目的:观察转化生长因子β1(transform ing growth factorβ1,TGF-β1)和骨形成蛋白2(bone morphogeneticprote in 2,BMP2)联合应用对体外培养的鼠牙乳头成牙本质细胞分化的影响。方法:取17 d胎龄小鼠下颌第一磨牙牙胚,胰蛋白酶消化分离...目的:观察转化生长因子β1(transform ing growth factorβ1,TGF-β1)和骨形成蛋白2(bone morphogeneticprote in 2,BMP2)联合应用对体外培养的鼠牙乳头成牙本质细胞分化的影响。方法:取17 d胎龄小鼠下颌第一磨牙牙胚,胰蛋白酶消化分离牙乳头,置半固态培养基培养6 d,半固态培养基中单独加入重组TGFβ1、BMP2,或分别与肝素联合,或TGFβ1和BMP2联合,组织学观察牙乳头的形态学变化。结果:TGF-β1或BMP2单独加入时未见细胞极化,但基质分泌增加。TGF-β1或BMP2加肝素可诱导牙乳头周边细胞发生极化,并分泌胞外基质。半固态培养基中同时加入TGF-β1和BMP2的牙乳头培养6 d后,牙乳头周边细胞出现极化和功能性分化,牙乳头周边胞外基质沉积明显,且可见牙乳头尖形态的维持及从牙乳头尖至牙乳头基底部成牙本质细胞分化梯度的维持。结论:本研究结果证实,在没有内釉上皮和完整的基底膜存在的情况下,TGF-β1和BMP2加肝素可诱导培养的牙乳头出现成牙本质细胞分化,并促进胞外基质的分泌。TGF-β1和BMP2均能诱导成牙本质细胞的细胞学分化和分泌功能,二者联合应用可协同发挥作用增强诱导效果。展开更多
