Objective:Ovarian cancer(OC)ranks among the leading causes of mortality among the female cancers worldwide.Numerous studies have explored the development and progression of OC at multiple genetic regulatory levels.How...Objective:Ovarian cancer(OC)ranks among the leading causes of mortality among the female cancers worldwide.Numerous studies have explored the development and progression of OC at multiple genetic regulatory levels.However,relatively few studies have explored the impact of post-translational modifications(PTM)on OC progression,which is essential for uncovering new therapeutic targets.This study aimed to systematically identify the key PTM types involved in OCprogression,and to explore and evaluate their translational potential as therapeutic targets.Methods:First,we utilized multiple general PTM antibodies to compare gross PTM levels between normal ovarian and OC tissues from clinical females.After identifying lactylation as the PTM with the most significant differences,we selected representative samples for label-free mass spectrometry to identify specific lactylation sites.Next,we transfected A2780(OC)cells with either wild-type(WT)or mutant(K192A[Q])poly(ADP-ribose)polymerase 1(PARP1)conjugated to enhanced green fluorescent protein(EGFP)with a StrepⅡpeptide tag and assessed various cellular indexes related to cell proliferation(clonogenicity assay),migration(scratch wound healing assay),and reactive oxygen species levels.Results:Pan-lactylation was significantly upregulated in clinical OC samples,with PARP1 lactylation at K192 being one of the most common modifications.The growth and migration of A2780 cells were markedly suppressed by overexpressing PARP1-WT but not mutant PARP1.Overexpressing PARP1 significantly downregulated the phosphorylation of extracellular signal-regulated kinases 1/2(ERK1/2).Conclusion:This study uncovered a novel PTM of PARP1 in OC,lactylation,and demonstrated that lactylation at K192 is crucial in regulating OC cell growth and migration via the ERK1/2 pathway.Further investigations are required to elucidate the broader functional implications of PARP1 lactylation and its therapeutic potential.展开更多

Background Cotton crop is infested by numerous arthropod pests from sowing to harvesting,causing substantial direct and indirect yield losses.Knowledge of seasonal population trends and the relative occurrence of pest...Background Cotton crop is infested by numerous arthropod pests from sowing to harvesting,causing substantial direct and indirect yield losses.Knowledge of seasonal population trends and the relative occurrence of pests and their natural enemies is required to minimize the pest population and yield losses.In the current study,analysis of the seasonal population trend of pests and natural enemies and their relative occurrence on cultivars of three cotton species in Central India has been carried out.Results A higher number and diversity of sucking pests were observed during the vegetative cotton growth stage(60 days after sowing),declining as the crop matured.With the exception of cotton jassid(Amrasca biguttula biguttula Ishida),which caused significant crop damage mainly from August to September;populations of other sucking insects seldom reached economic threshold levels(ETL)throughout the studied period.The bollworm complex populations were minimal,except for the pink bollworm(Pectinophora gossypiella Saunders),which re-emerged as a menace to cotton crops during the cotton cropping season 2017–2018 due to resistance development against Bt-cotton.A reasonably good number of predatory arthropods,including coccinellids,lacewings,and spiders,were found actively preying on the arthropod pest complex of the cotton crop during the early vegetative growth stage.Linear regression indicates a significant relationship between green boll infestations and pink bollworm moths in pheromone traps.Multiple linear regression analyse showed mean weekly weather at one-or two-week lag periods had a significant impact on sucking pest population(cotton aphid,cotton jassid,cotton whitefly,and onion thrips)fluctuation.Gossypium hirsutum cultivars RCH 2 and DCH 32,and G.barbadense cultivar Suvin were found susceptible to cotton jassid and onion thrips.Phule Dhanvantary,an G.arboreum cotton cultivar,demonstrated the highest tolerance among all evaluated cultivars against all sucking pests.Conclusion These findings have important implications for pest management in cotton crops.Susceptible cultivars warrant more attention for plant protection measures,making them more input-intensive.The choice of appropriate cultivars can help minimize input costs,thereby increasing net returns for cotton farmers.展开更多

However, there is currently no non-destructive and quantitative detection method for migration of plasticizers in propellant liner. In this study, we developed a HTPB sensing liner by incorporating conductive fillers-namely carbon black(CB), carbon nanotubes(CNTs), and graphene nanoplatelets(GNP)-into the HTPB matrix. The synergistic interaction between CNTs and GNP facilitates the formation of a tunneling conductive network that imparts electrical conductivity to the HTPB liner. To elucidate the functional relationship between conductivity and nitroglycerin(NG) migration, we applied the HTPB sensing liner onto double base propellant surfaces and measured both the conductivity of the sensing layer and NG migration during a 71°C accelerated aging experiment. The results shows that when CNTs/GNP content reaches 3wt%, there is an exponential correlation between conductivity and NG migration with a fitting degree of 0.9652;the average response sensitivity of ΔR/R0 relative to NG migration is calculated as 41.69, with an average deviation of merely5.67% between NG migrations derived from conductivity fittings compared to those obtained via TGA testing results. Overall, this sensing liner exhibits excellent capabilities for detecting NG migration nondestructively and quantitatively while offering a novel approach for assessing interfacial component migrations as well as debonding defects in propellants-a promising avenue for future self-monitoring strategies regarding propellant integrity.展开更多
Background Cotton is an important cash crop in China and a key component of the global textile market.Verticil-lium wilt is a major factor affecting cotton yield.Single nucleotide polymorphism(SNP)markers and phenotyp...Background Cotton is an important cash crop in China and a key component of the global textile market.Verticil-lium wilt is a major factor affecting cotton yield.Single nucleotide polymorphism(SNP)markers and phenotypic data can be used to identify genetic markers and loci associated with cotton resistance to Verticillium wilt.We used eight upland cotton parent materials in this study to construct a multiparent advanced generation inter-cross(MAGIC)population comprising 320 lines.The Verticillium wilt resistance of the MAGIC population was identified in the green-house in 2019,and the average relative disease index(ARDI)was calculated.A genome-wide association study(GWAS)was performed to discover SNP markers/genes associated with Verticillium wilt resistance.Results ARDI of the MAGIC population showed wide variation,ranging from 16.7 to 79.4 across three replicates.This variation reflected a diverse range of resistance to Verticillium wilt within the population.Analysis of distribution pat-terns across the environments revealed consistent trends,with coefficients of variation between 12.25%and 21.96%.Families with higher ARDI values,indicating stronger resistance,were more common,likely due to genetic diver-sity and environmental factors.Population structure analysis divided the MAGIC population into three subgroups,with Group I showing higher genetic variation and Groups II and III displaying more uniform resistance performance.Principal component analysis(PCA)confirmed these divisions,highlighting the genetic diversity underlying Verticil-lium wilt resistance.Through GWAS,we identified 19 SNPs significantly associated with Verticillium wilt resistance,distributed across three chromosomes.The screening of candidate genes was performed on the transcriptome derived from resistant and susceptible cultivars,combined with gene annotation and tissue expression patterns,and two key candidate genes,Ghir_A01G006660 and Ghir_A02G008980,were found to be potentially associated with Verticillium wilt resistance.This suggests that these two candidate genes may play an important role in responding to Verticillium wilt.Conclusion This study aims to dissect the genetic basis of Verticillium wilt resistance in cotton by using a MAGIC population and GWAS.The study seeks to provide valuable genetic resources for marker-assisted breeding and enhance the understanding of resistance mechanisms to improve cotton resilience against Verticillium wilt.展开更多

