【目的】本研究对APSIM-Sugar模型在云南半湿润半干旱蔗区的适应性进行评价。【方法】从研究地区2个代表站点甘蔗大田试验获取调参验证数据集,基于标准品种Q117模型参数,采用试错法对8个关键品种参数进行优化和模拟精度评价。【结果】... 【目的】本研究对APSIM-Sugar模型在云南半湿润半干旱蔗区的适应性进行评价。【方法】从研究地区2个代表站点甘蔗大田试验获取调参验证数据集,基于标准品种Q117模型参数,采用试错法对8个关键品种参数进行优化和模拟精度评价。【结果】确定了Q208和ROC22两个甘蔗新品种的模型参数优化值,评估了模型在云南蔗区的产量糖分模拟误差范围。结果显示蔗糖产量实测值与模型模拟值显著相关,绝对误差小于2 t/hm^2,斜率在0. 8~1. 26,R^2大于0. 8。【结论】经过校验的APSIM-Sugar模型能较好模拟云南半湿润半干旱蔗区甘蔗的生长发育过程,并响应环境变化造成的产量和糖分差异,但是在多年宿根和极端环境条件下的模拟精度仍有待提高。模型品种参数和输入数据的不确定性是造成模型模拟结果不确定性的重要原因。 展开更多

The effect of microwave treatment on the tensile properties of treated sugar palm fibre with 6% NaOH reinforced thermoplastic polyurethane composites was investigated. Firstly, the sugar palm fibres were treated by 6% alkali solution. Then, microwave treatment was used to treat the alkali treated sugar palm fibres. Three types of temperatures(i.e. 70, 80 and 90℃) were applied in microwave treatment. The extruder and hot press machines were used to mixing the sugar palm fibres and polyurethane resin, and fabricate the composites. Tensile properties(i.e. tensile strength, tensile modulus and elongation at break) were studied by following the ASTM D-638 standard. The highest tensile strength was recorded 18.42 MPa with microwave temperature at 70℃ and 6% alkali pre-treatment. Therefore, the temperature 70℃ of microwave treatment may consider the best degree cent grate. 展开更多

Some characteristics of nitrate reductase from sugar beet leaves shown in this paper were as follows:The nitrate reductase from sugar beet leaves required NADH as an electron donor.Accordingly,the nitrate reductase was classified as NADH-dependent(E.C.1 .6.6.1).The Km value of the nitrate reductase for NADH and NO-3 were 0.86 m mol and 0.18μp mol respectively.The optimum pH in reaction mixture solution for nitrate reduction activity was 7.5.The effect of variable concentrations of inorganic phosphorus in the reaction buffer on nitratereductase activity was investigated.When the inorganic phosphorus concentration was below35 mmol,the nitrate reductase activity was increased with increase of inorganic phosphorus concentration.Conversely,when the inorganic phosphorus concentration was Over 35 mmol.the nitrate reductase activity was inhibited.The nitrate reductase activity assayed in vitro was 3.2 and 5.6 times of that assayed in vivo under the condition of exogenous and endogenous ground substance respectively. 展开更多

It is practical, economic and sometimes essential to derive rules or conclusions by performing lesser runs of experiments. In this part, a methodology based on 2 f factorial design was brought up to derive guidelines to simulate growth curve and production of sugars by Spirulina (Arthrospira) maxima . The growth curve or accumulation process of sugars was idealized by sets of straight lines limited by phase transfers of growth or accumulation of sugars. Normal analyses of the critical values of the transfers were used to derive their linear relationships with the initial conditions of the experimental factors. These linear functions were called guidelines and were used to simulate the growth curve or accumulation of sugars. Generalization of the guideline technique was determined by the kinetic limitation of nutrient nitrogen or sulfur that was dependent upon their stoichiometric deficiency directly derived from their initial values in the medium. This method uses the initial conditions of culture and does not need measurements of concentrations of nitrate, sulfate and pigments during cultivation. It is a practical and useful alternative way to trace and predict approximately the growth curve and production of sugars by S. maxima . 展开更多

With the advancement of agricultural mechanization,soil compaction has become a serious environmental problem.Soil compaction can increase soil bulk density and firmness,reduce porosity and permeability,and deteriorate soil structure,ultimately inhibit sugar beet growth and reduce both root yield and sugar content.However,few farmers recognize the link between soil compaction and these adverse effects.Soil compaction has a cumulative effect,with significant differences observed in the vertical range of compaction accumulation.The most significant soil compaction occurs in the topsoil of 0-10 cm,and the influence depth can reach 70 cm,but it is small in deep soil,and the inflection point is at a soil depth of 10 cm.The degree of soil compaction is related to soil type,water content,tractor shaft load,tyre type,tyre pressure and operation speed,etc.Therefore,in the production process of sugar beet,it is advisable to avoid high-humidity operations,use low pressure tyres,reduce the number of tractor-units passes over the farmland,and implement agricultural and agronomic measures to minimize soil compaction.These practices will help protect the soil environment and ensure sustainable production of sugar beets. 展开更多

