中心组合设计效应面法优化荆芥中胡薄荷酮的提取工艺

Optimization of Extraction Process of Kurmenone from Schizonepeta chinensis by Central Composite Design Response Surface Method

摘要 目的通过优化荆芥水提液的关键制备参数,实现指标成分胡薄荷酮的最高效提取。方法使用Design Expert 8.0软件,中心组合设计法设计实验方案,参数为液料比、提取时间和提取循环,所制备水提液中胡薄荷酮含量为考察指标;含量测定采用高效液相色谱法,色谱柱为Waters XDB C_(18)(规格:柱长250 mm,内径:4.6 mm,粒径:5μm),流动相为乙腈-水(40∶60),流速1.0 mL·min^(-1),柱温25℃,检测波长254 nm,进样体积10μL。结果Design Expert 8.0软件给出20组参数,采用这些参数进行的实验结果显示,当液料比为1∶30、提取时间为60 min,提取循环为2次时,提取方案为最优,此时水提液中胡薄荷酮含量为0.3011 mg·mL^(-1)。结论中心组合设计响应面法能够更加准确、高效地找到制备荆芥水提液的最优工艺条件。 OBJECTIVE To optimize the key preparation parameters of water extract of Schizonepeta chinensis to achieve the most efficient extraction of index component kurmenone.METHODS Design Expert 8.01 software was used to Design the experimental scheme using the central combinational Design method.The parameters were liquid-solid ratio,extraction time and extraction cycle.The content of kurmenone in the prepared water extract was used as the index.For content determination,HPLC was performed on a Waters XDB C_(18) column(column length 250 mm,inner diameter 4.6 mm,particle size 5 μm) with mobile phase consisted of acetonitrile-water(40∶60) at the flow rate of 1.0 mL·min^(-1).The column temperature was 25 ℃,and the detection wavelength was 254 nm.The injection volume was 10 μL.RESULTS The Design Expert 8.01 software provides 20 sets of parameters,and the results of experiments using these parameters show that when the liquid-solid ratio was 1∶30,the extraction time was 60 minutes,and the extraction cycle was 2 times,the extraction scheme was optimal,and the content of kurmenone in the water extract was 0.3011 mg·mL^(-1).CONCLUSION The central composite design response surface methodology can find the optimal process conditions for the preparation of water extract of Schizonepeta chinensis more accurately and efficiently.
