a central role in the delivery of sulfur to a variety of metabolic pathways in this organism. For insight into the sulfur metabolic mechanism of the bacteria, an integral three-dimensional (3D) molecular structure of the protein encoded by this gene was built by homology modeling techniques, refined by molecular dynamics simulations, assessed by PROFILE-3D and PROSTAT programs and further used to search bind sites, carry out flexible docking with cofactor pyridoxal 5'-phosphate(PLP) and substrate cysteine and hereby detect its key residues. Through these procedures, the detail conformations of PLP-IscS(P-I) and cysteine-PLP-IscS(C-P-I) complexes were obtained. In P-I complex, the residues of Lys208, His106, Thr78, Ser205, His207, Asp182 and Gln185 have large interaction energies and/or hydrogen bonds fixation with PLP. In C-P-I complex, the amino group in cysteine is very near His106, Lys208 and PLP, the interaction energies for cysteine with them are very high. The above results are well consistent with those experimental facts of the homologues from other sources. Interestingly, the four residues of Glul05, Glu79, Ser203 and Hisl80 in P-I docking and the residue of Lys213 in C-P-I docking also have great interaction energies, which are fitly conservation in IscSs from all kinds of sources but have not been identified before. From these results, this gene can be confirmed at 3D level to encode the iron-sulfur cluster assembly protein IscS and subsequently play a sulfur traffic role. Furthermore, the substrate cysteine can be presumed to be effectively recruited into the active site. Finally, the above detected key residues can be conjectured to be directly responsible for the bind and/or catalysis of PLP and cysteine. 展开更多

Thermogravimetric study of medical transfusion tube (MTT) waste containing polyvinyl chloride (PVC) was carried out using the thermogravimetric analyser (TGA) with N2, at different heating rates of 5, 10, 20, 30, 50 ℃/min. The purpose is to obtain pyrolysis characteristics and kinetic parameters of medical waste. The experimental results indicate that the pyrolysis behavior of the MTT sample is in agreement with its main ingredient of PVC, appearing two stair stepping stages. The influence of the additives in MTT on pyrolysis behavior was also revealed, which could improve MTT pyrolysis at lower temperature in the first stage, and cause obvious unsmoothness and asymmetry of the second DTG peak. Four n-order kinetic models of Coats-Redfern, Ozawa, Kissinger and Freeman-carroll were used to get the kinetic parameters. Furthermore, a novel "two-step four-reaction model" was established to simulate the whole continuous process. The different methods and the kinetic parameters thus obtained were discussed and compared with each other in literatures. The reasons of deviation among kinetic values were tried to be elucidated. The new established model could more satisfactorily describe the pyrolysis process of MTT, being more mechanistic and conveniently serving for the engineering. 展开更多

Homology modeling and evolutionary trace analysis of superoxide dismutase from extremophile Acidithiobacillus ferrooxidans 被引量:1

The gene sod in Acidithiobacillusferrooxidans may play a crucial role in its tolerance to the extremely acidic, toxic and oxidative environment of bioleaching. For insight into the anti-toxic mechanism of the bacteria, a three-dimensional (3D) molecular structure of the protein encoded by this gene was built by homology modeling techniques, refined by molecular dynamics simulations, assessed by PROFILE-3D and PROSTAT programs and its key residues were further detected by evolutionary trace analysis. Through these procedures, some trace residues were identified and spatially clustered. Among them, the residues of Asn38, Glyl03 and Glul61 are randomly scattered throughout the mapped structure; interestingly, the other residues are all distinctly clustered in a subgroup near Fe atom. From these results, this gene can be confirmed at 3D level to encode the Fe-depending superoxide dismutase and subsequently play an anti-toxic role. Furthermore, the detected key residues around Fe binding site can be conjectured to be directly responsible for Fe binding and catalytic function. 展开更多

Thermal decomposition of 21 kinds of binary mixtures between typical medical compositions was investigated under nitrogen conditions by dynamic thermogravimetric analysis(TGA) at 25–800 °C. The weighed sum method(WSM) coupled with thermal analysis was applied to study the interaction between components. Then, co-pyrolysis kinetic model of the binary mixtures(tube for transfusion(TFT) and gauze) was established to verify the reliability of conclusions. The results show the follows. 1) Strong or weak interactions are shown between binary mixtures containing polyvinyl chloride(PVC), the main ingredient of TFT. The addition of other medical waste could enhance first stage decomposition of TFT. While, the secondary stage pyrolysis may be suppressed or enhanced or not affected by the addition. 2) There exists no interaction between catheter and other component, the DTG peak temperature representing Ca CO3 decomposition in catheter fraction is obviously lower than that of pure catheter; while,the shape of DTG peak keeps unchanged. 3) No evident reaction occurs between the other mix-samples, it is considered that their co-pyrolysis characteristics are linear superposition of mono-component pyrolysis characteristics. 展开更多

In the hydraulic transporting process of cutter-suction mining natural gas hydrate, when the temperature-pressure equilibrium of gas hydrate is broken, gas hydrates dissociate into gas. As a result, solid-liquid two-phase flow(hydrate and water) transforms into gas-solid-liquid three-phase flow(methane, hydrate and water) inside the pipeline. The Euler model and CFD-PBM model were used to simulate gas-solid-liquid three-phase flow. Numerical simulation results show that the gas and solid phase gradually accumulate to the center of the pipe. Flow velocity decreases from center to boundary of the pipe along the radial direction. Comparison of numerical simulation results of two models reveals that the flow state simulated by CFD-PBM model is more uniform than that simulated by Euler model, and the main behavior of the bubble is small bubbles coalescence to large one. Comparison of numerical simulation and experimental investigation shows that the values of flow velocity and gas fraction in CFD-PBM model agree with experimental data better than those in Euler model. The proposed PBM model provides a more accurate and effective way to estimate three-phase flow state of transporting gas hydrate within the submarine pipeline. 展开更多

In the background subtraction stage, moving objects were extracted. Then, the initial classification for moving shadow pixels and foreground object pixels was performed by using color invariant features. In the shadow model learning stage, instead of a single Gaussian distribution, it was assumed that the density function computed on the values of chromaticity difference or bright difference, can be modeled as a mixture of Gaussian consisting of two density functions. Meanwhile, the Gaussian parameter estimation was performed by using EM algorithm. The estimates were used to obtain shadow mask according to two constraints. Finally, experiments were carried out. The visual experiment results confirm the effectiveness of proposed method. Quantitative results in terms of the shadow detection rate and the shadow discrimination rate(the maximum values are 85.79% and 97.56%, respectively) show that the proposed approach achieves a satisfying result with post-processing step. 展开更多

Cardiac arrest(CA)is a critical condition in the field of cardiovascular medicine.Despite successful resuscitation,patients continue to have a high mortality rate,largely due to post CA syndrome(PCAS).However,the injury and pathophysiological mechanisms underlying PCAS remain unclear.Experimental animal models are valuable tools for exploring the etiology,pathogenesis,and potential interventions for CA and PCAS.Current CA animal models include electrical induction of ventricular fibrillation(VF),myocardial infarction,high potassium,asphyxia,and hemorrhagic shock.Although these models do not fully replicate the complexity of clinical CA,the mechanistic insights they provide remain highly relevant,including post-CA brain injury(PCABI),post-CA myocardial dysfunction(PAMD),systemic ischaemia/reperfusion injury(IRI),and the persistent precipitating pathology.Summarizing the methods of establishing CA models,the challenges encountered in the modeling process,and the mechanisms of PCAS can provide a foundation for developing standardized CA modeling protocols. 展开更多
