水稻发芽期耐冷基因Cold6的克隆

【目的】挖掘鉴定水稻耐冷基因,创制耐冷种质。【方法】对粳稻品种笹锦进行了甲基磺酸乙酯(EMS)诱变,筛选发芽期耐冷性显著提高的突变体,通过遗传分析、精细定位、转基因验证、表达模式分析和单倍型分析对候选基因进行克隆和功能分析。... 【目的】挖掘鉴定水稻耐冷基因,创制耐冷种质。【方法】对粳稻品种笹锦进行了甲基磺酸乙酯(EMS)诱变,筛选发芽期耐冷性显著提高的突变体,通过遗传分析、精细定位、转基因验证、表达模式分析和单倍型分析对候选基因进行克隆和功能分析。【结果】筛选到一个发芽期耐冷性显著提高的突变体M34。遗传分析表明耐冷性状由隐性单基因控制,精细定位将目标基因锁定为Cold6。通过CRISPR/Cas9基因编辑构建Cold6敲除突变体验证Cold6参与发芽期耐冷性调控。Cold6在水稻各组织器官均有表达,其中穗部表达最高,编码SNF2等多个结构域。进化树分析显示Cold6可能存在籼粳分化,单倍型分析鉴定出Cold6可分为10个单倍型,其中单倍型Ⅰ~Ⅲ为主要单倍型,单倍型Ⅰ主要为粳稻,单倍型Ⅱ和Ⅲ主要为籼稻。【结论】本研究克隆了Cold6,一个新的水稻发芽期耐冷性调控位点,编码SNF2蛋白,对Cold6分子机理的研究不但可以加深水稻中SNF2家族基因功能的认知,还可为北方粳稻耐冷分子育种提供理论线索和种质资源。 展开更多

Effects of applying electroshocking treatment at different stages of cold rolling-quenching-tempering process on microstructure and properties of M50 steel

Electroshocking treatment(EST),an efficient and rapid material treatment method,promotes microstructure evolution and improves mechanical properties.This study incorporates EST into the conventional cold rolling-quenching tempering process of M50 steel and investigates the influence and mechanism of applying EST at different stages of the process on the microstructure and mechanical properties.Scanning electron microscope(SEM),transmission electron microscope(TEM),and X-ray diffraction(XRD)were used to characterize the effect of EST on microstructure.The results show that EST can refine the grains of M50(average reduction of 10.1%in grain size),homogenize the grain size distribution,reduce the dislocation density(20.9%in average),promote the dissolution of carbides in the matrix and distribute them more uniformly along the grain boundaries,resulting in the improvement of mechanical properties.The mechanical properties of the specimen with the process flow of rolling-quenching-tempering-electroshocking showed excellent performance,with an increase in hardness of 1.4%,tensile strength of 17.7%,and elongation at break of 24.3%as compared to the specimen without EST.The tensile properties of the specimen with the process flow of rolling electroshocking-quenching-tempering showed the best performance,with an increase in tensile strength of 30.0%and elongation at break of 30.7%as compared to the specimen without EST. 展开更多

同点正交配置磁环和电偶极子(Co-centered orthogonal loop and dipole,COLD)是一种最常用的二分量电磁矢量传感器,但是COLD传感器没有充分利用磁环和电偶极子分量的空间信息。本文针对由COLD传感器组成的均匀线阵(Uniform linear array... 同点正交配置磁环和电偶极子(Co-centered orthogonal loop and dipole,COLD)是一种最常用的二分量电磁矢量传感器,但是COLD传感器没有充分利用磁环和电偶极子分量的空间信息。本文针对由COLD传感器组成的均匀线阵(Uniform linear array,ULA),将所有磁环和电偶极子分量分别沿两个正交方向均匀拉伸,形成L形阵,扩展阵列的空间孔径,并提出了基于广义旋转不变的降维多重信号分类算法(Dimension reduction multiple signal classification method based on generalized rotational invariance,GRIDR-MUSIC)。所提算法利用L形阵的几何构形,将导向矢量分隔成三部分,通过两个正交ULA的广义旋转不变结构,分别估计各个部分,使得波达角(Direction of arrival)和极化参数仅需一维谱峰搜索就可以估计得到,且无需参数匹配。最后,仿真实验验证了所提算法的有效性。 展开更多

