甲状腺乳头状癌组织中炎性因子的表达及其临床意义 被引量:3

目的:探讨甲状腺乳头状癌组织中炎性因子的表达及其与甲状腺乳头状癌临床病理特征的关系,为指导临床用药和判断预后奠定实验基础。方法:选取穿刺确诊为甲状腺乳头状癌的74例住院患者作为实验组,另选同期26名体检健康者作为正常对照组。... 目的:探讨甲状腺乳头状癌组织中炎性因子的表达及其与甲状腺乳头状癌临床病理特征的关系,为指导临床用药和判断预后奠定实验基础。方法:选取穿刺确诊为甲状腺乳头状癌的74例住院患者作为实验组,另选同期26名体检健康者作为正常对照组。2组受试者均空腹抽取静脉血,实验组患者于术后留取癌组织、癌旁正常组织(距癌组织2cm)及术后7d再次空腹抽取静脉血。采用酶联免疫法检测血清、癌旁正常组织及癌组织中白细胞介素1α(IL-1α)、IL-1β和环氧合酶-2(COX-2)蛋白的表达水平;采用实时荧光定量法和免疫组织化学方法检测甲状腺乳头状癌组织及癌旁正常组织中IL-1α、IL-1β和COX-2的mRNA和蛋白表达水平;分析IL-1α、IL-1β和COX-2表达与甲状腺乳头状癌的临床分期、病理类型及有无淋巴结转移的关系。结果:实验组患者血清中IL-1α、IL-1β和COX-2的蛋白表达水平明显高于正常对照组(P<0.01);甲状腺乳头状癌患者癌组织中IL-1α、IL-1β和COX-2的蛋白表达水平、蛋白阳性表达强度及mRNA表达水平明显高于癌旁正常组织(P<0.01)。在甲状腺乳头状癌组织中IL-1α与IL-1β、IL-1α与COX-2、IL-1β与COX-2均呈正相关关系(r=0.64,P=0.035;r=0.71,P=0.042;r=0.69,P=0.038)。结论:甲状腺乳头状癌组织中IL-1α、IL-1β和COX-2表达水平上调,提示炎性因子参与甲状腺乳头状癌浸润和转移,其应用有望成为新的肿瘤治疗策略和手段。 展开更多

Response of white blood cells and fibroblasts in the gingival fluid and the mixed saliva of the patients a long time after prosthetic repair, relying on the content of proinflammatory interleukins IL-1 β and IL-6, anti-inflammatory interleukins IL-4 and IL-10, the factor of tumor necrosis TNF-α and lactoferrin, has been investigated. 下调IL35通过上调自噬相关蛋白表达而抑制肝细胞癌侵袭和迁移 被引量:4

目的探讨白细胞介素35(interleukin 35,IL35)在肝癌中的表达及其临床意义、潜在的分子机制。方法收集手术确诊的肝细胞肝癌和癌旁组织40对。采用RT-qPCR和Western blot检测IL35在肝癌和癌旁组织中的表达,并分析IL35与各临床指标的相关性... 目的探讨白细胞介素35(interleukin 35,IL35)在肝癌中的表达及其临床意义、潜在的分子机制。方法收集手术确诊的肝细胞肝癌和癌旁组织40对。采用RT-qPCR和Western blot检测IL35在肝癌和癌旁组织中的表达,并分析IL35与各临床指标的相关性。Western blot检测IL35在肝癌细胞中的表达。慢病毒转染下调IL35。CCK-8和集落形成检测细胞增殖,Transwell实验检测侵袭和迁移,Western blot检测细胞自噬和EMT相关指标。结果IL35高表达于肝癌组织中(P<0.01);与HL7702肝细胞相比,IL35蛋白表达在Hep3B、Huh7、SMMC7721、Bel7402细胞中升高,且Hep3B表达最高(P<0.01);高表达与血管侵犯相关(P=0.0033);Transwell实验表明下调L35能有效抑制肝癌细胞侵袭和转移(P<0.01);CCK8和平板克隆实验表明,下调IL35能有效抑制肝癌细胞增殖(P<0.01);Western blot结果显示,下调IL35能促进LC3BII、Beclin1表达,抑制p62表达(P<0.01),同时促进E-cadherin表达并抑制N-cadherin、Vimentin的表达(P<0.01)。结论IL35高表达于肝细胞肝癌组织和肝癌细胞中,其表达与肝癌的血管侵犯相关;下调IL35能通过诱导自噬达到抑制肝癌细胞的侵袭和迁移的目的。 展开更多

目的:通过观察通利三焦、清热利湿方对IgA肾病大鼠肾组织miRNA-148b及血清IL-4表达水平的影响,探求本方干预IgA肾病的作用机制.方法:将48只健康清洁级雄性SD大鼠适应性喂养一周后,尿蛋白阴性者进入实验,按随机数字表法分为4组:正常组、... 目的:通过观察通利三焦、清热利湿方对IgA肾病大鼠肾组织miRNA-148b及血清IL-4表达水平的影响,探求本方干预IgA肾病的作用机制.方法:将48只健康清洁级雄性SD大鼠适应性喂养一周后,尿蛋白阴性者进入实验,按随机数字表法分为4组:正常组、模型组、贝那普利组与中药组,每组12只,使用牛血清白蛋白灌胃+脂多糖+四氯化碳+蓖麻油免疫诱导的方法制备实验性IgA肾病大鼠模型,予模型组、贝那普利组、中药组3组隔日灌胃牛血清白蛋白、每周皮下注射蓖麻油与四氯化碳、每4周尾静脉注射脂多糖,正常组予等体积的蒸馏水与生理盐水代替,造模持续8周.于第9周、第13周末观察BUN、Scr、24h-UTP的水平、光镜及电镜下肾脏组织病理改变及IgA免疫荧光检测结果确定造模是否成功,造模成功后,中药组予通利三焦、清热利湿方中药灌胃干预,贝那普利组每日予质量1.8 mg的贝那普利灌胃给药,正常组与模型组予等体积蒸馏水灌胃.干预治疗4周后运用ELISA法检测血清中IL-4含量变化,用RT-PCR法检测大鼠肾脏组织中IL-4、miRNA-148b的表达.结果:模型组大鼠出现精神萎靡、纳食减少、毛色黯淡粗糙等表现,且尿蛋白异常增多,HE、PASM染色可见系膜细胞增生,肾间质炎性细胞浸润明显,毛细血管袢变窄,IgA免疫荧光呈团块状荧光沉积,模型制备成功.模型组尿蛋白较正常组显著增多(P<0.05);贝那普利组与中药组尿蛋白较模型组改善(P<0.05),其中中药组改善效果优于贝那普利组(P<0.05).模型组光镜下系膜细胞及基质增生明显增多,肾小球基底膜增厚,毛细血管袢部分闭塞;荧光下见团块状荧光表达;电镜下足突中度融合,免疫复合物沉积.贝那普利组与中药组肾组织病理改变较模型组改善.模型组肾组织miRNA-148b及血清IL-4表达水平较正常组升高(P<0.05);与模型组比较,贝那普利组与中药组上述指标均显著下降(P<0.05),其中中药组改善效果优于贝那普利组(P<0.05).结论:通利三焦、清热利湿方可能通过下调肾组织miRNA-148b及血清IL-4表达水平发挥治疗IgA肾病的作用. 展开更多
