【Objectives】Si and microbial application could relieve the crop replanting problems(CRPs).We further studied the change of key microorganisms that are related to the beneficial effects,aiming at provide reference fo...【Objectives】Si and microbial application could relieve the crop replanting problems(CRPs).We further studied the change of key microorganisms that are related to the beneficial effects,aiming at provide reference for the manufacture and application of both microbial agents and Si fertilizer in food lily production.【Methods】A field experiment was conducted over a three-year period,from March 2019 to March 2022.The experimental field had been continuously cultivated with lily for 9 years.Three treatments were established:silicon fertilizer(SF),microbial agents(“Special 8^(TM)”,MF),and combined application of silicon fertilizer and microbial agents(SMF).A control group with blank soil(CK)was also included.At seedling stage of Lanzhou lilies in 2020 and 2021,the shoot and bulb dry weight,and the plant height and stem diameter of Lanzhou lilies were investigated for calculation of seedling index.In July 2020,20 plants were selected in each plot,and root zone soils were sampled at a depth of 20 cm,10 cm away from the roots,and then mixed to form a composite sample.The soil available Si and organic matter content were analyzed,and the fungal community structure and some specific microbial groups in soils were determined with high-throughput sequencing of ITS.【Results】All the three treatments significantly enhanced the lily plant growth and the seedling index,compared to CK.Besides,SF and MF treatments increased the relative abundances(RA)and diversity of fungal communities,and altered the community structures.The RA of some specific groups were found to be significantly correlated with the seedling index and/or soil available Si.Of them,the RA of the genera Fusarium,Dactylonectria,Humicola,Stilbella,and the species Humicola_grisea showed a positive correlation,while that of the genera Mortierella,Stilbella,Holtermanniella,and the species Mortierella_fatshederae showed a negative correlation with seedling index.The genera Fusarium,Stilbella,the species Humicola_grisea,and Dactylonectria_estremocensis showed a positive correlation,while the genura Stilbella,and the species Mortierella fatshederae showed a negative correlation with available Si content.In the co-occurence network of top twenty fungal genera and top sixteen bacterial genera(RA>0.2%),Holtermanniella was the only genus that interacted with the bacteria and negatively correlated with bacterial genus Blastococcus.Holtermanniella was also the most densely connected genera,followed by the genus Fusarium,Didymella and Humicola.In addition,the genus Holtermanniella was the key species connecting fungal and bacterial community in soil.Fungal functional prediction revealed that SF,MF and SMF treatments decreased plant pathogens guilds and increased the beneficial guilds Ectomycorrhizal,plant saprophyte,leaf saprophyte,and arbuscular mycorrhizal compared to CK.【Conclusions】Combined application of silicon fertilizer and microbial agents can alleviate continuous replanting problems of Lanzhou lilies through restoring the fungal community diversity,and promoting plant residue depredation,thus reducing soil born disease incidence.The beneficial genus Humicola and its one species H.grisea acts as bioconversion,and the genus Acremonium acts as plant pathogen inhibitor.展开更多
The enhancement of chalcopyrite bioleaching with an enriched microbial community by acidified seawater was studied,and the enhancing mechanism was analyzed.The microbial community was enriched at the Dabaoshan mine si...The enhancement of chalcopyrite bioleaching with an enriched microbial community by acidified seawater was studied,and the enhancing mechanism was analyzed.The microbial community was enriched at the Dabaoshan mine site,and the treated ore sample had high concentrations of chalcopyrite and galena.The experimental results show that copper extraction from chalcopyrite with an enriched microbial community in seawater was promoted from 13.1%to 62.1%by acidification in comparison with that without acidification.Further analyses of the solutions,solid residues and microbial compositions by scanning electron microscopy,X-ray diffraction,Raman spectroscopy,Fourier transform infrared spectroscopy and 16 S rDNA sequencing revealed the promoting effects of acidified seawater.This acidification can increase the biodissolution of chalcopyrite to increase the concentration of iron ions and maintain the redox potential in the range of 360−410 mV.The latter produces an optimal redox environment conducive to chalcopyrite dissolution via Cu_(2)S.The adaptability of the microbial community to a high-salt environment is improved.Chloride ions at 580 mmol/L improve the leaching kinetics of chalcopyrite by increasing the porosity and noncrystallinity of the intermediate elemental sulfur.This study provides a promising way to bioleaching copper minerals using seawater for areas with freshwater shortages.展开更多
Microbial fuel cells have already been used as biosensors to monitor assimilable organic carbon(AOC).However,their signal production from AOC is known to be completely suppressed by dissoved oxygen(DO).In this study,t...Microbial fuel cells have already been used as biosensors to monitor assimilable organic carbon(AOC).However,their signal production from AOC is known to be completely suppressed by dissoved oxygen(DO).In this study,two identical microbial electrolysis cell(MEC)based biosensors were inoculated with marine sediment and operated at two different anodic potentials,namely-300 mV and+250 mV relative to Ag/AgCl.The MEC biosensor operated under positive anodic potential conditions had electrochemically active microbial communities on the anode,including members of the Shewanellaceae,Pseudoalteromonadaceae,and Clostridiaceae families.However,the strictly anaerobic members of the Desulfuromonadaceae,Desulfobulbaceae and Desulfobacteraceae families were found only in the negative anodic potential MEC biosensor.The positive anodic potential MEC biosensor showed several other advantages as well,such as faster start-up,significantly higher maximum current production,fivefold improvement in the AOC detection limit,and tolerance of low dissolved oxygen,compared to those obtained from the negative anodic potential MEC biosensor.The developed positive anodic potential MEC biosensor can thus be used as a real-time and inexpensive detector of AOC concentrations in high saline and low DO seawater.展开更多
MoS_(2)/CuS composite catalysts were successfully synthesized using a one-step hydrothermal method with sodium molybdate dihydrate,thiourea,oxalic acid,and copper nitrate trihydrate as raw materials.The hydrogen pro-d...MoS_(2)/CuS composite catalysts were successfully synthesized using a one-step hydrothermal method with sodium molybdate dihydrate,thiourea,oxalic acid,and copper nitrate trihydrate as raw materials.The hydrogen pro-duction performance of MoS_(2)/CuS prepared with different molar ratios of Mo to Cu precursors(n_(Mo)∶n_(Cu))as cathodic catalysts was investigated in the two-chamber microbial electrolytic cell(MEC).X-ray diffraction(XRD),X-ray pho-toelectron spectroscopy(XPS),scanning electron microscopy(SEM),transmission electron microscope(TEM),linear scanning voltammetry(LSV),electrochemical impedance analysis(EIS),and cyclic voltammetry(CV)were used to characterize the synthesized catalysts for testing and analyzing the hydrogen-producing performance.The results showed that the hydrogen evolution performance of MoS_(2)/CuS-20%(nMo∶nCu=5∶1)was better than that of platinum(Pt)mesh,and the hydrogen production rate of MoS_(2)/CuS-20%as a cathode in MEC was(0.2031±0.0237)m^(3)_(H_(2))·m^(-3)·d^(-1) for 72 h at an applied voltage of 0.8 V,which was slightly higher than that of Pt mesh of(0.1886±0.0134)m^(3)_(H_(2))·m^(-3)·d^(-1).The addition of a certain amount of CuS not only regulates the electron transfer ability of MoS_(2) but also increases the density of active sites.展开更多

