“闹大”与“柔化”:民间环保组织的行动策略——以绿色潇湘为例 被引量:22

民间环保组织的行动使命是力求通过与地方政府建构"合作"关系来协助治理日渐严峻的环境问题。基于对"绿色潇湘"这一民间环保组织的研究发现,民间环保组织要获得与地方政府"合作"机会,进而配合地方政府对... 民间环保组织的行动使命是力求通过与地方政府建构"合作"关系来协助治理日渐严峻的环境问题。基于对"绿色潇湘"这一民间环保组织的研究发现,民间环保组织要获得与地方政府"合作"机会,进而配合地方政府对环境问题实施有效治理策略,一般会采取"闹大"与"柔化"相结合的行动策略。为了协助配合地方政府开展环境治理行动,民间环保组织基于新媒体平台开展"线上造势"、借助志愿者行动"线下做事"、契合政策导向"顺势而为"实施"闹大"策略。为了维持与地方政府的有效合作,民间环保组织秉持"站在政府对面而非对立面"的谋划合作、"唱白脸和唱红脸"的双面配合、"做阳谋而不做阴谋"的提出问题、"本地人解决本地问题"的处事方式的"柔化"策略。"柔化"是"闹大"的延续,是在有限的政治空间内寻找最大利益平衡点的行动策略转型,进而推动环境治理实践取得成效。 展开更多

For rechargeable aqueous zinc-ion batteries(ZIBs),the design of nanocomposites comprised of electrochemically active materials and carbon materials with novel structures has great prom-ise in addressing the issue of electrical conductivity and structural stability in the electrode materials during electrochemical cycling.We report the production of a novel flexible electrode material,by anchoring MnO_(2) nanosheets on a B,N co-doped carbon nanotube ar-ray(BNCNTs)grown on carbon cloth(BNCNTs@MnO_(2)),which was fabricated by in-situ pyrolysis and hydrothermal growth.The generated BNCNTs were strongly bonded to the surface of the car-bon fibers in the carbon cloth which provides both excellent elec-tron transport and ion diffusion,and improves the stability and dur-ability of the cathode.Importantly,the BNCNTs offer more active sites for the hydrothermal growth of MnO_(2),ensuring a uniform dis-tribution.Electrochemical tests show that BNCNTs@MnO_(2) delivers a high specific capacity of 310.7 mAh g^(−1) at 0.1 A g^(−1),along with excellent rate capability and outstanding cycling stability,with a 79.7% capacity retention after 8000 cycles at 3 A g^(−1). 展开更多

目的基于miR-135a/FOXO1/PINK1通路探讨柔肝化纤颗粒通过调控线粒体自噬来抑制肝星状细胞活化与增殖的影响及其机制。方法HSC-T6分为空白组、H_(2)O_(2)组、miR-135a-NC组、miR-135a-mimic组、miR-135a-in-hibitor-NC+H 2 O2组、miR-135... 目的基于miR-135a/FOXO1/PINK1通路探讨柔肝化纤颗粒通过调控线粒体自噬来抑制肝星状细胞活化与增殖的影响及其机制。方法HSC-T6分为空白组、H_(2)O_(2)组、miR-135a-NC组、miR-135a-mimic组、miR-135a-in-hibitor-NC+H 2 O2组、miR-135a-inhibitor+H_(2)O_(2)组、柔肝化纤颗粒含药血清对照组、H_(2)O_(2)+含药血清对照组、H_(2)O_(2)+含药血清组、H_(2)O_(2)+含药血清+miR-135a-NC组及H_(2)O_(2)+含药血清+miR-135a-mimic组。分别采用Real-time PCR、Western Blot、ELISA及流式细胞术检测miR-135a、FOXO1、PINK1、Parkin、LC3Ⅱ、Smad2、p-Smad2、转化生长因子-β1(transforming growth factor beta 1,TGF-β1)、NF-κB p65、p-NF-κB p65、α-SMA、Ⅰ型胶原、Ⅲ型胶原、TNF-α表达及线粒体膜电位、活性氧(reactive oxygen species,ROS)生成。结果给予H_(2)O_(2)及过表达miR-135a可显著上调HSC-T6 miR-135a、α-SMA、Ⅰ型胶原、Ⅲ型胶原、p-Smad2、TGF-β1、p-NF-κB p65、TNF-α表达及ROS生成(P<0.01);下调FOXO1、PINK1、Parkin、LC3Ⅱ表达与线粒体膜电位(P<0.01)。给予柔肝化纤颗粒及抑制miR-135a表达可显著下调HSC-T6 miR-135a、α-SMA、Ⅰ型胶原、Ⅲ型胶原、p-Smad2、TGF-β1、p-NF-κB p65、TNF-α表达及ROS生成(P<0.01);上调FOXO1、PINK1、Parkin、LC3Ⅱ表达与线粒体膜电位(P<0.01)。给予柔肝化纤颗粒同时过表达miR-135a可抑制柔肝化纤颗粒对HSC-T6的影响(P<0.01)。结论线粒体自噬可抑制HSC-T6活化,其机制与线粒体自噬抑制ROS生成,进而抑制TGF-β1/Smad2通路及炎症反应有关;柔肝化纤颗粒亦可抑制HSC-T6活化,其机制与柔肝化纤颗粒抑制miR-135a表达,活化FOXO1/PINK1通路,从而促进线粒体自噬有关。 展开更多

