目的 探讨血管内皮生长因子在前列腺癌和前列腺增生发生与发展过程中的潜在生物学作用及其与肿瘤细胞分化程度的关系。方法 采用免疫组织细胞化学SABC技术 ,对 31例前列腺癌和 17例前列腺增生组织中血管内皮生长因子的表达与分布状况... 目的 探讨血管内皮生长因子在前列腺癌和前列腺增生发生与发展过程中的潜在生物学作用及其与肿瘤细胞分化程度的关系。方法 采用免疫组织细胞化学SABC技术 ,对 31例前列腺癌和 17例前列腺增生组织中血管内皮生长因子的表达与分布状况及其与肿瘤细胞分级的关系进行了检测与分析。结果 血管内皮生长因子在前列腺癌和前列腺增生组织中均有表达 ;在前列腺上皮细胞胞浆、基质细胞和间质中呈弥散分布 ,血管上皮细胞及其周围基质细胞呈强阳性表达。半定量分析结果显示 ,前列腺癌组织中表达强度高于前列腺增生组织 ;未发现其表达强度与前列腺癌细胞分级 (或分化程度 )有统计学关联。结论 该研究提示 ,血管内皮生长因子在前列腺癌和前列腺增生组织中分布广泛 。 展开更多

目的 探讨IGF - 1R在前列腺腺癌和前列腺增生组织中的分布及其细胞生物学意义。方法 采用免疫组织化学技术ABC方法 ,对 2 3例前列腺腺癌和 15例前列腺增生组织中IGF - 1R的表达情况进行了检测分析。结果 前列腺腺癌组织和前列腺增生... 目的 探讨IGF - 1R在前列腺腺癌和前列腺增生组织中的分布及其细胞生物学意义。方法 采用免疫组织化学技术ABC方法 ,对 2 3例前列腺腺癌和 15例前列腺增生组织中IGF - 1R的表达情况进行了检测分析。结果 前列腺腺癌组织和前列腺增生组织中IGF - 1R表达均呈阳性 ,IGF - 1R主要分布于前列腺上皮细胞膜 ,基质细胞中亦有散在、少量的分布 ;半定量分析结果显示 ,前列腺增生组织中IGF -1R表达 (或染色 )强度高于前列腺癌组织 ;ICF - 1R表达强度与前腺癌细胞分级呈负相关趋势 ,肿瘤细胞分级越高 (或分化越差 )IGF - 1R表达越弱。结论 研究提示 ,IGF - 1R (或IGFs系统 )在前列腺上皮细胞转化和肿瘤细胞增殖分化过程中具有一定作用。 展开更多

Objective: To investigate the expression of aquaporin 1 in cervical squamous carcinomas (CSC) and cervical precancerous lesions, and the relationship between the tumor clinicopathological parameters, prognosis and the expression of AQP1. Methods: Immunohistochemical method (EliVision) was used to detect the expression of AQP1 in samples from 106 patients [20 with normal cervical tissue, 30 with cervical intraepithelial neoplasia (stage Ⅰ and Ⅱ) and 56 with CSC]. Survival analysis was performed by Kaplan-Meier method. Results: AQP1 protein was expressed in vascular endothelia of all samples. It showed upregulation of AQP1 expression in CSC. There was a significant difference between CSC and normal cervical tissues (P〈0.05). AQP1 was expressed in some tumor cells and unexpressed in normal squamous epithelial cells. And APQl-expressing tumor cells were positively related to lymph node metastasis. Patients with APQl-expressing tumor cells had the lower survival rate than the ones without. Conclusion: Abnormal expression of AQP1 plays an important role in the development of CSC. Positive expression of AQP1 in tumor cells maybe enhances tumor metastasis and could be used as a marker for tumor prognosis. 展开更多

The mutation of p53 gene was detected in Syrian hamster embryo(SHE) cells neoplastically initiated with a particles.The level of the p53 mRNA in transformant was obviously higher than that in non-irradiated counterpart,as measured by Northern blot analysis of total RNA.A pair of primers were designed based on p53 cDNA sequence to produce the whole length of coding sequence about 1.2 kilobase(Kb) by reverse transcription of mRNA followed by the polymerase chain reaction(RTPCR),but the length of fragment amplified from transformant mRNA was about 0.3Kb,remarkably shorter than that from normal SHE cells.Imrnunohistochemical analysis of p53 protein showed that no heavy staining was found on slice of tumor derived from transformant inoculated in nude mice with hamster specific p53 monocloned antibody HD200.The results implied that p53 gene had been mutated by deletion,which might lead to loss of p53 protein expression but the increased expression of p53 remained in a particle-induced SHE transformant. 展开更多

