红景天药材中两种黄酮化合物的提取工艺优化及含量测定研究 被引量:1

摘要 目的:优化红景天药材中山萘酚和槲皮素的提取工艺,并建立高效液相色谱测定含量的方法。方法:以山萘酚和槲皮素含量为检测指标,以提取溶剂、液料比、提取方法、提取时间为因素,采用四因素三水平L9(34)正交设计优选出山萘酚、槲皮素的最佳提取条件,并进行含量测定方法学验证。结果:最佳提取条件:用液料比为10:1的甲醇-盐酸混合液(甲醇体积分数为50%,盐酸浓度为2.0 mol/L)在恒温水浴(90℃)中回流提取60 min,得到山萘酚含量为1.25 mg/g,槲皮素含量为0.75 mg/g。结论:提取工艺合理可行,稳定可靠,含量测定方法重现性好,回收率高。 Objective:To optimize the extraction process of kaempferol and quercetin in Rhodiola and develop HPLC method for determining the content.Method:The optimum extraction conditions of kaempferol and quercetin were optimized by orthogonal design of four factors and three levels L9(34),with the contents of kaempferol and quercetin as the detection indexes and the extraction solvent,liquid to solid ratio,extraction method and extraction time as factors.The content determination methodology was verified.Result:The optimum extraction conditions were as follows:use a methanol-hydrochloric acid mixture(methanol volume fraction was 50%and hydrochloric acid concentration was 2.0 mol/L)with a solid-liquid ratio of 10:1 and reflux extraction for 60 min in a constant temperature water bath(90℃).The content of kaempferol and quercetin were 1.25 mg/g and 0.75 mg/g respectively.Conclusion:The extraction process is reasonable,feasible,stable and reliable.The content determination method has good reproducibility and high recovery rate.
