Glutathione-S-transferase(GST,EC2.5.1.18)multifunctional protease is important for detoxification,defense against biotic and abiotic stresses,and secondary metabolic material transport for plant growth and development...Glutathione-S-transferase(GST,EC2.5.1.18)multifunctional protease is important for detoxification,defense against biotic and abiotic stresses,and secondary metabolic material transport for plant growth and development.In this study,71 members of the BpGST family were identified from the entire Betula platyphylla Suk.genome.Most of the members encode proteins with amino acid lengths ranging from 101 to 875 and were localized to the cytoplasm by a prediction.BpGSTs can be divided into seven subfamilies,with a majority of birch U and F subfamily members according to gene structure,conserved motifs and evolutionary analysis.GST family genes showed collinearity with 22 genes in Oryza sativa L.,and three genes in Arabidopsis thaliana;promoter cis-acting elements predicted that the GST gene family is functional in growth,hormone regulation,and abiotic stress response.Most members of the F subfamily of GST(BpGSTFs)were expressed in roots,stems,leaves,and petioles,with the most expression observed in leaves.On the basis of the expression profiles of F subfamily genes(BpGSTF1 to BpGSTF13)during salt,mannitol and ABA stress,BpGSTF proteins seem to have multiple functions depending on the type of abiotic stress;for instance,BpGSTs may function at different times during abiotic stress.This study enhances understanding of the GST gene family and provides a basis for further exploration of their function in birch.展开更多
Small heat shock proteins(sHSPs)act as molecular chaperones that can prevent the accumulation of damaged proteins during abiotic stress,especially heat shock,but the mechanism is not clear.To study the function of sHS...Small heat shock proteins(sHSPs)act as molecular chaperones that can prevent the accumulation of damaged proteins during abiotic stress,especially heat shock,but the mechanism is not clear.To study the function of sHSPs in Lenzites gibbosa,a common polypore in northern temperate forests that causes spongy white rot of broadleaf trees,under temperature stress,L.gibbosa mycelia were grown at 25℃ for 9 d,treated at 33℃ for 15,30,60,and 120 min before sequencing the transcriptomes.From among 32 heat shock protein(HSP)genes found in the screen of the transcriptome data,a highly expressed gene was cloned and named Lghsp17.4.RT-qPCR was used to analyze the expression of the gene Lghsp17.4 under heat shock and dye stress.Both treatments induced higher expression of Lghsp17.4 at the transcriptional level,indicating that Lghsp17.4 might function in the response to heat stress and dye degradation.We previously found that L.gibbosa generally had a heat shock reaction(HSR)during degradation of aromatic compounds,and HSPs were always produced with manganese peroxidases(MnPs)and other lignin-degrading enzymes.Therefore,we measured the activity of MnPs in L.gibbosa after 33℃ heat shock to analyze the relationship between MnPs expression and Lghsp17.4 expression.Heat shocks of 0–30 min increased MnPs activity,and the change in MnPs activity were closely positively correlated with the expression levels of Lghsp17.4 over time,indicating a potential connection and interaction between LgHSP17.4 and MnPs during the HSR in L.gibbosa.Thus,LgHSP17.4 might have a positive regulatory effect on the HSR in L.gibbosa and be a critical component of a stress resistance mechanism.展开更多
Heat shock transcription factors(Hsfs)have important roles during plant growth and development and responses to abiotic stresses.The identification and func-tion of Hsf genes have been thoroughly studied in various he...Heat shock transcription factors(Hsfs)have important roles during plant growth and development and responses to abiotic stresses.The identification and func-tion of Hsf genes have been thoroughly studied in various herbaceous plant species,but not woody species,especially Phoebe bournei,an endangered,unique species in China.In this study,17 members of the Hsf gene family were identi-fied from P.bournei using bioinformatic methods.Phyloge-netic analysis indicated that PbHsf genes were grouped into three subfamilies:A,B,and C.Conserved motifs,three-dimensional structure,and physicochemical properties of the PbHsf proteins were also analyzed.The structure of the PbHsf genes varied in the number of exons and introns.Pre-diction of cis-acting elements in the promoter region indi-cated that PbHsf genes are likely involved in responses to plant hormones and stresses.A collinearity analysis dem-onstrated that expansions of the PbHsf gene family mainly take place via segmental duplication.The expression levels of PbHsf genes varied across different plant tissues.On the basis of the expression profiles of five representative PbHsf genes during heat,cold,salt,and drought stress,PbHsf pro-teins seem to have multiple functions depending on the type of abiotic stress.This systematic,genome-wide investigation of PbHsf genes in P.bournei and their expression patterns provides valuable insights and information for further func-tional dissection of Hsf proteins in this endangered,unique species.展开更多
The estimation of pain intensity is critical for medical diagnosis and treatment of patients.With the development of image monitoring technology and artificial intelligence,automatic pain assessment based on facial ex...The estimation of pain intensity is critical for medical diagnosis and treatment of patients.With the development of image monitoring technology and artificial intelligence,automatic pain assessment based on facial expression and behavioral analysis shows a potential value in clinical applications.This paper reports a framework of convolutional neural network with global and local attention mechanism(GLA-CNN)for the effective detection of pain intensity at four-level thresholds using facial expression images.GLA-CNN includes two modules,namely global attention network(GANet)and local attention network(LANet).LANet is responsible for extracting representative local patch features of faces,while GANet extracts whole facial features to compensate for the ignored correlative features between patches.In the end,the global correlational and local subtle features are fused for the final estimation of pain intensity.Experiments under the UNBC-McMaster Shoulder Pain database demonstrate that GLA-CNN outperforms other state-of-the-art methods.Additionally,a visualization analysis is conducted to present the feature map of GLA-CNN,intuitively showing that it can extract not only local pain features but also global correlative facial ones.Our study demonstrates that pain assessment based on facial expression is a non-invasive and feasible method,and can be employed as an auxiliary pain assessment tool in clinical practice.展开更多

