摘要
目的 观察四磨汤对肝脾气滞证功能性消化不良(functional dyspepsia, FD)大鼠白介素4(interleukin-4,IL-4)/免疫球蛋白E(immunoglobulin E,IgE)-IgE的高亲和力受体(IgE high affinity receptor, FcεRI)信号通路的影响,探究四磨汤改善肝脾气滞证FD的作用及可能机制。方法 用随机数字表法将60只大鼠分为6组,分别为空白组、模型组、莫沙必利组、四磨汤高、中、低剂量组。除空白组外的组别选取不可预知的慢性应激法复制肝脾气滞证FD大鼠模型,造模成功后治疗组大鼠每天同一时间予以相应治疗药物灌胃,模型组大鼠则予以蒸馏水灌胃,每日不限饮食饮水,连续灌胃14天。最后检测各组大鼠血清IL-4、IgE、FcεRI、五羟色胺(5-hydroxytryptamine, 5-HT)、白三烯(leukotriene, LT)含量,十二指肠IL-4 mRNA、FcεRI mRNA表达以及Tryptase、FcεRI蛋白的表达。结果 与空白组相比,模型组大鼠血清IL-4、IgE、FcεRI、5-HT、LT含量均明显增加(P<0.05),十二指肠IL-4 mRNA、FcεRI mRNA表达以及Tryptase、FcεRI蛋白的表达均增加(P<0.05);与模型组相比,治疗组大鼠血清IL-4、FcεRI、5-HT及LT含量均明显下降(P<0.05),除四磨汤低剂量组外的各治疗组血清IgE含量均明显下降(P<0.05)、十二指肠IL-4 mRNA、FcεRI mRNA表达及Tryptase、FcεRI蛋白的表达均显著下降(P<0.05)。结论 四磨汤对肝脾气滞证FD有效且能显著缓解肝脾气滞证FD的十二指肠低度炎症,其机制可能与抑制IL-4/IgE-FcεRI信号通路以减少相关活性物质的释放有关。
Objective To observe the effect of Simo Decoction on IL-4/IgE-FcεRI signaling pathway in rats with functional dyspepsia(FD)of liver-spleen qi stagnation syndrome,and to explore the effect of Simo Decoction on FD of liver-spleen qi stagnation syndrome and its possible mechanism.Methods 60 rats were randomly divided into six groups:blank group,model group,mosapride group,high,middle and low dose groups of Simo Decoction.Except the blank group,the FD rat model of liver-spleen qi stagnation syndrome was established by unpredictable chronic stress.At the same time,the rats in the treatment group were given gastric gavage with corresponding drugs,while the rats in the model group were given gastric gavage with distilled water.Daily diet and drinking water,continuous gavage for 14 days.Finally,the levels of serum interleukin-4(IL-4),immunoglobulin E(IgE),IgE High affinity receptor(FcεRI),5-hydroxytryptamine(5-HT)and leukotriene(LT)were measured,the expression of duodenum IL-4 mRNA,FcεRI mRNA,Tryptase and FcεRI protein were also detected.Results Compared with the blank group,the levels of IL-4,IgE,FcεRI,5-HT,and LT in the serum of the model group were increased significantly(P<0.05),and the expressions of IL-4 mRNA,FcεRI mRNA and Tryptase,FcεRI protein in the duodenum of the model group were increased significantly(P<0.05).Compared with the model group,the levels of IL-4,FcεRI,5-HT,and LT in the serum of treatment groups were decreased significantly(P<0.05),the levels of IgE in the serum of treatment groups except the low-dose group of Simo Decoction were decreased significantly(P<0.05),the expression of duodenum IL-4 mRNA,FcεRI mRNA,Tryptase and FcεRI protein were decreased in the treatment groups(P<0.05).Conclusion Simo Decoction is effective in the treatment of FD with liver-spleen qi stagnation syndrome and can significantly relieve the duodenum low-grade inflammation of FD with liver-spleen qi stagnation syndrome.The mechanism may be related to the inhibition of IL-4/IgE-FcεRI signaling pathway to reduce the release of related active substances.
