OBJECTIVE To explore the antipost-traumatic-stress-disorder(PTSD) effects and its probable mechanism of YQA14,a dopamine D3 receptor antagonist.METHODS Two PTSD animal models,the rat single prolonged stress(SPS) model...OBJECTIVE To explore the antipost-traumatic-stress-disorder(PTSD) effects and its probable mechanism of YQA14,a dopamine D3 receptor antagonist.METHODS Two PTSD animal models,the rat single prolonged stress(SPS) model and the mouse pre-shock model,were used in this experiment.In the SPS model,adult male Sprague-Dawley(SD) rats were randomly divided into control group,model group,positive group and YQA14 groups with different dosages.In the mouse pre-chock model,dopamine D3 receptor knockout(KO) and wild type(WT) mice were randomly divided into control group,model group,positive group and YQA14 group.After the establishment of animal models,the saline,sertraline(ig) and YQA14(ip)were administered to the animals in the control,model,positive control and test groups respectively.The open field test(OFT) was used to evaluate the locomotor activity while the contextual freezing(CF) measurement and elevated plus maze(EPM) test were used to evaluate the PTSD-like behaviors.RESULTS In the rat SPS model,neither SPS nor drug treatment affected the locomotor activity in rats.However,SPS rats showed significant PTSD-like behaviors with enhanced freezing time in CF(P<0.01) and decreased percentage of entries into open arms and time spent in open arms in EPM(P<0.05,P<0.01).Moreover,compared with the model group,the repeated administration of YQA14(3.125,6.25 and 12.5 mg·kg-1)significantly reduced the freezing time(P<0.01)and increased the percentages of entries into open arms and time spent in open arms(P<0.05).In the mouse pre-shock model,when both model groups showed significant higher freezing time compared with the respective control groups(P<0.05,P<0.01),YQA14 selectively alleviated the freezing time on WT mice(P<0.05) while had no effect on KO mice.In the EPM tests,the WT mice model group showed a significant reduction in the percentage of entries into open arms and time spent in open arms(P<0.05) while D3 R KO mice model group didn′ t show any reduction,compared with respective control groups.Furthermore,daily administration of YQA14 at 12.5 mg·kg-1 both significantly reduced the percentages of entries into and time spent in open arms(P<0.05) but not D3 R KO mice.None of the locomotor activity were significantly affected.CONCLUSION YQA14 could significantly alleviate the PTSD-like behaviors in rodents and the effects were mediated by the blockade of brain D3 receptors.展开更多
OBJECTIVE Post-traumatic stress disorder(PTSD)is characterized by poor adapta⁃tion to a traumatic experience and disturbances in fear memory regulation,and currently lacks effective medication.Cannabidiol(CBD)is the p...OBJECTIVE Post-traumatic stress disorder(PTSD)is characterized by poor adapta⁃tion to a traumatic experience and disturbances in fear memory regulation,and currently lacks effective medication.Cannabidiol(CBD)is the primary component of the Cannabis sativa plant;it does not have any psychoactive effects and has been implicated in modulating fear learning in mammals.The present study investigated the effect of CBD on PTSD-like behaviors in a mouse pre-shock model,the effect of CBD in the modulation of trauma-related fear memory,a crucial process leading to core symptoms of PTSD.METHODS Pre-shock model was applied in which mice were submitted to training with two days of 0.8 mA×12 times of foot-shock,and PTSD-like behaviors was evaluated during 3 and 26 d,including freezing time to the conditioned context,open field test,elevated plus maze test and social interaction test.RESULTS CBD(10 mg·kg^(-1))administration alleviated main PTSD-like symptoms in the mouse pre-shock model by attenuating trauma-related fear memory,decreasing anxiety-like behavior,and increasing social interaction behavior.However,sertraline(15 mg·kg^(-1))was only effective when adminis⁃tered throughout the test period.Furthermore,CBD reduced the formation,retrieval,and recon⁃solidation of trauma-related fear memory,whereas sertraline only reduced fear-memory retrieval.Neither CBD nor sertraline influenced the acquisi⁃tion of trauma-related fear memory.CONCLU⁃SION CBD produced anti-PTSD-like actions in mice,and could disrupt trauma-related fear mem⁃ory by interfering with multiple aspects of fear memory processing in mice.These findings indi⁃cate that CBD may be a promising candidate for treating PTSD.展开更多

目的:调查汶川地震后,极重灾区社区居民创伤后应激障碍发生率及影响因素,为进一步开展灾后心理卫生服务提供依据。方法:采用整群随机抽样的方法,从安县京安小区、什邡红白镇两个临时安置点社区居民2685人中,随机抽取820名社区居...目的:调查汶川地震后,极重灾区社区居民创伤后应激障碍发生率及影响因素,为进一步开展灾后心理卫生服务提供依据。方法:采用整群随机抽样的方法,从安县京安小区、什邡红白镇两个临时安置点社区居民2685人中,随机抽取820名社区居民登记人口学背景、受灾状况,并使用PTSD检查量表平民版(PTSD Checklist—Civilian Version,PCL—C)和领悟社会支持量表(Perceived Social Support Scale,PSSS)进行评估,依据DSM-Ⅳ中PTSD的诊断标准确诊PTSD患者。结果:在完成的820份调查评估资料中,785份PCL—C和PSSS问卷有效。男女比例为1:1.41;平均年龄(43±18)岁;26.0%患有躯体疾病,28.4%有地震前饮酒习惯,23.6%有震后饮酒行为。不同年龄居民PSSS总分及2因子分差异均无统计学意义(P〉0.05)。PTSD的临床检出率为12.4%(97/785),女性PTSD发生率高于男性(15.3%vs.8.3%,P=0.003),地震中有曾被掩埋经历者PTSD发生率高于无被掩埋经历者(26.3%vs.11.6%,P=0.018),有丧亲者的FFSD发生率高于无丧亲者(18.7%vs.11.1%,P=0.015)。以PTSD诊断是否成立为因变量,一般资料为自变量进行Logistic回归分析显示:女性(OR=2.070,95%CI:1.274—3.365)、有曾被掩埋经历者(OR=2.806,95%CI:1.249~6.306)、亲人丧失(OR=1.537,95%CI:1.087—2.173)及PSSS评分偏低(OR=0.960,95%CI:0.941~0.979)是PTSD发生的危险因素。结论:在灾后社区重建过程中,临时安置点社区居民存在较高的创伤后应激障碍的发生率,女性、有曾被掩埋经历、亲人丧失及个人感受到的社会支持偏低是PTSD发生的危险因素。展开更多