The application of soil phosphorus activator was able to increase the quantity of bacteria and fungi in soil, but its effect on actinomycetes in soil was not significant. The application of microbial inoculants increased the urease and sucrase activities in soil over the growing season, but only at the maturing stage soil acid phosphatase activity was enhanced with the applying soil phosphorus activator. The application of soil phosphorus activator increased alkali-hydrolyzable nitrogen and available phosphorus contents in soil, but did not increase available potassium content in soil. The optimal microbial inoculant application rate as applied as soil phosphorus activator was 7.5 kg hm-2.展开更多

Also aimed to observe the correlation between soil microbial biomass and soil DNA. The study concluded that type of fertilizer might alter the soil microbial biomass and DNA contents. In soil treated with organic fertilizers resulted in higher concentrations of microbial biomass and DNA contents in summer as compared to spring dute to increase in temperature. Correspondingly, in case of inorganic fertilizer, concentrations of soil microbial biomass and DNA detected higher in summer instead of spring. The statistical correlation between soil microbial biomass, DNA and ODR in spring and summer along with organic and inorganic fertilizers were calculated highly significant (p〉0.01). This study demonstrated the impact of fertilizers and seasonal variations on soil microbial biomass and also revealed significant correlation between soil microbial biomass and soil DNA.展开更多

Wastewater was fed to the upflow ML-MFC to remove chemical oxygen demand (COD), phosphorus and nitrogen with simultaneous electricity generation. The effluent from the cathode compartment of the upflow ML-MFC was then continuously fed to an external photobioreactor for removing the remaining phosphorus and nitrogen using microalgae. Alone, the upflow ML-MFC produces a maximum power density of 481 mW/m 3 , and obtains 77.9% COD, 23.5% total phosphorus (TP) and 97.6% NH4+-N removals. When combined with the photobioreactor, the system achieves 99.3% TP and 99.0% NH4+-N total removal. These results show both the effectiveness and the potential application of the coupled system to continuously treat domestic wastewater and simultaneously generate electricity and biomass.展开更多

