Fourier transform infrared spectroscopy(FTIR) and circular dichroism(CD) were used to investigate the conformational changes of heated whey protein(WP) and the corresponding changes in the hydrolysates immunoreactivit...Fourier transform infrared spectroscopy(FTIR) and circular dichroism(CD) were used to investigate the conformational changes of heated whey protein(WP) and the corresponding changes in the hydrolysates immunoreactivity were determined by competitive enzyme-linked immunosorbent assay(ELISA).Results showed that the contents of α-helix and β-sheet of WP did not decrease much under mild heating conditions and the antigenicity was relatively high;when the heating intensity increased(70 ℃ for 25 min or 75 ℃ for 20 min),the content of α-helix and β-sheet decreased to the minimum,so was the antigenicity;However,when the WP was heated at even higher temperature and for a longer time,the β-sheet associated with protein aggregation begun to increase and the antigenicity increased correspondingly.It was concluded that the conformations of heated WP and the antigenicity of its hydrolysates are related and the optimum structure for decreasing the hydrolysates antigeniity is the least content of α-helix and β-sheet.Establishing the relationship between the WP secondary structure and WP hydrolysates antigenicity is significant to supply the reference for antigenicity reduction by enzymolysis.展开更多
As an acidic glycoprotein,carcinoembryonic antigen(CEA)is of great value as a broad-spectrum tumor marker in the differential diagnosis and surveillance of malignant tumors.In this work,we report an electrochemical ap...As an acidic glycoprotein,carcinoembryonic antigen(CEA)is of great value as a broad-spectrum tumor marker in the differential diagnosis and surveillance of malignant tumors.In this work,we report an electrochemical aptasensor for the ultrasensitive and highly selective detection of CEA,taking advantage of the dual amplification by the boronate affinity-assisted electrochemically controlled atom transfer radical polymerization(BA-eATRP).Specifically,the BAeATRP-based electrochemical aptasensing of CEA involves the capture of target antigens by nucleic acid aptamers,the covalent crosslinking of ATRP initiators to CEA antigens via the selective interactions between the phenylboronic acid(PBA)group and the cis-diol group of the monosaccharide residues,and the collection of the ferrocene(Fc)reporters via the eATRP of ferrocenylmethyl methacrylate(FcMMA).As CEA is decorated with hundreds of cis-diol groups,the BA-based crosslinking can result in the labeling of each CEA with hundreds of ATRP initiators;furthermore,the eATRP of FcMMA results in the surface-initiated growth of long-chain ferrocenyl polymers,leading to the tethering of each ATRP initiator-conjugated site with hundreds to thousands of Fc reporters.Such that,the BAeATRP can result in the efficient labeling of each CEA with a plenty of Fc reporters.Under the optimized conditions,the BA-eATRP-based strategy enables the highly selective aptasensing of CEA at a concentration as low as 0.34 pg·mL^(-1),with a linear range of 1.01000 pg·mL^(-1).Besides,this aptasensor has been successfully applied to the quantitative analysis of CEA in human serum.The BA-eATRP-based electrochemical aptasensor is cost-effective and simple in operation,holding broad application prospect in the ultrasensitive and highly selective detection of CEA.展开更多

To explore the relat...Interleukin-8 homolog (vIL-8) is a chemokine encoded by the genome of Marek's disease virus. Chicken IL-8 (cIL8) is an important chemokine of chickens which plays a role in antiviral activity. To explore the relationship between vIL-8 and cIL8 can help to discern the function of vIL-8. In this study, the amino acid sequences of vIL-8 and cIL8 were aligned. The cIL8 gene was expressed in E. coli and the cIL8 fusion protein was obtained, after induction with IPTG. The protein was separated by SDS-PAGE and the band of interest was excised and minced for mouse immunization. An immunofluorescence test was used to detect vIL-8 expression in insect cells. The results showed that both vIL-8 and cIL8 were typical CXC chemokines in structure, and they had similar key amino acids, known to be important for receptor binding. The result of the immunofluorescence test showed that the mouse anti-cIL8 serum could react with vIL-8 expressed by insect cells. Therefore, vIL-8 shares common antigenic determinants with cIL8, and they may have a common receptor. This feature of vIL-8 suggests that it may participate in the immune evasion of the virus.展开更多