OBJECTIVE To investigate the role and mechanism of G protein-coupled receptor kinase 2(GRK2)involving in hepatocel ular carcinoma(HCC)progression.METHODS Cel Counting Kit 8 and tumor colony formation assay were design...OBJECTIVE To investigate the role and mechanism of G protein-coupled receptor kinase 2(GRK2)involving in hepatocel ular carcinoma(HCC)progression.METHODS Cel Counting Kit 8 and tumor colony formation assay were designed to detect HCC cell proliferation,wound healing assay was to detect HCC migration.The correlation between GRK2 and early growth response-1(EGR1)were detected by RT-PCR and real-time PCR assays.Co-immunoprecipitation and Western blot assay were adopted to detect the relationship between GRK2and insulin-like growth factor 1 receptor(IGF-1R)signaling pathway.RESULTS In this study we find that GRK2plays an inhibition role in IGF1-induced HCC cell proliferation and migration.Overexpression of GRK2 causes a decrease in EGR1 expression,while knockdown of GRK2 leads to the dramatically increase in EGR1 expression in the treatment of IGF1.Through co-immunoprecipitation and Western blot assay,we confirm that GRK2can interact with IGF-1R and inhibiting IGF1-induced activation of IGF1R signaling pathway.Silencing EGR1attenuates GRK2 overexpression-caused inhibition of cell proliferation,tumor colony number and migrationactivity,while overexpressing of EGR1 restores the antiproliferative and migratory effect by GRK2 overexpression in HCCLM3 cells.CONCLUSION Taken together,these results suggest that GRK2 may inhibit IGF1-induced HCC cell growth and migration through down-regulation of EGR1 and indicate that enforced GRK2 may offer a potential therapeutic approach against HCC.展开更多

Four levels of SPs supplements(0, 0.25%, 0.50% and 0.75% dry matter basis) were tested. p H, NH3-N, Neutral Detergent Fiber(NDF), Acid Detergent Fiber(ADF), Dry Matter Digestibility(DMD) and the Total Volatile Fatty Acid(TVFA) were measured at 6, 24, 48 h of in vitro mixed incubation the fluids. Microbial populations were determined at 24 h and Microbial Proteins(MCP) were determined at 24 and 48 h. The gas production after 48 h in vitro incubation presented linear growth(p<0.05) and the highest content was the level of 0.5% SPs(dry matter basis). NH3-N concentration reached the highest concentration with 0.75%(dry matter basis) at 48 h. p H linearly increased(linear, p<0.05) from 6 to 48 h. The digestion of DMD increased with increasing doses of SPs at 24 h. NDF and ADF linearly(p<0.01) decreased by adding SPs. The concentration of TVFA linearly increased(p<0.05) at 6, 24 and 48 h, and reached the highest concentration at the level of 0.75%(dry matter basis). SPs decreased(p<0.05) the molar proportion of acetate and propionate, respectively at 24 and 48 h. Acetat to Propionate ratio(A/P; linear, p<0.05) increased at 48 h, and reached the greatest value at the level of 0.75%(dry matter basis) at 48 h. The populations of rumen Ruminococcus flavefaciens(R. flavefaciens), Ruminococcusalbus(R. albus), Fibrobacter succinogenes(F. succinogenes), Butyrivibrio fibrisolvens(B. fibrisolvens), Streptococcus bovis(S.bovis), Ruminobacter amylophilus(R. amylophilus) and Succinimonas amylolytica(S. amylolytica) were analyzed based on the total rumen bacterial 16 S ribosomal deoxyribonucleic acid(r DNA). The relative abundance of R. flavefaciens and R. albus increased at 24 h, but the relative abundance of F. succinogenes decreased at this time. The incubation of SPs had no effect on the abundance of S. bovis and R. amylophilus. The relative abundance of B. fibrisolvens and S. amylolytica reached the greatest value(p<0.05) at middle doses of SPs inclusion at 24 h. The value of MCPs linearly increased(p<0.05) at 24 and 48 h. These results showed that SPs could improve in vitro fermentation and nutrient digestion of feed substrates, and had the ability to modulate the ruminal fermentation pattern by regulating the composition of functional rumen microbes. Hence, SPs might be a potential feed additive applied in the diets of ruminants.展开更多

