摘要
本实验以三穗鸭为材料,采用PCR扩增并采用直接测序技术对三穗鸭DRD4基因进行SNP多态位点检测,共筛查出4个SNP位点:exon2-C12406T、exon3-A13144G、intron2-C12777T、intron2-T12789C。其中exon2-C12406T、exon3-A13144G位点突变为同义突变。对DRD4进行生物信息学分析表明,突变前后的等位基因频率估算有差异,但其m RNA二级结构预测,蛋白质二级、三级结构预测分析均无差异。
In this experiment, the polymorphism of DRD4 gene in Sansui duck was analyzed by PCR. Four SNPs including exon2-C12406 T, exon3-A13144 G, intron2-C12777 T and intron2-T12789 C were detected. Two of which SNPs(exon2-C12406 T, exon3-A13144G) were synonymous mutations. The results for bioinformatic analysis indicated that the synonymous mutations could cause the change of allele frequency. But no differences were found after m RNA secondary structure, secondary and tertiaryprotein structure were predicted.