ThGLP belongs to true germin subfamily through phylogenetic analyses. Gene expression profiles in roots and leaves were evaluated using real-time quantitative RT-PCR. The results show that the gene was highly induced by drought, salt, low temperature, CdCl2 and abscisic acid treatments. Our results demonstrate that the ThGLP gene is expressed in leaves and roots, is involved in different abiotic stress re-sponses and controlled by an ABA-dependent signaling pathway.展开更多

Methods: The total RNAs of matched BE and normal esophagus mucosa of BE patients were isolated using one step Trizol method. Matched RNAs were qualified using 10 g/L agarose gel electrophoresis. cRNAs were synthesized, fluorescence labeled and purified after total RNAs were purified. The RNAs of BE and normal esophagus mucosa were hybridized with Agilent oligomicroarray (30 968 probes). The fluorescence intensity features were detected by Agilent scanner and quantified by feature extraction software. Results: (1) The total RNA, reverse transcription product and fluorescence labeled cRNA were all of high quality; (2) There were 142 up-regulated genes and 284 down-regulated genes among 2-fold DEGs. Conclusion: Microarray-based studies are feasible in endoscopically obtained tissues. Many BE-associated genes are screened by the high-throughput gene chip. The development and progression of BE is a complicated process involving multiple genes and multiple procedures, and functional study of these genes may help to identify the key genes or pathways involved in the pathogenesis and development of BE.展开更多

Consequently, the copy number of specific gene was increased substantially, leading to the improvement of expression efficiency.Using this approach, a recombinant plasmid , designed as PLYD, was constructed and transformated into the Escherichia coli strain DH5α. Upon induction , the desired protein was synthesized in a considerable level and accumulated up to 63% of the total cell proteins. The present study revealed that tandem repeating of expression cartridge provided a convenient means to improve expression level efficiently.展开更多