Effects of valsartan on oxidative stress and the atherogenesis

Objective: To investigate the therapy effect of valsartan on oxidative stress and the formation of atherosclerosis of rabbit. Methods: An atherosclerotic rabbit model was established by feeding high cholesterol diet supplemented by bovine serum albumin injection bolus. The rabbits were randomly divided into the control, model, and valsartan treated group, six rabbits in each group. Blood samples were collected at the end of 8 weeks for examination of serum lipid levels and MDA levels; the aortas were harvested for histological morphometry analysis, vascular cell adhesion molecule-1 (VCAM-1) immunohistochemical analysis and in situ superoxide detection to reflect the activity of NAD(P)H oxidase. Results: Rabbits fed with high cholesterol diet showed higher serum lipids levels than those fed with normal diet(P<0.01). Treatment with valsartan (10 mg/kg per day) did not alter serum lipids levels. But the serum MDA level and ratio of lesion to intima area reduced significantly compared with model group(P<0.05). The expression of VCAM-1 decreased significantly in the valsartan treated group than in the model group (P<0.05).In addition, in situ superoxide detection also show the markedly reduction of superoxide as a result of valsartan treatment. Conclusion: These results indicate that the valsartan treatment can reduce the atherosclerotic progression, the mechanisms of which may include the inhibiting the NAD(P)H oxidase activity to produce superoxide and the downregulating the expression of redox sensitive genes in the downstream, such as VCAM-1. 展开更多

Objective: To detect the different morphologic features, developmental regulation, potential of proliferation and differentiation of neonatal rat cochlea progenitor spheres. Methods: We isolated the cochlea sensory epithelium cells from neonatal rats and cultured them in nonadherent conditions to acquire different morphologic spheres. Then we observed the diameter and compositional change of cell colonies in distinct sphere types on day 3, 6, 9 and 12, and summarized the regularity of development and their conversion. We also detected the proliferative activity of distinct spheres by immunohistochemical staining of Abcg2, Nestin and BrdU. After induced spontaneous differentiation, the spheres were detected in the changes of the marker of hair cell, MyosinVIIA; by immunocytochemical staining, we revealed the potential of how different spheres were converted into hair cell-like cells. Results: The acquired three types of suspended spheres are solid, transitional, and hollow. There's morphologic significance among them and they can covert into the other type of spheres among them. The ability of self-renewing and proliferation in distinct spheres vary and all of them have the potential of spontaneously differentiation into hair cell-like cells. Conclusion: All the type of spheres not only has the potential of proliferation and differentiation, but also hasthe potential of spontaneous differentiation into hair cell-like cells. Distinct types of cell spheres neither originate from different progenitor cell subcolonies nor are different stages of the same cell spheres. Solid spheres are most practically useful. 展开更多

Objective: To construct the EGFR targeted non-viral vector GE7 system and explore the in vitro effect of p21WAF-1/CIPI gene on growth of human glioma cells mediated by the GE7 system. Methods: The EGFR targeted non-viral vector GE7 gene delivery system was constructed. The malignant human glioma cell line U251MG was transfected in vitro with β-galactosidase gene ( reporter gene) and p21WAF-1/CIPI gee (therapeutic gene) using the GE7 system. By means of X-gal staining, MTS and FACS, the transfection efficiency of exogenous gene and apoptosis rate of tumor cells were examined. The expression of p21WAF-1/ CIPI gene in transfected U251MG cell was examined by immunohistochemis-try staining. Results: The highest transfer rate of exogenous gene was 70% . After transfection with p21WAF-1/CIPI gene, the expression of WAF-1 increased remarkably and steadily; the growth of U251MG cells were inhibited evidently. FACS examination showed G1 arrest. The average apoptosis rate was 25.2%. Conclusion: GE7 system has the ability to transfer exogenous gene to targeted cells efficiently, and expression of p21WAF-1/CIPI gene can induce apoptosis of glioma cell and inhibit its growth. 展开更多

Methods: The mononuclear cells were separated from the peripheral, spleen and uterus or placenta blood. Flow cytometry was employed to analyze the percent of Treg cells in total T cells in different stages of pregnancy. Immunohistochemical staining was used to make sure the distribution of Treg in spleen in different stages of pregnancy. Results: The results of immunohistochemical staining showed that compared with spleen Treg cells in normal unpregnant mice, spleen Treg cells on day 7 and 14 of pregnancy significantly increased. After splenectomy, peripheral blood and placenta Treg cells on day 7 of pregnancy markedly decreased as compared with the normal pregnancy(P<0.01). And the cells on day 14of pregnancy were markedly recovered as compared with the normal pregnancy. Conclusion: Our study indicated that the spleen and its Treg cells might play important roles in transient tolerance during pregnancy. 展开更多
