摘要
了解巨须裂腹鱼(Schizothorax macropogon)在藏木鱼道和加查鱼道过坝的基因交流需求及其受到的过滤作用,为鱼道精细化运行管理提供数据支撑和参考依据。2020—2021年通过分析采自藏木水电站上游、藏木鱼道、藏木水电站下游、加查鱼道和加查水电站下游等5个群体共217尾巨须裂腹鱼的线粒体Cyt b基因序列,计算遗传多样性,获得其遗传结构特征。结果表明:(1)藏木水电站和加查水电站上游与下游巨须裂腹鱼群体遗传多样性水平相近,单倍型多样性范围为0.821~0.879,核苷酸多样性范围为0.00226~0.00244,两两群体间基因交流频繁(45.41<|Nm|<2083.83),未发生遗传分化(-0.0110<FST<0.0109;P>0.05),应作为同一个管理单元进行保护;巨须裂腹鱼在藏木水电站和加查水电站具有过坝进行基因交流的需求,从遗传多样性保护的角度,被列为过鱼对象是合理的。(2)遗传多样性分析结果显示,藏木鱼道和加查鱼道对巨须裂腹鱼没有明显过滤作用。
Schizothorax macropogon is primarily distributed in the mainstream and tributaries of the upper and middle Yarlung Zangbo River and it is a primary target fish species for the fish passages at Zangmu and Jiacha hydropower stations.Based on Cyt b gene sequence analyses,we determined the genetic diversity and genetic structure of five S.macropogon populations from upstream of Zangmu hydropower station to below Jiacha hydropower station,including the fishways of both stations.We also analyzed the attenuation of S.macropogon populations by the fishways and the need for assisting gene flow in upstream populations.During 2020⁃2021,a total of 217 S.macropogon samples from the five geographic populations were collected for DNA extraction and Cyt b gene sequencing.Results show that:(1)The genetic diversity of the five S.macropogon populations were similar,with a haplotype diversity range of 0.821⁃0.879 and a nucleotide diversity range of 0.00226⁃0.00244.The gene flow between populations was frequent(45.41<|Nm|<2083.83),with no detected genetic divergence(-0.0110<FST<0.0109;P>0.05).The five geographic populations were therefore defined as one genetic management unit.Gene flow in S.macropogon passing through Zangmu fishway and Jiacha fishway is necessary to avoid inbreeding.Thus,it is reasonable to select S.macropogon as a target species for the two fishways.(2)There was no obvious attenuation of the S.macropogon haplotype by the fishways.These results provide data support and a theoretical reference for selecting target fish species and optimizing fishway operation.