Background:The aim of this work is to detect and compare the peripheral blood mi RNA expression profiles in patients with severe traumatic brain injury(s TBI)2,12,24,48,and 72 h after injury at high altitude and to pr...Background:The aim of this work is to detect and compare the peripheral blood mi RNA expression profiles in patients with severe traumatic brain injury(s TBI)2,12,24,48,and 72 h after injury at high altitude and to predict the target genes of differential expressed mi RNAs.Methods:Twenty s TBI patients from high-altitude areas were randomly selected according to the inclusion and exclusion criteria and were divided into five groups:the 2-h group,12-h group,24-h group,48-h group,and 72-h group.Peripheral blood mi RNA expression profiles were detected using real-time quantitative PCR(q RT-PCR).Results:The expression levels of mi R-18 a,mi R-203,mi R-146 a,mi R-149,mi R-23 b,and mi R-let-7 b in peripheral blood showed significant differences between the 2-h group and the 12-h group.The expression levels of mi R-203,mi R-146 a,mi R-149,mi R-23 b,and mi R-let-7 f in peripheral blood were up-regulated in the 24-h group.In the 48-h group,the expression levels of mi R-181 d,mi R-29 a,and mi R-18 b were upregulated.In the 72-h group,the expression levels of mi R-203,mi R-146 a,mi R-149,mi R-23 b,and mi R-let-7 f changed.The main target genes of the differentiation expressed mi RNAs were genes that regulate inflammatory responses,apoptosis,and DNA damage/repair.Conclusions:mi RNAs may be involved in the pathogenesis of s TBI by dynamically regulating the target genes that regulate inflammatory responses,apoptosis,and DNA damage/repair pathways.展开更多
CBF/DREB proteins play a critical role in abiotic stress-mediated gene expression and represent attractive regulons for plant breeding programs.However,no study has been conducted for CBF/DREB protein-related genes in...CBF/DREB proteins play a critical role in abiotic stress-mediated gene expression and represent attractive regulons for plant breeding programs.However,no study has been conducted for CBF/DREB protein-related genes in jujube(Ziziphus jujuba Mill.).In this study,twenty-five ZjDREB genes were identified and annotated from the jujube(Z.jujuba‘Dongzao’)genome.Detailed analysis,including gene classification,annotation,phylogenetic evaluation,conserved motif determination and expression profiling were performed on all genes.Phylogenetic analysis showed that ZjDREB proteins were divided into five subgroups(A1–A5),but lacking a subgroup A6 corresponding to AtDREBs.The ZjDREB genes were distributed in nine of twelve chromosomes in the genome.Additionally,the expression patterns of the DREB genes under different abiotic stresses were investigated using q RT-PCR.Nineteen ZjDREB genes were down-regulated under low temperature,in contrast six ZjDREB genes(01,03,05,11,23 and 24)were up-regulated.Under drought,salinity and high temperature conditions,expression of ZjDREB03,09,10,14,15,17 and 20 genes were induced and showed similar expression patterns,suggesting that various stress conditions share common elements in the signaling pathway.The results suggest that the family of DREB genes play an important role in abiotic stresses in jujube,and provide a foundation for further functional studies of this important class of transcriptional regulators.展开更多

