摘要
为了从母系遗传的角度探讨北京油鸡的遗传多样性和系统进化,研究测定了165只北京油鸡线粒体DNA(mtDNA)D-loop区全序列,分析其遗传多样性并结合NCBI下载的43个其他鸡种序列构建了系统发育树。结果显示,北京油鸡mtDNA D-loop区全序列长度1231~1232 bp,存在一处碱基缺失;共发现26处单核苷酸变异位点,组成了13种单倍型;核苷酸多样性为0.00427±0.00074,单倍型多样性为0.563±0.047,核苷酸平均差异数为5.258;北京油鸡13种单倍型可划分为A、B、C、E四个分支,其中C分支为优势分支,占检测个体的73.3%。结果表明,北京油鸡线粒体遗传多样性呈中等水平,保种效果较好。
To investigate the genetic diversity and phylogeny of Beijing You chicken(BJY)from the perspective of maternal inheritance,the whole sequences of mitochondrial DNA(mtDNA)D-loop region of 165 BJY chickens were detected.Then,the polymorphism was analyzed and phylogenetic trees were constructed together with D-loop sequences of 43 other chicken breeds downloaded from the National Center for Biotechnology Information(NCBI).The results showed that the full length of mtDNA Dloop region was 1231 to 1232 bp in BJY chickens,with one single base deletion.Totally,26 single nucleotide mutation sites and 13 haplotypes were found.The nucleotide diversity,haplotype diversity and average nucleotide differences of BJY chickens were 0.00427±0.00074,0.563±0.047 and 5.258,respectively.The 13 haplotypes of BJY chickens could be clustered into four clades,A,B,C and E,among which C was the dominant clade accounting for 73.3%of the detected individuals.The results indicated that the genetic diversity of BJY chickens was under a moderate level,indicating that it was well conserved.
