摘要
为了提升彝族药酢浆草药材质量标准,参考现行质量标准,对酢浆草药材进行性状鉴别;测定并评价酢浆草药材水分、总灰分、酸不溶性灰分、浸出物等质量控制指标;对酢浆草药材茎横切面和粉末进行显微鉴别;优化薄层色谱鉴别方法;采用高效液相色谱法测定酢浆草中异牡荆苷含量,色谱柱为ZORBAX SB C18(250 mm×4.6 mm,5μm),以乙腈-0.1%磷酸溶液(体积比17∶83)为流动相,流速1 mL/min,柱温30℃,紫外检测波长340 nm,进样量10μL。酢浆草药材性状符合规定,显微鉴别特征明显,优化后的薄层鉴别方法色谱图斑点清晰,分离度好,异牡荆苷质量浓度和峰面积线性关系均良好,精密度、重复性、稳定性等均符合规定。该方法为彝族药酢浆草药材质量控制和应用开发提供了理论依据和技术支撑。
To improve the quality standards of Yi medicine Oxalis corniculata,the characteristics of Oxalis corniculata were identified,The quality control indicators such as moisture,total ash,acid insoluble ash,and extract of Oxalis corniculata were measured and evaluated.Microscopic identification was carried out on the transverse section and powder of the stem of the vinegar herb.The thin-layer chromatography(TLC) identification method was optimized.HPLC was used to determine the content of Isovitexin in Oxalis corniculata.A ZORBAX SB C18 column(250 mm×4.6 mm,5 μm) was used,with acetonitrile and 0.1% phosphoric acid solution(the volume ratio of 17:83) as the mobile phase,a flow rate of 1 mL/min,column temperature of 30 ℃,UV detection wavelength of 340 nm,and injection volume of 10 μL.The characteristics of the medicinal herb Oxalis corniculata chinensis comply with regulations,with obvious microscopic identification features.The optimized TLC identification method has clear chromatographic spots and good separation.The linear relationship between the mass concentration and peak area of Isovitexin is good,and the precision,repeatability,stability,etc.all meet the regulations.The method provides theoretical basis and technical support for quality control and evaluation of Oxalis corniculata.