Methods Mutated exon 5 of PTEN gene was examined in normal ovary(n = 5), ovarian cyst (n =5), ovarian borderline tumor (n = 9), epithelial ovarian cancer(n = 60), and ovarian cancer cell line (n = 1)by polymerase chain reaction-single strand conformation polymorphism (PCR-SSCP). mRNA expression of PTEN gene was evaluated in corresponding tissues and cell line by reverse transcription polymerase chain reaction(RT-PCR). The mutation and mRNA expression of PTEN gene were compared with clini-copathological features of ovarian cancer. Results Mutated exon 5 of PTEN gene was detected only in 5(7.1%)cases of epithelial ovarian cancer. mRNA expression level of PTEN gene in ovarian borderline tumor or ovarian cancer was lower than that in normal ovary or ovarian cyst(P < 0.05). The level of PTEN gene mRNA expression was negatively correlated with clinicopathological staging of ovarian cancer, whereas positively correlated with histological differentiation (P < 0.05). mRNA expression level of PTEN gene in ovarian endometrioid cancer was significantly lower than that in ovarian serous or mucinous cancer (P < 0.05=. Conclusions Mutation of PTEN gene occurs in ovarian cancer. Down-regulated expression of PTEN is probably an important molecular event in tumorigenesis of ovarian cancer. Abnormal expression of PTEN gene is involved in progression of ovarian cancer. Reduced expression of PTEN gene is closely associated with tumorigenesis and pathobiological behaviors of ovarian endometrioid cancer.展开更多

Cold-resistance pathways that operate in model plants such as Arabidopsis thaliana and Oryza sativa have been studied extensively.It has been found that CBF genes play an important role in plant cold resistance.Liriod...Cold-resistance pathways that operate in model plants such as Arabidopsis thaliana and Oryza sativa have been studied extensively.It has been found that CBF genes play an important role in plant cold resistance.Liriodendron chinense,a tree known for its graceful tree shape and widely spread in south China,has weak cold tolerance.However,little is known about its response to cold.To further study the function of L.chinense CBF gene family,we started by characterizing all members of this gene family in the L.chinense genome and their expression profiling.Phylogenetic analysis found that 14 CBF genes in L.chinense are more closely related to their homologues in woody plants and A.thaliana than those in O.sativa.Cis-acting elements and GO analysis showed that some LcCBF genes participated in the biological process of cold stress response.The transcriptomic and RT-qPCR data showed that most of LcCBF genes displayed an initially increasing and subsequently decreasing trend during cold stress course and the expression profile of each member was different.Some LcCBF genes exhibited a different abundance in callus,root,stem and leaf tissues.The structure and expression characteristics of LcCBF genes imply that they may have similar and different functions in response to cold stress conditions.The identification and analysis of LcCBF gene family have laid the foundation for future studies into L.chinense cold stress mechanisms and for the cultivation of cold-resistance cultivars.展开更多

The experiment was designed to study the association of cerebral ischemia and reperfusion with endothelin- 1 (ET- 1) gene expression of rat brains and time-dependent changes of ET- 1 gene expression during cerebral ischemia.Materials and methods. Thirty- three male SD rats were divided into dot blot hybridization(n = 27) and in silu hybridization groups(n= 6). The focal cerebral ischemia and reperfusion models were made with suture embolism of middle cerebral artery. Dot blot hybridization groups were redivided into control and ischemic subgroups (ischemia for 0. 5 , 1 , 1. 5 , 3 , 6 , 12 , 24 , 48 and 72 h respectively). In situ hybridization groups were redivided into ischemia and reperfusion groups. After 24 h ischemia and 24 h reperfusion,ET1 gene expressions were investigated with in situ hybridization and the resuhs were analyzed with IBAS 2000 Image Analysis System.Results. Dot blot hybridization showed that ET-1 mRNA of cerebral cortex and caudate- putamen was increased at 6 h of ischemia and reached peak at 24 h (3. 9 and 3. 7 fold respectively) ,and at 72 h of ischemia it remained at high levels(3. 5 and 2. 1 fold respectively). In silu hybridization showed that the levels of ET- 1 mRNA of cerebral cortex and caudate-putamen were also markedly increased both in 24 h ischemia and 24 h reperfusion groups (P<0. 01 , P<0. 05 respectively) .Conclusions. ET-1 gene expression in focal ischemic brain tissue were markedly and progressively increased during cerebral ischemia and reperfusion and downregulation of ET- 1 gene expression may be a new approach to the treatment of ischemic cerebrovascular diseases.展开更多

