以新疆石河子地区月季花为试材,采用Illumina(Mi Seq)平台进行26S r DNA高通量测序分析、α多样性分析、可培养酵母菌的分离纯化技术(稀释涂布法和孢子弹射法)、26S r DNA D1/D区测序鉴定、糖发酵及碳同化等生理生化实验,研究了月季花...以新疆石河子地区月季花为试材,采用Illumina(Mi Seq)平台进行26S r DNA高通量测序分析、α多样性分析、可培养酵母菌的分离纯化技术(稀释涂布法和孢子弹射法)、26S r DNA D1/D区测序鉴定、糖发酵及碳同化等生理生化实验,研究了月季花上附生酵母菌(可培养和不可培养)菌群多样性,并对系统进化进行分析,以期为有效开发利用新疆酵母菌资源奠定基础,并为探索花蜜微生物与植物相互作用提供理论依据。结果表明:经26S r DNA高通量测序筛选后得到41 337个优化序列,划分入3 078个OTU,月季附生酵母菌群落结构丰富,鉴定为3个门、10个纲、13个目、22个科、37个属、90个种。担子菌门(Basidiomycota)为优势门、占18.9%,线黑粉菌属(Filobasidium)为优势属、占12.2%,酵母菌种群的丰度存在差异;可培养的酵母菌共鉴定出3个酵母菌属4个已知种群,分别为季也蒙毕赤酵母(Meyerozyma guilliermondii)、粘性红酵母(Rhodotorula mucilaginosa)、乌兹别克斯坦隐球酵母(Cryptococcus uzbekistanensis)、浅白隐球酵母(Cryptococcus albidus),优势酵母属为隐球酵母属(Cryptococcus)。展开更多