Four levels of SPs supplements(0, 0.25%, 0.50% and 0.75% dry matter basis) were tested. p H, NH3-N, Neutral Detergent Fiber(NDF), Acid Detergent Fiber(ADF), Dry Matter Digestibility(DMD) and the Total Volatile Fatty Acid(TVFA) were measured at 6, 24, 48 h of in vitro mixed incubation the fluids. Microbial populations were determined at 24 h and Microbial Proteins(MCP) were determined at 24 and 48 h. The gas production after 48 h in vitro incubation presented linear growth(p<0.05) and the highest content was the level of 0.5% SPs(dry matter basis). NH3-N concentration reached the highest concentration with 0.75%(dry matter basis) at 48 h. p H linearly increased(linear, p<0.05) from 6 to 48 h. The digestion of DMD increased with increasing doses of SPs at 24 h. NDF and ADF linearly(p<0.01) decreased by adding SPs. The concentration of TVFA linearly increased(p<0.05) at 6, 24 and 48 h, and reached the highest concentration at the level of 0.75%(dry matter basis). SPs decreased(p<0.05) the molar proportion of acetate and propionate, respectively at 24 and 48 h. Acetat to Propionate ratio(A/P; linear, p<0.05) increased at 48 h, and reached the greatest value at the level of 0.75%(dry matter basis) at 48 h. The populations of rumen Ruminococcus flavefaciens(R. flavefaciens), Ruminococcusalbus(R. albus), Fibrobacter succinogenes(F. succinogenes), Butyrivibrio fibrisolvens(B. fibrisolvens), Streptococcus bovis(S.bovis), Ruminobacter amylophilus(R. amylophilus) and Succinimonas amylolytica(S. amylolytica) were analyzed based on the total rumen bacterial 16 S ribosomal deoxyribonucleic acid(r DNA). The relative abundance of R. flavefaciens and R. albus increased at 24 h, but the relative abundance of F. succinogenes decreased at this time. The incubation of SPs had no effect on the abundance of S. bovis and R. amylophilus. The relative abundance of B. fibrisolvens and S. amylolytica reached the greatest value(p<0.05) at middle doses of SPs inclusion at 24 h. The value of MCPs linearly increased(p<0.05) at 24 and 48 h. These results showed that SPs could improve in vitro fermentation and nutrient digestion of feed substrates, and had the ability to modulate the ruminal fermentation pattern by regulating the composition of functional rumen microbes. Hence, SPs might be a potential feed additive applied in the diets of ruminants.展开更多

Microbial community diversities in the drainage from three mines(Dexing Copper Mine,Qibaoshan Copper Mine and Yaogangxian Tungsten Mine,China)were analyzed using 16S rDNA PCR-RFLP approach.The efficiencies of chalcopy...Microbial community diversities in the drainage from three mines(Dexing Copper Mine,Qibaoshan Copper Mine and Yaogangxian Tungsten Mine,China)were analyzed using 16S rDNA PCR-RFLP approach.The efficiencies of chalcopyrite bioleaching were compared using enrichment of the three cultures.Phylogenetic analysis indicates that the dominant microorganisms are clustered with the Proteobacteria,the remaining is affiliated with Nitrospira,Acidobacteria and Actinobacteria.At the genus level,Acidithiobacillus is the dominant group in both YTW and QBS samples,while Spingomonas is dominant in YGX sample.Moreover,the principal component analysis(PCA)reveals that QBS and YTW have similar geochemical character and microbial communities.The results also show that pH value and tungsten concentration play a key role in microbial community distribution and relative abundance.The bioleaching efficiency of the enrichment cultures from YTW and QBS is similar.After 15 d,the bioleaching rates of low grade chalcopyrite(0.99%)are both up to 99.5% when using 10 g/L pulp density due to the similar microbial composition of YTW and QBS.Moreover,the leaching efficiencies of enrichment cultures containing multiple bioleaching microorganisms are higher than that of pure culture Acidithiobacillus ferrooxidans.展开更多

The nitrogen removal rate(NRR) finally improved to 15.77 kg/m3/d by shortening hydraulic retention time(HRT) to 1.06 h. Well-shaped red anammox granules were extensively enriched inside the reactor. The results of nitrogen removal kinetics indicated that the present bioreactor has great nitrogen removal potential, because the maximum rate of substrate utilization(Umax) predicted by Stover-Kincannon model is suggested as 55.68 kg/(m3·d). Analysis of the microbial community showed that the anammox genus Candidatus Kuenenia dominated the bacterial communities. The relative abundance of Candidatus Kuenenia rose from 12.29% to 36.95% after progressively shorter HRT and higher influent substrate concentrations, illustrating the stability of nitrogen removal performance and biomass enrichment offered by the UASB in carrying out high-rate anammox process.展开更多

The single factor tests were used to investigate the effects of ratio of strains, inoculum size, culture temperature and culture time on the production of microbial oils. The best processing conditions were obtained by orthogonal test through measuring the content of microbial oils in product. The result shows that the microbial oils content of the co-fermented product under the ratio of Trichoderma.koningii to Trichosporon cutaneum of 1:1, the inoculum size of strains of 11% at 28 ℃ for 6 d is 7.15 g/L. It is shown that the production of microbial oils co-fermented by Trichoderma.koningii and Trichosporon cutaneum with Daqu distiller's grains is possible. The research provides a new idea for the reuse of Daqu distiller's grains and also provides a new way for the development of microbial oils.展开更多
The energy metabolism structure of microbial community plays an important role in the process of biohydrometallurgy.In this article,an artificial microbial community composed of three strains(Acidithiobacillus ferroox...The energy metabolism structure of microbial community plays an important role in the process of biohydrometallurgy.In this article,an artificial microbial community composed of three strains(Acidithiobacillus ferrooxidans,Leptospirillum ferriphilum and Acidithiobacillus thiooxidans)was used to leach three kinds of chalcopyrites with different iron-sulfur ratios.After 36 d of leaching,the chalcopyrite with iron-sulfur ratio of about 1:1 achieved the highest copper extraction(69.62%).In the early stage,iron oxidizing bacteria predominated,and the expression of rus and rio was 8 times higher than that in the late stage.In the late stage,sulfur oxidizing bacteria predominated,and the expression of tetH and HdrAB was 4 times higher than that in the early stage.Furthermore,the three bioleaching systems above were added with elemental sulfur(3 g/L);the chalcopyrite with iron-sulfur ratio of about 2:1 achieved the highest copper extraction(80.63%).The results suggest that the energy metabolism structure of the microbial community could be changed by changing the iron-sulfur ratio during the leaching process for improving the leaching efficiency of chalcopyrite.展开更多