Methods Interleukin(IL)-2 and interferon-3, represent Thl type cytokines. IL-4, IL-6, IL-10, and IL-13 represent Th2 type cytokines. The gene expressions of Th1/Th2 cytokines in human glioma cells, glioma infiltrating lymphocytes, and glioma cell lines were detected by reverse transcription polymerase chain reaction (RT-PCR). The biological activity of cytokines in the supematant of glioma cell lines was assayed by enzyme-linked immunosorbent assay (ELISA) method. Results The total positive rates of Th1 and Th2 type cytokines gene in human glioma cells were 14.77% and 75%. The total positive rates of Th1 and Th2 type cytokines gene in glioma infiltrating lymphocytes were 22.73% and 68.17%. There was obviously predominant expression of Th2 type cytokines in human glioma tissues, glioma infiltrating lymphocytes, and glioma cell lines. There was no unbalanced expression of Th1/Th2 cytokines in normal brain tissues. Conclusion There is a predominant expression of Th2 type cytokines in human glioma cells. The switching of Th1/Th2 cytokines gene may play an important role in the occurring and developing of human gliomas.展开更多

文摘Heat shock transcription factors(Hsfs)have important roles during plant growth and development and responses to abiotic stresses.The identification and func-tion of Hsf genes have been thoroughly studied in various herbaceous plant species,but not woody species,especially Phoebe bournei,an endangered,unique species in China.In this study,17 members of the Hsf gene family were identi-fied from P.bournei using bioinformatic methods.Phyloge-netic analysis indicated that PbHsf genes were grouped into three subfamilies:A,B,and C.Conserved motifs,three-dimensional structure,and physicochemical properties of the PbHsf proteins were also analyzed.The structure of the PbHsf genes varied in the number of exons and introns.Pre-diction of cis-acting elements in the promoter region indi-cated that PbHsf genes are likely involved in responses to plant hormones and stresses.A collinearity analysis dem-onstrated that expansions of the PbHsf gene family mainly take place via segmental duplication.The expression levels of PbHsf genes varied across different plant tissues.On the basis of the expression profiles of five representative PbHsf genes during heat,cold,salt,and drought stress,PbHsf pro-teins seem to have multiple functions depending on the type of abiotic stress.This systematic,genome-wide investigation of PbHsf genes in P.bournei and their expression patterns provides valuable insights and information for further func-tional dissection of Hsf proteins in this endangered,unique species.

文摘The estimation of pain intensity is critical for medical diagnosis and treatment of patients.With the development of image monitoring technology and artificial intelligence,automatic pain assessment based on facial expression and behavioral analysis shows a potential value in clinical applications.This paper reports a framework of convolutional neural network with global and local attention mechanism(GLA-CNN)for the effective detection of pain intensity at four-level thresholds using facial expression images.GLA-CNN includes two modules,namely global attention network(GANet)and local attention network(LANet).LANet is responsible for extracting representative local patch features of faces,while GANet extracts whole facial features to compensate for the ignored correlative features between patches.In the end,the global correlational and local subtle features are fused for the final estimation of pain intensity.Experiments under the UNBC-McMaster Shoulder Pain database demonstrate that GLA-CNN outperforms other state-of-the-art methods.Additionally,a visualization analysis is conducted to present the feature map of GLA-CNN,intuitively showing that it can extract not only local pain features but also global correlative facial ones.Our study demonstrates that pain assessment based on facial expression is a non-invasive and feasible method,and can be employed as an auxiliary pain assessment tool in clinical practice.

文摘Germin and Germin-like protein (GLP) have various proposed roles in plant developmental stages and stress- related processes. A novel GLP cDNA clone was isolated from a cDNA library of Tamarix hispida. ThGLP, coded 225aa, possesses conserved motif of plant germin and Germin-like protein. ThGLP belongs to true germin subfamily through phylogenetic analyses. Gene expression profiles in roots and leaves were evaluated using real-time quantitative RT-PCR. The results show that the gene was highly induced by drought, salt, low temperature, CdCl2 and abscisic acid treatments. Our results demonstrate that the ThGLP gene is expressed in leaves and roots, is involved in different abiotic stress re-sponses and controlled by an ABA-dependent signaling pathway.