Various kinds of base oils were applied to cold rolling aluminum strips on a test mill for evaluationof the influences of these base oils,aromatics contents and viscosity of base oils on their lubricating performances and surface reflectivity of rolled strips at annealing. Results showed that low friction coefficient androlling force were obtained by using the normal paraffins,whereas their contaminations on the annealed stripsurface were the same as those of other saturated hydrocarbons. Aromatics in base oil affected the stir face reflectivity of annealed strips, but the decrease of aromatics in base oil was ineffective to improve rolled stripssurface quality when it is less than 1 %. Base oil viscosity has the great influences on the lubricating performances and surface reflectivity of annealed strips just in this condition. 展开更多

This study was to investigate the effects of cold stress on the contents of total antioxidant capacity (T-AOC) malondialdehyde (MDA) and superoxide dismutase (SOD) in duodenum, jejunum and ileum of chickens. A total of 80 15-dayold male chickens were treated by cold stress with the duration of the acute cold stress being 1, 3, 6, 12, and 24 h, and the chronic cold stress was 5, 10, and 20 days, respectively. Cold stress temperature was (12±1)℃. The chemical colorimetric method was used to detect the changes of the T-AOC, SOD activities and MDA contents. The results showed that compared with the corresponding control group, effects of acute cold stress on the T-AOC in duodenum, jejunum and ileum of chickens significantly (P〈0.05) increased firstly and then decreased. Under chronic cold stress, the T-AOC significantly (P〈0.05) decreased. Under acute cold stress and chronic cold stress, the MDA contents significantly (P〈0.05) increased in duodenum, jejunum and ileum of chickens. The effects of acute cold stress on the SOD activities in duodenum, jejunum and ileum of chickens significantly (P〈0.05) increased firstly and then decreased Under chronic cold stress the SOD activities significantly (P〈0.05) decreased in jejunum and ileum, but significantly (P〈0.05) decreased firstly and then increased in duodenum. 展开更多

G蛋白偶联受体是重要的信号传导因子,在低温信号应答中发挥着重要作用。COLD1(chilling tolerance divergence 1)编码1个G蛋白信号调节因子,可能作为低温信号感受器发挥作用。以采后黄瓜作为试验材料,克隆CsCOLD1的全长序列,分析其序列特性及其在低温处理期间的表达变化。结果显示,CsCOLD1基因全长1443 bp,编码480个氨基酸残基;CsCOLD1蛋白定位在细胞膜、叶绿体上,含有9次跨膜结构,是一个跨膜蛋白;黄瓜CsCOLD1与拟南芥等双子叶植物COLD1蛋白的氨基酸序列同源性较高,两者COLD1的进化关系较近,葫芦科植物COLD1的GPHR_N与ABA_GPCR结构域高度保守;在6℃低温处理下CsCOLD1表达量在叶片中基本没有变化;在5、10℃低温处理下,CsCOLD1的表达量在采后黄瓜中显著下调。虽然未在CsCOLD1启动子中鉴定到低温响应元件,但检测到乙烯响应元件、MYB和MYC转录因子结合位点,表明该基因是一个逆境胁迫相关基因。CsCOLD1蛋白的氨基酸序列中含有多个磷酸化位点,表明其可能在磷酸化后发挥功能。 展开更多

In this study, the process, weld combinations, laser-CMT hybrid welding and applications of CMT welding are critically reviewed. Microstructure and other weld characteristics have been discussed at length for various base metal combinations. Particularly, the welding of aluminium and steel with better results has been possible with CMT Welding. The results reviewed in this article indicate that the CMT-Laser hybrid welding is more preferable to Laser or Laser hybrid welding. CMT welding has found applications in automobile industries, defence sectors and power plants as a method of additive manufacturing. 展开更多