Microorganisms,one of the key factors affecting the bioleaching process,change the components of extracellular polymeric substance(EPS)and community structure to survive in leaching environments.In this work,Fourier t...Microorganisms,one of the key factors affecting the bioleaching process,change the components of extracellular polymeric substance(EPS)and community structure to survive in leaching environments.In this work,Fourier transform infrared(FTIR),X-ray powder diffraction(XRD)and 16S rDNA high-throughput sequence analyses were used to reveal the microbial changes in planktonic and sessile phases during bioleaching.The results showed the occupation of sessile cells decreased from 66.2%to(10±3)%.After bioleaching,the planktonic and sessile cells have similar EPS,but they are different from the original cells.Pyrite dissolution mainly occurs at the early and late stages with the decreasing of particle diameter,by 50%and 40%,respectively.The 16S rDNA gene based sequence analysis results in total of 1117420 Reads across the six samples,presented among 7 phyla,9 classes,17 orders,23 families and 31 genera.Genera Leptospirillum and Sulfobacillus are the main bacteria at the early and middle stages,and Leptospirillum is the main genus at the end of bioleaching.Aquabacterium and Acidovorax are special genera in sessile cells and Weissella is special in planktonic ones.展开更多
The effects of different amounts of carbon and nitrogen sources on the soil microbial biomass carbon,dissolved organic carbon and related enzyme activities were studied by the simulation experiment of rice straw retur...The effects of different amounts of carbon and nitrogen sources on the soil microbial biomass carbon,dissolved organic carbon and related enzyme activities were studied by the simulation experiment of rice straw returning to the field,and the mechanism of the decomposition of rice straw returning to the field was discussed.Completely randomized experiment of the two factors of the three levels was designed,and a total of nine treatments of indoor soil incubation tests were conducted.Full amount of rice straw was applied to the soil in this simulation experiment and different amounts of brown sugar and urea were added in the three levels of 0(no carbon source and nitrogen source),1(low levels of carbon and nitrogen sources)and 2(high levels of carbon and nitrogen sources),respectively.The results showed that the addition of different amounts of carbon and nitrogen sources to the rice straw could increase the soil carbon content.Compared with T0N0,the microbial biomass carbon of T2N2 was increased significantly by 170.48%;the dissolved organic carbon content of T1N2 was significantly increased by 58.14%and the free humic acid carbon contents of T0N2,T1N1 and T2N0 were significantly increased by 56.16%and 45.55%and 47.80%,respectively;however,there were no significant differences among those of treatments at later incubation periods.The addition of different carbon and nitrogen sources could promote the soil enzyme activities.During the incubation period,all of the soil enzyme activities of adding sugar and urea were higher than those of T0N0 treatment.Therefore,the addition of different amounts of carbon and nitrogen sources to rice straw returning could improve soil microbial biomass carbon content,dissolved organic carbon and soil enzyme activities.展开更多

文摘【Objectives】Si and microbial application could relieve the crop replanting problems(CRPs).We further studied the change of key microorganisms that are related to the beneficial effects,aiming at provide reference for the manufacture and application of both microbial agents and Si fertilizer in food lily production.【Methods】A field experiment was conducted over a three-year period,from March 2019 to March 2022.The experimental field had been continuously cultivated with lily for 9 years.Three treatments were established:silicon fertilizer(SF),microbial agents(“Special 8^(TM)”,MF),and combined application of silicon fertilizer and microbial agents(SMF).A control group with blank soil(CK)was also included.At seedling stage of Lanzhou lilies in 2020 and 2021,the shoot and bulb dry weight,and the plant height and stem diameter of Lanzhou lilies were investigated for calculation of seedling index.In July 2020,20 plants were selected in each plot,and root zone soils were sampled at a depth of 20 cm,10 cm away from the roots,and then mixed to form a composite sample.The soil available Si and organic matter content were analyzed,and the fungal community structure and some specific microbial groups in soils were determined with high-throughput sequencing of ITS.【Results】All the three treatments significantly enhanced the lily plant growth and the seedling index,compared to CK.Besides,SF and MF treatments increased the relative abundances(RA)and diversity of fungal communities,and altered the community structures.The RA of some specific groups were found to be significantly correlated with the seedling index and/or soil available Si.Of them,the RA of the genera Fusarium,Dactylonectria,Humicola,Stilbella,and the species Humicola_grisea showed a positive correlation,while that of the genera Mortierella,Stilbella,Holtermanniella,and the species Mortierella_fatshederae showed a negative correlation with seedling index.The genera Fusarium,Stilbella,the species Humicola_grisea,and Dactylonectria_estremocensis showed a positive correlation,while the genura Stilbella,and the species Mortierella fatshederae showed a negative correlation with available Si content.In the co-occurence network of top twenty fungal genera and top sixteen bacterial genera(RA>0.2%),Holtermanniella was the only genus that interacted with the bacteria and negatively correlated with bacterial genus Blastococcus.Holtermanniella was also the most densely connected genera,followed by the genus Fusarium,Didymella and Humicola.In addition,the genus Holtermanniella was the key species connecting fungal and bacterial community in soil.Fungal functional prediction revealed that SF,MF and SMF treatments decreased plant pathogens guilds and increased the beneficial guilds Ectomycorrhizal,plant saprophyte,leaf saprophyte,and arbuscular mycorrhizal compared to CK.【Conclusions】Combined application of silicon fertilizer and microbial agents can alleviate continuous replanting problems of Lanzhou lilies through restoring the fungal community diversity,and promoting plant residue depredation,thus reducing soil born disease incidence.The beneficial genus Humicola and its one species H.grisea acts as bioconversion,and the genus Acremonium acts as plant pathogen inhibitor.