文摘In this paper, the theoretical expressions of wood thermal conductiv ity in the choral and radical direction are derived from the micro-structure of wood by applying some basic principles in physical mechanics. The thermal conduc tivities of about twenty species of trees were calculated by means of the expres sions and compared with its experimental values under the same condition. The av erage relative error is about 5%, so the calculation result is satisfactory.

文摘From viewpoint of chemical element and microstructure of wood, this paper makes a discussion on thermal diffusivity of wood and two theoretical expressions of thermal diffusivity for the choral and radial directions were derived. The thermal diffusivities of the choral and radial directions for about 20 species of trees were calculated with the derived theoretical expressions and compared with the experimental values. The average error of the theoretical values of thermal diffusivity was 7.5% for choral direction and 6.2% for radial direction.
文摘This is a research report on the interrelationship among understanding, memory and oral expression in listening comprehension training. The research investigated 150 non-English majors through a questionnaire in a higher vocational college. The result indicates that the interrelationship among understanding, memory and oral expression in listening classes which are determined by the listening materials and teaching method directly influences students' listening proficiency and speaking ability. It is suggested that some strategies to be used to foster students' abilities in these three areas so as to improve their listening ability.
文摘Objective: To analyze the differential expression genes (DEGs) between Barrett’s esophagus (BE) and normal esophagus mucosa and explore the target genes related to the development and progression of BE. Methods: The total RNAs of matched BE and normal esophagus mucosa of BE patients were isolated using one step Trizol method. Matched RNAs were qualified using 10 g/L agarose gel electrophoresis. cRNAs were synthesized, fluorescence labeled and purified after total RNAs were purified. The RNAs of BE and normal esophagus mucosa were hybridized with Agilent oligomicroarray (30 968 probes). The fluorescence intensity features were detected by Agilent scanner and quantified by feature extraction software. Results: (1) The total RNA, reverse transcription product and fluorescence labeled cRNA were all of high quality; (2) There were 142 up-regulated genes and 284 down-regulated genes among 2-fold DEGs. Conclusion: Microarray-based studies are feasible in endoscopically obtained tissues. Many BE-associated genes are screened by the high-throughput gene chip. The development and progression of BE is a complicated process involving multiple genes and multiple procedures, and functional study of these genes may help to identify the key genes or pathways involved in the pathogenesis and development of BE.
文摘A novel strategy to enhance the expression efficiency of cloned target gene in Escherichia coli was developed. The whole expression cartridge , consisting of promoter. SD sequence , target gene and transcription terminator, was tandem repeatedly engineered into a expression plasmid. Consequently, the copy number of specific gene was increased substantially, leading to the improvement of expression efficiency.Using this approach, a recombinant plasmid , designed as PLYD, was constructed and transformated into the Escherichia coli strain DH5α. Upon induction , the desired protein was synthesized in a considerable level and accumulated up to 63% of the total cell proteins. The present study revealed that tandem repeating of expression cartridge provided a convenient means to improve expression level efficiently.

