摘要
本研究建立乌头汤有效成分含量测定的HPLC方法。检测条件如下,流动相10 mmol/L磷酸二氢铵,磷酸调pH为2.1-乙腈,色谱柱Waters XSelect C18(4.6 mm 250 mm,5μm),检测波长210 nm和235 nm,柱温35℃,流速1 mL/min。结果表明,该方法下,各物质间分离度良好,与阴性对照品相比较均无干扰,符合方法学考察中各项指标要求。HPLC方法可有效分离乌头汤中12种不同结构类型成分,稳定高效,操作简便,结果准确,可作为乌头汤有效成分的含量测定方法。与此同时将乌头碱、次乌头碱和新乌头碱等毒性成分一并纳入检测范围,以确保用药安全,可为今后乌头汤新药开发及生产的质量标准研究提供科学依据。
The study established a high performance liquid chromatography(HPLC) method for the determination of effective components in wutou decoction. The detection conditions were as follows, mobile phase was 10 mmol/L ammonium dihydrogen phosphate, pH was adjusted to 2.1 with phosphoric acid-acetonitrile, chromatographic column was Waters XSelect C18(4.6 mm 250 mm, 5 μm), detection wavelength were 210 nm and 235 nm, column temperature was 35 ℃, and the flow rate was 1 mL/min. The results showed that the separation degree of each compound was superior, and there was no interference compared with the negative control, which met the requirements of various indexes in the methodological investigation. The HPLC method could effectively separate 12 different structural types of components in wutou decoction. It was stable, efficient, simple and accurate. It could be used as the content determination method of effective components in wutou decoction. At the same time, toxic components such as aconitine, hypaconitine and mesaconitine were included in the detection range to ensure drug safety. It could provide a scientific basis for the quality standard research of new drug development and production of wutou decoction in the future.