Complex traits are multifactorial traits controlled by polygenic host factors.These trait-related phenotypic characteristics and performance including body weight,blood chemistry,immune cell profiles,as well host susc...Complex traits are multifactorial traits controlled by polygenic host factors.These trait-related phenotypic characteristics and performance including body weight,blood chemistry,immune cell profiles,as well host susceptibility to infectious and chronic diseases.In recent years,tremendous efforts were invested aiming to map the host genetic factors attribute to these traits and subsequently clone the gene/s underlying these loci.In parallel to human studies,a number of mouse models and approaches were developed aimed to enhance the mapping process and the gene cloning.These include of using resources such as F2,backcross,advanced intercross lines,outbred populations,consomic,congenic and recombinant inbred lines(RIL).The constraints of these approaches were the limited resolution mapping of genomic regions of the quantitative trait loci(QTL)associated with the trait of interests,and the limited genetic diversity observed in the parental founders.To overcome these limitations,a new genetically highly diverse recombinant inbred lines of mouse population was established,namely the Collaborative Cross(CC),created from full reciprocal mating of 8 divergent strains of mice:A/J,C57BL/6J,129S1/SvI mJ,NOD/LtJ,NZO/HiL tJ,CAST/Ei,PWK/PhJ,and WSB/EiJ.By intercrossing these eight founders to generate the different CC lines,the genetic makeup of the newly developed resource is completely different from the eight parental lines,and will show heterosis,which subsequently will response differently comparing with their original founders.Finally,our results suggest that it is not essential to defining the phenotypic response of the eight parental lines,prior of assessing the CC lines,because it is believed that genetic interaction of the new genetic makeup of the new lines will reveal new phenotypic response,which completely different from the parental lines.In this report,we present to the community the power of the CC for dissecting variety of complex traits including host susceptibility to infectious and chronic diseases as well body performance traits.Based on our results from a variety of studies,we recommend to the community,that the best strategy of using this population is to aim of phenotyping about 50 and more of CC lines,with limited number of biological replicates(3-4 mice per line),and subsequently using the publicly available high dense genotype information of the CC lines as well the sequence database of the eight founders,it will be possible performing QTL mapping to a unprecedented precision genomic regions less than 1 MB,subsequently lead to identify potential strong candidate genes.These achievements are believed cannot be obtained with any other currently available mouse resource populations.展开更多

This study aimed to represent the genetic differentiation among different geographic populations of soybean aphid in Northeast China. In order to investigate and assess the genetic diversity, genetic differentiation, molecular variance, population structure, ecological importance and evolutionary history of A. glycines, we sequenced a fragment of one protein-coding gene, the cytochrome c oxidase I/of mitochondrial DNA gene. The results showed that four haplotypes were defined among CO 11 gene of 180 sequences of soybean aphid in Northeast China including H1 shared by all the populations. Lower haplotype diversity (Hd=0.3590± 0.0420) and nucleotide diversity (Pi=0.0012±0.0002) were observed and high gene flow was detected in every two populations, while most of the variation (80.81%) arose from variability within A. glycines from individuals. Low genetic differentiation and high gene flow (Nm=2.106) indicated a high migration rate between the populations, which might reveal that gene flow in different geographic populations did not affect by geographical distance. The phylogenetic tree and the haplotype network ofA. glycines were obtained based on sequences of CO Ⅱ gene, there were no significant genealogical branches or clusters recognized in NJ tree, and no clear distribution, delineation of haplotypes were demonstrated in the haplotype network according to geographical location. This study rejected the vicariance hypothesis: geographic isolation could be a barrier and it restricted A. glycines gene flow among 10 populations.展开更多

The migration mode transition of cancer cell enhances its invasive capability and the drug resistance,where physical confinement of cell microenvironment has been revealed to induce the mesenchymal-amoeboid transition...The migration mode transition of cancer cell enhances its invasive capability and the drug resistance,where physical confinement of cell microenvironment has been revealed to induce the mesenchymal-amoeboid transition(MAT).However,most existing studies are performed in PDMS microchannels,of which the stiffness is much higher than that of most mammalian tissues.Therefore,the amoeboid migration transition observed in these studies is actually induced by the synergistic effect of matrix stiffness and confinement.Since the stiffness of cell microenvironment has been reported to influence the cell migration in 2D substrate,the decoupling of stiffness and confinement effects is thus in need for elucidating the underlying mechanism of MAT.However,it is technically challenging to construct microchannels with physiologically relevant stiffness and channel size,where existing microchannel platforms with physiological relevance stiffness are all with>10μm channel width.Such size is too wide to mimic the physical confinement that migrating cancer cells confront in vivo,and also larger than the width of PDMS channel,in which the MAT of cancer cell was observed.Therefore,an in vitro cell migration platform,which could mimic both stiffness and confinement of the native physical microenvironment during cancer metastasis,could profoundly contribute to researches on cancer cell migration and cellular mechanotransduction.In this paper,we overcome the limitations of engineering soft materials in microscale by combining the collagen-alginate hydrogel with photolithography.This enables us to improve the accuracy of molded microchannel,and thus successfully construct a 3D microchannel platform,which matches the stiffness and width ranges of native environmental confinement that migrating cancer cells confront in vivo.The stiffness(0.3~20 kPa),confinement(channel width:3.5~14μm)and the adhesion ligand density of the microchannel can be tuned independently.Interestingly,using this platform,we observed that the migration speed of cancer cell is influenced by the synergistic effect of channel stiffness and width,and the increasing stiffness reverses the effect of channel width on the migration speed of cancer cells.In addition,MAT has a strong correlation with the channel stiffness.These findings make us reconsider the widely accepted hypothesis:physical confinement can induce MAT.Actually,this transition can only occur in stiff confined microenvironment not in soft one.For soft microchannels,the compliance of the channel walls could cause little cell/nucleus deformation,and the MAT could not be induced.To further investigate the mechanism of MAT,we developed a computational model to simulate the effect of nucleus deformation on MAT.With the model,we found that deforming the cell nuclear by decreasing the nucleus stiffness will reduce the cellmigration speed.This implies that nuclear stiffness plays an important role in the regulation of cancer migration speed and thus MAT in microchannels.The effect of channel stiffness on MAT and migration speed as observed in our experiment could partially explain previous findings reported in the literature,where the increasing matrix stiffness of tumor microenvironment promotes cancer metastasis.Our observations thus highlight the critical role of cell nuclear deformation not only in MAT,but also in regulating cellular mechanotransduction and cell-ECM interactions.This developed platform is capable of mimicking the native physical microenvironment during metastasis,providing a powerful tool for high-throughput screening applications and investigating the interaction between cancer migration and biophysical microenvironment.展开更多

