摘要
目的MRI介导的低频聚焦超声联合纳米微泡对提高MSCs治疗脑梗死大鼠的效果研究。方法建立MCAO大鼠模型,制备纳米微泡,标记MSCs的超顺磁氧化铁(SPIO)并采取低频聚焦超声联合微泡注射和MSCs注射治疗。对治疗前后大鼠的神经功能评分脑梗死病大鼠黑质TH阳性和DAT阳性神经元的凋亡情况;提取总蛋白,用酶联免疫吸附法(ELISA)检测营养因子VEGF、NT-3、NGF和BDNF的含量进行对比分析。结果MRI介导的低频聚焦超声联合纳米微泡对提高MSCs治疗脑梗死大鼠可以显著提高大鼠的神经功能,MRI介导的低频聚焦超声联合纳米微泡对提高MSCs治疗减少脑梗死病大鼠黑质TH阳性和DAT阳性神经元的凋亡,减少了氧化应激对神经元的损伤,营养因子VEGF、NT-3、NGF和BDNF的含量也显著增高。结论MRI介导的低频聚焦超声联合纳米微泡对提高MSCs治疗脑梗死大鼠可以促进脑梗死的神经修复以改善预后,减轻社会负担和经济负担,具有重要的临床价值和社会意义。
Objective To study the effect of MRI-mediated low frequency focused ultrasound combined with nano-microbubbles on improving MSCs treatment in rats with cerebral infarction.Methods MCAO rat model was established,nano-microbubbles were prepared,the superparamagnetic iron oxide(SPIO)of MSCs was labeled,and low frequency focused ultrasound combined with microbubble injection and MSCs injection were used for treatment.The neurological function scores of rats before and after treatment were evaluated according to the apoptosis of THpositive and DAT-positive neurons in substantia nigra of rats with cerebral infarction.Total protein was extracted,and the contents of nutritional factors VEGF,NT-3,NGF and BDNF were detected by enzyme-linked immunosorbent assay(ELISA)for comparative analysis.Results MRI-mediated low frequency focused ultrasound combined with nano-microbubbles could significantly improve the neurological function of rats with cerebral infarction by improving MSCs treatment.MRI-mediated low frequency focused ultrasound combined with nano-microbubbles improved MSCs treatment,reduced the apoptosis of TH-positive and DAT-positive neurons in substantia nigra of rats with cerebral infarction,decreased the damage of oxidative stress to neurons,and significantly increased the contents of nutritional factors VEGF,NT-3,NGF and BDNF.Conclusion MRI-mediated low frequency focused ultrasound combined with nano-microbubbles meant for improving MSCs treatment in rats with cerebral infarction can promote nerve repair of cerebral infarction to ameliorate prognosis and reduce social and economic burden.It has important clinical value and great social significance.
