摘要
目的:优化紫苏叶多糖提取条件,探究其抗氧化活性,为紫苏叶多糖提取及有效利用提供理论依据。方法:采用单因素试验和响应面法优化紫苏叶多糖超声辅助提取工艺,并探究紫苏叶多糖的体外抗氧化活性。结果:单因素分析中,料液比1∶35(g∶mL)、超声温度60℃、超声时间50 min和超声功率250 W时,紫苏叶多糖得率最高,分别为13.6%、14.7%、14.8%、13.8%;响应面法优化结果显示,料液比1∶38(g∶mL)、超声温度59℃、超声时间50 min和超声功率250 W时,紫苏叶多糖得率最高,为14.471%±0.168%;多糖溶液浓度为2.0 mg·mL^(-1)时对DPPH·和ABTS·^(+)的清除能力分别为83.06%和77.96%。结论:紫苏叶多糖对DPPH·和ABTS·^(+)均有一定的清除能力,具有良好的抗氧化能力。
Objective:To optimize the extraction conditions of polysaccharide of leaves of Perilla frutescens and investigate its antioxidant activity,to provide theoretical basis for the extraction and effective utilization of polysaccharide of leaves of Perilla frutescens.Method:One-factor test and response surface method were used to optimize the ultrasound-assisted extraction process of polysaccharides of leaves of Perilla frutescens and to investigate the in vitro antioxidant activity of polysaccharides of leaves of Perilla frutescens.Result:The highest yield of polysaccharides of leaves of Perilla frutescens was obtained at single factor analysis with the solid-liquid ratio of 1∶35(g∶mL),ultrasonication temperature of 60℃,ultrasonication time of 50 min,and ultrasonication power of 250 W,which was 13.6%,14.7%,14.8%,13.8%respectively.The optimal results of response surface method showed tha when the solid-liquid ratio was 1∶38(g∶mL),the ultrasonication temperature was 59℃,the ultrasonic time was 50 min and the ultrasonication power was 250 W,the polysaccharide yield of leaves of Perilla frutescens was the highest,which was 14.471%±0.168%.When the concentration of polysaccharide solution was 2.0 mg·mL^(-1),the scavenging ability of DPPH·and ABTS·^(+)was 83.06%and 77.96%,respectively.Conclusion:Polysaccharide of leaves of Perilla frutescens had certain scavenging ability on DPPH·and ABTS·^(+),and had good antioxidant ability.