Based on the basic theory of disaggregate model and its modelling method, nine grades were selected as the alternatives of trip time, the variables affecting time choice and the method getting their values were determined, and a multinomial logit (MNL) model was developed. Another 1 200 trip data of rural population were selected to testify the model's validity. The result shows that the maximum absolute error of each period between calculated value and statistic is 3.6%, so MNL model has high calculation accuracy.展开更多
Background: Gossypium arboreum is a diploid species cultivated in the Old World. It possesses favorable characters that are valuable for developing superior cotton cultivars.Method: A set of 197 Gossypium arboreum acc...Background: Gossypium arboreum is a diploid species cultivated in the Old World. It possesses favorable characters that are valuable for developing superior cotton cultivars.Method: A set of 197 Gossypium arboreum accessions were genotyped using 80 genome-wide SSR markers to establish patterns of the genetic diversity and population structure. These accessions were collected from three major G. arboreum growing areas in China. A total of 255 alleles across 80 markers were identified in the genetic diversity analysis.Results: Three subgroups were found using the population structure analysis, corresponding to the Yangtze River Valley, North China, and Southwest China zones of G.arboreum growing areas in China. Average genetic distance and Polymorphic information content value of G. arboreum population were 0.34 and 0.47, respectively, indicating high genetic diversity in the G. arboreum germplasm pool. The Phylogenetic analysis results concurred with the subgroups identified by Structure analysis with a few exceptions. Variations among and within three groups were observed to be 13.61% and 86.39%, respectively.Conclusion: The information regarding genetic diversity and population structure from this study is useful for genetic and genomic analysis and systematic utilization of economically important traits in G. arboreum.展开更多
The purpose of this study was to explore the effects of recombinant human intestinal alkaline phosphatase(recIAP) on human neutrophils in vitro, and the migration, phagocytosis, apoptosis in presence and absence of LP...The purpose of this study was to explore the effects of recombinant human intestinal alkaline phosphatase(recIAP) on human neutrophils in vitro, and the migration, phagocytosis, apoptosis in presence and absence of LPS. In this study, freshly extracted human neutrophils were used to establish an inflammatory cell model, and the control group, recIAP group, LPS group and recIAP +LPS group were set up to stimulate the model. The migration of neutrophils was detected by agarose gel drop method. Fluorescent particles and fluorescent probes were added to different treatment groups, and the phagocytic rate of neutrophils and the release of reactive oxygen species(ROS) from neutrophils were detected by flow cytometry. The apoptosis rate of neutrophils was detected by flow cytometry according to Annexin V-FITC apoptosis detection kit. The results showed that regardless of the presence or absence of LPS, recIAP could inhibit the migration of neutrophils, phagocytosis and the release of ROS. In addition, recIAP could weaken the inhibitory effect of LPS on neutrophils apoptosis.展开更多
Background:Genetic improvement in fiber quality is one of the main challenges for cotton breeders.Quantitative trait loci(QTL)mapping provides a powerful approach to dissect the molecular mechanism in fiber quality tr...Background:Genetic improvement in fiber quality is one of the main challenges for cotton breeders.Quantitative trait loci(QTL)mapping provides a powerful approach to dissect the molecular mechanism in fiber quality traits.In present study,F14 recombinant inbred line(RIL)population was backcrossed to paternal parent for a paternal backcross(BC/P)population,deriving from one upland cotton hybrid.Three repetitive BC/P field trials and one maternal backcross(BC/M)field trial were performed including both two BC populations and the original RIL population.Results:In total,24 novel QTLs are detected for fiber quality traits and among which 13 QTLs validated previous results.Thirty-five QTLs in BC/P populations explain 5.01%–22.09%of phenotype variation(PV).Among the 35 QTLs,23 QTLs are detected in BC/P population alone.Present study provides novel alleles of male parent for fiber quality traits with positive genetic effects.Particularly,qFS-Chr3–1 explains 22.09%of PV in BC/P population,which increaseds 0.48 cN·tex−1 for fiber strength.A total of 7,2,8,2 and 6 QTLs explain over 10.00%of PV for fiber length,fiber uniformity,fiber strength,fiber elongation and fiber micronaire,respectively.In RIL population,six common QTLs are detected in more than one environment:qFL-Chr1–2,qFS-Chr5–1,qFS-Chr9–1,qFS-Chr21–1,qFM-Chr9–1 and qFM-Chr9–2.Two common QTLs of qFE-Chr2–2(TMB2386-SWU12343)and qFM-Chr9–1(NAU2873-CGR6771)explain 22.42%and 21.91%of PV.The region between NAU4034 and TMB1296 harbor 30 genes(379 kb)in A05 and 42 genes(49 kb)in D05 for fiber length along the QTL qFL-Chr5–1 in BC/P population,respectively.In addition,a total of 142 and 46 epistatic QTLs and QTL×environments(E-QTLs and QQEs)are identified in recombinant inbred lines in paternal backcross(RIL-P)and paternal backcross(BC/P)populations,respectively.Conclusions:The present studies provide informative basis for improving cotton fiber quality in different populations.展开更多
文摘Objective:Ovarian cancer(OC)ranks among the leading causes of mortality among the female cancers worldwide.Numerous studies have explored the development and progression of OC at multiple genetic regulatory levels.However,relatively few studies have explored the impact of post-translational modifications(PTM)on OC progression,which is essential for uncovering new therapeutic targets.This study aimed to systematically identify the key PTM types involved in OCprogression,and to explore and evaluate their translational potential as therapeutic targets.Methods:First,we utilized multiple general PTM antibodies to compare gross PTM levels between normal ovarian and OC tissues from clinical females.After identifying lactylation as the PTM with the most significant differences,we selected representative samples for label-free mass spectrometry to identify specific lactylation sites.Next,we transfected A2780(OC)cells with either wild-type(WT)or mutant(K192A[Q])poly(ADP-ribose)polymerase 1(PARP1)conjugated to enhanced green fluorescent protein(EGFP)with a StrepⅡpeptide tag and assessed various cellular indexes related to cell proliferation(clonogenicity assay),migration(scratch wound healing assay),and reactive oxygen species levels.Results:Pan-lactylation was significantly upregulated in clinical OC samples,with PARP1 lactylation at K192 being one of the most common modifications.The growth and migration of A2780 cells were markedly suppressed by overexpressing PARP1-WT but not mutant PARP1.Overexpressing PARP1 significantly downregulated the phosphorylation of extracellular signal-regulated kinases 1/2(ERK1/2).Conclusion:This study uncovered a novel PTM of PARP1 in OC,lactylation,and demonstrated that lactylation at K192 is crucial in regulating OC cell growth and migration via the ERK1/2 pathway.Further investigations are required to elucidate the broader functional implications of PARP1 lactylation and its therapeutic potential.