The content of sugar is influenced by sucrose synthase (SS) activity in roots. The effects of nitrogen level in the aminonitrate ratio on SS activity of leaves and roots, roots yield and sugar content in sugar beet were studied in the field experiment by nutrient solution culture. The results showed that SS activity in leaves was lower than that in roots. With nitrogen level increasing, SS decomposition activity enhanced, and synthesis activity reduced. SS activity was regulated by different nitrogen forms and the ratio of NO3 and NH4^+. SS synthesis activity was enhanced as NH4^+ increasing when NO3 : NH4^+≥ 1, and it decreased as increasing NH4^+ when NO3 : NH4^+≤ 1, and it was the highest when NO3 : NH4^+=1. SS decomposition activity was enhanced as NO3- increasing. Sucrose content in root was lowed as nitrogen level increasing, but it was enhanced as NH4^+ increasing in the same nitrogen level. Root and sugar yield were the highest in the medium nitrogen level and NO3 : NH4^+=1. The result in field experiment corresponded with that in the nutrient fluid culture. It provides a basis for using reasonably nitrogen fertilizer in sugar beet production. 展开更多

The experiment of glutamate synthase activity (GOGATA) in both leaf blades and roots under different nitrogen levels was carried out at Northeast Agricultural University in 1993. The result showed that GOGATA rose rapidly to reach its peak from seedling stage to foliage rapid growth stage, and then declined gradually. GOGATA was enhanced with increasing nitrogen levels and had significant positive correlation with nitrogen levels at the middle stage of growth GOGATA in leaf blades was the strongest compared with crowns, petioles and roots, thus, it could represent the highest enzyme activity of the whole plant. GOGATA had quadratic curvilinear correlation with root yield and sugar production. GOGATA in leaf blades had significant positive correlation with α-NH 2-N at the foliage rapid growth stage while GOGATA in roots existed this relation at the latter stage of growth. GOGATA in roots had significant negative correlation with sugar content at harvest. 展开更多

This experiment used pumpkin Longyuanlixiang as material to research the effect of different pruning ways with one branch, two branches, and three branches on the sugar-decreasing matters during the ripeness of pumpkin (Cucurbita moschata) fruits. The results showed that pectin contents of pumpkin fruits during the ripeness all increased at first and then dropped in every treatment. The pectin contents under the treatments of one branch and two branches reached the high apex after pollinated for 30 d as well as for 35 d under the treatment of three branches. Pentose and chromium contents of fruits had the same trend under different pruning ways and increased during the ripenes. Three kinds of sugar-decreasing matters contents under treatment of two branches were bigger than treatment of one branch, while treatment of three branches was the lowest. 展开更多

The experiment of Glutamate Dehydrogenase (GDH) activity in various plant parts under different nitrogen levels in frame culture during the whole period of growth was carried out on campus of Northeast Agricltural University in 1993. The result showed that GDH activity in leaf blades under four nitrogen applied levels rose rapidly to the acme from the seedling to foliage rapid growth stage, then diminished rapidly to the lower level at the latter stage of foliage rapid growth. This level was kept to harvest. GDH activity in roots at each growth stage under all nitrogen levels exhibited little disparity and did not show ostensible regularity of changes. GDH activity in leaf blades was stimulated with nitrogen, however, it reduced with nitrogen fertilizer applying further. GDH activity in leaf blades was the biggest compared with crowns, petioles and roots, which suggested that it could represent the highest enzyme activityof the whole plant. 展开更多

The supernatant of the fermented broth was mixed with colloidal chitin and heated in water-bath at 37℃ for 30 min, then 3,5-dinitrosalicylic acid reagent (DNS) was added to the mixture, and let them react for 10 min in water-bath. 为明确甜菜夜蛾Spodoptera exigua短神经肽F(sNPF)基因对其取食行为、能量代谢及饥饿耐受性的调控作用,本研究克隆并分析了该基因功能。结果表明:甜菜夜蛾sNPF基因开放阅读框(ORF)为548 bp,编码182个氨基酸;其蛋白序列与鳞翅目夜蛾科其... 为明确甜菜夜蛾Spodoptera exigua短神经肽F(sNPF)基因对其取食行为、能量代谢及饥饿耐受性的调控作用,本研究克隆并分析了该基因功能。结果表明:甜菜夜蛾sNPF基因开放阅读框(ORF)为548 bp,编码182个氨基酸;其蛋白序列与鳞翅目夜蛾科其他物种sNPF蛋白氨基酸序列相似性达96.7%~100%,具有高度保守性。组织表达分析显示,sNPF在4龄幼虫期表达量最高,且在头部特异性高表达。通过RNA干扰技术(注射双链sNPF干扰RNA,dssNPF)研究发现:与对照(注射dsGFP)相比,干扰后24、48、72 h甜菜夜蛾4龄幼虫取食量分别显著降低13.15%、17.59%和20.13%;同期体重累计增长率分别显著下降10.14%、23.08%和34.59%。饥饿胁迫试验进一步揭示,干扰sNPF基因导致4龄幼虫48 h和72 h死亡率显著升高至54%和100%;同时,幼虫体内糖原含量显著减少37.53%,而海藻糖(升高66.42%)、海藻糖合成酶(升高24.26%)、胰岛素(升高53.59%)含量均显著上升。上述结果提示,sNPF基因通过调控取食行为、能量物质积累与代谢途径(如糖原消耗加速、海藻糖合成增强等),直接影响甜菜夜蛾的生长发育及饥饿耐受能力,在其生存适应过程中发挥关键作用。本研究为靶向sNPF基因开发新型甜菜夜蛾绿色防控技术提供了重要理论依据。 展开更多