文摘Background:The aim of this work is to detect and compare the peripheral blood mi RNA expression profiles in patients with severe traumatic brain injury(s TBI)2,12,24,48,and 72 h after injury at high altitude and to predict the target genes of differential expressed mi RNAs.Methods:Twenty s TBI patients from high-altitude areas were randomly selected according to the inclusion and exclusion criteria and were divided into five groups:the 2-h group,12-h group,24-h group,48-h group,and 72-h group.Peripheral blood mi RNA expression profiles were detected using real-time quantitative PCR(q RT-PCR).Results:The expression levels of mi R-18 a,mi R-203,mi R-146 a,mi R-149,mi R-23 b,and mi R-let-7 b in peripheral blood showed significant differences between the 2-h group and the 12-h group.The expression levels of mi R-203,mi R-146 a,mi R-149,mi R-23 b,and mi R-let-7 f in peripheral blood were up-regulated in the 24-h group.In the 48-h group,the expression levels of mi R-181 d,mi R-29 a,and mi R-18 b were upregulated.In the 72-h group,the expression levels of mi R-203,mi R-146 a,mi R-149,mi R-23 b,and mi R-let-7 f changed.The main target genes of the differentiation expressed mi RNAs were genes that regulate inflammatory responses,apoptosis,and DNA damage/repair.Conclusions:mi RNAs may be involved in the pathogenesis of s TBI by dynamically regulating the target genes that regulate inflammatory responses,apoptosis,and DNA damage/repair pathways.

文摘CBF/DREB proteins play a critical role in abiotic stress-mediated gene expression and represent attractive regulons for plant breeding programs.However,no study has been conducted for CBF/DREB protein-related genes in jujube(Ziziphus jujuba Mill.).In this study,twenty-five ZjDREB genes were identified and annotated from the jujube(Z.jujuba‘Dongzao’)genome.Detailed analysis,including gene classification,annotation,phylogenetic evaluation,conserved motif determination and expression profiling were performed on all genes.Phylogenetic analysis showed that ZjDREB proteins were divided into five subgroups(A1–A5),but lacking a subgroup A6 corresponding to AtDREBs.The ZjDREB genes were distributed in nine of twelve chromosomes in the genome.Additionally,the expression patterns of the DREB genes under different abiotic stresses were investigated using q RT-PCR.Nineteen ZjDREB genes were down-regulated under low temperature,in contrast six ZjDREB genes(01,03,05,11,23 and 24)were up-regulated.Under drought,salinity and high temperature conditions,expression of ZjDREB03,09,10,14,15,17 and 20 genes were induced and showed similar expression patterns,suggesting that various stress conditions share common elements in the signaling pathway.The results suggest that the family of DREB genes play an important role in abiotic stresses in jujube,and provide a foundation for further functional studies of this important class of transcriptional regulators.
文摘Objective To investigate the mutation and expression of tumor suppressor gene-PTEN mRNA and explore their roles in tumorigenesis and progression of ovarian cancer. Methods Mutated exon 5 of PTEN gene was examined in normal ovary(n = 5), ovarian cyst (n =5), ovarian borderline tumor (n = 9), epithelial ovarian cancer(n = 60), and ovarian cancer cell line (n = 1)by polymerase chain reaction-single strand conformation polymorphism (PCR-SSCP). mRNA expression of PTEN gene was evaluated in corresponding tissues and cell line by reverse transcription polymerase chain reaction(RT-PCR). The mutation and mRNA expression of PTEN gene were compared with clini-copathological features of ovarian cancer. Results Mutated exon 5 of PTEN gene was detected only in 5(7.1%)cases of epithelial ovarian cancer. mRNA expression level of PTEN gene in ovarian borderline tumor or ovarian cancer was lower than that in normal ovary or ovarian cyst(P < 0.05). The level of PTEN gene mRNA expression was negatively correlated with clinicopathological staging of ovarian cancer, whereas positively correlated with histological differentiation (P < 0.05). mRNA expression level of PTEN gene in ovarian endometrioid cancer was significantly lower than that in ovarian serous or mucinous cancer (P < 0.05=. Conclusions Mutation of PTEN gene occurs in ovarian cancer. Down-regulated expression of PTEN is probably an important molecular event in tumorigenesis of ovarian cancer. Abnormal expression of PTEN gene is involved in progression of ovarian cancer. Reduced expression of PTEN gene is closely associated with tumorigenesis and pathobiological behaviors of ovarian endometrioid cancer.