文摘In this paper,we investigate the periodic traveling wave solutions problem for a single population model with advection and distributed delay.By the bifurcation analysis method,we can obtain periodic traveling wave solutions for this model under the influence of advection term and distributed delay.The obtained results indicate that weak kernel and strong kernel can both deduce the existence of periodic traveling wave solutions.Finally,we apply the main results in this paper to Logistic model and Nicholson’s blowflies model.

文摘Background Cotton crop is infested by numerous arthropod pests from sowing to harvesting,causing substantial direct and indirect yield losses.Knowledge of seasonal population trends and the relative occurrence of pests and their natural enemies is required to minimize the pest population and yield losses.In the current study,analysis of the seasonal population trend of pests and natural enemies and their relative occurrence on cultivars of three cotton species in Central India has been carried out.Results A higher number and diversity of sucking pests were observed during the vegetative cotton growth stage(60 days after sowing),declining as the crop matured.With the exception of cotton jassid(Amrasca biguttula biguttula Ishida),which caused significant crop damage mainly from August to September;populations of other sucking insects seldom reached economic threshold levels(ETL)throughout the studied period.The bollworm complex populations were minimal,except for the pink bollworm(Pectinophora gossypiella Saunders),which re-emerged as a menace to cotton crops during the cotton cropping season 2017–2018 due to resistance development against Bt-cotton.A reasonably good number of predatory arthropods,including coccinellids,lacewings,and spiders,were found actively preying on the arthropod pest complex of the cotton crop during the early vegetative growth stage.Linear regression indicates a significant relationship between green boll infestations and pink bollworm moths in pheromone traps.Multiple linear regression analyse showed mean weekly weather at one-or two-week lag periods had a significant impact on sucking pest population(cotton aphid,cotton jassid,cotton whitefly,and onion thrips)fluctuation.Gossypium hirsutum cultivars RCH 2 and DCH 32,and G.barbadense cultivar Suvin were found susceptible to cotton jassid and onion thrips.Phule Dhanvantary,an G.arboreum cotton cultivar,demonstrated the highest tolerance among all evaluated cultivars against all sucking pests.Conclusion These findings have important implications for pest management in cotton crops.Susceptible cultivars warrant more attention for plant protection measures,making them more input-intensive.The choice of appropriate cultivars can help minimize input costs,thereby increasing net returns for cotton farmers.

文摘During the storage of composite propellants, the migration of plasticizers and other unbonded additives at the interfaces of liner adhesives has garnered significant attention in understanding liner failure mechanisms, aging processes, and safety performance. However, there is currently no non-destructive and quantitative detection method for migration of plasticizers in propellant liner. In this study, we developed a HTPB sensing liner by incorporating conductive fillers-namely carbon black(CB), carbon nanotubes(CNTs), and graphene nanoplatelets(GNP)-into the HTPB matrix. The synergistic interaction between CNTs and GNP facilitates the formation of a tunneling conductive network that imparts electrical conductivity to the HTPB liner. To elucidate the functional relationship between conductivity and nitroglycerin(NG) migration, we applied the HTPB sensing liner onto double base propellant surfaces and measured both the conductivity of the sensing layer and NG migration during a 71°C accelerated aging experiment. The results shows that when CNTs/GNP content reaches 3wt%, there is an exponential correlation between conductivity and NG migration with a fitting degree of 0.9652;the average response sensitivity of ΔR/R0 relative to NG migration is calculated as 41.69, with an average deviation of merely5.67% between NG migrations derived from conductivity fittings compared to those obtained via TGA testing results. Overall, this sensing liner exhibits excellent capabilities for detecting NG migration nondestructively and quantitatively while offering a novel approach for assessing interfacial component migrations as well as debonding defects in propellants-a promising avenue for future self-monitoring strategies regarding propellant integrity.

文摘Background Cotton is an important cash crop in China and a key component of the global textile market.Verticil-lium wilt is a major factor affecting cotton yield.Single nucleotide polymorphism(SNP)markers and phenotypic data can be used to identify genetic markers and loci associated with cotton resistance to Verticillium wilt.We used eight upland cotton parent materials in this study to construct a multiparent advanced generation inter-cross(MAGIC)population comprising 320 lines.The Verticillium wilt resistance of the MAGIC population was identified in the green-house in 2019,and the average relative disease index(ARDI)was calculated.A genome-wide association study(GWAS)was performed to discover SNP markers/genes associated with Verticillium wilt resistance.Results ARDI of the MAGIC population showed wide variation,ranging from 16.7 to 79.4 across three replicates.This variation reflected a diverse range of resistance to Verticillium wilt within the population.Analysis of distribution pat-terns across the environments revealed consistent trends,with coefficients of variation between 12.25%and 21.96%.Families with higher ARDI values,indicating stronger resistance,were more common,likely due to genetic diver-sity and environmental factors.Population structure analysis divided the MAGIC population into three subgroups,with Group I showing higher genetic variation and Groups II and III displaying more uniform resistance performance.Principal component analysis(PCA)confirmed these divisions,highlighting the genetic diversity underlying Verticil-lium wilt resistance.Through GWAS,we identified 19 SNPs significantly associated with Verticillium wilt resistance,distributed across three chromosomes.The screening of candidate genes was performed on the transcriptome derived from resistant and susceptible cultivars,combined with gene annotation and tissue expression patterns,and two key candidate genes,Ghir_A01G006660 and Ghir_A02G008980,were found to be potentially associated with Verticillium wilt resistance.This suggests that these two candidate genes may play an important role in responding to Verticillium wilt.Conclusion This study aims to dissect the genetic basis of Verticillium wilt resistance in cotton by using a MAGIC population and GWAS.The study seeks to provide valuable genetic resources for marker-assisted breeding and enhance the understanding of resistance mechanisms to improve cotton resilience against Verticillium wilt.