文摘The enhancement of chalcopyrite bioleaching with an enriched microbial community by acidified seawater was studied,and the enhancing mechanism was analyzed.The microbial community was enriched at the Dabaoshan mine site,and the treated ore sample had high concentrations of chalcopyrite and galena.The experimental results show that copper extraction from chalcopyrite with an enriched microbial community in seawater was promoted from 13.1%to 62.1%by acidification in comparison with that without acidification.Further analyses of the solutions,solid residues and microbial compositions by scanning electron microscopy,X-ray diffraction,Raman spectroscopy,Fourier transform infrared spectroscopy and 16 S rDNA sequencing revealed the promoting effects of acidified seawater.This acidification can increase the biodissolution of chalcopyrite to increase the concentration of iron ions and maintain the redox potential in the range of 360−410 mV.The latter produces an optimal redox environment conducive to chalcopyrite dissolution via Cu_(2)S.The adaptability of the microbial community to a high-salt environment is improved.Chloride ions at 580 mmol/L improve the leaching kinetics of chalcopyrite by increasing the porosity and noncrystallinity of the intermediate elemental sulfur.This study provides a promising way to bioleaching copper minerals using seawater for areas with freshwater shortages.

文摘Microbial fuel cells have already been used as biosensors to monitor assimilable organic carbon(AOC).However,their signal production from AOC is known to be completely suppressed by dissoved oxygen(DO).In this study,two identical microbial electrolysis cell(MEC)based biosensors were inoculated with marine sediment and operated at two different anodic potentials,namely-300 mV and+250 mV relative to Ag/AgCl.The MEC biosensor operated under positive anodic potential conditions had electrochemically active microbial communities on the anode,including members of the Shewanellaceae,Pseudoalteromonadaceae,and Clostridiaceae families.However,the strictly anaerobic members of the Desulfuromonadaceae,Desulfobulbaceae and Desulfobacteraceae families were found only in the negative anodic potential MEC biosensor.The positive anodic potential MEC biosensor showed several other advantages as well,such as faster start-up,significantly higher maximum current production,fivefold improvement in the AOC detection limit,and tolerance of low dissolved oxygen,compared to those obtained from the negative anodic potential MEC biosensor.The developed positive anodic potential MEC biosensor can thus be used as a real-time and inexpensive detector of AOC concentrations in high saline and low DO seawater.
文摘MoS_(2)/CuS composite catalysts were successfully synthesized using a one-step hydrothermal method with sodium molybdate dihydrate,thiourea,oxalic acid,and copper nitrate trihydrate as raw materials.The hydrogen pro-duction performance of MoS_(2)/CuS prepared with different molar ratios of Mo to Cu precursors(n_(Mo)∶n_(Cu))as cathodic catalysts was investigated in the two-chamber microbial electrolytic cell(MEC).X-ray diffraction(XRD),X-ray pho-toelectron spectroscopy(XPS),scanning electron microscopy(SEM),transmission electron microscope(TEM),linear scanning voltammetry(LSV),electrochemical impedance analysis(EIS),and cyclic voltammetry(CV)were used to characterize the synthesized catalysts for testing and analyzing the hydrogen-producing performance.The results showed that the hydrogen evolution performance of MoS_(2)/CuS-20%(nMo∶nCu=5∶1)was better than that of platinum(Pt)mesh,and the hydrogen production rate of MoS_(2)/CuS-20%as a cathode in MEC was(0.2031±0.0237)m^(3)_(H_(2))·m^(-3)·d^(-1) for 72 h at an applied voltage of 0.8 V,which was slightly higher than that of Pt mesh of(0.1886±0.0134)m^(3)_(H_(2))·m^(-3)·d^(-1).The addition of a certain amount of CuS not only regulates the electron transfer ability of MoS_(2) but also increases the density of active sites.

