摘要
制备乌榄叶黄酮类化合物;通过不同方法研究其体外抗氧化活性;建立D-半乳糖小鼠氧化损伤模型,研究乌榄叶对机体抗氧化能力影响。结果表明:在体外试验中,乌榄叶黄酮类化合物的抗氧化能力与维生素C相当,对DPPH·、ABTS^+·、羟自由基具有显著的清除作用,且具有较强的还原力、抗脂质体过氧化能力(IC50=0.065 mg/mL)和氧化自由基吸收能力(oxygen radical absorbance capacity,ORAC)。在体内实验中,与模型组相比,乌榄叶黄酮类化合物显著升高肝脏、肾脏和胰腺指数(P<0.05);有效降低血清中丙二醛(malondialdehyde,MDA)含量,增加总抗氧化能力(total antioxidant capacity assay kit,T-AOC)、增强谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)、超氧化物歧化酶(superoxide dismutase,SOD)的活性(P<0.05,P<0.01);减少肝脏组织损伤。乌榄叶黄酮类化合物在体内外均具有较强的抗氧化活性,因此,可用作制药和食品工业中的天然抗氧化剂和抗老化剂。
Preparation total flavonoids from Canarium pimela leaves (TFCPL) and determined the antioxidant capacity of TFCPL from different methods in vitro and its effects on D-galactose induced oxidative damage mice. The results showed that antioxidant activity of TFCPL was similar to that of vitamin C in vitro. And it had effective activity on scavenging DPPH·, ABTS+·, OH free radica, reducing power, inhibition lipid peroxidation (IC50=0.065mg/mL) and oxygen radical absorbance capacity (ORAC). In vivo, TFCPL increased the indices of liver,kidney and pancreas (P<0.05). TFCPL might dose-dependently decrease the malondialdehyde (MDA) content and increase the total antioxidant capacity assay kit (T-AOC), glutathione peroxidase (GSH-Px), superoxide dismutase (SOD) activities (P<0.05, P<0.01) in serum, minimizing damage to the liver. TFCPL had activity of antioxidant in vivo and in vitro. Therefore, TFCPL could be used as a natural antioxidant and anti-aging agent in the pharmaceutical and food industries.
