Penicillium simplicissimum was cultured and preserved on the potato dextrose agar(PDA)medium.PDA-RBBR(Remazal Brilliant Blue R)medium was used for the screening of the strains,which is able to produce enzymes.After th...Penicillium simplicissimum was cultured and preserved on the potato dextrose agar(PDA)medium.PDA-RBBR(Remazal Brilliant Blue R)medium was used for the screening of the strains,which is able to produce enzymes.After the mutation process in Penicillium simplicissimum induced by chemical reagent and ultraviolet radiation,a high laccase-producing strains Penicillium simplicissimum was obtained.When 5 m L diethyl sulfate(2%)was mixed along with 5 m L spore suspension for 30 min,chemical mutagenesis reached its best condition.And the optimum conditions of UV mutagenesis were that spore suspension was irradiated for 4 min under 15 W UV lamp at a distance of 30 cm.The highest activity of C_5E_4 strains was 4.80 U/g over 18%higher than the maximum laccase activity of original microorganism.Five generations of the mutant strains were cultured,and the laccase activity of the strains was measured.The result showed that C_5E_4 strains can product laccase of the five subcultures stably.展开更多

Magnetic multiwalled carbon nanotubes (MMCNT) which possess excellent capability of electron transfer were prepared by chemical coprecipitation method. Scanning electron microscope (SEM) and vibrating sample magnetometer (VSM) were used to identify its surfacetopography and magnetization, respectively. Laccase was immobilized on the MMCNT modified magnetic carbon paste electrode by the aid of chitosan/silica (CS) hybrid membrane. Using current-time detection method, the biosensor shows a linear response related to the concentration of catechol in the range from 10-7 to 0.165×10-3 mol/L. The corresponding detection limit is 3.34×10-8 mol/L based on signal-to-noise ratios (S/N) ≥3 under the optimized conditions. In addition, its response current retains 90% of the original after being stored for 45 d. The results indicate that this proposed strategy can be expected to develop other enzyme-based biosensors.展开更多

Mendelian randomization(MR)is widely used in causal mediation analysis to control unmeasured confounding effects,which is valid under some strong assumptions.It is thus of great interest to assess the impact of violat...Mendelian randomization(MR)is widely used in causal mediation analysis to control unmeasured confounding effects,which is valid under some strong assumptions.It is thus of great interest to assess the impact of violations of these MR assumptions through sensitivity analysis.Sensitivity analyses have been conducted for simple MR-based causal average effect analyses,but they are not available for MR-based mediation analysis studies,and we aim to fill this gap in this paper.We propose to use two sensitivity parameters to quantify the effect due to the deviation of the IV assumptions.With these two sensitivity parameters,we derive consistent indirect causal effect estimators and establish their asymptotic propersties.Our theoretical results can be used in MR-based mediation analysis to study the impact of violations of MR as-sumptions.The finite sample performance of the proposed method is illustrated through simulation studies,sensitivity ana-lysis,and application to a real genome-wide association study.展开更多

Objective To establish a loop-mediated isothermal amplification( LAMP) method for detecting diarrhea pathogens( Shigella and Salmonella) in rhesus monkeys and evaluate the application of the LAMP method for detecting ...Objective To establish a loop-mediated isothermal amplification( LAMP) method for detecting diarrhea pathogens( Shigella and Salmonella) in rhesus monkeys and evaluate the application of the LAMP method for detecting bacterial diseases in nonhuman primate laboratory animals. Materials and Methods A total of 205 fecal samples of rhesus monkeys were detected in this LAMP assay. The specificity and sensitivity of LAMP for Shigella and Salmonella were analyzed,and real-time polymerase chain reaction( REAL-TIME PCR) assay was employed as control. Results The LAMP method established here needed only 45 min to complete the reaction at 63℃. Its detection limit was 10 pg / μL and with a high specificity. The positive rate of Shigella and Salmonella was 1. 5% and 6. 3%,respectively. Conclusions Here we have established a fast and simple Shigella and Salmonella LAMP detection method that has strong specificity and high sensitivity and is suitable for rapid detection of bacterial disease in macaques. The development of this rapid detection kit is underway,and it will be helpful to the diarrhea detection.展开更多