文摘Simulated heap bioleaching of low-grade high pyrite-bearing chalcocite ore was conducted at 40 °C with aeration of CO_2 and N_2.Ore samples were collected at day 43,64,85,106 and subjected to microbial community analysis by 16S rRNA gene clone library.Phylogenetic analyses of 16S rDNA fragments revealed that the retrieved sequences are mainly related to genus Acidithiobacillus,Leptospirillum and Sulfobacillus.Aeration of CO_2 and N_2 significantly impacted the microbial community composition.When CO_2 was aerated,the proportion of genus Acidithiobacillus considerably increased,whereas the proportion of genus Leptospirillum and genus Sulfobacillus declined.However,with the aeration of N_2,the proportion of genus Acidithiobacillus and Leptospirillum increased,but genus Sulfobacillus decreased.When there was no aeration,the microbial community was similar to the inocula with the proportion of genus Leptospirillum mounted.These results indicated that the limitation of oxygen could change the bioleaching microbial community and the aeration of CO_2 and N_2 was favourable for the growth of sulfur-oxidizer(At.caldus) and iron-oxidizer(L.ferriphilum) respectively,which could be used for the regulation of microorganisms' role in mineral bioleaching.
文摘The experiment was conducted to study the impact of application of microbial inoculants, compared with no microbial fertilizer, on enzyme activity, microbial biomass and available nutrient contents in paddy soil in Heilongjiang Province. The application of soil phosphorus activator was able to increase the quantity of bacteria and fungi in soil, but its effect on actinomycetes in soil was not significant. The application of microbial inoculants increased the urease and sucrase activities in soil over the growing season, but only at the maturing stage soil acid phosphatase activity was enhanced with the applying soil phosphorus activator. The application of soil phosphorus activator increased alkali-hydrolyzable nitrogen and available phosphorus contents in soil, but did not increase available potassium content in soil. The optimal microbial inoculant application rate as applied as soil phosphorus activator was 7.5 kg hm-2.

文摘The present study was conducted to see the short term impact of organic and inorganic fertilizers on soil microbial biomass both in spring and summer. Also aimed to observe the correlation between soil microbial biomass and soil DNA. The study concluded that type of fertilizer might alter the soil microbial biomass and DNA contents. In soil treated with organic fertilizers resulted in higher concentrations of microbial biomass and DNA contents in summer as compared to spring dute to increase in temperature. Correspondingly, in case of inorganic fertilizer, concentrations of soil microbial biomass and DNA detected higher in summer instead of spring. The statistical correlation between soil microbial biomass, DNA and ODR in spring and summer along with organic and inorganic fertilizers were calculated highly significant (p〉0.01). This study demonstrated the impact of fertilizers and seasonal variations on soil microbial biomass and also revealed significant correlation between soil microbial biomass and soil DNA.

文摘A coupled system consisting of an upflow membrane-less microbial fuel cell (upflow ML-MFC) and a photobioreactor was developed, and its effectiveness for continuous wastewater treatment and electricity production was evaluated. Wastewater was fed to the upflow ML-MFC to remove chemical oxygen demand (COD), phosphorus and nitrogen with simultaneous electricity generation. The effluent from the cathode compartment of the upflow ML-MFC was then continuously fed to an external photobioreactor for removing the remaining phosphorus and nitrogen using microalgae. Alone, the upflow ML-MFC produces a maximum power density of 481 mW/m 3 , and obtains 77.9% COD, 23.5% total phosphorus (TP) and 97.6% NH4+-N removals. When combined with the photobioreactor, the system achieves 99.3% TP and 99.0% NH4+-N total removal. These results show both the effectiveness and the potential application of the coupled system to continuously treat domestic wastewater and simultaneously generate electricity and biomass.
文摘A 31 week feeding trial was conducted to investigated the effects of dietary supplementation of microbial phytase and cellulase on performance,nutrients utilization and tibia quality of laying hens fed maize and soybean meal diets.192 18 week old Hisex layers were used in the trial A 2×2×2 factorial design was used in the experiment with three factors of two levels each:0 38% and 0 16% of dietary non phytate P(nP).0 and 300 U·kg -1 of phytase (Ph),and 0 and 0 10% of cellulase (Ce).The results showed that supplementation of 300 U·kg -1 phytase significantly improved utilization of dietary crude ash,CP,Ca,total P and copper (P<0 05),and improved tibia breaking strength (P<0 05).No effect of phytase on performance was observed.Addition of 0 10% cellulase decreased feed intake (P<0 05),increased utilization of CF (P<0 05) and Ca(P<0 01),and decreased total tibia ash weight (P<0 05).300 U·kg -1 phytase and 0 10% cellulase exhibited obvious positive interactions to enhance utilization of dietary phytic P and copper (P<0 05).0 16% nP did not reduce performance of the layers,but improved egg shell quality at 20 wks,increased utilization of dietary total P,phytic P and Copper (P<0 01),decreased utilization of dietary CP,increased tibia breaking strength and Ca,Mn contents of tibia(P<0 01)