【目的】克隆木薯糖转运蛋白MeSWEET10-8基因,并对其进行功能分析,为探究MeSWEET10-8基因的糖转运调控机制及其与植物病原菌互作功能提供理论参考。【方法】以木薯品种华南9号为材料,以接种木薯黄单胞菌(Xanthomonas axonopodis pv.mani... 【目的】克隆木薯糖转运蛋白MeSWEET10-8基因,并对其进行功能分析,为探究MeSWEET10-8基因的糖转运调控机制及其与植物病原菌互作功能提供理论参考。【方法】以木薯品种华南9号为材料,以接种木薯黄单胞菌(Xanthomonas axonopodis pv.manihotis,Xam)成熟叶片的cDNA为模板,PCR扩增MeSWEET10-8基因,对其进行生物信息学分析,并通过MEGA 11.0构建不同物种SWEET蛋白的系统发育树;结合亚细胞定位、糖转运试验、转录组测序(RNA-seq)分析基因功能;实时荧光定量PCR检测接种Xam对MeSWEET10-8基因表达量的影响。【结果】从木薯感病的成熟叶片中克隆获得MeSWEET10-8基因的编码区(CDS)序列,长度为840 bp,与Phytozome数据库的参考序列(登录号:Manes.14G047700)完全一致,其编码蛋白的相对分子质量为31.81 kD,理论等电点(pI)为8.18,不包含信号肽,具有2个保守的MtN3_Slv功能域和7个跨膜结构域,启动子区包含5种光响应元件(Box 4、G-box、GA-motif、GT1-motif、TCT-motif)、1种水杨酸(SA)响应元件(TCA-element)、1种脱落酸(ABA)响应元件(ABRE)、1种茉莉酸甲酯(MeJA)响应元件(TGACG-motif)、2种赤霉素(GA)响应元件(CGTCA-motif、GARE-motif)、3种生物和非生物胁迫响应元件(ARE、LTR、TC-rich repeats)。系统发育分析结果显示,MeSWEET10-8蛋白与拟南芥AtSWEET10亲缘关系最近,与感病蛋白OsSWEET13和OsSWEET14同属于CladeⅢ分支。亚细胞定位和糖转运试验证明MeSWEET10-8蛋白定位于细胞膜上,具有转运木糖的功能。RNA-seq结果显示,MeSWEET10-8基因在木薯茎部组织中表达水平较高,叶柄和根顶端分生组织次之,在叶和贮藏根中则不表达。实时荧光定量PCR检测结果显示,MeSWEET10-8基因受Xam诱导,其相对表达量在接种前期逐渐升高,在72 h时达最高值,之后开始下降。【结论】木薯MeSWEET10-8蛋白具有转运木糖的功能,在Xam的侵染下也被诱导表达,表明MeSWEET10-8蛋白参与植物与病原菌互作过程,且其可能对植物茎部发育起到重要的生理作用。 展开更多

为了实现对烟草花中主要活性成分(还原糖、总黄酮、绿原酸、芸香苷和西柏三烯二醇)的快速无损检测,以全国196份不同产地、不同品种的烟草花为研究对象,采用近红外光谱技术建立了5种活性成分的分析模型。通过筛选光谱预处理方法,优化谱... 为了实现对烟草花中主要活性成分(还原糖、总黄酮、绿原酸、芸香苷和西柏三烯二醇)的快速无损检测,以全国196份不同产地、不同品种的烟草花为研究对象,采用近红外光谱技术建立了5种活性成分的分析模型。通过筛选光谱预处理方法,优化谱区范围和主成分数,进一步提升了模型性能。利用内部交叉验证,得到了各成分的最优模型。结果表明:还原糖、总黄酮、绿原酸、芸香苷和西柏三烯二醇5种活性成分的近红外校正模型中,校正集相关系数(R_(c))为0.957~0.986,校正均方根误差(RMSEC)为0.025%~0.995%;交叉验证相关系数(R_(cv))为0.901~0.979,交叉验证均方根误差(RMSECV)为0.030%~1.350%。外部验证结果显示,5种活性成分模型的相对标准偏差(RSD)均小于10%,且t检验表明,近红外光谱法预测值与传统化学分析值之间无显著差异。这表明该方法具有较高的准确性和可靠性,能够快速无损地检测烟草花中5种主要活性成分,为烟草花的高效开发利用提供了技术支撑。 展开更多
