摘要
JAZ蛋白是植物茉莉酸信号途径中的负调控因子,通过与多种不同的蛋白相互作用进而参与调控不同的生物学过程。HblJAZ3蛋白是橡胶树乳管细胞茉莉酸信号传导的重要组分,其编码基因受割胶和茉莉酸上调表达。为了进一步阐明HblJAZ3在乳管细胞中的功能,特别是参与茉莉酸对橡胶生物合成调控的作用机制,本文利用酵母双杂交技术筛选HblJAZ3的互作蛋白。结果初步表明,HblJAZ3可与NBR1类蛋白、17.5 ku和18.1ku Ⅰ类小热激蛋白、含C2结构域蛋白、SRPP1、HEV2.1、蛋白酶体6型亚基α、E3泛素化连接酶、C3HC4类环锌指蛋白和MYC2类蛋白相互作用。表明这些互作蛋白可能介导HblJAZ3对茉莉酸响应的基因表达调控、自身蛋白稳定性的调节、被自噬降解途径和泛素化标记的26S蛋白酶体降解的过程。
JAZ proteins are repressive components of jasmonic acid (JA) signaling in plants, which involved in regulating various biological processes via interacting with distinct proteins. HblJAZ3 is an important component involved in JA signaling transduction in Hevea brasiliensis laticifer and its encoding gene was induced by tapping and exogenous JA treatment. To elucidate the possible function of HblJAZ3 in the laticifer, especially the roles in JA-mediated natural rubber biosynthesis, the yeast two-hybridization method was used to screen its interacting targets. The results revealed that NBRl_like, 17.5 and 18.1 ku class I heat shock protein-like, C2 domain- containing protein, SRPP1, HEV2.1, proteasome subunit alpha type-6, E3 ubiquitin-protein ligase, C3HC4-type RING zinc finger protein and MYC2-1ike protein were able to interact with HblJAZ3 protein in yeasts. These HblJAZ3-interacting partners may mediate the expression of genes responding to JA via HblJAZ3 protein and the stability of HblJAZ3 protein, and mediate degradation of HblJAZ3 protein via autophagy degradation pathway and 26S proteasome degradation pathway as well.