文摘This study was conducted to investigate the effect of Soybean Peptides(SPs) on ruminal fermentation and microbial diversity in vitro. Four levels of SPs supplements(0, 0.25%, 0.50% and 0.75% dry matter basis) were tested. p H, NH3-N, Neutral Detergent Fiber(NDF), Acid Detergent Fiber(ADF), Dry Matter Digestibility(DMD) and the Total Volatile Fatty Acid(TVFA) were measured at 6, 24, 48 h of in vitro mixed incubation the fluids. Microbial populations were determined at 24 h and Microbial Proteins(MCP) were determined at 24 and 48 h. The gas production after 48 h in vitro incubation presented linear growth(p<0.05) and the highest content was the level of 0.5% SPs(dry matter basis). NH3-N concentration reached the highest concentration with 0.75%(dry matter basis) at 48 h. p H linearly increased(linear, p<0.05) from 6 to 48 h. The digestion of DMD increased with increasing doses of SPs at 24 h. NDF and ADF linearly(p<0.01) decreased by adding SPs. The concentration of TVFA linearly increased(p<0.05) at 6, 24 and 48 h, and reached the highest concentration at the level of 0.75%(dry matter basis). SPs decreased(p<0.05) the molar proportion of acetate and propionate, respectively at 24 and 48 h. Acetat to Propionate ratio(A/P; linear, p<0.05) increased at 48 h, and reached the greatest value at the level of 0.75%(dry matter basis) at 48 h. The populations of rumen Ruminococcus flavefaciens(R. flavefaciens), Ruminococcusalbus(R. albus), Fibrobacter succinogenes(F. succinogenes), Butyrivibrio fibrisolvens(B. fibrisolvens), Streptococcus bovis(S.bovis), Ruminobacter amylophilus(R. amylophilus) and Succinimonas amylolytica(S. amylolytica) were analyzed based on the total rumen bacterial 16 S ribosomal deoxyribonucleic acid(r DNA). The relative abundance of R. flavefaciens and R. albus increased at 24 h, but the relative abundance of F. succinogenes decreased at this time. The incubation of SPs had no effect on the abundance of S. bovis and R. amylophilus. The relative abundance of B. fibrisolvens and S. amylolytica reached the greatest value(p<0.05) at middle doses of SPs inclusion at 24 h. The value of MCPs linearly increased(p<0.05) at 24 and 48 h. These results showed that SPs could improve in vitro fermentation and nutrient digestion of feed substrates, and had the ability to modulate the ruminal fermentation pattern by regulating the composition of functional rumen microbes. Hence, SPs might be a potential feed additive applied in the diets of ruminants.
文摘Under field conditions, an experiment was conducted to study the effects of ammonification bacteria, potassium bacteria and phosphorus bacteria on nutrient availability in soil and yield of rice in the cold region of China and compared to the conventional fertilization. Results showed that DF1P2 treatment (ammonifiers 1.5× 108 cfu· m2, phosphorus bacteria 1.5× 108 cfu. m2, and potassium bacteria 1.5× 108 cfu· m2) increased available nutrient concentrations in soil, increased the concentrations of N, P, and K in plant organs and increased the rice yield and was the most significantly among all the treatments. This treatment could be recommended as the best suitable biological fertilizer application rate for the rice production in the cold region of China.

文摘Microbial community diversities in the drainage from three mines(Dexing Copper Mine,Qibaoshan Copper Mine and Yaogangxian Tungsten Mine,China)were analyzed using 16S rDNA PCR-RFLP approach.The efficiencies of chalcopyrite bioleaching were compared using enrichment of the three cultures.Phylogenetic analysis indicates that the dominant microorganisms are clustered with the Proteobacteria,the remaining is affiliated with Nitrospira,Acidobacteria and Actinobacteria.At the genus level,Acidithiobacillus is the dominant group in both YTW and QBS samples,while Spingomonas is dominant in YGX sample.Moreover,the principal component analysis(PCA)reveals that QBS and YTW have similar geochemical character and microbial communities.The results also show that pH value and tungsten concentration play a key role in microbial community distribution and relative abundance.The bioleaching efficiency of the enrichment cultures from YTW and QBS is similar.After 15 d,the bioleaching rates of low grade chalcopyrite(0.99%)are both up to 99.5% when using 10 g/L pulp density due to the similar microbial composition of YTW and QBS.Moreover,the leaching efficiencies of enrichment cultures containing multiple bioleaching microorganisms are higher than that of pure culture Acidithiobacillus ferrooxidans.

文摘In this study, a lab-scale upflow anaerobic sludge blanket(UASB) reactor was applied to studying the high-rate nitrogen removal of granule-based anammox process. The nitrogen removal rate(NRR) finally improved to 15.77 kg/m3/d by shortening hydraulic retention time(HRT) to 1.06 h. Well-shaped red anammox granules were extensively enriched inside the reactor. The results of nitrogen removal kinetics indicated that the present bioreactor has great nitrogen removal potential, because the maximum rate of substrate utilization(Umax) predicted by Stover-Kincannon model is suggested as 55.68 kg/(m3·d). Analysis of the microbial community showed that the anammox genus Candidatus Kuenenia dominated the bacterial communities. The relative abundance of Candidatus Kuenenia rose from 12.29% to 36.95% after progressively shorter HRT and higher influent substrate concentrations, illustrating the stability of nitrogen removal performance and biomass enrichment offered by the UASB in carrying out high-rate anammox process.

文摘Abstract: Daqu distiller's grains were co-fermented as raw materials by Trichoderma.koningii and Trichosporon cutaneum to produce microbial oils, which can provide raw materials for bio-diesel development. The single factor tests were used to investigate the effects of ratio of strains, inoculum size, culture temperature and culture time on the production of microbial oils. The best processing conditions were obtained by orthogonal test through measuring the content of microbial oils in product. The result shows that the microbial oils content of the co-fermented product under the ratio of Trichoderma.koningii to Trichosporon cutaneum of 1:1, the inoculum size of strains of 11% at 28 ℃ for 6 d is 7.15 g/L. It is shown that the production of microbial oils co-fermented by Trichoderma.koningii and Trichosporon cutaneum with Daqu distiller's grains is possible. The research provides a new idea for the reuse of Daqu distiller's grains and also provides a new way for the development of microbial oils.

