摘要
目的运用网络药理学研究大柴胡汤治疗2型糖尿病的作用机制。方法通过中药系统药理数据库及分析平台(TCMSP)筛选大柴胡汤主要化学成分和靶点,通过GeneCards获得2型糖尿病相关靶点基因,将药物靶点基因和疾病靶点映射而得到大柴胡汤治疗2型糖尿病的预测靶点。利用Cytoscape 3.6.0软件构建中药化合物-靶点网络与蛋白互作网络(PPI)。利用DAVID 6.8数据库进行交集基因的基因本体论(GO)分析和京都基因与基因组百科全书(KEGG)通路富集分析。结果大柴胡汤治疗2型糖尿病有效成分98个,相关靶点260个,关键靶点35个。GO分析包括生物学进程625个,细胞组分71个和分子功能129个。KEGG通路富集分析包括134条信号通路,主要与TNF信号通路、PI3K-Akt信号通路、p53信号通路、细胞凋亡信号通路等密切相关。结论大柴胡汤主要通过作用于炎症反应、胰岛素抵抗、胰岛β细胞凋亡、血管增生等多方面治疗2型糖尿病。
Objective To explore the mechanism of Dachaihu Decoction in the treatment of type 2 diabetes mellitus based on network pharmacology.Methods The main chemical components and targets of Dachaihu Decoction were obtained by Tradition Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP),and target genes of type 2 diabetes mellitus were obtained by GeneCards.The target genes of drug and disease were mapped to predict target genes of Dachaihu Decoction for type 2 diabetes mellitus.Cytoscape 3.6.0 software was used to construct the compound-target network and protein-protein interaction network(PPI)of tradition Chinese medicine.Gene ontology(GO)analysis of potential genes and enrichment analysis of kyoto encyclopedia of genes and genomes(KEGG)pathway were performed by DAVID 6.8 database.Results There were 98 active ingredients,260 related targets,35 key targets in Dachaihu Decoction on type 2 diabetes mellitus.GO analysis included 625 biological processes,71 cellular components and 129 molecular functions.The enrichment analysis of KEGG pathway included 134 signaling pathways,which were mainly related to TNF signaling pathway,PI3K-Akt signaling pathway,p53 signaling pathway,and apoptosis signaling pathway.Conclusion Dachaihu Decoction plays an important role in the treatment of type 2 diabetes mellitus mainly by acting on inflammation,insulin resistance,apoptosis in pancreatic beta-cells and angiogenesis.