文摘The detection of hypersonic targets usually confronts range migration(RM)issue before coherent integration(CI).The traditional methods aiming at correcting RM to obtain CI mainly considers the narrow-band radar condition.However,with the increasing requirement of far-range detection,the time bandwidth product,which is corresponding to radar’s mean power,should be promoted in actual application.Thus,the echo signal generates the scale effect(SE)at large time bandwidth product situation,influencing the intra and inter pulse integration performance.To eliminate SE and correct RM,this paper proposes an effective algorithm,i.e.,scaled location rotation transform(ScLRT).The ScLRT can remove SE to obtain the matching pulse compression(PC)as well as correct RM to complete CI via the location rotation transform,being implemented by seeking the actual rotation angle.Compared to the traditional coherent detection algorithms,Sc LRT can address the SE problem to achieve better detection/estimation capabilities.At last,this paper gives several simulations to assess the viability of ScLRT.

Taking the activity migrations of Wangshun Mountain piedmont fault toward Lishan piedmont fault and Weinan platform front fault, Dabaopi-Niujiaojian fault toward Shenyusi-Xiaojiazhai fault, among a serial of NE-trending faults from Baoji city to Jingyang County as examples, their migration time and process are analyzed and discussed in the present paper. It is useful for further understanding the structure development and physiognomy evolution history of Weihe basin.
文摘Based on arbitrarily wide-angle wave equations,a reverse-time propagation scheme is developed by substituting the partial derivatives of depth and time with central differences. The partial derivative of horizontal direction is replaced with high order difference. The imaging condition is computed by solving the eikonal equations. On the basis of above techniques,a prestack reverse-time depth migration algorithm is developed. The processing exam-ples of synthetic data show that the method can remove unwanted internal reflections and decrease the migration noise. The method also has the advantage of fidelity and is applicable of dip angle reflector imaging.

文摘OBJECTIVE To investigate the role and mechanism of G protein-coupled receptor kinase 2(GRK2)involving in hepatocel ular carcinoma(HCC)progression.METHODS Cel Counting Kit 8 and tumor colony formation assay were designed to detect HCC cell proliferation,wound healing assay was to detect HCC migration.The correlation between GRK2 and early growth response-1(EGR1)were detected by RT-PCR and real-time PCR assays.Co-immunoprecipitation and Western blot assay were adopted to detect the relationship between GRK2and insulin-like growth factor 1 receptor(IGF-1R)signaling pathway.RESULTS In this study we find that GRK2plays an inhibition role in IGF1-induced HCC cell proliferation and migration.Overexpression of GRK2 causes a decrease in EGR1 expression,while knockdown of GRK2 leads to the dramatically increase in EGR1 expression in the treatment of IGF1.Through co-immunoprecipitation and Western blot assay,we confirm that GRK2can interact with IGF-1R and inhibiting IGF1-induced activation of IGF1R signaling pathway.Silencing EGR1attenuates GRK2 overexpression-caused inhibition of cell proliferation,tumor colony number and migrationactivity,while overexpressing of EGR1 restores the antiproliferative and migratory effect by GRK2 overexpression in HCCLM3 cells.CONCLUSION Taken together,these results suggest that GRK2 may inhibit IGF1-induced HCC cell growth and migration through down-regulation of EGR1 and indicate that enforced GRK2 may offer a potential therapeutic approach against HCC.

In the past, the research on the convergence of GAs was based on Holland's model theorem. The diversity of the evolutionary population and the convergence of GAs are studied by using the concept of negentropy based on the discussion of the characteristic of GA. Some test functions are used to test the convergence of GAs, and good results have been obtained. It is shown that the global optimization may be obtained by selecting appropriate parameters of simple GAs if the evolution time is enough.

文摘This study was conducted to investigate the effect of Soybean Peptides(SPs) on ruminal fermentation and microbial diversity in vitro. Four levels of SPs supplements(0, 0.25%, 0.50% and 0.75% dry matter basis) were tested. p H, NH3-N, Neutral Detergent Fiber(NDF), Acid Detergent Fiber(ADF), Dry Matter Digestibility(DMD) and the Total Volatile Fatty Acid(TVFA) were measured at 6, 24, 48 h of in vitro mixed incubation the fluids. Microbial populations were determined at 24 h and Microbial Proteins(MCP) were determined at 24 and 48 h. The gas production after 48 h in vitro incubation presented linear growth(p<0.05) and the highest content was the level of 0.5% SPs(dry matter basis). NH3-N concentration reached the highest concentration with 0.75%(dry matter basis) at 48 h. p H linearly increased(linear, p<0.05) from 6 to 48 h. The digestion of DMD increased with increasing doses of SPs at 24 h. NDF and ADF linearly(p<0.01) decreased by adding SPs. The concentration of TVFA linearly increased(p<0.05) at 6, 24 and 48 h, and reached the highest concentration at the level of 0.75%(dry matter basis). SPs decreased(p<0.05) the molar proportion of acetate and propionate, respectively at 24 and 48 h. Acetat to Propionate ratio(A/P; linear, p<0.05) increased at 48 h, and reached the greatest value at the level of 0.75%(dry matter basis) at 48 h. The populations of rumen Ruminococcus flavefaciens(R. flavefaciens), Ruminococcusalbus(R. albus), Fibrobacter succinogenes(F. succinogenes), Butyrivibrio fibrisolvens(B. fibrisolvens), Streptococcus bovis(S.bovis), Ruminobacter amylophilus(R. amylophilus) and Succinimonas amylolytica(S. amylolytica) were analyzed based on the total rumen bacterial 16 S ribosomal deoxyribonucleic acid(r DNA). The relative abundance of R. flavefaciens and R. albus increased at 24 h, but the relative abundance of F. succinogenes decreased at this time. The incubation of SPs had no effect on the abundance of S. bovis and R. amylophilus. The relative abundance of B. fibrisolvens and S. amylolytica reached the greatest value(p<0.05) at middle doses of SPs inclusion at 24 h. The value of MCPs linearly increased(p<0.05) at 24 and 48 h. These results showed that SPs could improve in vitro fermentation and nutrient digestion of feed substrates, and had the ability to modulate the ruminal fermentation pattern by regulating the composition of functional rumen microbes. Hence, SPs might be a potential feed additive applied in the diets of ruminants.
文摘The interactions on gold active and migratory quantities and rates between tuffaceous slate and solu tions with different compositions were experimentally studied at 200 ℃, 20 MPa, in a high pressure apparatus. After reaction, tuffaceous slate became light colored and soft, and its mass density reduced. The amount of gold extracted from tuffaceous slate ranges widely, from 0 027 to 0 234 μg/g. Chlorine solution may activate appreciable amount of gold, and the gold migratory rate is high enough, from 50 70% to 92 30%, which reveals that sulphur and chlorine work together in solutions to accelerate gold activation and migration, and to realize gold mineralization in favorable places.
文摘Complex traits are multifactorial traits controlled by polygenic host factors.These trait-related phenotypic characteristics and performance including body weight,blood chemistry,immune cell profiles,as well host susceptibility to infectious and chronic diseases.In recent years,tremendous efforts were invested aiming to map the host genetic factors attribute to these traits and subsequently clone the gene/s underlying these loci.In parallel to human studies,a number of mouse models and approaches were developed aimed to enhance the mapping process and the gene cloning.These include of using resources such as F2,backcross,advanced intercross lines,outbred populations,consomic,congenic and recombinant inbred lines(RIL).The constraints of these approaches were the limited resolution mapping of genomic regions of the quantitative trait loci(QTL)associated with the trait of interests,and the limited genetic diversity observed in the parental founders.To overcome these limitations,a new genetically highly diverse recombinant inbred lines of mouse population was established,namely the Collaborative Cross(CC),created from full reciprocal mating of 8 divergent strains of mice:A/J,C57BL/6J,129S1/SvI mJ,NOD/LtJ,NZO/HiL tJ,CAST/Ei,PWK/PhJ,and WSB/EiJ.By intercrossing these eight founders to generate the different CC lines,the genetic makeup of the newly developed resource is completely different from the eight parental lines,and will show heterosis,which subsequently will response differently comparing with their original founders.Finally,our results suggest that it is not essential to defining the phenotypic response of the eight parental lines,prior of assessing the CC lines,because it is believed that genetic interaction of the new genetic makeup of the new lines will reveal new phenotypic response,which completely different from the parental lines.In this report,we present to the community the power of the CC for dissecting variety of complex traits including host susceptibility to infectious and chronic diseases as well body performance traits.Based on our results from a variety of studies,we recommend to the community,that the best strategy of using this population is to aim of phenotyping about 50 and more of CC lines,with limited number of biological replicates(3-4 mice per line),and subsequently using the publicly available high dense genotype information of the CC lines as well the sequence database of the eight founders,it will be possible performing QTL mapping to a unprecedented precision genomic regions less than 1 MB,subsequently lead to identify potential strong candidate genes.These achievements are believed cannot be obtained with any other currently available mouse resource populations.

