摘要
为弄清湖南省某鸡场发病病因,对送检的病死鸡进行了解剖、组织细菌的分离,并对分离菌株进行了生化鉴定、16SrDNA扩增测序分析,且进一步测序分析了其荚膜和脂多糖PCR分型。解剖结果发现,6只鸡均有心包积液、肺脏实变、肝脏表面有许多针尖状灰白色的坏死点,脾脏和肠无明显症状。对采集组织脏器肝脏、肺脏、脾脏,接种TSA平皿后,均有长出卵圆形菌落。纯化后革兰氏染色为G-短小杆菌,经细菌鉴定仪鉴定为多杀性巴氏杆菌。16SrDNA测序结果显示其与巴氏杆菌美国株CCUG17978的同源性为100%;通过荚膜和脂多糖PCR分型鉴定为荚膜A型和脂多糖L1型。经使用巴氏杆菌较为敏感的磺胺类药物对应性治疗后,鸡群恢复健康。该鸡场发病系荚膜A型和脂多糖L1型巴氏杆菌感染。
In order to find out the cause of the disease in an infected chicken farm in Hunan Province,the dead chickens were dissected and the tissue bacteria were isolated.The isolates were identified by biochemical assay,16SrDNA amplification and sequencing,and the capsular and lipopolysaccharide PCR typing were further analyzed.Autopsy results showed that all the 6 chickens had pericardial effusion,lung consolidation,and many needle-like gray and white necrotic spots on the surface of the liver,and no obvious symptoms in the spleen and intestine.After inoculation with TSA platen,oval-shaped colonies grew in the collected tissue organs,such as liver,lung and spleen.After purification,G-Pumilus was detected by Gram staining and Pasteurella polycida was identified by bacterial identification apparatus.16SrDNA sequencing showed 100%homology with CCUG17978,a US strain of Pasteurella.Capsular type A and lipopolysaccharide type L1 were identified by PCR.The chickens recovered after corresponding treatment with sulfonamides,which are more sensitive to pasteurella.The chicken farm was infected with capsular type A and lipopolysaccharide type L1 pasteurella.