The study was designated to explore the physiological mechanism of cold tolerance enhanced by phosphate in rice. An experiment was conducted to investigate the effects of different levels of phosphate fertilizer on cold tolerance and its related physiological parameters in rice seedings (chilling-sensitive cv. Changbai 9 and chilling-tolerant cv. Jijing 81) under low temperature stress. At the same time, the identification of cold tolerance was conducted. Compared with the normal temperature treatment, the relative chlorophyll content, photosynthesis rate, Fv/Fm and qP decreased and index of unsaturated fatty acid increased in rice under low temperature stress. The effect of chilling-sensitive cultivars was more than that of chilling-tolerant cultivars, more phosphorus fertilizer properly improved seedling quality of rice, slowed relative chlorophyll content dropping degree of rice seeding, increased photosynthesis rate, Fv/Fm, qP and index of unsaturated fatty acids, and enhanced the ability to chilling-tolerant cultivars under low temperature. The effect on chilling-tolerant cultivars was significantly higher than that on chilling sensitive cultivars by applying more phosphorus fertilizer. Phosphate regulated photosynthetic physiology and membrane fluidity to reduce injury by low temperature, and increasd the cold tolerance capacity of rice. 展开更多

Winter wheat (Triticum aestivum) cultivars Dongnongdongmai 1 with strong cold/freezing tolerance and Jimai 22 with weak cold/freezing tolerance were used for investigating the difference of microstructure and ultrastructure between leaves of two cultivars under low temperature stress (5℃ and -15 ℃) using optical and electron microscope. The results showed that there was no obvious difference between leaves of Dongnongdongmai 1 and Jimai 22 in microstructure. However, the difference between those leaves was distinct in ultrastructure. The grana lamella and stroma lamella were stacked regularly and arranged parallelly along the long axis of chloroplasts in cv. Dongnongdongmai 1, while the arrangement directions of thylakoids in Jimai 22's leaves were so irregular as to form various angles with the long axis of chloroplasts. At -15℃, the mitochondrias were swelled to be round and the structure of cristaes became blurry in both cultivars' leaves, while some cristaes of Jimai 22 disappeared. The homogeneity of microstructure of cold closed-die forged gears can highly affect their service performance. The homogeneity of microstructure and Vickers hardness in cold closed-die forged gear of 20 Cr Mn Ti alloy is comprehensively studied by using optical microscopy and Vickers hardness tester. The results show that the distribution homogeneity of the aspect ratio of grain and Vickers hardness is the same. In the circumferential direction of the gear tooth, the distribution of the aspect ratio of grain and Vickers hardness is inhomogeneous and they gradually decrease from the surface to the center of the tooth. In the radial direction, the distribution of the aspect ratio of grain and Vickers hardness is inhomogeneous on the surface of the gear tooth; while it is relatively homogeneous in the center of the gear tooth. In the axial direction of the gear tooth, the distribution of the aspect ratio of grain and Vickers hardness is relatively homogeneous from the small-end to the large-end of the gear tooth. 展开更多

The dynamic model of cold rolling mill based on strip flatness and thickness integrated control was proposed,containing the following sub-models:the rolling process model,the dynamic model of rolls along axial direction,and the compensation model.Based on the rule of volume flow rate,the dynamic rolling process model was built.The work roll and backup roll were taken as elastic continuous bodies,the effect of shear and moment of inertia were taken into consideration,and then the dynamic model of rolls was built.The two models were coupled together,and the dynamic model of rolling mill was built.In the dynamic model,the thermal expansion of the rolls,the wear of the rolls and other related parameters can not be considered.In order to compensate the dynamic model,the coupled static model of rolls and strip was applied.Then,according to the inner relationship of these models,the dynamic model and the compensation model were coupled,and the dynamic model of rolling mill based on the strip flatness and thickness integrated control was built.The dynamic simulation of the rolling process was made,and the dynamic thickness and the dynamic flatness information were obtained.This model not only provides a theory basis for the virtual rolling,but also provides a platform for the application of advanced control theory. 展开更多
