摘要
目的:采用热蛋白质组学(TPP)技术构建“高通量靶点筛选-靶标关联-临床新应用”研究体系,精准剖析灯盏细辛提取物在脑病治疗中的作用机制及临床应用价值,挖掘其潜在新功效。方法:采用TPP技术,高通量筛选灯盏细辛提取物在人小胶质细胞HCM3中的作用靶点;通过IPA数据库和Genvestigator平台对灯盏细辛提取物的作用靶点与下游靶标、疾病进行关联分析,挖掘潜在适应症。结果:采用TPP技术筛选出灯盏细辛提取物的40个作用靶点,包括23个直接靶点和17个间接靶点;经IPA数据库与Genvestigator平台联合分析发现,其中一些靶点参与阿尔茨海默病、帕金森病的治疗过程,提示其在脑胶质瘤治疗和神经再生方面具有潜在应用价值。结论:基于“高通量靶点筛选-靶标关联-临床新应用”的中药靶标研究方法,从靶标层面阐明了灯盏细辛提取物治疗脑病的作用机制和临床新功效。
Objective:This study aims to accurately investigate the mechanism and clinical significance of Erigeron breviscapus extract in treating encephalopathy and uncover its new effects by establishing a high-throughput target screening-target association-new clinical application research system through thermal proteome profiling(TPP)technology.Methods:TPP technology was utilized to conduct a high-throughput screening of the targets of E.breviscapus extract in human microglia clone 3(HCM3).The ingenuity pathway analysis(IPA)database and Genvestigator platform were used to analyze the association between the target and downstream target and disease of the E.breviscapus extract,so as to explore potential indications.Results:TPP technology identified 40 targets from the E.breviscapus extracts,comprising 23 direct targets and 17 indirect targets.A combined analysis using the IPA database and Genvestigator platform indicated that these targets were involved in the treatment of Alzheimer′sdisease and Parkinson′sdisease and had potential applications in glioma treatment and nerve regeneration.Conclusion:The traditional Chinese medicine target research method based on high-throughput target screening-target association-new clinical application reveals the mechanism of action and new clinical efficacy of E.breviscapus extract in treating encephalopathy at the target level.