文摘Aphis glycines (Hemiptera: Aphididae) is considered as a cosmopolitan pest of cultivated soybean, major difficulties in its control measures may be due to its higher genetic diversity; however, the knowledge about population genetic diversity of this species is limited. This study aimed to represent the genetic differentiation among different geographic populations of soybean aphid in Northeast China. In order to investigate and assess the genetic diversity, genetic differentiation, molecular variance, population structure, ecological importance and evolutionary history of A. glycines, we sequenced a fragment of one protein-coding gene, the cytochrome c oxidase I/of mitochondrial DNA gene. The results showed that four haplotypes were defined among CO 11 gene of 180 sequences of soybean aphid in Northeast China including H1 shared by all the populations. Lower haplotype diversity (Hd=0.3590± 0.0420) and nucleotide diversity (Pi=0.0012±0.0002) were observed and high gene flow was detected in every two populations, while most of the variation (80.81%) arose from variability within A. glycines from individuals. Low genetic differentiation and high gene flow (Nm=2.106) indicated a high migration rate between the populations, which might reveal that gene flow in different geographic populations did not affect by geographical distance. The phylogenetic tree and the haplotype network ofA. glycines were obtained based on sequences of CO Ⅱ gene, there were no significant genealogical branches or clusters recognized in NJ tree, and no clear distribution, delineation of haplotypes were demonstrated in the haplotype network according to geographical location. 文摘Three observation methods were used to investigate the existing form and the behavior of rare earth during the sintering process of high activity mischmetal (RE, with lanthanum and cerium) doped WC-8%Co-0.048%RE(mass fraction) alloy with low carbon-containing level by scanning electron microscopy (SEM) and energy dispersive X-ray spectroscopy (EDXS), considering the fact that the addition amount of rare earth in the alloy is very minute. The directional migration process and mechanism of cerium were discussed. First, the sinter skin (surface) is observed. oxide on the sinter skin, and lanthanum in these cerium observed, and lanthanum containing phase/micro-zone in It is shown that there exists a dispersedly distributed cerium containing enrichment positions is very minute. Secondly, the polished section is the alloy is identified. Finally, based on the fact that the fracture of cemented carbide is resulted from the heterogeneous phase or other defects within the microstructure, the fracture surface is observed and cerium containing phase/micro-zone in the fracture source approximately 260 μm from the surface is identified. These combined observations reveal adequately the fact that lanthanum and cerium get separated and cerium predominantly migrates towards the surface during the sintering process.