文摘The energy metabolism structure of microbial community plays an important role in the process of biohydrometallurgy.In this article,an artificial microbial community composed of three strains(Acidithiobacillus ferrooxidans,Leptospirillum ferriphilum and Acidithiobacillus thiooxidans)was used to leach three kinds of chalcopyrites with different iron-sulfur ratios.After 36 d of leaching,the chalcopyrite with iron-sulfur ratio of about 1:1 achieved the highest copper extraction(69.62%).In the early stage,iron oxidizing bacteria predominated,and the expression of rus and rio was 8 times higher than that in the late stage.In the late stage,sulfur oxidizing bacteria predominated,and the expression of tetH and HdrAB was 4 times higher than that in the early stage.Furthermore,the three bioleaching systems above were added with elemental sulfur(3 g/L);the chalcopyrite with iron-sulfur ratio of about 2:1 achieved the highest copper extraction(80.63%).The results suggest that the energy metabolism structure of the microbial community could be changed by changing the iron-sulfur ratio during the leaching process for improving the leaching efficiency of chalcopyrite.

文摘In this study,conductive polymer polyaniline(PANI)is employed to modify the anodes of benthic microbial fuel cells(BMFC).Four electrochemical methods are used to synthesize the polyaniline anodes;the results show that the PANI modification,especially the pulse potential method for PANI synthesis could obviously improve the cell energy output and reduce the anode internal resistance.The anode is modified by PANI doped with Fe or Mn to further improve the BMFC performance.A maximum power density of 17.51 mW/m2 is obtained by PANI-Fe anode BMFC,which is 8.1 times higher than that of control.The PANI-Mn anode BMFC also gives a favorable maximum power density(16.78 mW/m2).Fe or Mn modification has better effect in improving the conductivity of polyaniline,thus improving the energy output of BMFCs.This work applying PANI composite anode into BMFC brings new development prospect and could promote the practical application of BMFC.

文摘Microorganisms,one of the key factors affecting the bioleaching process,change the components of extracellular polymeric substance(EPS)and community structure to survive in leaching environments.In this work,Fourier transform infrared(FTIR),X-ray powder diffraction(XRD)and 16S rDNA high-throughput sequence analyses were used to reveal the microbial changes in planktonic and sessile phases during bioleaching.The results showed the occupation of sessile cells decreased from 66.2%to(10±3)%.After bioleaching,the planktonic and sessile cells have similar EPS,but they are different from the original cells.Pyrite dissolution mainly occurs at the early and late stages with the decreasing of particle diameter,by 50%and 40%,respectively.The 16S rDNA gene based sequence analysis results in total of 1117420 Reads across the six samples,presented among 7 phyla,9 classes,17 orders,23 families and 31 genera.Genera Leptospirillum and Sulfobacillus are the main bacteria at the early and middle stages,and Leptospirillum is the main genus at the end of bioleaching.Aquabacterium and Acidovorax are special genera in sessile cells and Weissella is special in planktonic ones.

文摘The effects of different amounts of carbon and nitrogen sources on the soil microbial biomass carbon,dissolved organic carbon and related enzyme activities were studied by the simulation experiment of rice straw returning to the field,and the mechanism of the decomposition of rice straw returning to the field was discussed.Completely randomized experiment of the two factors of the three levels was designed,and a total of nine treatments of indoor soil incubation tests were conducted.Full amount of rice straw was applied to the soil in this simulation experiment and different amounts of brown sugar and urea were added in the three levels of 0(no carbon source and nitrogen source),1(low levels of carbon and nitrogen sources)and 2(high levels of carbon and nitrogen sources),respectively.The results showed that the addition of different amounts of carbon and nitrogen sources to the rice straw could increase the soil carbon content.Compared with T0N0,the microbial biomass carbon of T2N2 was increased significantly by 170.48%;the dissolved organic carbon content of T1N2 was significantly increased by 58.14%and the free humic acid carbon contents of T0N2,T1N1 and T2N0 were significantly increased by 56.16%and 45.55%and 47.80%,respectively;however,there were no significant differences among those of treatments at later incubation periods.The addition of different carbon and nitrogen sources could promote the soil enzyme activities.During the incubation period,all of the soil enzyme activities of adding sugar and urea were higher than those of T0N0 treatment.Therefore,the addition of different amounts of carbon and nitrogen sources to rice straw returning could improve soil microbial biomass carbon content,dissolved organic carbon and soil enzyme activities.

文摘Dynamic changes of a microbial community for lignocellulose degradation were explored in details. Community composition and development were investigated by the means of denaturing gradient gel electrophoresis (DGGE), and results showed that the microbial community was constituted of 14 kinds of bacteria and presented the fluctuation in some degrees with fermentation. Furthmore, the result of cluster analysis of DGGE pattern was accordant with growth curve, and the degradation process was divided into three stages: initial stage (0-12 h), intermediate stage (24-144 h) and end stage (144-216 h).
