头孢噻肟钠对鲟源嗜水气单胞菌和弗氏柠檬酸杆菌的体外药效学研究

In vitro pharmacodynamic study of cefotaxime sodium on Aeromonas hydrophila and Citrobacter freundii from sturgeon

摘要 为筛选出针对中华鲟细菌性疾病的治疗药物,并为其使用提供参考,针对鲟源嗜水气单胞菌和弗氏柠檬酸杆菌进行了药物敏感性试验,并测定了头孢噻肟钠对2种菌株的最小抑菌浓度(MIC),建立了1、2、4、8倍MIC下头孢噻肟钠对2种菌株的杀菌动力学曲线,测定了该药对2种细菌的抗菌后效应(PAE)。药敏试验结果显示,2种菌株对4种头孢菌素(头孢噻肟、头孢曲松、头孢吡肟和头孢哌酮)敏感或中度敏感,对多黏菌素B、复方新诺明、庆大霉素、红霉素、氟苯尼考、恩诺沙星等9种药物存在不同程度的耐药。头孢噻肟钠对2种菌株的MIC均为1 mg/L。杀菌动力学曲线显示,头孢噻肟钠对2种菌株的抑菌效果表现出强时间依赖性及浓度依赖性。PAE结果显示,2、4、8倍MIC的头孢噻肟钠对弗氏柠檬酸杆菌的体外PAE分别为(0.22±0.12)、(0.39±0.09)、(0.67±0.20)h,对嗜水气单胞菌的体外PAE分别为(1.24±0.17)、(2.64±0.31)、(2.96±0.28)h。研究结果表明,头孢噻肟钠对鲟源嗜水气单胞菌和弗氏柠檬酸杆菌具有较好的体外抑制效果。 To screen potential therapeutic drug for bacterial diseases in Acipenser sinensis caused by Aeromonas hy-drophila and Citrobacter freundii,and to provide a theoretical reference for its application,drug sensitivity tests were conducted for A.hydrophila and C.freundii isolated from sturgeon,and the minimum inhibitory concentra-tion(MIC)of cefotaxime sodium against the two strains was determined.The bactericidal kinetic curves of cefo-taxime sodium against the two strains were established at 1×MIC,2×MIC,4×MIC,and 8×MIC concentrations,and the post-antibiotic effect(PAE)was determined.The results of the drug sensitivity test showed that both strains were sensitive or moderately sensitive to four cephalosporins(cefotaxime,ceftiaxone,cefepime,and cefoperazone),and had varying degrees of resistance to nine drugs including polymyxin B,cotrimoxazole,genta-micin,erythromycin,florfenicol and enrofloxacin.The MIC of cefotaxime sodium against C.freundii and A.hy-drophila was both 1 mg/L.The bactericidal kinetic curves showed that the antibacterial effect of cefotaxime sodium on the two strains was strongly time-and concentration-dependent.The in vitro PAE of cefotaxime sodium at 2×MIC,4×MIC,and 8×MIC concentration against C.freundii was(0.22±0.12)h,(0.39±0.09)h,and(0.67±0.20)h respectively.The in vitro PAE for A.hydrophila were(1.24±0.17)h,(2.64±0.31)h,and(2.96±0.28)h,respectively.The findings indicate that cefotaxime sodium exerts a strong in vitro inhibitory effect on A.hydrophila and C.freundii isolated from sturgeon.