文摘The migration mode transition of cancer cell enhances its invasive capability and the drug resistance,where physical confinement of cell microenvironment has been revealed to induce the mesenchymal-amoeboid transition(MAT).However,most existing studies are performed in PDMS microchannels,of which the stiffness is much higher than that of most mammalian tissues.Therefore,the amoeboid migration transition observed in these studies is actually induced by the synergistic effect of matrix stiffness and confinement.Since the stiffness of cell microenvironment has been reported to influence the cell migration in 2D substrate,the decoupling of stiffness and confinement effects is thus in need for elucidating the underlying mechanism of MAT.However,it is technically challenging to construct microchannels with physiologically relevant stiffness and channel size,where existing microchannel platforms with physiological relevance stiffness are all with>10μm channel width.Such size is too wide to mimic the physical confinement that migrating cancer cells confront in vivo,and also larger than the width of PDMS channel,in which the MAT of cancer cell was observed.Therefore,an in vitro cell migration platform,which could mimic both stiffness and confinement of the native physical microenvironment during cancer metastasis,could profoundly contribute to researches on cancer cell migration and cellular mechanotransduction.In this paper,we overcome the limitations of engineering soft materials in microscale by combining the collagen-alginate hydrogel with photolithography.This enables us to improve the accuracy of molded microchannel,and thus successfully construct a 3D microchannel platform,which matches the stiffness and width ranges of native environmental confinement that migrating cancer cells confront in vivo.The stiffness(0.3~20 kPa),confinement(channel width:3.5~14μm)and the adhesion ligand density of the microchannel can be tuned independently.Interestingly,using this platform,we observed that the migration speed of cancer cell is influenced by the synergistic effect of channel stiffness and width,and the increasing stiffness reverses the effect of channel width on the migration speed of cancer cells.In addition,MAT has a strong correlation with the channel stiffness.These findings make us reconsider the widely accepted hypothesis:physical confinement can induce MAT.Actually,this transition can only occur in stiff confined microenvironment not in soft one.For soft microchannels,the compliance of the channel walls could cause little cell/nucleus deformation,and the MAT could not be induced.To further investigate the mechanism of MAT,we developed a computational model to simulate the effect of nucleus deformation on MAT.With the model,we found that deforming the cell nuclear by decreasing the nucleus stiffness will reduce the cellmigration speed.This implies that nuclear stiffness plays an important role in the regulation of cancer migration speed and thus MAT in microchannels.The effect of channel stiffness on MAT and migration speed as observed in our experiment could partially explain previous findings reported in the literature,where the increasing matrix stiffness of tumor microenvironment promotes cancer metastasis.Our observations thus highlight the critical role of cell nuclear deformation not only in MAT,but also in regulating cellular mechanotransduction and cell-ECM interactions.This developed platform is capable of mimicking the native physical microenvironment during metastasis,providing a powerful tool for high-throughput screening applications and investigating the interaction between cancer migration and biophysical microenvironment.

文摘Referring to the 1 248 survey data of rural population in 14 provinces of China, the influencing factors of trip time choice were analyzed. Based on the basic theory of disaggregate model and its modelling method, nine grades were selected as the alternatives of trip time, the variables affecting time choice and the method getting their values were determined, and a multinomial logit (MNL) model was developed. Another 1 200 trip data of rural population were selected to testify the model's validity. The result shows that the maximum absolute error of each period between calculated value and statistic is 3.6%, so MNL model has high calculation accuracy.

文摘Background: Gossypium arboreum is a diploid species cultivated in the Old World. It possesses favorable characters that are valuable for developing superior cotton cultivars.Method: A set of 197 Gossypium arboreum accessions were genotyped using 80 genome-wide SSR markers to establish patterns of the genetic diversity and population structure. These accessions were collected from three major G. arboreum growing areas in China. A total of 255 alleles across 80 markers were identified in the genetic diversity analysis.Results: Three subgroups were found using the population structure analysis, corresponding to the Yangtze River Valley, North China, and Southwest China zones of G.arboreum growing areas in China. Average genetic distance and Polymorphic information content value of G. arboreum population were 0.34 and 0.47, respectively, indicating high genetic diversity in the G. arboreum germplasm pool. The Phylogenetic analysis results concurred with the subgroups identified by Structure analysis with a few exceptions. Variations among and within three groups were observed to be 13.61% and 86.39%, respectively.Conclusion: The information regarding genetic diversity and population structure from this study is useful for genetic and genomic analysis and systematic utilization of economically important traits in G. arboreum.

文摘The purpose of this study was to explore the effects of recombinant human intestinal alkaline phosphatase(recIAP) on human neutrophils in vitro, and the migration, phagocytosis, apoptosis in presence and absence of LPS. In this study, freshly extracted human neutrophils were used to establish an inflammatory cell model, and the control group, recIAP group, LPS group and recIAP +LPS group were set up to stimulate the model. The migration of neutrophils was detected by agarose gel drop method. Fluorescent particles and fluorescent probes were added to different treatment groups, and the phagocytic rate of neutrophils and the release of reactive oxygen species(ROS) from neutrophils were detected by flow cytometry. The apoptosis rate of neutrophils was detected by flow cytometry according to Annexin V-FITC apoptosis detection kit. The results showed that regardless of the presence or absence of LPS, recIAP could inhibit the migration of neutrophils, phagocytosis and the release of ROS. In addition, recIAP could weaken the inhibitory effect of LPS on neutrophils apoptosis.

文摘Background:Genetic improvement in fiber quality is one of the main challenges for cotton breeders.Quantitative trait loci(QTL)mapping provides a powerful approach to dissect the molecular mechanism in fiber quality traits.In present study,F14 recombinant inbred line(RIL)population was backcrossed to paternal parent for a paternal backcross(BC/P)population,deriving from one upland cotton hybrid.Three repetitive BC/P field trials and one maternal backcross(BC/M)field trial were performed including both two BC populations and the original RIL population.Results:In total,24 novel QTLs are detected for fiber quality traits and among which 13 QTLs validated previous results.Thirty-five QTLs in BC/P populations explain 5.01%–22.09%of phenotype variation(PV).Among the 35 QTLs,23 QTLs are detected in BC/P population alone.Present study provides novel alleles of male parent for fiber quality traits with positive genetic effects.Particularly,qFS-Chr3–1 explains 22.09%of PV in BC/P population,which increaseds 0.48 cN·tex−1 for fiber strength.A total of 7,2,8,2 and 6 QTLs explain over 10.00%of PV for fiber length,fiber uniformity,fiber strength,fiber elongation and fiber micronaire,respectively.In RIL population,six common QTLs are detected in more than one environment:qFL-Chr1–2,qFS-Chr5–1,qFS-Chr9–1,qFS-Chr21–1,qFM-Chr9–1 and qFM-Chr9–2.Two common QTLs of qFE-Chr2–2(TMB2386-SWU12343)and qFM-Chr9–1(NAU2873-CGR6771)explain 22.42%and 21.91%of PV.The region between NAU4034 and TMB1296 harbor 30 genes(379 kb)in A05 and 42 genes(49 kb)in D05 for fiber length along the QTL qFL-Chr5–1 in BC/P population,respectively.In addition,a total of 142 and 46 epistatic QTLs and QTL×environments(E-QTLs and QQEs)are identified in recombinant inbred lines in paternal backcross(RIL-P)and paternal backcross(BC/P)populations,respectively.Conclusions:The present studies provide informative basis for improving cotton fiber quality in different populations.
