摘要
目的探讨麝香对裸鼠人胃癌组织中肿瘤标志物人表皮生长因子受体2(human epidermal growth factor receptor 2,HER_2)和血管内皮生长因子(vascular endothelial growth factor,VEGF)水平的影响。方法将建立异位移植瘤模型成功后的35只裸鼠按随机数表法分为模型组,氟尿嘧啶组,麝香低、中、高剂量组共5组,每组7只。采用质量浓度为0. 9的氯化钠溶液灌胃,2 ml/(只·d)、腹腔注射5-Fu 0. 125 g/(kg·d)、麝香溶液0. 01、0. 02、0. 04 g/(kg·d)灌胃,21 d后眼眶取血,处死动物,剥取肿瘤。采用免疫组化检测瘤组织HER_2、VEGF的表达,采用酶联免疫法检测血清肿瘤标志物癌胚抗原(CEA)、糖类抗原125 (CA125)、CA50、CA19-9水平。结果 HER_2平均灰度值,各用药组均较模型组偏低(P <0. 05); HER_2平均面密度结果,各用药组均较模型组减少(P <0. 05); VEGF平均灰度值,麝香高剂量组较模型组偏低(P <0. 05); VEGF平均面密度,麝香高剂量组较模型组减少(P <0. 05)。给药各组CEA、CA125、CA50、CA19-9水平均下降。CEA结果显示,除麝香低剂量组外,其他用药组均低于模型组(P <0. 05); CA125结果显示,各治疗组均比模型组偏低(P <0. 05); CA50及CA19-9结果相似,除麝香低剂量组外,其他用药组较模型组偏低(P <0. 05)。结论麝香对胃癌裸鼠异位移植瘤具有一定的抗肿瘤作用,能在一定程度上减慢肿瘤的生长,麝香抑瘤的作用机制可能与通过下调肿瘤组织VEGF及HER表达活性相关。
Objective To study the effects of musk on tumor markers and human epidermal growth factor receptor2(HER2)and vascular endothelial growth factor(VEGF)levels in human gastric cancer tissues in nude mice.Methods Thirty-five thtransplantation tumor models of nude mice which had successfully built gastric cancer cell lines were randomly divided into control group,fluorouracil group,low,medium and high dose musk groups,each group of seven.Each group was used respectively saline lavage for a rat(2ml/d),5-Fu intraperitoneal injection[0.125g/(kg·d)],musk solution lavage[0.01,0.02,0.04g/(kg·d)].The orbital blood was obtained after21days,rats were put to death and tumor tissues were striped.The expression of HER2and VEGF was detected by using immunohistochemical detection and serum tumor markers of carcino-embryonic antigen(CEA),carbohydrate antigen125(CA125),CA50,CA19-9were detected by enzyme-linked immunoso rbent assay.Results The mean gray values and the average area density of HER2showed that fluorouracil group and musk group were significantly lower than those in the model group(P<0.05).The mean gray value and the mean area density of VEGF in the musk high-dose group was significantly lower than that of the model group(P<0.05).The values of CEA,CA125,CA50and CA19-9were decreased in all groups.The results of CEA showed that there were significant differences between the high,middle and low dose groups(P<0.05).The results of CA125showed that there were significant differences between the groups of fluorouracil and musk in high,middle and low dose groups(P<0.05).The results of CA50and CA19-9were similar,and they showed that the musk high,middle dose and fluorouracil groups were significantly decreased than the model control group(P<0.05).Conclusion Musk for tumor growth in nude mice has a certain anti-tumor effect,to some extent,which may be related to the down-regulation of vascular endothelial growth factor and human epidermal growth factor receptor expression in tumor tissues.
