摘要
目的应用网络药理学推测藤梨根的抗胃癌活性成分及作用机制。方法以“mihoutaogen”作为关键词在中药系统药理学数据库与分析平台(TCMSP)进行查询,筛选藤梨根的主要活性成分和靶点;以“stomach cancer”为关键词在人类基因数据库(GeneCards)搜索疾病相关基因;通过String网站构建藤梨根抗胃癌的蛋白互作图(PPI);运用软件Cytoscape 3.6.0对核心靶点进行拓扑分析并对信号通路进行富集分析。结果共筛选出藤梨根主要活性成分6种,作用靶点137个,胃癌病变靶点149个,药物有效成分和疾病共有靶点78个。推测藤梨根抗胃癌靶点主要有EGF、TP53、VEGFA、EGFR和IL6。结论推测藤梨根抗胃癌的主要活性成分,探讨其作用机制,为今后临床研究提供理论依据。
OBJECTIVE On the basis of network pharmacology,the anti-gastric cancer active components of Actinidia chinensis Root and their mechanism of action were selected and predicted.METHODS“Mihoutaogen”was used as a key word to query the pharmacological database and analysis platform of Chinese medicine system(TCMSP),and the main chemical components and the corresponding targets of the root were screened out.The disease-related genes were searched from GeneCards using“stomach cancer”as keyword.The protein interaction network map(PPI)of Fuji pear root against stomach cancer was made through the STRING website.Cytoscape 3.6.0 software was used to carry out topology analysis of the core targets,and further enrichment analysis of the signal pathways.RESULTS 6 main active ingredients and 149 targets of Actinidia chinensis root and 149 targets of gastric cancer were screened.There were 78 targets for both active ingredients and diseases.The main targets of Actinidia chinensis root against gastric cancer were EGF,EGFR,IL6,VEGFA and TP53.CONCLUSION The main active compounds of Actinidia chinensis root against gastric cancer were predicted preliminarily and its mechanism was explored which provided theoretical basis for future clinical research.
