玫瑰茄多糖对HeLa细胞凋亡相关蛋白的影响 被引量:2

Effect of Polysaccharide from Hibiscus Sabdariffa on Apoptosis-Related Proteins in HeLa Cells

摘要 为探究玫瑰茄多糖对人宫颈癌HeLa细胞凋亡相关蛋白的影响,采用水提醇沉法和DEAE-52纤维素交换树脂法制备玫瑰茄多糖,western-blot法检测HeLa细胞中Bax、Bcl-2、pro-Caspase-3、Caspase-3蛋白的含量.结果表明:与空白对照组相比,玫瑰茄多糖对HeLa细胞增殖有抑制作用,且具有浓度依赖性(P<0.05,P<0.01).采用高剂量(4 mg/mL)的玫瑰茄多糖处理HeLa细胞后,细胞中Bax、Caspase-3蛋白的含量显著提高(P<0.01),Bcl-2、pro-Caspase-3蛋白的含量明显降低(P<0.05,P<0.01).这说明玫瑰茄多糖可通过线粒体途径诱导HeLa细胞凋亡. In order to explore the effect of polysaccharide from Hibiscus sabdariffa on the apoptosis-related proteins in HeLa cells,polysaccharide was prepared by the water extraction and alcohol precipitation method and DEAE-52 cellulose exchange resin method.The protein of Bax,Bcl-2,pro-Caspase-3,Caspase-3,which is involved in the apoptosis in HeLa cells,was detected by the western-blot method.The results show as follows:compared with the blank control group,polysaccharides have an inhibitory effect on the proliferation of HeLa cells in a concentration-dependent manner(P<0.05,P<0.01).After treating HeLa cells with a high dose of polysaccharide(4 mg/mL),it is found that the proteins of Bax and Caspase-3 in the cells significantly increase(P<0.01),and the proteins of Bcl-2 and pro-Caspase-3 significantly decrease(P<0.05,P<0.01).It indicates that polysaccharide from H.sabdariffa can induce HeLa cell apoptosis through the mitochondrial pathway.
