Objective:To investigate the effects of silkworm pupa(Bombyx mori) protein(SPP) on cell proliferation,apoptosis and energy metabolism in human colon cancer cells DLD-1.Methods:CCK-8 was used to detect cell proliferation rate after 72 h of cell culture for the control group(normal cultured DLD-1 cells) and SPP dose groups;Annexin-V/PI was applied to observe cell apoptosis;XFe24 Extracellular Flux Analyzer was used to detect cell mitochondrial respiratory function and glycolytic function.Results:Comparing with the control,SPP significantly inhibited the proliferation of DLD-1 cells with all the dosage tested(P <0.01);flow cytometry showed that SPP significantly promoted apoptosis(P<0.05).Additionally,SPP could significantly inhibited mitochondrial metabolism and glycolysis of DLD-1 cells and decreased cell energy metabolism in all groups treated with different doses.Conclusion:SPP can cause oxidative damage,promote apoptosis,and reduce mitochondrial respiratory and glycolysis rate in colon cancer DLD-1 cells,which reveals that SPP has the potential to serve as the anti-cancer drugs in the future,but further experimental evidence is needed. 展开更多

The incidence rate of colon cancer ranks the third among malignant tumors worldwide.Gloeostereum incarnatum(GI),a fungus with homology of medicine and food,has multiple pharmacological activities.It was investigated in this study that the anti-colon cancer effect of polysaccharides purifi ed from GI(GIPS)in ApcMinC/Gpt mice(an in situ colon cancer mouse model).Eight-week administration with GIPS at doses of 30 or 90 mg/kg strongly inhibited tumor growth including the reduction on numbers and the suppression of the size without infl uencing the animals’body weight and organ functions.According to the proteomics performing by antibody array,among 308 detected cytokines,GIPS significantly regulated 89 of them.Compared with vehicle-treated mice,GIPS effectively reduced the levels of interleukin(IL)-1β,IL-4,IL-6,IL-17,IL-22,tumor necrosis factor(TNF)-α,matrix metalloproteinase(MMP)-2,and enhanced the levels of IL-15 and IL-18 in serum and/or colon tissues,which suggested its anti-inflammation of GIPS.GIPS suppressed nuclear aggregation of β-catenin,affected the expression of WNT1 and related proteins,thereby regulated the activation of the Wnt signaling.Altogether,GIPS can inhibit the growth of colon cancer,at least partially,via inhibiting the Wnt/β-catenin signaling pathway. 展开更多

Human consumption of linoleic acid(LA,18:2ω-6,abundant in vegetable oils)is very high.Animal experiments showed that excessive LA intake increased azoxymethane-induced colon tumorigenesis,however,the impact of excessive LA on colon cancer in human is not conclusive,making it difficult to make dietary recommendations for optimal intake of LA.Understanding the molecular mechanisms of LA on colon tumorigenesis could help to clarify its health effect,and facilitate development of mechanismbased strategies for preventing colon cancer.Recent studies show that the previously unappreciated cytochrome P450 monooxygenase-mediated eicosanoid pathway is upregulated in colon cancer and plays critical roles in its pathogenesis,and could contribute to the effects of dietary LA,as well asω-3 fatty acids,on colon tumorigenesis.In this review,we will discuss recent studies about the roles of cytochrome P450 monooxygenases in fatty acid metabolism and its roles in colonic inflammation and colon cancer,and how this information could help us to clarify the health impacts of dietary fatty acids. 展开更多