文摘Cold-resistance pathways that operate in model plants such as Arabidopsis thaliana and Oryza sativa have been studied extensively.It has been found that CBF genes play an important role in plant cold resistance.Liriodendron chinense,a tree known for its graceful tree shape and widely spread in south China,has weak cold tolerance.However,little is known about its response to cold.To further study the function of L.chinense CBF gene family,we started by characterizing all members of this gene family in the L.chinense genome and their expression profiling.Phylogenetic analysis found that 14 CBF genes in L.chinense are more closely related to their homologues in woody plants and A.thaliana than those in O.sativa.Cis-acting elements and GO analysis showed that some LcCBF genes participated in the biological process of cold stress response.The transcriptomic and RT-qPCR data showed that most of LcCBF genes displayed an initially increasing and subsequently decreasing trend during cold stress course and the expression profile of each member was different.Some LcCBF genes exhibited a different abundance in callus,root,stem and leaf tissues.The structure and expression characteristics of LcCBF genes imply that they may have similar and different functions in response to cold stress conditions.The identification and analysis of LcCBF gene family have laid the foundation for future studies into L.chinense cold stress mechanisms and for the cultivation of cold-resistance cultivars.

The experiment was designed to study the association of cerebral ischemia and reperfusion with endothelin- 1 (ET- 1) gene expression of rat brains and time-dependent changes of ET- 1 gene expression during cerebral ischemia.Materials and methods. Thirty- three male SD rats were divided into dot blot hybridization(n = 27) and in silu hybridization groups(n= 6). The focal cerebral ischemia and reperfusion models were made with suture embolism of middle cerebral artery. Dot blot hybridization groups were redivided into control and ischemic subgroups (ischemia for 0. 5 , 1 , 1. 5 , 3 , 6 , 12 , 24 , 48 and 72 h respectively). In situ hybridization groups were redivided into ischemia and reperfusion groups. After 24 h ischemia and 24 h reperfusion,ET1 gene expressions were investigated with in situ hybridization and the resuhs were analyzed with IBAS 2000 Image Analysis System.Results. Dot blot hybridization showed that ET-1 mRNA of cerebral cortex and caudate- putamen was increased at 6 h of ischemia and reached peak at 24 h (3. 9 and 3. 7 fold respectively) ,and at 72 h of ischemia it remained at high levels(3. 5 and 2. 1 fold respectively). In silu hybridization showed that the levels of ET- 1 mRNA of cerebral cortex and caudate-putamen were also markedly increased both in 24 h ischemia and 24 h reperfusion groups (P<0. 01 , P<0. 05 respectively) .Conclusions. ET-1 gene expression in focal ischemic brain tissue were markedly and progressively increased during cerebral ischemia and reperfusion and downregulation of ET- 1 gene expression may be a new approach to the treatment of ischemic cerebrovascular diseases.
文摘Objective To study the expression and switching of Thl/Th2 cytokines gene in human gliomas and its effects on occurring and developing of human gliomas. Methods Interleukin(IL)-2 and interferon-3, represent Thl type cytokines. IL-4, IL-6, IL-10, and IL-13 represent Th2 type cytokines. The gene expressions of Th1/Th2 cytokines in human glioma cells, glioma infiltrating lymphocytes, and glioma cell lines were detected by reverse transcription polymerase chain reaction (RT-PCR). The biological activity of cytokines in the supematant of glioma cell lines was assayed by enzyme-linked immunosorbent assay (ELISA) method. Results The total positive rates of Th1 and Th2 type cytokines gene in human glioma cells were 14.77% and 75%. The total positive rates of Th1 and Th2 type cytokines gene in glioma infiltrating lymphocytes were 22.73% and 68.17%. There was obviously predominant expression of Th2 type cytokines in human glioma tissues, glioma infiltrating lymphocytes, and glioma cell lines. There was no unbalanced expression of Th1/Th2 cytokines in normal brain tissues. Conclusion There is a predominant expression of Th2 type cytokines in human glioma cells. The switching of Th1/Th2 cytokines gene may play an important role in the occurring and developing of human gliomas.