目的探讨柔肝化纤颗粒对大鼠骨髓间充质干细胞(BMSCs)归巢于肝脏的影响及治疗肝纤维化效果。方法选取SD雄性大鼠52只,分为空白对照组(10只)、模型组(42只),建模成功后将肝纤维化大鼠模型随机分为模型组、BMSCs组、柔肝化纤颗粒组、柔肝... 目的探讨柔肝化纤颗粒对大鼠骨髓间充质干细胞(BMSCs)归巢于肝脏的影响及治疗肝纤维化效果。方法选取SD雄性大鼠52只,分为空白对照组(10只)、模型组(42只),建模成功后将肝纤维化大鼠模型随机分为模型组、BMSCs组、柔肝化纤颗粒组、柔肝化纤颗粒+BMSCs组。干预4周后,分别采用qPCR、Western Blot检测各组肝组织AFP、ALB、CK18基因表达与SDF-1、CXCR4、HGF、EGF基因及蛋白表达、HE染色、Masson染色检测肝组织纤维化程度、PAS法检测肝脏储存糖原功能及冷冻切片检测携带绿色荧光蛋白的BMSCs向肝组织归巢情况。结果动物实验方面,与模型组相比,使用柔肝化纤颗粒联合BMSCs移植组肝脏组织AFP、ALB、CK18表达上调,HGF、EGF表达上调,肝脏内携带绿色荧光蛋白的BMSC数量明显增加,肝纤维化程度下降。结论柔肝化纤颗粒能动员BMSCs归巢于纤维化肝组织,并能够修复受损肝组织,其机制可能与SDF-1/CXCR4轴有关。 展开更多

Design parameter optimization of beam foundation on soft soil layer with nonlinear finite element 被引量:2

Finite element method was performed to investigate the influences of beam stiffness, foundation width and cushion thickness on the beating capacity of beam foundation on underlying weak laminated clay. The comparison between numerical results and results from field test including plate-bearing test and foundation settlement observation shows reasonable agreement. According to the numerical results, the beam width, length, cross section and cushion thickness were optimized. The results show that the stresses in subgrade soil decrease greatly with increasing the cushion thickness and width of foundation. However, the foundation settlement and influencing depth of displacement also increase correspondingly under conditions of relatively thinner cushion thickness. For the foundations on underlying weak layer, increasing foundation width merely might be inadequate for improving the bearing capacity, and the appropriate width and cushion thickness depend on the response of subgrade. A comparison between rigid and flexible beams was also discussed. The influence of a flexible beam foundation on subgrade is relatively smaller under the same loading conditions, and the flexible beam foundation appears more adaptable to various subgrades. The proposed flexible beam foundation was adopted in engineering. According to the calculation results, beam width of 2.4 m and cushion thickness of 0.8 m are proposed, and a flexible beam foundation is applied in the optimized design, which is confirmed reasonable by the actual engineering. 展开更多

To improve the computational efficiency of the reliability-based design optimization(RBDO) of flexible mechanism, particle swarm optimization-advanced extremum response surface method(PSO-AERSM) was proposed by integrating particle swarm optimization(PSO) algorithm and advanced extremum response surface method(AERSM). Firstly, the AERSM was developed and its mathematical model was established based on artificial neural network, and the PSO algorithm was investigated. And then the RBDO model of flexible mechanism was presented based on AERSM and PSO. Finally, regarding cross-sectional area as design variable, the reliability optimization of flexible mechanism was implemented subject to reliability degree and uncertainties based on the proposed approach. The optimization results show that the cross-section sizes obviously reduce by 22.96 mm^2 while keeping reliability degree. Through the comparison of methods, it is demonstrated that the AERSM holds high computational efficiency while keeping computational precision for the RBDO of flexible mechanism, and PSO algorithm minimizes the response of the objective function. The efforts of this work provide a useful sight for the reliability optimization of flexible mechanism, and enrich and develop the reliability theory as well. 展开更多
