摘要
以兰属然洛组织培养根状茎和组织培养苗为试验材料,研究5种不同发光二极管(LED)光照[红光(R)、红光∶蓝光(B)=6∶4(R6B4)、红光∶蓝光=5∶5(R5B5)、红光∶蓝光=4∶6(R4B6)、蓝光]对兰属然洛根状茎增殖分化和组织培养苗壮苗生根阶段生长和生理生化性质的影响。结果表明,在根状茎增殖分化阶段,与荧光灯对照(CK)相比,红光∶蓝光=5∶5(R5B5)处理兰属然洛组织培养根状茎粗度显著增加、分化率最高;可溶性蛋白含量随蓝光比例的增加呈现先降低后上升的趋势,而可溶性糖含量则呈现先增加后降低趋势,R6B4处理最高;过氧化氢含量、活性氧含量随蓝光比例增加而减少,而过氧化氢酶活性随蓝光比例增加而增强,R5B5处理的过氧化氢含量与CK相比差异不显著,而活性氧含量和过氧化氢酶活性与CK相比显著增加。在壮苗生根阶段,R5B5处理的组织培养苗叶长、叶宽、假鳞茎宽、叶片数、株高、根长、根粗、地上部鲜重以及叶绿素a含量、叶绿素b含量、类胡萝卜素含量和总叶绿素含量较高;组织培养苗可溶性糖含量均随着红光比例的降低先升高后降低,与CK相比R5B5处理降低最小;R5B5处理CRY2基因的相对表达量最高。综上所述,不同LED光照处理对兰属然洛根状茎的增殖分化以及组织培养苗的生长、叶绿素含量、可溶性蛋白含量、可溶性糖含量、抗氧化系统、CRY2基因表达均有不同程度的影响,以R5B5处理的效果最好。
Using Cymbidium Ranluo tissue-cultured rhizomes and tissue-cultured seedlings as experimental materials,this study investigated the effects of five LED illumination treatments on rhizome proliferation/differentiation and seedling growth/physicochemical properties during the seedling-strengthening and rooting stages.The five treatments were red light(R),red light∶blue light=6∶4(R6B4),red light∶blue light=5∶5(R5B5),red light∶blue light=4∶6(R4B6),and blue light(B).The results showed that during the rhizome proliferation and differentiation stage,compared with the fluorescent lamp control(CK),the diameter of Cymbidium Ranluo tissue-cultured rhizomes increased significantly and the differentiation rate was the highest under R5B5 treatment.The soluble protein content decreased firstly and then increased with the increase of blue light ratio,while the soluble sugar content increased firstly and then decreased,which was the highest under R6B4 treatment.The content of hydrogen peroxide and reactive oxygen species decreased with the increase of blue light ratio,while the activity of catalase increased with the increase of blue light ratio.There was no significant difference in hydrogen peroxide content between the R5B5 treatment and the CK,while the reactive oxygen species content and catalase activity increased significantly compared to CK.During the seedling-strengthening and rooting stages,R5B5-treated tissue-cultured seedlings exhibited higher values in leaf length,leaf width,pseudobulb width,leaf number,plant height,root length,root diameter,fresh weight of aboveground parts,chlorophyll a content,chlorophyll b content,carotenoid content and total chlorophyll content.The soluble sugar content of the tissue-cultured seedlings first increased and then decreased with the reduction of the red light ratio,with the R5B5 treatment showing the smallest decrease compared to the CK.The relative expression of CRY2 gene was the highest under R5B5 treatment.In summary,different LED illumination treatments have varying effects on the rhizome proliferation and differentiation of Cymbidium Ranluo,as well as on the growth,chlorophyll content,soluble protein content,soluble sugar content,antioxidant system,and CRY2 gene expression of the tissue-cultured seedlings,with the R5B5 treatment showing the best results.
